The sprA and sprB genes, encoding the chymotrypsin-like proteases Streptomyces griseus protease A (SGPA) and Streptomyces griseus protease B (SGPB), and the sprT gene that encodes Streptomyces griseus trypsin (SGT) were cloned from S. griseus and were overexpressed in various strains of S. griseus. When the sprT gene was introduced into S. griseus, trypsin activity increased 2-fold in the A-factor deficient mutant strain, S. griseus HH1, and increased 4-fold in the wild strain, S. grise us IFO 13350. However, there was no detectable increase of chymotrypsin activity in the transformants of S. griseus with either sprA or sprB, in contrast to the results obtained from S. lividans as a heterologous host. To solve the negative gene dosage effects in S. griseus, either the sprA or the sprB genes with their own ribosome binding sites were linked to the downstream of the entire sprT gene, and the coexpression efficiency was examined in S. lividans and S. griseus. The transformants of S. lividans with either pWHM3-TA (sprT+sprA) or pWHM3TB (sprT+sprB) showed 3-fold increase of trypsin activity over that of the control, however, only the transformant of pWHM3-TB demonstrated 7-fold increase in chymotrypsin activity, indicating that the pWHM3-TB has a successful construction for the overexpression of chymotrypsin in Streptomyces. When the coexpression vectors were introduced into S. griseus IFO 13350, the trypsin level sharply increased by more than 4-fold, however, the chymotrypsin level did not increase. These results strongly suggest that the overexpression of the sprA and sprB genes is tightly regulated in S. griseus.
Quality of fresh-cut tomato slices was compared during cold storage under various modified atmosphere packaging conditions. Chilling injury of slices in containers sealed with Film A was higher than with Film B; these films had oxygen transmission rates of 87.4 and 60.0 ml h-1 m-2 nun-1 at $5^{\circ}C$ and $99\%$ RH, respectively. While slices in containers with an initial atmospheric composition of air, $4\%$ CO2 + 1 or $20\%\;O_2, \;8\%\;CO_2+1$ or $20\%\;O_2$, or $12\%\; CO_2+\;20\%\;O_2$ showed fungal growth, slices in containers with $12\%\;CO_2 +\;1\%\;O_2$ did not. Low ethylene in containers enhanced chilling injury. Modified atmosphere packaging provided good quality tomato slices with a shelf-life of 2 weeks or more at $5^{\circ}C$. Experiments were conducted to compare changes in quality of slices of red tomato (Lycopersicon esculentum Mill. 'Sunbeam') fruit from plants grown using black polyethylene or hairy vetch mulches under various foliar disease management systems including: no fungicide applications (NF), a disease forecasting model (Tom-Cast), and weekly fungicide applications (WF), during storage at $5^{\circ}C$ under a modified atmosphere. Slices were analyzed for firmness, soluble solids content (SCC), titratable acidity (TA), pH, electrolyte leakage, fungi, yeasts, and chilling injury. With both NF and Tom-Cast fungicide treatments, slices from tomato fruit grown with hairy vetch (Vicia villosa Roth) mulch were firmer than those from tomato fruit grown with black polyethylene mulch after 12 days storage. Ethylene Production of slices from fruit grown using hairy vetch mulch under Tom-Cast was about 1.5- and 5-fold higher than that of slices from WF and NF fungicide treatments after 12 days, respectively. Objectives: The occurrence characteristics of BTEXS and phytoncides were investigated by type of urban forest. Methods: Four types of urban green space (Hongneung Forest, Mt. Chunjang, residential park, and traffic island) and Gwangneung Forest were selected. Monitoring of phytoncides and BTEXS was conducted considering the activity times of urban residents (five times per day) using a Tenax TA tube and suction pump in June 2017 (one day). Results: Phytoncide concentrations were ranked as Gwangneung Forest>Hongneung Forest>Mt. Cheonjang>traffic island>residential park. Relatively high concentrations of phytoncides were also identified in the urban forest. There was no significant difference between Gwangneung Forest and the urban forest. BTEXS concentrations were ranked as traffic island>residential park>Hongneung Forest>Gwangneung Forest>Mt. Cheonjang. Traffic island and residential park showed high levels of BTEXS depending on the inflow of vehicles. The difference in concentration by time was significant for the traffic island in particular. Pollutant levels in Hongneung Forest were as low as in Gwangneung Forest. Conclusion: The concentrations of phytoncides and BTEXS were different by types of urban green space, and the potential for health and hygiene of urban forests were able to be investigated. This study is expected to provide as basic data for the creation of urban forest spaces in the future.
This study aimed to evaluate the efficacy of fructose-1,6-bis phosphate aldolase (SMALDO) DNA vaccination against Schistosoma mansoni infection using different routes of injection. The SMALDO has been cloned into the eukaryotic expression vector pcDNA3.1/V5-His TOPO-TA and was used in injecting Swiss albino mice intramuscularly (IM), subcutaneously (SC), or intraperitoneally (IP) ($50{\mu}g/mouse$). Mice vaccinated with non-recombinant pcDNA3.1 served as controls. Each group was immunized 4 times at weeks 0, 2, 4, and 6. Two weeks after the last booster dose, all mice groups were infected with 80 S. mansoni cercariae via tail immersion. At week 8 post-infection, animals were sacrificed for assessment of parasitological and histopathological parameters. High anti-SMALDO IgG antibody titers were detected in sera of all vaccinated groups (P<0.01) compared to the control group. Both the IP and SC vaccination routes resulted in a significant reduction in worm burden (46.2% and 28.9%, respectively, P<0.01). This was accompanied by a significant reduction in hepatic and intestinal egg counts (41.7% and 40.2%, respectively, P<0.01) in the IP group only. The number of dead eggs was significantly increased in both IP and IM groups (P<0.01). IP vaccination recorded the highest significant reduction in granuloma number and diameter (54.7% and 29.2%, respectively, P<0.01) and significant increase in dead miracidia (P<0.01). In conclusion, changing the injection route of SMALDO DNA vaccination significantly influenced the efficacy of vaccination. SMALDO DNA vaccination via IP route could be a promising protective and antipathology vaccine candidate against S. mansoni infection.

The purpose of this study was to study accumlated residual soils which may be one of the causes for yellowing of worn cloths. Wear and wash tests of white cotton undershirts were repeated at 30 households sellected at random over a period of 60 days. Laundry conditions were similar to home laundry habits in a fact-finding survey, using a powdery heavy duty detergent containing no enzymes or enzymes. The subjects in this study were survey of laundry actual condition, the undershirts from prior to and after the final washing was measured residual soils, $L^*a^*b^*$ value and mellowness index of CIE system. D3ta were analysed by simple correlation analysis of wear and wash cycle, residual soils, whiteness The results obtained were summarized as follows: 1. Using pattern of washing machine, Presoaking was no singinificant differnece in general characteristics of survey respondent. Laundry frequency was significant difference in income level, occupation of housewives whether or not. Use of cold and hot water was significant difference in residence shape. 2. The analyzed consequences of recognition and actual behavior in connection with laundry were found variables each other to have independence or not. 3. Amount of residual sebum soils is using non-enzyme detergent were much more than in using enzyme detergent, increased linearly with increase of the number of wear and wash cycles. 4. Residual protein soils with increase of the number wear and wash cycles less than in laundering more easy than sebum soils. Since accumulated residual sebum soils were much more than residual protein soils. 5. Increase of residual soils was raised mellowness index and diminshed whiteness. yellowness index of residual sebum soils was higher than protein soils. If increase of whiteness will be incresed, amount of residual sebum soils will be decreased sebum soils. Because amount of residual sebum soils much more than protein soils, yellowness index of residual sebum soils was more higher than that of protein soils.
The purpose of this study was to determine the biomechanical effect of wearing the mouthguard on the lower limb during drop landing. Nine male university students who have no musculoskeletal disorder were recruited as the subjects. Linear velocity, angular velocity, vertical GRF, loading rate, joint moment, and lower extremity muscle activity were determined for each subject. For each dependent variable, paired t-test was performed to test if significant difference existed between with mouthguard (WM) and without mouthguard (WOM) conditions (p<.05). The results showed that linear velocity, angular velocity, vertical GRF and loading rate were no significant difference between the two groups. The inversion moment of the ankle joint was increased in WM compared to WOM. Average IEMG values from BF, TA, and LG in WM were significantly greater than corresponding values in WOM during IP phase. This indicates that wearing mouthguard played a vital role in muscle tuning for maintaining joint stability of the lower limb and preventing injury.

스퍼터링 소스는 전자기 박막 등 기능성 박막을 비롯하여 결질피막, 장식성 피막등의 제조에 이용되는 것으로 각종 증발원 중에서 가장 널리 사용되는 증발원이다. 70년대 이후 스퍼터링 소스는 마그네트론 스퍼터링으로 대표되는 방식이 사용되어 왔으며 지금까지도 가장 일반적인 방식이 되어 왔다. 마그네트론 스퍼터링 증발원은 증발율에서는 기술적인 향상이 이루어진 반면 이온화율의 향상은 그다지 이루어지지 않아 경질피막과 같은 화합물 피막의 특성 향상에는 한계를 드러내게 되었다. 그러다가 186년 Window 등 이 자장의 세기를 변형시킨 비평형 마그네트론 소스(Unbalanced Magnetron;UBM)를 처음 발표하여 이온화율의 향상이 가능하다는 것이 알려지면서 이에 대한 많은 연구가 진행되었다. UBM 소스는 마그네트론 스퍼터링 소스의 외부에 전자석을 설치하여 기판에 흐르는 이온의 양을 증가시킴으로써 소스와 기판사이의 거리를 증가시킬 수 있고 따라서, 복잡한 형상의 부품코팅이 가능하며 피막 특성을 향상시킬 수 있는 장점이 있다. 본 연구에서는 UBM 스퍼터링 소스를 설계, 제작하여 그 특성을 다양한 측면에서 조사하였다. 특히, 자작의 최적 설계를 통해 전자석의 조건을 도출하였음, Dual UBM 소스의 특성을 동시에 조사하였다. 자기장의 simulation에는 Quick field 프로그램을 이용하였고 기존의 방식과의 비교를 통해 최적의 조건을 도출하였다. 이를 바탕으로 inner pole의 크기를 30mm, outer pole의 크기를 26mm로 고정하여 설계하였고, 외부에 전자석이 설치된 UBM 소스를 제작하였다. 본 UBM 소스는 4" 타겟을 사용할 수 있으며 전자석의 조건을 10A까지 변화시켜 자기장의 세기를 변화시킬 수 있게 하였다. 제작된 소스의 동작조건 설정과 최적화를 위한 스퍼터링 장치를 함께 제작하여 UBM 소스의 최적 동작 조건을 도출하였다. 전자석의 전류가 4.5A일 때 Inner Pole과 Outer Pole의 자기장의 세기가 도일함을 알 수 있었다. 기판과 타겟의 거리가 200mm일 경우에 기판에 흐르는 전류밀도는 2mA/cm2이상이 됨을 확인하였다. 이 결과는 기존의 마그네트론 소스가 기판과 타겟사이의 거리가 100mm일 때 1mA/cm2 정도가 되는 것과 비교하면 이온화율이 획기적으로 향상된 것임을 알 수 있다.수 있다.

This is a retrospective study on the nature and severity of zygomatic bone fractures. This study was based on a series of 358 patients with zygomatic bone fractures who treated as in-patient at Soonchunhyang Chonan Hospital during the period of Jan. 1993 through Dec. 1996. The results obtained are as follows : 1. The ratio of men to women was 4.41:1. The age frequency was highest in the second, third decade. The yearly distribution was the highest in 1993. The monthly distribution was highest in Feb. and Jun. 2. The most frequent cause was the traffic accident.(66.2%) 3. The most frequent type of fractures was the class I(undisplaced fx.).(25%) 4. The most frequent associated oral and maxillofacial injuries was soft tissue.(46.9%) 5. The most frequent associated systemic injuries was upper, lower extremity.(38%) 6. In A.I.S of oral and maxillofacial area, the mean score of A.I.S was $2.5{\sim}3$. In I.S.S, the highest mean score was in the ped TA.(19) 7. In respect of treatment, the most common method was open reduction with rigid fixation on fronto-zygomatic suture area.(47.8%) 8. The incidence of complication was 12% and the most common complication was malunion.(6.1%)
Termites play an important role in the degradation of dead plant materials and have acquired endogenous and symbiotic cellulose digestion capabilities. The feruloyl esterase enzyme (FAE) gene amplified from the metagenomic DNA of Coptotermes formosanus gut was cloned in the TA cloning vector and subcloned into a pET32a expression vector. The Ft3-7 gene has 84% sequence identity with Clostridium saccharolyticum and shows amino acid sequence identity with predicted xylanase/chitin deacetylase and endo-1,4-beta-xylanase. The sequence analysis reveals that probably Ft3-7 could be a new gene and that its molecular mass was 18.5 kDa. The activity of the recombinant enzyme (Ft3-7) produced in Escherichia coli (E.coli) was 21.4 U with substrate ethyl ferulate and its specific activity was 24.6 U/mg protein. The optimum pH and temperature for enzyme activity were 7.0 and $37^{\circ}C$, respectively. The substrate utilization preferences and sequence similarity of the Ft3-7 place it in the type-D sub-class of FAE.
