Preparation and Characterization of Carbon Nanofiber from Liquid Phase Carbon Source (액상법에 의한 Carbon Nanofiber 제조 및 특성 분석)

  • Nanostructured carbon materials have been found to have applications in fuel cell electrodes, field emitters, electronic devices, sensors and electromagnetic absorbers, etc. Especially, the CNF (carbon nanofiber) can be expected to play an important role in catalyst supporters for fuel cell electrodes and chemical reactions. In this study, we synthesized CNF from a liquid phase carbon source by a solvothermal method. In addition, we studied the parameters for the preparation of CNF by controlling heating and cooling rates, synthesis temperature and time. We characterized the CNF by SEM/TEM, XRD, Raman spectroscopy and EDS. We found that the heating and cooling rate have strong effects on the CNF formation and growth. We were able to prepare the best CNF at the heating rate of $10^{\circ}$/min, at $450^{\circ}$ for 60 minutes, and at the cooling rate of $4^{\circ}$/min. As a result of Raman spectra, we found that the sample showed two characteristic Raman bands at ${\sim}1350cm^{-1}$ (D band) and ${\sim}1600cm^{-1}$ (G band). The G band indicates the original graphite feature, but the D band has been explained as a disorder feature of the carbon structure. The diameter and length of the CNF was about $15{\sim}20nm$, and over $1{\mu}$, respectively.

Physicochemical Property Changes on Respiratory System of Rats After Intratracheal Instillation Exposure to Korea Chrysotile and Anthophyllite (국내산 백석면과 안소필라이트의 물리화학적 특성과 호흡기계 내 변화 연구)

  • Objectives: To assess the hazard of Korea chrysotile and anthophylite, fibers were analyzed for their physicochemical properties by transmission electron microscope equipped with energy dispersive X-ray spectrometer (TEM-EDS). Methods: To evaluate the biopersistence of 2 domestic asbestos, Sprague-Dawely rats were exposed to 2 mg asbestos by intratracheal instillation. Each asbestos (chrysotile ; $8,814,244{\times}10^6$ fibers/mg, average size $0.08{\mu}m{\times}4.39{\mu}m$, anthophyllite ; $5,182{\times}10^6$ fibers/mg, average size $0.95{\mu}m{\times}7.29{\mu}m$) were evaluated after a single intratracheal instillation. At times from 1 week to 4 weeks after exposure, the numbers of asbestos fivers in the bronchoalveolar lavage fluid and in the lung were calculated. Results: Anthophyllite fivers continuously have retained for 4 weeks but chrysotile fivers were rarely found at 4 weeks after exposure in the bronchoalveolar lavage fluid. Chrysotile fivers at 4 weeks after treatment were not observed but anthophyllite was easily observed in the lung with phase contrast microscopy. According to electron microscopic observation of asbestos in the lung, within 1 week after the administration of chrysotile fivers were decreased rapidly but anthophyllite fivers were very little change for 4 weeks. When chrysotile fivers have been lost Fe in 1 week, there were no significant changes in anthophyllite fivers in the lung. Conclusions: These findings indicate that after a long time exposure to chrysotile, asbestos bodies can not be found in the bronchoalveolar lavage fluid.

  • An in vitro immune effector mechanism against the target encysted metacercariae of Paragonimus westermani was demonstrated in the rat system. Peritoneal exudate cells, mainly macrophages from normal rats, showed adherence to and killing of encysted metacercariae of p. westermani in the presence of complement-independent serum from rats infected with Paragonimus metacercariae. These reactions were specific for the excysted metacercariae, as tissue-migrating juvenile worms were not affected. Damage of encysted metacercariae of p. westermani due to antibody and macrophages was assessed by morphological observation, by cell adherence reaction and by the use of vital dyes. frypan blue dye exclusion proved to be a reliable indicator of judging metacercarial viability. Electron microscopic studies demonstrated that macrophages reacted with fusty material on the tegumental surface and fine structures in the syncytium of the parasites. The tubular tunnels formed between the basement membrane and muscle layers of the damaged parasites were also noticeable. The relevance of these findings to cellular immunity in the early paragonimiasis was discussed.

Radioprotective Effects of Propolis on the Mouse Testis Exposed to X-ray. (프로폴리스가 X-선에 노출된 마우스 정소에 미치는 방사선 방어 효과)

  • The propolis is natural product produced by honeybees and is known to have many biologically useful properties such as anti-microbial, anti-oxidative and anti-tumorigenic activity. However, its radio-protective property has not been well studied. To investigate radio-protective effect of propolis on mouse testis, mice were supplemented with propolis after 5 Gy irradiation. The histological changes of testis were detected by TEM. The results indicate that propolis may protect tissue deformation which is induced by 5 Gy of ionizing radiation. Furthermore, to elucidate the potential molecular mechanisms involved in radio-protective property of propolis, we performed microarray experiments using oligo DNA microarray. We found 65 up-regulated genes and 224 down-regulated genes, whose expression levels were affected more than 2-fold by propolis treatment in mice irradiated at 5 Gy. We confirmed microarray data with reverse transcription-PCR using gene specific primers. The results of RT-PCR are highly correlated with those of microarray. These results may help understanding molecular mechanisms of radioprotective effects by propolis in mouse model.

  • Paenibacillus is a gram-positive, spore-forming aerobes that was previously classified as a Bacillus species. Paenibacillus sp. CK214 was highly motile on LB agar plates and showed typical colonial morphology of Paenibacillus. However, its motility was defective in the absence of glucose. Electron microscopic observation revealed that the cells of CK214 cultured on LB agar plates were peritrichously flagellated but not flagellated in the presence of glucose. Flagellar filaments were purified by centrifugation after shearing off from the CK214 cells with vigorous pipetting. The purified protein was composed of a single flagellin with an apparent molecular size of 29 kDa. Recognition of the protein by anti-Edwardsiella tarda flagellin protein antibody demonstrates that the protein is a flagellin protein. A decreased level of flagellin protein was detected in CK214 cells grown under glucose-supplemented media.

Development of a High-Resolution Electrocardiography for the Detection of Late Potentials (Late Potential의 검출을 위한 고해상도 심전계의 개발)

  • Most of the conventional electrocardiowaphs foil to detect signals other than P-QRS-T due to the limited SNR and bandwidth. High-resolution electrocardiography(HRECG) provides better SNR and wider bandwidth for the detection of micro-potentials with higher frequency components such as vontricular late potentials(LP). We have developed a HRECG using uncorrected XYZ lead for the detection of LPs. The overall gain of the amplifier is 4000 and the bandwidth is 0.5-300Hz without using 60Hz notch filter. Three 16-bit A/D converters sample X, Y, and Z signals simultaneously with a sampling frequency of 2000Hz. Sampled data are transmitted to a PC via a DMA-controlled, optically-coupled serial communication channel. In order to further reduce the noise, we implemented a signal averaging algorithm that averaged many instances of aligned beats. The beat alignment was carried out through the use of a template matching technique that finds a location maximizing cross-correlation with a given beat tem- plate. Beat alignment error was reduced to $\pm$0.25ms. FIR high-pass filter with cut-off frequency of 40Hz was applied to remove the low frequency components of the averaged X, Y, and Z signals. QRS onset and end point were determined from the vector magnitude of the sigrlaIL and some parameters needed to detect the existence of LP were estimated. The entire system was designed for the easy application of the future research topics including the optimal lead system, filter design, new parameter extraction, etc. In the developed HRECG, without signal averaging, the noise level was less than 5$\mu$V$_rms RTI$. With signal averaging of at least 100 beats, the noise level was reduced to 0.5$\mu$V$_rms RTI$, which is low enough to detect LPs. The developed HRECG will provide a new advanced functionality to interpretive ECG analyzers.

Analysis of Soot Particle Morphology Using Rayleigh Debye Gans Scattering Theory (RDG 산란 이론을 이용한 그을음 탄소 입자의 형상 분석)

  • Soot particles generated by fossil fuel combustion normally have fractal morphology with aggregates consisting of small spherical particles. Thus, Rayleigh or Mie elastic light scattering theory is not feasible for determining the fractal properties of soot aggregates. This paper describes a detailed process for applying Rayleigh-Debye Gans (RDG) scattering theory to effectively extract the morphological properties of any nano-scale particles. The fractal geometry of soot aggregates produced from an isooctane diffusion flame was observed using ex situ transmission electron microscopy (TEM) after thermophoretic sampling. RDG scattering theory was then used to analyze their fractal morphology, and various properties were calculated, such as the diameter of individual soot particles, number density, and volume fraction. The results show indiscernible changes during the soot growth process, but a distinct decreasing trend was observed in the soot oxidation process. The fractal dimension of the soot aggregates was determined to be around 1.82, which is in good agreement with that produced for other types of fuel. Thus, it can be concluded that the value of the fractal dimension is independent of the fuel type.

Antimicrobial Characteristics of Scutellariae Radix Extract (황금추출물의 항균특성)

  • It was carried out for research and development of natural antimicrobial on Scutellariae Radix extract against food-borne infection bacteria .Scutellariae Radix extract showed remarkable antimicrobial activites against Bacillus cereus, Listeria monocytogenes, Escherichia coli, and Vivro parahaemolyticus when examined by disk method, it was very stable on the wide rane of temperature and pH,.The growth rates of Bacillus cereus, Listeria monocytogenes, Escherichia coli, and Vivro parahaemolyticus were decreased at the concentration of more than 500 ppm Scutellariae Radix extract, Indicating that the minimum inhibitory concentration (MIC)of the Bacillus cereus, Listeria monocytogenes, Escherichia coli, and Vivro parahaemolyticus to Scutellariae Radix extract were around 500 ppm . The morphological changes were observed by transmission electron microscope and scanning electron microscope and the microbial cells membran was destroyed by Scutellariae Radix extract. It was identified that the membrane integrity of the sensitive cells was disrupted by exposure to Scutellariae Radix extract as the $\beta$-galactosidase test on experimental substrate ONPG(o-nitrophenyl-$\beta$-D-galacto-pyranoside)

A Study on the Preferred Orientation Characteristics of AlN Thin Films by Reactive Evaporation Method using NH3 (NH3를 이용한 반응성 증착법에 의한 AlN 박막의 우선배향특성에 관한 연구)

  • Aluminum nitride(AlN) is a compound (III-V group) of hexagonal system with a crystal structure. Its Wurzite phase is a very wide band gap semiconductor material. It has not only a high thermal conductivity, a high electrical resistance, a high electrical insulating constant, a high breakdown voltage and an excellent mechanical strength but also stable thermal and chemical characteristics. This study is on the preferred orientation characteristics of AlN thin films by reactive evaporation using $NH_3$. We have manufactured an AlN thin film and then have checked the crystal structure and the preferred orientation by using an X-ray diffractometer and have also observed the microstructure with TEM and AlN chemical structure with FT-IR. We can manufacture an excellent AlN thin film by reactive evaporation using $NH_3$ under 873 K of substrate temperature. The AlN thin film growth is dependent on Al supplying and $NH_3$ has been found to be effective as a source of $N_2$. However, the nuclear structure of AlN did not occur randomly around the substrate a particle of the a-axis orientation in fast growth speed becomes an earlier crystal structure and is shown to have an a-axis preferred orientation. Therefore, reactive evaporation using $NH_3$ is not affected by provided $H_2$ amount and this can be an easy a-axis orientation method.

Effect of Hot-stamping Heat Treatment on the Microstructure of Al-Segregated Zone in TWB Laser Joints of Al-Si-coated Boron Steel and Zn-coated DP Steel (Al-Si 도금된 보론강과 Zn 도금된 DP강 TWB 레이저 용접부내의 Al-편석부 미세조직에 미치는 핫스탬핑 열처리의 영향)

  • Al-Si coated boron steel and Zn coated DP steel plates were laser-welded to manufacture a Tailor Welded Blank (TWB) for a car body frame. Hot-stamping heat treatment ($900^{\circ}C$, 5 min) was applied to the TWB weld, and the microstructural change and transformation mechanism were investigated in the Al-rich area near the bond line of the Al-Si coated steel side. There was Al-rich area with a single phase, $Fe_3(Al,Si)$, which was transformed to ${\alpha}-Fe$ (Ferrite) after the heat treatment. It could be explained that the $Fe_3(Al,Si)$ phase was transformed to ${\alpha}-Fe$ during heat treatment at $900^{\circ}C$ for 5 min and the resultant ${\alpha}-Fe$ phase was not transformed by rapid cooling. Before the heat treatment, the microstructures around the $Fe_3(Al,Si)$ phase consisted of martensite, bainite and ${\alpha}-Fe$ while they were transformed to martensite and ${\delta}-Fe$ after the heat treatment. Due to the heat treatment, Al was diffused to the $Fe_3(Al,Si)$ and this resulted in an increase of Al content to 0.7 wt% around the Al-rich area. If the weld was held at $900^{\circ}C$ for 5 min it was transformed to a mixture of austenite (${\gamma}$) and ${\delta}-Fe$, and only ${\gamma}$ was transformed to the martensite by water cooling while the ${\delta}-Fe$ was remained unchanged.
