마디풀(Polygonum aviculare L.)의 전초가 지질과산화(脂質過酸化) 및 간기능(肝機能)에 미치는 영향 (Anti-lipid Peroxidation and Liver Protective Effects of Polygonum aviculare L.)

  • Polygonum aviculare L. was extracted by methanol and then fractionated systematically with solvents and column chromatographic method in order to isolate ingredients with anti-lipid peroxidation and liver protectective effects. Among these fractions, ethylacetate soluble part(MWE) showed the strongest in the anti-lipid peroxidation. Furthermore, the crude subtraction(MWE 4) with Rf value near 0.42, which is separated from MWE by column chromatography using a solvent system, inhibited the lipid peroxidation of rat liver in vitro. Moreover, MWE 4 decreased GOT, GPT and TBA value compared with control and suggested high protective effects against hepatotoxicity induced by carbon tetrachloride in mice. The active compounds in MWE 4 were assumed to be flavonoid glucosides.

계육과 돈육으로 제조한 Gold Cut의 저장에 따른 품질변화 (Quality Changes of Cold Cuts Prepared from Pork and Chicken during Storage)

  • Cold cuts were prepared from the pork and chicken with polyphosphate salt and spices. Microorganism was more proliferated and acid value was more decreased In pork cold cut rather than chicken cold cut. Otherwise TBA value of chicken cold cut was more increased than that of pork cold cut. Textures of springeness and cohesiveness were not significantly different in both cold cuts. From sensory evaluation, color, appearance and odor were significantly different, but texture, flavor and total acceptability were not significantly different. From these results, if we could study some antioxidants furthermore. we think it possible to manufacture cold cut from chicken.

즉석 추어탕을 냉동 건조미꾸라지의 저장성 (Storage Stability of Freeze Dried Loach for Instant Choo-o-tang)

  • Storage stability of boiled and freeze dried loach and antioxidative effect of Zanthoxylum schinifolium were studied to confirm the possibility in development of instant choo o tang(Korean traditional loach soup). Packaging and storage temperature did not cause a measurable change in in vitro protein digestibility and trypsin indigestible substrate within 45 days of storage but remarkable quality changes were occurred in all samples stored after 60 days. Vacuum packaging and low temperature storage(4 oC) had some effect in retarding protein quality deterioration due to delaying polyunsaturated fatty acid oxidation. Maximum peroxide value and TBA value were reached in 15 days, and there were a slow(TBA value) and rapid reduction(POV) after peaks were reached. In contrast, increasing brown pigment development and fluorescence intensity continued until 90 days of storage. Treatment of ethanolic extracts from Zanthoxylum schinifolium prior to freeze drying could protect against lipid oxidation of freeze dried loach products.

생약성분을 첨가한 소시지의 저장성에 대한 연구 (The Effect of Medicinal Plants with Additives on Storage Characteristics of Sausage)

  • This study was conducted to investigate the effects of medicinal plants with additives on storage characteristics of sausage. The water activity of the sausage with the additives tended to decrease with the storage time: the highest value was shown in the Angelica added sausage samples, in both cases added with dried powder as well as with extracted powder. The Paeonia extract powder added sausage had the lowest pH value. The herbal plant added sausages showed the lowest cooking and storage losses among all. The values of TBA (thiobarbituric acid value) and VBN (volatile basic nitrogen) increased with storage time, but the values of the sample with the extract powder additive were lower than those of the freeze-dry powder additive. Total plate counts of bacteria increased with storage time fur all samples, but those for the samples with Pueraria, Platycodon, Angelica, Bupleurum and Paeonia additives exhibited lower values than the control. From the above results, the sausages with medicinal plant additives showed the superior storage safety and quality characteristics.

Antioxidative Activity of Korean Wild Leaf Vegetables : Pleurospemum Kamtschaticum, Aderophora Remotiflor and Aster Glheni

  • The purpose of this study was to evaluate the antioxidative potential of Korean wild leaf vegetables in vivo as well as in vitro. The antloxidative activities of Pleurospemum kamtschaticum, Aderophora remotiflor and Aster gfheni were evaluated as a reference for Spinacia oleracea. Fifty rats were find either a control diet or one of several vegetable diets for 4 weeks. The level of thiobarbituric acid reactive substance (TBARS) and the activity of catalase and superoxide dismutase (SOD) in the liver were compared. The levels of total cholesterol (TC), high density lipoprotein cholesterol (HDL-C) and triglyceride (TG) in plasma were also compared. Also, the contents of $\beta$-carotene, vitamin C, vitamin E and total phenolic compound, including flavonoid and thiobarbituric acid (TBA) value using linoleic acid model system, were measured in Korean wind leaf vegetables. The TBARS values of Aderophora remotiflor and Aster giheni tended to be lower than that of Spinacia olerncea. The SOD activity of Artrr glheffi was significantly higher than that of Spinacia oleracea and reached 265% that of the control animals, whereas there was no difference between the control animals and the vegetable diet animals in terms of catalase activity. Compared to the control animals, TG and TC levels were significantly solver and showed a lower artheriosclerotic index. TBA values of Pleurospemum Kamschaticum and Aderophora remptiflor were only 18% of control value. TBA value of Spinacia olerucra was 41% of control value. These results suggest that Aderophora remotinor and Aster glheni could have antioxidative potency in vivo as well as in vitro and potential value far functional flood to improve the plasma lipid profile. flavonoid and phenolic compounds could be the major contributing factor in the antioxidative potential of Aderophora remotiflor and Aster glheni.

목초액의 아질산염 소거 및 항산화 활성 (Nitrite-Scavenging and Antioxidant Activities of Wood Vinegar)

  • 목초액의 아질산염 소거, 전자공여작용 및 대두유에 농도별(0.5, 1 및 5%)로 첨가하여 항산화성을 조사하였다. 아질산염 소거 효과는 모든 농도에서 pH가 증가하면서 감소하는 경향을 보였고, pH 1.2의 경우 100 ${\mu}\ell$/mL의 농도에서 80%이상의 높은 활성을 보였다. 전자공여작용은 50 ${\mu}\ell$/mL의 농도에서 약 80%의 효과를 나타내었다. 목초액을 첨가하지 않은 대두유를 control로 사용하였고, 0.1% BHT를 첨가한 대두유는 비교군으로 사용하여 5$0^{\circ}C$에서 10일간 저장하면서 실험을 하였다. 과산화물가는 목초액 1%와 5% 첨가군이 control과 BHT 첨가군보다 낮게 나타났으며, TBA가는 5% 목초액에서 가장 낮았고, 1% 목초액, 0.1% BHT 및 0.5% 목초액 첨가군 순으로 나타났다. 산가는 5% 목초액 첨가군이 가장 낮았고, 0.1% BHT, 1% 및 0.5% 목초액의 순으로 낮게 나타났다.

Riboflavin Tetrabutylate가 약물대사 효소 및 지질 과산화효소에 미치는 영향 (Effect of Riboflavin Tetrabutylate on the Activity of Drug Metabolizing Enzyme and Lipid Peroxidation in Liver Microsomes of Rats)

  • Lipid peroxidation in vitro has been identified as a basic deteriorative reaction in cellular mechanism of aging processes, such as air pollution oxidant damage to cell and to the lung, chlorinated hydrocarbon hepatotoxicity. Many experimental evidences were reported by several investigators that lipid peroxidation could be one of the principle causes for the hepatotoxicity produced by $CCl_4$. It is now reasonably established that $CCl_4$ is activated to a free radical in vivo, that lipid peroxidation occurs very quickly in microsomes prepared from damaged livers, that the peroxidation is associated with loss of enzyme activity of microsomes, and that various antioxidants can protect animals against the hepatotoxic effect of $CCl_4$. Recent studies have drawn attention to some other feature of microsomal lipid peroxidation. Incubation of liver microsomes in the presence of NADPH has led to a loss of cytochrome $P_{450}$. However, the presence of an antioxidant prevented lipid peroxidation and preserved cytochrome $P_{450}$. Decrease of cytochrome $P_{450}$ in microsomes under in vitro incubation can be enhanced by $CCl_4 and these changes were parallel to a loss of microsomal polyunsaturated fatty acid and formation of malonaldehyde. The primary purpose of this experiment was to study the effect of riboflavin tetrabutylate on lipid peroxidation, specially, the relationship between lipid peroxidation and drug metabolizing enzyme system which is located in smooth endoplasmic recticulum as well as the effect of ritoflavin tetrabutylate on drug metabolizing enzyme system of animal treated with $CCl_4$. Albino rats were used for experimental animal. In order to induce drug metabolizing enzyme system, phenobarbital was injected intraperitoneally. $CCl_$ and riboflavin tetrabutylate were given intraperitoneally as solution in olive oil. Microsomal fraction was isolated from liver of animals and TBA value as well as the activity of drug metabolizing enzyme were measured in the microsomal fractions. The results are summerized as following. 1) The secobarbital induced sleeping time of $CCl_4$ treated rat was about 2 times longer than that of the control group. However, the pretreatment with riboflavin tetrabutylate inhibited completely the lengthened sleeping time due to $CCl_4$ treatment. Furthermore TBA value was significantly increased in $CCl_4$ treated rat in comparison to control group tut the increase of TBA value was prevented by the pretreatment with riboflavin tetrabutylate. On the other hand, the activity of hepatic drug metabolizing enzyme was decreased in $CCl_4$ group, however, the pretreatment with riboflavin tetrabutylate also prevented the decrease of the enzyme activity caused by $CCl_4$. 2) The effect of riboflavin tetrabutylate on TBA value and the activity of drug metabolizing enzyme in vitro was similar to in vivo results. Incubation of liver microsome from rat in the presence of $CCl_4$, $Fe^{++}$, or ascorbic acid has led to the marked increase of TBA value, however, the addition of riboflavin tetrabutylate in incubation mixture prevented significantly the increase of TBA value, suggesting the inhibition of lipid peroxidation. In accordance with TBA value, the activity of drug metabolizing enzyme was inhibited in the presence of $CCl_4$, $Fe^{++}$, ascorbic acid but the addition of riboflavin tetrabutylate protected the loss of the enzyme activity in microsome under in vitro incubation.

생강의 첨가가 분쇄막창의 저장특성에 미치는 영향 (Changes in Storage Characteristics of Pork Rectum by Addition of Ginger (Zingiber officinale Roscoe))

  • 신선한 암퇘지의 막창부위에 지방을 제거하고 파쇄기로 잘게 부수어 가열하지 않은 것과 $100^{\circ}C$의 autoclave에서 10분간 가열한 것에 분쇄한 생강을 1, 5 및 10%가 되게 첨가하여 polyethylene film으로 1차 포장한 후 알루미늄 호일로 2차 포장한 다음 $5^{\circ}C$에서 보관하면서 품질의 변화를 관찰하였다. 저장 중 pH는 첨가구 사이의 유의적인 차이를 나타내지 않았으나, 가열한 막창이 다소 낮은 값을 나타내었다. TBA가는 각각 0.1 mg/kg과 0.25 mg/kg으로 가열 막창이 생막창보다 높은 값을 나타내었으며, 두 처리구에서 모두 생강의 첨가가 TBA-value의 상승을 억제시켜 저장성을 연장시키는 것을 확인할 수 있었다. 휘발성 염기질소의 함량은 생강의 첨가로 감소하였으며, 30~40 mg%의 초기부패 판정기준을 고려할 때 생막창은 $5^{\circ}C$에서 4일, 가열막창은 6일 이내에 소비하는 것이 바람직할 것으로 판단되었다. VBN 함량을 기준으로 10% 생강의 첨가는 가열막창의 저장기간을 4일 가량 연장시킬 수 있는 것으로 확인되었다.

Bacillus polyfermenticus SCD 에탄올 추출물이 떡갈비의 품질 및 저장성에 미치는 영향 (Effects of Ethanol Extracts of Bacillus polyfermenticus SCD on Tteokgalbi Quality during Storage)

  • 본 연구에서 대조구는 항산화제를 첨가하지 않았고, ascorbic acid(0.06%)를 첨가한 처리구(T1), B. polyfermenticus SCD 에탄을 추출물(5%)을 첨가한 처리구(T2)와 ascorbic acid(0.03%) 및 B. polyfermenticus SCD 에탄을 추출물(2.5%)를 첨가한 처리구(T3)로 구분하여 실험을 실시하였다. 비타민 C 혹은 B. polyfermenticus SCD의 ethanol 추출액을 첨가한 떡갈비는 저장성이 향상되었다. 특히 지질산패도의 경우 처리구들은 대조구에 비하여 현저히 낮은 값을 보였을 뿐만 아니라, B. polyfermenticus SCD의 에탄을 추출물을 첨가한 처리구가 가장 낮은 값을 나타냈다. 휘발성 염기태질소값은 항산화제를 넣은 모든 처리구에서 낮은 결과를 보였다. 따라서, 기존에 많은 연구를 통해 항산화제로 많이 쓰여지고 있는 비타민 C와 더불어 B. polyfermenticus SCD의 에탄올 추출물이 뛰어난 천연항산화 능력을 보였다

방아 추출물의 항산화 효과 (Antioxidative Activity of Extract from Bangah Herb)

  • 방아의 ether추출물을 중성, 페놀성, 산성 및 염기성 획분으로 분획한 다음 그들의 항산화 활성을 수소 공여능, 과산화물가, TBA가 및 흰쥐 간 microsome을 이용한 비효소적 지질 과산화 억제 활성 등으로 측정하고, 항산화 활성이 비교적 높게 나타나는 페놀성 획분과 중성 획분을 다시 소획분으로 분획한 다음 그중에서 항산화 활성이 강한 소획분성분을 GC/MS 및 NMR로 분석, 동정하였다. 페놀성 획분은 DPPH에 의한 수소 공여능, POV, TBA가 및 흰쥐 간 microsome의 지질 과산화 억제 활성 등 모두에서 높게 나타났으며, 중성 획분은 흰쥐 간 microsome의 지질 과산화 억제 활성에서 높게 나타났다. 페놀성 획분을 소획분으로 분획하였을 때 소획분 P-1, P-2 및 P-3에서 항산화 활성이 강하였으며, 특히 P-2는 합성 항산화제인 BHT나 천연 항산화물질로 알려져 있는 caffeic acid보다도 강하였다. 소획분 P-2를 TMS 유도체로 만들어 GC로 분석하였을 때 major peak 성분인 Peak I, II, III 및 IV 모두 TMS m/z 73를 base peak로 가지고 있어 4개의 peak 모두 -OH, -COOH기를 가지고 있는 것으로 확인할 수 있었다. 중성 획분의 소획분 중에는 N-2에서 강한 활성을 나타내었고 소획분 N-2의 주성분을GC/MS 및 NMR로 분석 한 결과 estragole로 확인되었다. Estragole의 흰쥐 간 microsome의 지질 과산화 50% 억제 농도는 $20{\sim}50\;{\mu}g/ml$로서 BHT보다는 약하였으니 caffeic acid, gallic acid와는 비슷한 수준의 항산화 활성을 나타내었다.
