The seasonal and spatial changes in dissolved organic carbon(DOC) in Lake Kasumigaura, a shallow and eutrophic lake, were analyzed from October 1992 to October 1995. The proportion of T-DOC was classified by labile(L-DOC) and refractory DOC(R-DOC) on the basis of long-term incubation, fractionated the molecular weight of T-DOC by ultrafiltration. The porewater DOC were measured at sedimental surface of the central basin in order to evaluate the DOC released from the sediment. The proportion of L-DOC and R-DOC were accounted for about 15% and 85% of T-DOC in the central basin, respectively. The molecular weight(MW) distribution occupied some 60% of the low and medium MW. The horizontal variation of T-DOC concentrations trended to higher in the central basin than in the inlet of influent rivers, because of contribution by autochthonous organic carbon loading. The seasonal variation of T-DOC showed to higher summer than winter in the inlet of influent, but at the central basin it fluctuated little seasonally. During the high increase of porewater DOC in 1994 evaluated the high release possibility from the sediment surface (10cm). The present study suggests that autochthonous organic carbon loading must be controlled for improving the water quality of the eutrophic lakes.
Seasonal and vertical distribution of water quality were investigated from May 2001 to June 2002 in Kumoh reservoir located nearby Kumi City, Kyungpook. Kumoh reservoir that lost the role of agricultural irrigation is currently of rapid eutrophication. The vertical distribution of DO was observed clinograde with hypolimnetic anoxic zone. T-P concentrations at the surface ranged from 0.008 to 0.152 mgP/L and T-N concentrations ranged from 1.4 to 3.0 mgN/L. The vertical and seasonal variation of T-N was smaller than T-P. DOC concentrations, indicator of organic matter pollution, ranged from 2.8 to 5.4 mgC/L. Apportionment of Total-DOC (T-DOC) indicated that 14% of T-DOC was attributed to Labile-DOC(L-DOC) and the rest was due to Refractory-DOC(R-DOC). The values of TSI(Trophic State Index) ranged between 44 and 52 indicating that Kumoh reservoir is under mesotrophic condition. The results of this study indicate that Kumoh reservoir is likely to be under influence of eutrophication and thus water quality will be aggravated. Therefore, the Kumoh reservoir requires further treatment to improve water quality and a plan of the reusing water resource should be developed.

This study was conducted to evaluate the characteristics of dissolved orgamc matters based on their origins. The dissolved organic carbon(DOC) represents an index for dissolved organic matter and basically regarded as a source of organic pollution. The monthly variations and vertical profiles of dissolved organic carbon(DOC) in Kumoh reservoir were surveyed from May 2001 to April 2002. In addition, other areas such as river, reservoir, sewage and industrial wastewater were also surveyed in summer 2001. Kumoh reservoir was divided with depth into three layers .: epilimnion, metalimnion and hypolimnion. The proportion of total DOC(T-DOC) was classified by labile DOC(L-DOC) and refractory DOC(R-DOC) on the basis of long-term incubation. DOC of freshwater and Kumoh reservoir was ranged to be 1.6~4.1 mgC/L and 2.1~4.0 mgC/L, respectively. L-DOC accounted for 3~30% of DOC from watershed. Therefore, refractory dissolved organic carbon(R-DOC) was major component of DOC in the watershed. The decomposition rate(k) ranged from 0.008 $d^{-1}$ to 0.083 $d^{-1}$ in Kumoh reservoir. The highest decomposition rate(k) was observed at River Hoein III freshwater. Therefore, modified total organic carbon analyzer is needed to be applied for effective management of dissolved organic matter.
The effect of ozone oxidation on bio-degradability of leachate was studied. Ozone oxidation process was used as pre-treatment process to enhance performance of biological process in treating landfill leachate. Optimum ozone injection rate and contact time in this experiment was $160{\;}mg{\;}O_/{\ell}{\cdot}hr$ and 45 minutes, respectively. Bio-degradability was enhanced 5.08% by ozone oxidation. The ratio of ozone demand/DOC concentration was $0.049~0.091{\;}mg{\;}O_3/mg{\;}DOC$. The increase of bio-degradability depending on ozone injection rate(D) and contact time(T) can be expressed as follows ; The rate of bio-degradation of DOC was increased proportionally with the increase of ozone injection rate and contact time irrespective of landfill site age. The increase of bio-degradability by ozone addition was not satisfactory. It is hard to expect significant increase in bio-degradability by ozone treatment only. Thus, it is evaluated that ozone oxidation can not increase biodegradability significantly in concentrated wastewater composed of complex organic compound such as leachate.

The fractionation characteristics of organic matter were investigated in inflow and effluent of each other pollution sources and river. While the DOC/TOC ratio in the influent of public sewage treatment plant and livestock disposal facilities was above 0.58, the POC/TOC ratio of human livestock Night soil treatment plant and stormwater runoff was more than 0.7. The TOC removal efficiency of public sewage treatment plant and human livestock Night soil treatment plant were 88.5 % and 99.6 %, respectively. Although the concentration distribution of organic matter pollution most of total organic carbon (TOC) in effluent of pollution sources accounted for dissolved organic carbon (DOC) type (DOC/TOC ratio >0.89) and Refractory-DOC (RDOC)/TOC ratio was higher (>0.65). The fractionation characteristics of organic matter in river were similar with that of sewage treatment plant and TOC concentration showed the positive correlation with DOC ($r^2=0.93$) and RDOC ($r^2=0.89$) concentration. The decay rate of Labile DOC (LDOC) (avg. $0.128day^{-1}$) was higher than labile particulate organic carbon (LPOC) ($0.082day^{-1}$), while that of DOC ($0.008day^{-1}$) was lower than POC ($0.039day^{-1}$) (paired t-test, p < 0.001, n = 5). These study results suggested that it should consider important both TOC and DOC as the target indicator to control refractory organic matter in pollution sources.
Expression of human Cu.Zn-superoxide dismutase (SOD) with activity comparable to human erythrocyte enzyme was achieved in E. coli B21(DE3) by using the pET-17b expression vector containing a T7 promoter. Recombinant human SOD was found in the cytosol of disrupted bacterial cells and represented > 25% of the total bacterial proteins. The protein produced by the E. coli cells was purified using a combination of ammonium sulfate precipitation, Sephacryl S-100 gel filtration and DEAE-Sephacel ion exchange chromatography. The recombinant Cu,Zn-SOD and human erythrocyte enzyme were compared using dismutation activity, SDS-PAGE and immunoblotting analysis. The mass of the subunits was determined to be 15,809 by using a electrospray mass spectrometer. The copper specific chelator. diethyldithiocarbamate (DOC) reacted with the recombinant Cu,Zn-SOD. At $50{\mu}M$ and $100{\mu}M$ concentrations of DOC, the dismutation activity was not inhibited for one hour but gradually reduced after one hour. This result suggests that the reaction of DOC with the enzyme occurred in two distinct phases (phase I and phase II). During phase I of this reaction, one DOC reacted with the copper center, with retention of the dismutation activity while the second DOC displaced the copper, with a loss of activity in phase II.
This study was to investigate the effects of carbon loadings, temperature and expansion ratio on the waterborne organic removal by the biologically active GAC fluidized bed on a laboratory scale. The raw water to be treated comes from midstream of Han river. BACFB(Biological Activated Carbon Fluidized Bed) process was very effective to remove the biodegradable fraction of dissolved organic matter. The more carbon weighed, the more DOC removed in a range from 16.7 to 133.3 g/l. DOC and UV$_{254}$ were removed more than 40% and 20% above 20$\circ$C respectively. Between 5$\circ$C and 10$\circ$C, DOC and UV$_{254}$ were eliminated about 30% and 15% respectively. In general, even if the temperature was higher, DOC removal was a little sensitive, probably influenced by GAC's residual adsorption capacity. UV$_{254}$ reduction was little fluctuated in accordance with water temperature. The gradual increase in expansion ratio from 10% to 75% didn't greatly affect on the removal of DOC and UV$_{254}$. The expansion ratio, therefore, is not a key factor over the critical expansion ratio.
In this study, the relationship between organic carbon distribution characteristics and decomposition rate classified according to the particle size and biological degradation characteristics in water was investigated for the Nam river and Geumho river. The average concentrations of TOC in the Nam river and Geumho river were 2.7±1.2 mg/L and 5.0±1.2 mg/L, respectively, but the composition ratios for each type of organic carbon were similar. An average value of 80.9% of TOC was present as DOC and 72.8% of DOC consisted of Refractory-DOC (RDOC). In addition, the change in the RDOC composition ratio according to temporal and spatial distribution was the smallest. There was no difference in the decomposition rate of organic carbon except for TOC by the site (p≥0.108, one-way ANOVA), and the decomposition rates of Labile-POC (LPOC) and LDOC were similar at 0.139±0.102 and 0.137±0.149 day-1, respectively (p=0.110, paired t-test). The coefficient of variation (CV) of the decomposition rate of DOC (average 8.1%), which had the smallest composition ratio of organic carbon, was 1.1, showing the largest temporal variation. The TOC, POC, and DOC decomposition rates showed a significant correlation with the ratio of the initial concentration to the concentration after 25 days of decomposition (OC25/OC0) (r2=0.89~0.94, p<0.001), and the decomposition rates of LPOC and LDOC were significantly correlated with the ratio of the initial concentration to the concentration after 5 days of decomposition (LOC5/LOC0) (r2=0.67~0.75). This suggests that it is possible to estimate the decomposition rate through the concentration of each type of organic carbon.

The number of vehicles employing diesel engines is rapidly rising. Accompanying this trend, application of an after-treatment system is strictly required as a result of reinforced exhaust regulations. The Diesel Particulate Filter (DPF) system is considered as the most efficient method to reduce particulate matter (PM), but the improvement of a regeneration performance at any engine operation point presents a considerable challenge by itself. Therefore, the present study evaluates the effect of fuel injection characteristics on regeneration performance in a DOC and a catalyzed CR-DPF system. The temperature distribution on the rear surface of the DOC and the exhaust gas emission were analyzed in accordance with fuel injection strategies and engine operating conditions. A temperature increase more than BPT of DPF system was obtained with a small amount fuel injection although the exhaust gas temperature was low and flow rate was high. This increase of temperature at the DPF inlet cause PM to oxidize completely by oxygen. In the case of multi-step injection, the abrupt temperature changes of DOC inlet didn't occur and THC slip also could not be observed. However, in the case of pulse type injection, the abrupt injection of much fuel results in the decrease of DOC inlet temperatures and the instantaneous slip of THC was observed.
