요양시설 노인의 피부건강상태 및 피부수분 관련 요인에 관한 연구 (A Study on Skin Health Status and Related Factors of Skin Hydration in Institutionalized Elderly)

  • Purpose: This study was performed to identify skin health status and the factors influencing skin hydration in the institutionalized elderly. Methods: The subjects were 139 elders living in 2 nursing homes in Seoul. Data for skin health status were collected by measurement of skin hydration, sebum, trans-epidermal water loss and pH using Corneometer, Sebumeter, Tewameter and Skin pH-meter on face, flank, arm and leg. Baseline data were collected by a structured questionnaire. The data were analyzed by SPSS/WIN 12.0 program. Results: Skin hydration and pH were relatively low especially on arms and legs but sebum and TEWL were not low, relatively. There were significant differences in skin health status according to gender, smoking, alcohol intake, and bath cleanser. There was significant positive correlation between skin-hydration and bath-time, TEWL and bath-frequency. Smoking, bath-cleanser, bath-time. pH and gender were identified as the significant influencing factors of skin hydration. Conclusion: Smoking, bath-time and pH need to be considered as important control factors for increasing skin health status in the institutionalized elderly.

20대 한국인과 중국인의 피부특성 연구 (A Study of Skin characteristics of the Chinese and Korean people in their 20's)

  • 피부란 인체의 외부표면을 덮고 있는 조직으로서 물리적 화학적으로 외부로부터 신체를 보호하는 동시에 전신의 대사에 필요한 생화학적 기능을 수행하는, 생명유지에 없어서는 안 되는 기관이다. 피부의 해부학적인 구조와 생리학적인 기능은 부위, 성별, 연령, 인종 및 영양 상태 등에 따라 많은 차이를 보인다. 본 연구에서는 한국의 20대와 한국 거주하는 중국의 20대 피부의 보습능을 측정하였다. 수분량의 경우 중국인이 한국인보다 유의미하게 높은 것으로 나타났고, 피부수분증발량 또한 중국인이 한국인보다 낮게 나타났다. 측정결과 20대 한국인과 중국인에 대하여 동일한 주거환경 하에서의 피부 보습능을 측정한 결과, 중국인이 좋은 것으로 나타났다.

천연 밀단백질/계면활성제 복합체의 세정에 있어 피부보호 (The Cutaneous Protection for Detergent Formulation of Nature Wheat Protein Surfactant Complexes)

  • The cutaneous tolerability of detergent formulations can be improved by means of suitable additives. They complex the surfactant molecules lowering the concentration of their free monomeric species. Proteins derivatives used as additives for detergency are usually prepared by partial hydrolysis of plant reserve proteins. The main purpose of the hydrolytic cleavage is to make them water soluble and suitable for liquid products. Water solubility and stability are obtained by means of complexation with surfactants which also increase their actual hydrophobicity, an important parameter affecting cosmetic properties of proteins. Transepidermal water loss (TEWL) and electric capacitance (EC) have been adopted as investigation technigues to evaluate the skin integrity/damage in vitro tests, The performance of native wheat protein / surfactant complexes has been compared with traditional protein hydrolysates as detergent additives. The results show a noticeable reduction of skin irritation in surfactant formulations with addition of native wheat protein.

개에서 sodium lauryl sulphate에 의한 손상 피부에 대한 각질세포간 지질의 국소적용 효과 (Effect of Topical Application of Intercellular Lipids on Sodium Lauryl Sulphate-Damaged Skin Barrier Function in Dogs)

  • 세라마이드, 콜레스테롤, 자유지방산은 각질세포간 주요 지질로써 피부장벽의 생성과 유지에 있어 중요한 역할을 담당한다. 그러나, 개에서 각질세포간 지질에 대한 역할은 거의 알려져 있지 않다. 본 연구는 개에서 1.25% sodium lauryl sulphate (SLS)에 의한 피부 장벽손상 유발후 2% 세라마이드, 2%콜레스테롤, 2% 리놀레익산과 이들 세가지 지질의 혼합제 (혼합제)의 장벽손상 복구를 평가하고자 실시하였다. 피부장벽기능은 표피경유수분소실(TEWL), 피부 수화도, 피부 산도, 피부 두께 측정을 통하여 평가하였고 최종적으로 조직학적 분석과 투과 전자 현미경 (TEM)을 통하여 피부 장벽구조를 평가하였다. SLS는 개의 피부에 효과적으로 피부장벽 손상을 유발하였다. 표피경유수분소실은 세라마이드와 혼합제 적용부위에서 계면활성제 및 대조군과 비교하여 유의적인 감소를 나타내었다 (p<0.05, p<0.01). 표피경유수분소실은 12일째에 모든 지질 적용군에서 유의적으로 감소하였으나 특히 세라마이드와 혼합제에서 콜레스테롤과 리놀레익산보다 유의적으로 감소하였다 (p<0.01). 실험 기간 동안, 피부 수화도는 세라마이드와 혼합제에서 유의적 증가를 보였다 (p<0.01). 조직학적 분석에서도 각각의 지질들은 피부손상을 회복시켰다. 특히 세라마이드와 혼합제의 효과가 탁월하였고 각질층의 두께도 대조군에 비해 유의적으로 증가되었다. 피부 산도는 세라마이드, 리놀레익산, 혼합제에서 유의적으로 감소하였다 (p<0.05). 따라서 세라마이드와 혼합제의 국소적용은 계면활성제에 의한 피부 장벽 손상에 효과가 탁월함을 확인하였으며 아토피와 같은 염증성 피부염과 외부 자극에 의한 피부 장벽 손상시에도 세라마이드 또는 혼합제의 병용이 효과적일 것으로 사료된다.

락툴로스 함유 듀올리고 섭취에 의한 피부개선 효과 (Skin Health Effect of DuOligo Intake Containing Lactulose)

  • 본 연구에서는 락툴로스(lactulose) 51.67%와 갈락토올리고당(galactooligo saccharides) 15.8%로 이루어진 듀올리고(DuOligo)의 섭취를 통하여 피부개선 효능을 조사하였다. 40~60 대의 건강한 여성 37명을 대상으로 대조군(덱스트린)과 실험군(듀올리고군)으로 나누어 각각 8주 동안 섭취 후, 수분보유량, 경표피수 분손실량, 멜라닌지수, 홍반지수와 주름지수를 측정하였다. 그 결과, 듀올리고 섭취 8주 후의 수분 보유량은 대조군에 비하여 수분 보유량이 38.22% 유의적으로 증가하였다(p < 0.01). 대조군의 경피수분손실량은 8주후 $3.39g/h/m^2$로 감소한데 비하여 듀올리고군의 경피수분손실량은 8주후 $5.32g/h/m^2$ 감소하였다. 대조군의 멜라닌 지수는 시간에 따라 그 값이 유의적으로 증가하였으나, 듀올리고군의 멜라닌 지수는 시간에 따른 유의적 차이가 나타나지 않았다. 또한, 듀올리고군은 대조군에 비하여 주름의 총 넓이, 총 주름의 길이, 주름의 수 및 주름의 깊이가 확연하게 감소하였다(p < 0.05). 듀올리고의 피부개선 기능성 소재로서의 응용 가능성을 확인하였다.

아토피 피부염 환자에서 황연해독탕(黃連解毒湯)이 함유된 한방화장품에 대한 임상 연구 (Clinical Research of Atopic Dermatitis Treated by Hwangryeonhaedok-Tang in Cosmetics)

  • This clinical research was conducted to test patients with Atopic Dermatitis by external application with Hwangryeonhaedok-Tang in cosmetics. We gave scores to 31 patients who visited the Dept. of Oriental Medical Opthalmology & Otolaryngology & Dermatology of Semyung University Jecheon Oriental Medical Hospital from May 15th, 2008 to June 26th. Fifteen patients were treated with the ointment that contain Hwangryeonhaedok-Tang (experimental group) and sixteen patients were treated with normal ointment that doesn't have Hwangryeonhaedok-Tang (control group) for 4 weeks. We observed change of total IgE, eosinophil count, Skin Temperature, Transepidermal Water Loss(TEWL), Skin Hydration and Skin pH. Also Clinical Index of Atopic Dermatitis(SCORAD Index) and global assesment of efficacy were used to evaluate the effects of Hwangryeonhaedok-Tang. Statistical analysis was performed by using frequency analysis and descriptive analysis. Statistical significance was achieved if the probability was less than 5%(p<0.05). After 4 weeks of external application treatment, SCORAD Index in experimental group was significant statistically decreased compared with control group. After 4 weeks of external application treatment, total IgE of both groups were decreased and eosinophil count in control group was decreased but experimental group was unstatistically decreased. Unstatistically, both groups didn't showed significant effect on Skin Temperature. Transepidermal Water Loss(TEWL) in both groups were increased but experimental group was significant decreased compared with control group. Skin Hydration in experimental group was significant statistically increased compared with control group. Statistically, both groups didn't showed significant effect on Skin pH. Only experimental group showed little increase. After 4 weeks of external application treatment, experimental group showed significant effect on global assesment of efficacy. Considering the above results, we can speculate that cosmetics with Hwangryeonhaedok-Tang has some therapeutical effects in mitigating the symptoms of Atopic Dermatitis.

포스파티딜세린의 각질세포 분화 유도를 통한 피부장벽 기능 강화 (Phosphatidylserine Enhances Skin Barrier Function Through Keratinocyte Differentiation)

  • 포스파티딜세린(Phosphatidylserine; PS)은 생체막에서 구조적인 역할을 담당하는 인지질로서, 생체 내 다양한 세포작용에 필수적인 신호전달 효소의 보조인자로서 작용하는 것으로 알려져 있다. 하지만, PS의 생리활성에 대한 연구는 거의 이루어지지 않았고, 특히 피부에서의 생리활성에 대한 연구는 전무한 실정이다. 본 연구에서는 무모생쥐의 피부에 tape-stripping으로 경표피수분손실(TEWL)의 증가를 유도한 후, PS를 도포함으로써 그 손실을 현저히 감소시켰다. 또한, PS 도포군의 피부에서 세라마이드 함량이 증가된 사실을 확인한 바 있다. PS 도포군에서 non-hydroxyl 세라마이드와 glucosyl 세라마이드의 함량이 비처리군과 비교하여 각각 1.4배와 1.6배로 증가하였다. PS는 또한 피부각질세포의 분화를 촉진하였다. 피부각질세포에 PS를 처리함으로써 세포 형태가 분화상을 띄고 있음을 현미경 상에서 확인하였고, 표피분화의 특이적 표지 단백질인 Involucrin (INV)과 Transglutaminase 1 (TG'ase 1)의 발현이 각각 3.5배와 3배로 현저히 증가하였음을 웨스턴 블랏을 통하여 확인하였다. 또한 무모생쥐 피부에 PS를 도포한 결과 INV와 loricrin 단백질 발현이 증가하였다. 본 연구는 PS가 피부에서 생리활성을 나타낸다는 최초의 증거를 제시하며, 구체적으로는 각질세포 분화를 촉진함으로써 피부 세라마이드 함량을 증가시키고 경표피 수분손실을 감소시켜 궁극적으로 피부장벽을 강화하는 작용을 한다는 것을 보여준다.

The effects of Two Terpenoids, UA and ONA on Skin Barrier and Its Application

  • Ursolic acid (UA) and Oleanolic acid (ONA), known as urson, micromerol, prunol and malol, are pentacyclic triterpenoid compounds which naturally occur in a large number of vegetarian foods, medicinal herbs, and plants. They may occur in their free acid form or as aglycones for triterpenoid saponins, which are comprised of a triterpenoid aglycone, linked to one or more sugar moieties. Therefore UA and ON A are similar in pharmacological activity. Lately scientific research, which led to the identification of UA and ONA, revealed that several pharmacological effects, such as antitumor, hepatoprotective, anti-inflammatory, antimicrobial, and anti-hyperlipidemic could be attributed to UA and ONA. Here, we introduced the effects of UA and ONA on acute barrier disruption and normal epidermal permeability barrier function. To clarify the effects of UA and ONA on skin barrier recovery, both flank skin of 8-12 weeks hairless mice were topically treated with samples (2mg/ml) after tape stripping, then measured recovery rate using TEWL on hairless mice. The recovery rate increased in UA and ONA treated groups at 6h more than 20% compared to vehicle treated group (p <0.05). For verifying the effects of UA and ONA on normal epidermal barrier, hydration and TEWL were measured for 1 and 3 weeks after UA and ONA applications (2mg/ml per day). We also investigated the features of epidermis and dermis using electron microscopy (EM) and light microscopy (LM). Both samples increased hydration compared to Vehicle group from 1 week without TEWL alteration (p<0.005). EM examination using Ru04 and OsO4 fixation revealed that secretion and numbers of lamellar bodies and complete formation of lipid bilayers were most prominent (ONA$\geq$UA>Vehicle). LM finding showed that stratum corneum was slightly increased and especially epidermal thickening and flattening was observed (UA>ONA>Vehicle). Using Masson-trichrome and elastic fiber staining, we observed collagen thickening and elastic fiber increasing by UA and ONA treatments. In vitro results of collagen and elastin synthesis and elastase inhibitory experiments were also confirmed in vivo findings. This result suggested that the effects of UA and ONA related to not only skin barrier but also collagen and elastic fibers. Taken together, UA and ONA can be relevant candidates to improve barrier function and pertinent agents for cosmetic applications.

기허(氣虛), 혈허(血虛) 동물모형(動物模型)의 일반(一般) 특성(特性) 및 피부(皮膚) 특성(特性) 연구(硏究) (A Study on the Skin Characteristics of Qi Deficiency and Blood Deficiency Animal Model)

  • Back ground and Objective : There is a need for objectification and scientific verification of Pattern identification in Oriental medicine. The purpose of this study was to investigate the skin characteristics of Qi deficiency and Blood deficiency animal models. Material and Methods : Sprague-Dawley rats were divided into three groups: normal group, Qi deficiency group and Blood deficiency group. The Qi deficiency animal model was induced through restriction of food (75g/kg/day) for 20 days. Blood deficiency animal model was induced by bleeding from tail vein(0.3 ml/time) 8 times. The normal animal model was kept without any intervention. The general condition was observed by measuring body weight, body temperature, blood pressure, pulse rate, and hematological and biochemical parameters. The skin characteristics were observed by measuring the erythema index(EI), melanin index(Ml), transepidermal water loss(TEWL) and dermal microcirculation. Results : 1. In the Qi deficiency group, body weight was lower than the other groups. In the Qi deficiency group, blood pressure was lower than in the Normal group. There was no difference in body temperature and pulse rate between the three groups. 2. In the Qi deficiency group, blood sugar was lower than in the Blood deficiency group. There was no difference in triacylglycerol between the three groups. In the Qi deficiency group, the WBC count was lower than in the Blood deficiency group. RBC count was highest in the Qi deficiency group, Normal group and Blood deficiency group respectively. In the Qi deficiency group, Hb and Hct were higher than the other groups. 3. EI and MI were decreased in the Qi deficiency group, and EI showed a significant decrease. 4. EI and MI were increased in the Blood deficiency group, and MI showed a significant increase. 5. TEWL was significantly increased in the Qi deficiency group, while it was decreased in the Blood deficiency group, TEWL was highest in the Qi deficiency group, Normal group and Blood deficiency group respectively and all three groups showed significant difference. 6. In the Qi deficiency group, dermal microcirculation was lower than the other groups. Conclusion : The above results show that the erythema index decreases in the Qi deficiency model, and the melanin index increases in the Blood deficiency model. The Qi deficiency animal model shows an increase in transepidermal water loss, while the Blood deficiency animal model shows a decrease. Further studies should develop new models of Pattern Identification that are more specific.

석류농축액의 광노화에 의한 주름 개선 효과 (Effect of pomegranate concentration solution on photoaging)

  • Objective : Solar ultraviolet (UV) light can cause photoaging of human skin. Many researchers have focused on effective prevention to mitigate the aging process. This study was aimed to investigate anti-wrinkle effects of pomegranate concentration solution (PCS) using photoaged hairless mouse model. Method : To clarify the protective effect of PCS against UVB-induced photo-damage, water capacity was measured with transepidermal water loss (TEWL) in SKH-1 hairless mice. In addition, image of skin replicas was analyzed. Results : Our results showed that the PCS treatment protects skin against UVB-induced photoaging, based on the parameters including moisture capacity and TEWL. According to visual assessment of skin replica, application of 1 ml/kg PCS inhibited UV-induced wrinkle formation in mice skin as compared to the vehicle-applied control mice. Conclusion : Taken together, these results indicated that PCS could improve skin wrinkle formation induced by UVB irradiation in SHK-1 hairless mice. PCS could be applied as an anti-wrinkle agent.
