Antiobesity Activity of Chrysanthemum zawadskii Methanol Extract (구절초 추출물의 항비만 활성)

  • Chrysanthemum zawadskii, a herbaceous perennial plant belonging to the Compositae, grows wild in Asian countries, including Japan, China, and Korea. The biological, antioxidative, anti-inflammatory, and antibacterial activities of C. zawadskii have been reported, its antiobesity activity has not been elucidated. In the present study, the effect of C. zawadskii methanol extract (CZME) on pancreatic lipase enzyme activity, adipocyte differentiation, and adipogenesis was investigated using an in vitro assay and a cell model system. CZME effectively suppressed lipase enzyme activity in a dose-dependent manner. CZME also inhibited insulin, dexamethasone, 3-isobutyl-1-methylxanthine (MDI)-induced adipocyte differentiation, lipid accumulation, and the level of triglyceride in 3T3-L1 preadipocytes in a dose-dependent manner, without cytotoxicity. The antiobesity effect of CZME might be modulated by gene and protein expression of cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding proteins (C/EBP) α, C/EBPβ, and the peroxisome proliferator-activated receptor γ (PPAR γ). CZME also triggered lipolysis in a dose-dependent manner in MDI-induced 3T3-L1 preadipocytes. Taken together, these results provide important new insights into the antiobesity activities of C. zawadskii, showing that they involve pancreatic lipase inhibition, as well as antiadipogenic and lipolysis effects. CZME might be a promising source in the field of nutraceuticals. However, the active compounds that confer the antiobesity activities of CZME need to be identified.

Effects of Gynostemma pentaphyllum Callus Extract on Anti-inflammation, Anti-allergy, and Alleviation of Keratinocyte Damage Caused by S. aureus (돌외 캘러스 추출물의 항염, 항알러지 및 S. aureus에 의한 각질세포 손상 완화 효능)

  • This study was conducted to identify the efficacy related to skin barrier functions such as anti-inflammatory, anti-allergic, cornified envelope formation and the alleviation effect of keratinocyte damage caused by S. aureus using the callus extract, which was induced and extracted from the leaves of Gynostemma pentaphyllum in Ulleungdo Island. In order to confirm the anti-inflammatory effect of the G. pentaphyllum callus extract on the skin, the expression of inflammatory cytokines was investigated in primary epidermal keratinocytes (HEKa) activated with PAR-2 agonist, and the G. pentaphyllum callus extract showed IL-8, IL-25, and TSLP production inhibitory effect. β-Hexosaminidase assay using RBL-2H3 cells was performed to confirm the anti-allergic efficacy, and G. pentaphyllum callus extract showed the effect of inhibiting the release of β-hexosaminidase. In addition, G. pentaphyllum callus extract showed the cornified envelope formation effect in HaCaT cells, and through the co-culture experiment with HaCaT cells and S. aureus, it showed alleviation effect of keratinocyte damage caused by S. aureus. Therefore, G. pentaphyllum callus extract is considered to be a useful cosmetic material for improving skin barrier with anti-inflammatory, anti-allergic and alleviation effect of keratinocyte damage caused by S. aureus.

Comparative Study on Immunological Markers Between Human Immunodeficiency Virus(HIV)-Infected and Normal Persons in Korea (국내 Human Immunodeficiency Virus(HIV) 감염자와 정상인의 면역학적 표지인자 비교연구)

  • Several studies showed that the immunological factors such as CD4+ cell number, CD4%, CD8+ cell number and CD4/CD8 ratio and the serological factors such as, ${\beta}^2$-microglobulin(${\beta}^2$-MG), neopterin, soluble CD4, and soluble CD8 are related to the risk of development of AIDS. Especially, the CD4+ cell counts have been used to monitor progresson of HIV disease, to stratify, and to follow patients in clinical trials. Recently, the Centers for Disease Control and Prevention(CDCP) in USA has made the CD4+ cell count as a part of the classification of HIV disease. It is composed of 3 categories such as 1, 2, and 3 which asr $\geq$ 500/$mm^3$, 200/$mm^{3} $\geq$ and < 500/$mm^3$, and < 200/$mm^3$, respectively. In this study, to estimate the differences of immunological factors between HIV-infected and normal human groups in Korea, CD4+ T and CD8+ T cells, and the CD4/CD8 ratio were measured in 185 HIV-infected subjects and 140 healthy adult subjects. The lymphocyte subsets such as CD4+ T and CD8+ T were analysed by flow cytometer(FACStar) with two-color immunofluorescent stain using monoclonal antibodies such as anti-CD4 and anti-CD8 antibodies. The absolute numbers and percentages of CD4+ T and CD8+ T and the CD4/CD8 ratio of HIV infected persons were $462\pm{277}/mm^3$, $18.2\pm7.7%$, $1,170\pm{534}/mm^3$, $47.0\pm10.6%$ and $0,43\pm0.26 whereas those of uninfected persons were $886\pm{299}mm^3$, $32.9\pm{7.0%}, 730{\pm}259/mm^3$, $26.8\pm6.4%$ and $1.31\pm0.46$(P<0.01). In addition, estimating the reference values of peripheral blood lymphocyte subsets of Korean, the absolute numbers and percentages of CD4+ T and CD8+ T and the CD4/CD8 ratio of 140 healthy adults persons were measured and compared with those of foreigners. The reference ranges of CD4+ T cells, CD8+ T cells, CD4%, CD8%, and the CD4/CD8 ratio and 1.31$\pm$0.46, respectively. The significant differences were not observed when compared with those of foreigners. However a little difference was observed in the percentages of CD4+ T and the absolute numbers of CD8+ T between the normal values of Korean and those of foreigners were $43.6\pm8.9%$, $560\pm{230}/mm^3$. This result can also be useful as a basic data for the treatment and surveillance of HIV-infected patients in Korea.

SONICATED EXTRACT OF TREPONEMA DENTICOLA IMPAIRS THE LYMPHOCYTE PROLIFERATION (Treponema denticola sonicated extract에 의한 임파구 세포주기 차단 기전에 관한 연구)

  • 근관내 spirochetes의 존재유무가 명확하게 밝혀져 있지 않았으나 최근 PCR을 사용한 연구에서 Treponema denticola균주가 감염근관의 50% 이상의 경우에서 발견됨에 따라 이 세균이 치수 및 치근단 질환에 관여하는지에 대한 관심 이 높아졌다. 하지만 그 정확한 기전은 아직 밝혀져 있지 않다. 이와 관련하여 Shenker등이 T. denticola의 sonicated extract에서 순수분리된 단백질 (SIP)이 임파구 proliferation을 방해함을 보고한바 있다. 따라서 본 연구의 목적은 면역억제단백질 SIP이 어떤 기전에 의해서 임파구증식을 억제하는지를 밝히는 데 있다. 건강한 혈액 공여자로부터 추출해낸 T세포에 PHA (phytohemagglutinin)로 증식자극을 주게되는데 이 과정에서 SIP을 처리하거나 처리하지 않은 경우를 비교하여 세포주기 진행과정을 유세포분석기 (Becton-Dickinson FACS$^{tarplus}$) 를 통하여 평가하였다. 실험결과 세단계의 chromatography과정을 통해 순수정제된 SIP은 50kDa와 56kDa의 두가지 polypeptide로 구성되어 있고 0.25$\mu\textrm{g}$으로 처리된 T 임파구는 42.5%의 [$^3$H]thymidine incorporation 억제가 그리고, 0.5$\mu\textrm{g}$으로 처리한 경우는 75.1%의 억제가 일어나 dose-dependent한 양상이 나타났다. Propidium iodide와 유세포 분석기를 사용하여 세포주기를 분석한 결과 medium으로만 처리한 경우 97%이상의 임파구는 G$_0$/G$_1$ phase에 머물러 있었으나 PHA자극을 받은 경우 G$_0$/G$_1$ phase에서 58%, S phase에서 34.6%, G$_2$/M phase에서 7.4%로 분포되어 나타났다. SIP으로 전처리한 경우 세포 증식이 감소하여 0.25$\mu\textrm{g}$을 첨가한 경우 75.1%가 G$_0$/G$_1$ phase에 머물러 있었고 더 강한 농도의 0.5$\mu\textrm{g}$을 첨가한 경우는 87.7%가 G$_0$/G$_1$ phase에서 S phase로 진행되지 않고 머물러있었다. 따라서 SIP으로 전처리된 T 임파구는 그 증식이 G$_0$/G$_1$ phase에서 차단된 것으로 보인다. 이러한 면역억제현상이 in vitro 상태뿐 아니라 in vivo에서도 진행된다면 spirochete가 치수 및 치근단 질환의 병인론에 연관된 면역반응저하기전에 중요한 역할을 하는 것으로 추론할 수 있다.

Study on Keratinocyte Differentiation and Skin Barrier Function of Adeonphorae Radix Root Extracts (Adenophorae Radix 뿌리 추출물에 의한 Keratinocyte의 분화 및 피부장벽 기능에 대한 연구)

  • We have studied on the keratinocytes differentiation and skin barrier function using Adenophorae radix (A. radix) root extract, which was known to contain triterpenoid, saponin and starch. A. radix root extracts showed the $PPAR{\alpha}$ expression level of Wy-14,643 $0.5-1.0{\mu}M$ in CV-1 cells. The cornified envelop formation (CE) of human keratinocyte cell line (HaCaT) and normal human keratinocyte (NHK) showed a statistically significant increased compared to the control. When HaCaT cells were treated with A. radix root extract, transglutaminase (TGase-1) was significantly increased. As a result of clinical study of the simple cosmetic formulation containing A. radix root extract for about 2 weeks, TEWL values were significantly decreased and water contents were increased. The ceramides, which were obtained from the inner forearm, were also significantly increased statistically. We suggest that the A. radix root extract can be used as a preventive and therapeutic agent for skin diseases such as dry skin and atopy.

Development of an Immobilized Adsorbent for in situ Removal of Ammonium Ion from Mammalian Cell Culture Media and its Application to a Mammalian Cell Bioreactor: I. Development of Immobilized Adsorbent System (동물세포 배양액으로부터 암모늄 이온의 동시제거를 위한 고정화 흡착제의 개발과 동물세포 배양 시스템에의 응용: I. 고정화 흡착시스템 개발)

  • Three types of adsorbents were developed by immobilizing synthetic zeolite, Philipsite-Gismonine, in alginate, cellulose acetate and dialysis membrane for the in situ removal of ammonium ion which inhibits growth and productivity of animal cells such as CHO cells producing tPA. Ammonium ion removal efficiency and cell growth promoting effect with various immobilized adsorbents were evaluated and the membrane type was selected as an optimal immobilized adsorbent. The experiments were then simulated by adding 8mM ammonium chloride and immobilized adsorbent in order to validate the removal effect under high density cell cultures. The results showed increase in maximum cell density by three times, in cell viability, and in tPA productivity by 40%. And it was found that the promoting effects were more significant in case of high ammonium ion concentration system. It was also found that the optimum addition time for immobilized adsorbents was 48 hr in the absence of ammonium chloride addition and 72 hr in the presence of ammonium chloride addition.

  • In this study, we investigated in vitro immunostimulatory activity of fruit extracts from Pyrus ussuriensis var. hakunensis (Nakai) T.B. Lee (PUF) using mouse macrophage RAW264.7 cells. PUF increased the production of immunostimulatory factors such as NO, iNOS, IL-1β, IL-6 and TNF-α, and phagocytic activity in RAW264.7 cells. The inhibition of TLR2 and TLR4 blocked PUF-mediated production of immunostimulatory factors in RAW264.7 cells. In addition, the inhibition of MAPKs signaling pathway reduced PUF-mediated production of immunostimulatory factors. From these results, PUF may have immunostimulatory activity via TLR2/4-mediated activation of MAPKs signaling pathway. Therefore, PUF expected to be used as a potential immune-enhancing agent.

The Production of Tissue Type Plasminogen Activator from Normal Human Cell tine (정상 인체 세포로부터 조직 플라스미노겐 활성인자의 대량생산)

  • A method to produce tissue type Plasminogen Activator (tPA) from normal human fibroblast is developed by cultivating cells in serum free media containing heparin as an inducer. Optimal dose of this inducer was 30$\mu$g/m$\ell$. The composition of serum free medium was also defined to fit to the industrial scale cultivation. 1.42 ug of tPA per 10$^5$ viable cells per ml was produced. 1.1 gram of tPA can be produced every day from this cell line under normal perfusion chemostat operations assuming that same productivity is maintained when the process is sealed up. This method could reduce pro-duction costs and simplify purification processes by using serum free medium. Tissue type PA produced from this cell line has high ability of dissolving clots, based upon fibrin lysis test showing 50mm$^2$ of clearing zones in agarose gel plate. These results were reproducible and in good agreement with results of ELISA assay. tPA from normal human cells will be safer than that from melanoma and recombinant cells in human clinical trials.

Anti-adipogenic Activity of Cortex ulmi pumilae Extract in 3T3-L1 Preadipocytes (유근피 추출물의 3T3-L1지방전구세포의 분화 억제 효능에 관한 연구)

  • Cortex ulmi pumilae, the cortex of Ulmus davidiana var. japonica, has been used in traditional folk medicine for its anti-inflammatory effect. Although its various bioactivities such as anti-inflammatory, anti-microbial, and anti-cancer, have been reported, the anti-adipogenic activity of cortex ulmi pumilae remains unclarified. In the present study, we investigated the effect of cortex ulmi pumilae extract on adipocyte differentiation in 3T3-L1 preadipocytes. Treatment with cortex ulmi pumilae extract significantly reduced the formation of lipid droplets and triglyceride content in a dose-dependent manner; this is associated with an inhibition of the adipogenic transcription factors, CCAAT/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$), $C/EBP{\beta}$, and peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$). In addition, cortex ulmi pumilae extract treatment during the early stage of adipogenesis showed more efficient anti-adipogenic activity than treatment during other stages of adipogenesis. Cortex ulmi pumilae extract also inhibited cell proliferation and induced G1 arrest of 3T3-L1 cells in the early stage of adipogenesis. This was associated with upregulated expression of Cdk inhibitor p21 and downregulated expression of cyclin E and phospho-Rb, indicating that cortex ulmi pumilae extract blocks mitotic clonal expansion by cell cycle regulation. Taken together, these results suggest that cortex ulmi pumilae extract possesses anti-adipogenic activity through the inhibition of adipocyte differentiation by blocking mitotic clonal expansion.

Effects if Benzo(a)pyrene on Natural Killer Cell Activity of Mice (Benzo(a)pyrene이 마우스 자연살해세포 활설에 미치는 영향)

  • Benzo(a)pyrene(B(a)P), an extensively studied polycyclic aromatic hydrocarbon(PAH), is a common contaminant produced through the burning of fossil fuels, particularly coal, and from the exhaust products of internal combustion engines. It produces a wide range of toxicities, including carcinogenicity in experimental animals. B(a)P has been shown to suppress systemic immunity in experimental animals, which may contribute to the growth of the chemical-induced tumors. Using colorimetric MTT assay natural killer(NK) cell-mediated growth inhibition of tomor cell was measured in normal and B(a)P-exposed C57BL/6 mice. Non-adherent splenocytes of normal or B(a)P-exposed mice were cultured with Yac-1 cells at four different effector/target(E/T) cell ratios ranging from 200/1, 100/1, 50/1, and 25/1 in an assay volume of 0.1 ml. After the optical density of culture wells containing MTT solution was measured at a wavelength of 540 nm, the percentage of dead cells relative to the control target cell number was calculated. The NK activity of B(a)P-exposed mice was markedly lower than that of non-exposed mice group at all E/T ratios. These results indicated that suppression of NK cell activity may play a role in allowing for the growth of tumors.
