Identification of Bovine Lymphocyte Antigen DRB3.2 Alleles in Iranian Golpayegani Cattle by DNA Test

  • The bovine lymphocyte antigen (BoLA)-DRB3 gene encodes cell surface glycoproteins that initiate immune responses by presenting processed antigenic peptides to CD4 T helper cells. DRB3 is the most polymorphic bovine MHC class II gene which encodes the peptide-binding groove. Since different alleles favour the binding of different peptides, DRB3 has been extensively evaluated as a candidate marker for associations with various bovine diseases and immunological traits. For that reason, the genetic diversity of the bovine class II DRB3 locus was investigated by polymerase chain reaction-restriction fragment length polymorphism method (PCR-RFLP). This study describes genetic variability in the BoLA-DRB3 in Iranian Golpayegani Cattle. Iranian Golpayegani Cows (n = 50) were genotyped for bovine lymphocyte antigen (BoLA)-DRB3.2 allele by polymerase chain reaction and restriction fragment length polymorphism method. Bovine DNA was isolated from aliquots of whole blood. A two-step polymerase chain reaction followed by digestion with restriction endonucleases RsaI, HaeIII and BstYI was conducted on the DNA from Iranian Golpayegani Cattle. In the Iranian Golpayegani herd studied, we identified 19 alleles.DRB3.2${\times}$16 had the highest allelic frequency (14%), followed by DRB3.2${\times}$7 (11%). Six alleles (DRB3.2${\times}$25, ${\times}$24, ${\times}$22, ${\times}$20, ${\times}$15, ${\times}$3) had frequencies = 2%. Although additional studies are required to confirm the present findings, our results indicate that exon 2 of the BoLA-DRB3 gene is highly polymorphic in Iranian Golpayegani Cattle.

  • Listeriolysin O (LLO), one of the most immunogenic proteins of Listeria monocytogenes and its main virulence factor, mediates bacterial escape from the phagosome of the infected cell. Thus, its expression in a nonpathogenic bacterial host may enable effective delivery of heterologous antigens to the host cell cytosol and lead to their processing predominantly through the cytosolic MHC class I presentation pathway. The aim of this project was to characterize the delivery of a model antigen, chicken egg ovalbumin (OVA), to the cytosol of dendritic cells by recombinant Bacillus subtilis vegetative cells expressing LLO. Our work indicated that LLO produced by non-sporulating vegetative bacteria was able to support OVA epitope presentation by MHC I molecules on the surface of antigen presenting cells and consequently influence OVA-specific cytotoxic T cell activation. Additionally, it was proven that the genetic context of the epitope sequence is of great importance, as only the native full-sequence OVA fused to the N-terminal fragment of LLO was sufficient for effective epitope delivery and activation of $CD8^+$ lymphocytes. These results demonstrate the necessity for further verification of the fusion antigen potency of enhancing the MHC I presentation, and they prove that LLO-producing B. subtilis may represent a novel and attractive candidate for a vaccine vector.

  • Background: Bone marrow mesenchymal stem cells (MSC) inhibit the immune response of lymphocytes to specific antigens and dendritic cells (DC) are professional antigenpresenting cells whose function is to present antigen to naive T-lymphocytes with high efficiency and play a central role in the regulation of immune response. We studied the effects of MSC on DC to evaluate the relationship between MSC and DC in transplantation immunology. Methods: MSC were expanded from the bone marrow and DC were cultured from peripheral blood mononuclear cells (PBMNC) of 6 myelogenous leukemia after achieving complete response. Responder cells isolated from PBMNC and lysates of autologous leukemic cells are used as tumor antigen. The effect of MSC on the DC was analyzed by immunophenotype properties of DC and by proliferative capacity and the amount of cytokine production with activated PBMNC against the allogeneic lymphocytes. Also, cytotoxicity tests against leukemic cells studied to evaluate the immunologic effect of MSC on the DC. Results: MSC inhibit the CD83 and HLA-class II molecules of antigen-loaded DC. The proliferative capacity and the amount of INF-$\gamma$ production of lymphocytes to allogeneic lymphocytes were decreased in DC co-cultured with MSC. Also the cytotoxic activity of lymphocytes against leukemic cells was decreased in DC co-cultured with MSC. Conclusion: MSC inhibit the activation and immune response of DC induced by allogeneic or tumor antigen.

Characterization of CTL Clones Specific for Single Antigen, H60 Minor Histocompatibility Antigen

  • Background: Disparities of Minor H antigens can induce graft rejection after MHC-matched transplantation. H60 has been characterized as a dominant antigen expressed on hematopoietic cells and considered to be an ideal model antigen for study on graft-versus-leukemia effect. Methods: Splenocytes from C57BL/6 mice immunized with H60 congenic splenocytes were used for establishment of H60-specific CTL clones. Then the clones were characterized for proliferation capacity and cytotoxicity after stimulation with H60. Clone #14, #15, and #23 were tested for the TCR binding avidity to H60-peptide/$H-2K^b$ and analyzed for TCR sequences. Results: H60-specific CTL clones showed different levels of proliferation capacity and cytotoxic activity to H60-stimulation. Clones #14, #15, and #23 showed high proliferation activity, high cytotoxicity, and low activities on both aspects, respectively, and have TCRs with different binding avidities to H60-peptide/$H-2K^b$ with $t_{1/2}$ values of 4.87, 6.92, and 13.03 minutes, respectively. The TCR usages were $V{\alpha}12D-3-01+J{\alpha}11-01$ and $V{\beta}12-1-01+D{\beta}1-01+J2-7-01$ for clone #14, $V{\alpha}13D-1-02+J{\alpha}34-02$ and $V{\beta}13-1-02+D{\beta}2-01+J{\beta}2-7-01$ for clone #15, and $V{\alpha}16D+J{\alpha}45-01$ and $V{\beta}12-1-01+D{\beta}1-01+J{\beta}2-5-01$ for clone #23. Conclusion: The results will be useful for modeling GVL and generation TCR transgenic mouse.

Evaluation of schistosomula lung antigen preparation and soluble egg antigen vaccines on experimental schistosomiasis mansoni

  • Schistosomiasis causes significant morbidity and mortality worldwide. This study aimed to assess the effect of schistosomula lung antigen preparation (SLAP) and soluble egg antigen (SEA) on a murine schistosomiasis mansoni model. Ninety laboratory-bred male Swiss albino mice were divided into 6 groups. Two doses of the vaccine were given at 2-week intervals. All mice were subcutaneously infected with 80±10 Schistosoma mansoni cercariae 2 weeks after the last vaccination dose. They were sacrificed 7 weeks post-infection. Parasitological and histopathological studies were conducted to assess the effect of inoculated antigens (single or combined). The results showed that the combination of SLAP and SEA (combination group) led to a significant reduction in worm burden (65.56%), and liver and intestine egg count (59% and 60.59%, respectively). The oogram pattern revealed a reduction in immature and mature eggs (15±0.4 and 10±0.8, respectively) and an increased number of dead eggs in the combination group (P<0.001). In terms of histopathological changes, the combination group showed notably small compact fibrocellular egg granuloma and moderate fibrosis in the liver. A high percentage of destroyed ova was observed in the intestine of the combination group. This study demonstrates for the first time the prophylactic effect of combined SLAP and SEA vaccine. The vaccine induced a significant reduction in the parasitological and pathological impacts of schistosomiasis mansoni in hepatic and intestinal tissues, making it a promising vaccine candidate for controlling schistosomiasis.

SV40의 T항원으로 불사화한 지방줄기세포주로부터 생산한 배양액의 항염증 효능 (Anti-inflammatory Effect of Conditioned Medium From an Immortalized Adipose-derived Stem Cell Line by SV40 T Antigen)

  • 지방줄기세포는 다양한 세포로 분화할 수 있어서 세포치료제로 주목받고 있으나 세포 증식 속도가 느리고 세포의 수명이 짧아 임상 적용에 어려움이 있다. 본 연구팀은 선행연구에서 지방줄기세포의 증식속도를 향상시키고 세포 수명을 연장시키기 위해 SV40의 T항원 유전자를 사람의 ADSC에 도입하여 불사화시킨 ADSC 세포주를 제작하였으며 ADSC-T라고 명명하였다. 본 연구에서는 ADSC-T 세포의 분화능력을 검증하고, ADSC-T 세포를 배양하여 획득한 무혈청 배양액의 항염증 효능을 평가하였다. 그 결과, ADSC-T 세포주는 지방세포와 골세포로 분화할 수 있는 능력을 보유하고 있어 줄기세포의 가장 중요한 특징인 분화능력을 유지하고 있음을 알 수 있었다. 또한 ADSC-T의 배양액은 염증을 유발하는 NF-κB의 활성을 억제하였고, NF-κB의 표적 유전자인 COX-2와 iNOS의 발현도 억제하였다. 또한 ADSC-T 배양액은 ERK, JNK, p38의 인산화를 저해하여 세포 내 염증 유발 신호 전달 회로인 MAPKs 회로를 억제하는 것으로 나타났으며, 염증을 유발하는 cytokine인 TGF-β, TNF-α, IL-6, IL-13의 발현도 억제하는 효과를 보여 강한 항염증 효능이 있을 것으로 추정되었다. 실제로, 아토피 모델 쥐인 Nc/Nga 마우스를 DNCB로 처리하여 아토피 피부염을 유발한 뒤 ADSC-T 배양액을 도포한 결과 강한 치료 효과를 나타내었다. 이상의 결과로부터, 불사화된 ADSC-T 세포주는 지방줄기세포의 장점과 효능을 유지하고 있으며 향후 지방줄기세포의 세포 수급 문제를 해결할 수 있을 뿐 아니라 임상 적용이 가능한 배양액의 생산에도 널리 이용될 수 있을 것으로 사료된다.

Role of IL-23 and Th17 Cells in Airway Inflammation in Asthma

  • Asthma is characterized by chronic airway inflammation with intense eosinophil and lymphocyte infiltration, mucus hyperproduction, and airway hyperresponsiveness. Accumulating evidence indicates that antigen-specific Th2 cells and their cytokines such as IL-4, IL-5, and IL-13 orchestrate these pathognomonic features of asthma. In addition, we and others have recently shown that IL-17-producing $CD4^+$ T cells (Th17 cells) and IL-23, an IL-12-related cytokine that is essential for survival and functional maturation of Th17 cells, are involved in antigen-induced airway inflammation. In this review, our current understanding of the roles of IL-23 and Th17 cells in the pathogenesis of allergic airway inflammation will be summarized.

  • The effects of thyroxine (TX) or propylthiouracil (PPT) administration on the antibody forming activity agains t sheep red blood cell (SRBC) and Newcastle disease virus (NDV) were studied by using of hemagglutination and hemagglutination-inhibition techniques. Antibody titers to both SRBC and NDV increased significantly in the TX-treated group, whereas decreased in the PPT-treated group, compared with control. When TX was administered after antigen inoculatioon, antibody forming activity was significantly enhanced, compared with the TX administration before antigen inoculation.

Effect of thymus extract on the activation of cytotoxic and accessory functions of tumor-associated macrophages

  • The present investigation was under taken to study whether the tumor-associated macrophages (TAM) of Daltons lymphoma (DL), a spontaneous transplantable murine T cell lymphoma can be activated to tumoricidal state by crude thymus extract. Intraperitoneal administration of thymus extract to DL-bearing mice resulted in activation of TAM with an enhanced IL-1, TNF and antigen presenting ability. It was found that treatment with thymus extract could also enhance the phagocytic and cytotoxic activity of TAM. However, only a marginal increase in arginase activity was observed. Till date to the best of our knowledge the effect of crude thymus extract on the activation of tumor associated macrophages has not been investigated, this study provides a new piece of information in the area of thymus therapy.

The role of dendritic cells in tumor microenvironments and their uses as therapeutic targets

  • Dendritic cells (DC), which consist of several different subsets, specialize in antigen presentation and are critical for mediating the innate and adaptive immune responses. DC subsets can be classified into conventional, plasmacytoid, and monocyte-derived DC in the tumor microenvironment, and each subset plays a different role. Because of the role of intratumoral DCs in initiating antitumor immune responses with tumor-derived antigen presentation to T cells, DCs have been targeted in the treatment of cancer. By regulating the functionality of DCs, several DC-based immunotherapies have been developed, including administration of tumor-derived antigens and DC vaccines. In addition, DCs participate in the mechanisms of classical cancer therapies, such as radiation therapy and chemotherapy. Thus, regulating DCs is also important in improving current cancer therapies. Here, we will discuss the role of each DC subset in antitumor immune responses, and the current status of DC-related cancer therapies.
