  • Experiments were carried out to the effect of various amino acids on the production of tetraacetylphytosphingosine (TAPS). Among various amino acids were tested, cysteine decreased the production of TAPS at 7 days of flask culture. Especially, Serine is precursor of TAPS but didn't affect the production of TAPS. Glycine and glutamate increased the production of TAPS. Especially, glutamate increased 20% of cell mass and 37% of TAPS production.

  • Ceramides are a major lipid class known to play an essential role in maintaining skin function. Thus, efforts have been made to produce ceramides and ceramide precursors in large quantities for industrial applications. The yeast Wickerhamomyces ciferrii, a natural producer of the ceramide precursor tetraacetyl phytosphingosine (TAPS), has been isolated and engineered through various mutagenesis approaches aiming to enhance TAPS production. Herein, a high-throughput screening platform for isolating W. ciferrii mutants with improved TAPS production is described. A fluorescence-mediated reporter system that allows initial quantification of TAPS content in yeast cells based on BODIPY staining was developed. The optimal concentration of BODIPY for monitoring intracellular TAPS levels in W. ciferrii was 400 ㎍/L, as shown by a linear correlation between the actual TAPS levels and mean fluorescence intensities. Fluorescence-activated cell sorting was used for isolating high TAPS-producing strains from an ethyl methanesulfonate-induced mutant library. After several rounds of sorting, mutants exhibiting a high-TAPS phenotype were isolated, and the M40 strain with the highest TAPS titer was chosen for large-scale cultivation. The influence of different carbon sources for optimizing TAPS production was also evaluated using the M40 strain. A maximum production yield of 5.114 g/L of ceramide precursors, including TAPS and triacetyl phytosphingosine, was achieved with the supplementation of molasses. This novel platform enables rapid screening of high TAPS-producing strains using the common dye BODIPY and can be easily extended for the development of mutants with high productivity of ceramide precursors in yeast and other microorganisms.

  • Experiments were carried out to develop of mutant strain from Pichia ciferrii ATCC 14091 and investigate the effect of sodium acetate on the production of tetraacetylphytosphingosine (TAPS). A high-TAPS producing mutant was simply selected by staining with sudan black B and colony shape after the treatment of UV and NTG. Sodium acetate enhanced the TAPS production.

Proposed approach for determination of tributary areas for scattered pressure taps

  • In wind load calculations based on pressure measurements, the concept of 'tributary area' is usually used. The literature has less guidance for a systematic computational methodology for calculating tributary areas, in general, and for scattered pressure taps, in particular. To the best of the author's knowledge, there is no generic mathematical equation that helps calculate the tributary areas for irregular pressure taps. Traditionally, the drawing of tributary boundaries for scattered and intensively distributed taps may not be feasible (a time and resource consuming task). To alleviate this problem, this paper presents a proposed numerical approach for tributary area calculations on rectangular surfaces. The approach makes use of the available coordinates of the pressure taps and the dimensions of the surface. The proposed technique is illustrated by two application examples: first, quasi-regularly distributed pressure taps, and second, taps that have scattered distribution on a rectangular surface. The accuracy and the efficacy of the approach are assessed, and a comparison with a traditional method is presented.

Optimization of cell growth and TAPS production by Pichia ciferrii mutant in batch culture

  • Batch culture of mutant derived from Pichia ciferrii ATCC 14091 was investigated for producing the intracellular tetraacetylphytosphingosine (TAPS). Composite experimental design was used to optimize the composition of the culture medium for maximizing the productivity of TAPS. In this experiment, various culture parameters were investigated that were the effects of temperature, the initial culture pH, the carbon-to-nitrogen ratio, the concentration of trace elements, and the concentration of cofactors. The optimal temperature for cell growth and TAPS synthesis appeared to be %25^{\circ}C$. An initial pH value of 7.5 gave the best results. Under the best condition, the maximum TAPS concentration indicated 7.2 g/L and its productivity was 0.06 g/L-hr in a 2.5 L jar

Against a Lenition Account of Tapping: Evidence from Yonbyon Korean

  • The purpose of this study is to revisit the property of tapping, based on the data from Yonbyon Korean. Taps have been described as short segments derived from corresponding stops or trills. It is also widely assumed that tapping occurs due to lenition to minimize articulatory effort. However, Yonbyon Korean data show that taps can occur in strong as well as weak positions The results of the acoustic experiments conducted in this study show that in syllable-onset position, obstruent taps consistently appear from the underlying laterals, while in intervocalic position, sonorant taps similar to American English taps occur. The results of this study provide evidence against the uniform account of tapping as the result of lenition.

New GGNMOS I/O Cell Array for Improved Electrical Overstress Robustness

  • A 0.18-${\mu}m$ 3.3 V grounded-gate NMOS (GGNMOS) I/O cell array for timing controller (TCON) application is proposed for improving electrical overstress (EOS) robustness. The improved cell array consists of 20 GGNMOS, 4 inserted well taps, 2 end-well taps and shallow trench isolation (STI). Technology computer-aided design (TCAD) simulation results show that the inserted well taps and extended drain contact gate spacing (DCGS) is effective in preventing EOS failure, e.g. local burnout. Thermodynamic models for device simulation enable us to obtain lattice temperature distributions inside the cells. The peak value of the maximum lattice temperature in the improved GGNMOS cell array is lower than that in a conventional GGNMOS cell array. The inserted well taps also improve the uniformity of turn-on of GGNMOS cells. EOS test results show the validity of the simulation results on improvement of EOS robustness of the new GGNMOS I/O cell array.

Establishment of an easy Ic measurement method of HTS superconducting tapes using clipped voltage taps

  • The critical current, $I_c$ of HTS superconducting tapes can be measured by transport or contactless method. Practically, the transport method using the four-probe method is the most common. In this study, a simple test procedure by clipping the voltage lead taps have been introduced instead of soldering which reduces time and effort and thereby achieving a much faster measurement of $I_c$. When using a pair of iron clips, $I_c$ value decreased as compared with the measured one by standard method using soldered voltage taps and varies with the width of the clipped specimen part. However, when using a pure Cu clip, both by clipping and by soldering voltage taps give a comparable result and $I_c$ measured are equal and close to the samples specification. As a result, material to be used as voltage clip should be considered and should not influence the potential voltage between the leads during $I_c$ measurement. Furthermore, the simulation result of magnetic flux during $I_c$ measurement test showed that the decrease of $I_c$ observed in the experiment is due to the magnetic flux density, $B_y$ produced at the clipped part of the sample by the operating current with iron clips attached to the sample.

A deep learning framework for wind pressure super-resolution reconstruction

  • Strong wind is the main factors of wind-damage of high-rise buildings, which often creates largely economical losses and casualties. Wind pressure plays a critical role in wind effects on buildings. To obtain the high-resolution wind pressure field, it often requires massive pressure taps. In this study, two traditional methods, including bilinear and bicubic interpolation, and two deep learning techniques including Residual Networks (ResNet) and Generative Adversarial Networks (GANs), are employed to reconstruct wind pressure filed from limited pressure taps on the surface of an ideal building from TPU database. It was found that the GANs model exhibits the best performance in reconstructing the wind pressure field. Meanwhile, it was confirmed that k-means clustering based retained pressure taps as model input can significantly improve the reconstruction ability of GANs model. Finally, the generalization ability of k-means clustering based GANs model in reconstructing wind pressure field is verified by an actual engineering structure. Importantly, the k-means clustering based GANs model can achieve satisfactory reconstruction in wind pressure field under the inputs processing by k-means clustering, even the 20% of pressure taps. Therefore, it is expected to save a huge number of pressure taps under the field reconstruction and achieve timely and accurately reconstruction of wind pressure field under k-means clustering based GANs model.
