  • In the study, we Investigated the behavior and removal efficiency of organics, nitrogen. phosphorus with operating conditions in SBRs. Substrate used was synthetic wastewater in which the ratio of $COD_{cr}$. : N : P was 100 : 12 : 2. The cycling the in SBRs was adjusted at 6 hours and 8 hours, and then certainly Included anaerobic and aerobic conditions. Also, for each cycling time. we performed 2 series of experiment simultaneously which was set up 10 days and 20 days as SRT. The removal efficiency of $COD_{cr}$. was over 97% in all operating conditions. In the 6 hours cycling time, the removal efficiency of $PO_4^{3-}-P$ reached almost 100% in steady state. And then we could observe a typical phonemena of phosphorus release and uptake, and the removal efficiency of N was 67%, Residual N source was almost TKN and most of the rest remained as $NO_2-N$. Also the difference in both SRTs was not observed practically. In the 8 hours cycling time, dissolution of sludge appeared. and, $PO_4^{3-}-P was not nearly removed but nitrogen was removed up to 75%, And the residual nitrogen was accumulated as $NO_2^--N$.

  • Partial nitrification blocking of the oxidation of nitrite ($NO_{2}^{-}$) to nitrate ($NO_{3}^{-}$) has cost-efficient advantages such as lower oxygen and organics demand for nitrification and denitrification, respectively. A nitrifying membrane bioreactor of submerged type was operated for the treatment of synthetic ammonium wastewater with the purpose of nitrite build-up without affecting the efficiency of ammonium oxidation. A high ammonium concentration (1,000 mg/l) was completely converted to nitrate at up to 2 kg $N/m^3$ day under sufficient aeration. The control of pH under sufficient aeration was not a reliable strategy to maintain stable nitrite build-up. When the dissolved oxygen concentration was kept at 0.2-0.4 mg/l by adjusting the aeration rate, about 70% of nitrite content was obtained with ammonium oxidation efficiency higher than 93%. The increase of suction pressure due to membrane fouling was not significant under lowered aerating environment over a 6-month period of operation. The composition of nitrifier community, including relative abundance of nitrite oxidizers in a nitrite-accumulating condition, was quantified by fluorescence in situ hybridization analysis.

Enrichment of Electrochemically Active Bacteria Using a Three-Electrode Electrochemical Cell

  • Electrochemically active bacteria were successfully enriched in an electrochemical cell using a positively poised working electrode. The positively poised working electrode (+0.7 V vs. Ag/AgCl) was used as an electron acceptor for enrichment and growth of electrochemically active bacteria. When activated sludge and synthetic wastewater were fed to the electrochemical cell, a gradual increase in amperometric current was observed. After a period of time in which the amperometric current was stabilized (generally 8 days), linear correlations between the amperometric signals from the electrochemical cell and added BOD (biochemical oxygen demand) concentrations were established. Cyclic voltammetry of the enriched electrode also showed prominent electrochemical activity. When the enriched electrodes were examined with electron microscopy and confocal scanning laser microscopy, a biofilm on the enriched electrode surface and bacterium-like particles were observed. These experimental results indicate that the electrochemical system in this study is a useful tool for the enrichment of an electrochemically active bacterial consortium and could be used as a novel microbial biosensor.

Performance of Fusarium oxysporum EKT01/02 isolate in cyanide biodegradation system

  • This study reports a cyanide resistant and/or tolerant fungus, isolated from the rhizosphere of Zea mays contaminated with cyanide-based pesticides. The isolate was characterised using molecular biology. The effect of free cyanide and heavy metals on the growth of isolate in a synthetic gold mine wastewater was examined. The molecular analyses identified the isolate as Fusarium oxysporum EKT01/02 (KU985430/KU985431). The isolate had a free cyanide degradation efficiency of 77.6%. The results indicated greater growth impairment in culture containing Arsenic (optical density 1.28 and 1.458) and cyanide (optical density 1.315 and 1.385). Higher growth was observed in all cultures supplemented with extracellular polymeric substance. This study showed that the isolate possesses wide substrate utilisation mechanism that could be deployed in environmental engineering applications.

Study of chemical coagulation conditions for a disperse red dye removal from aqueous solutions

  • Coagulation process using aluminum sulfate ($Al_2(SO_4)_3$) and ferric chloride ($FeCl_3$) was employed as a treatment method for decolorization of a synthetic textile wastewater containing red dye in this paper. Factors such as initial pH, coagulant dosage, initial concentration, conductivity and mixing conditions that influence color removal efficiency were experimentally tested. It was found that $Al_2(SO_4)_3$ is more efficient than $FeCl_3$ as coagulant. When $40mgL^{-1}$ aluminum sulfate was used, results showed that color induced by the red dye was efficiently removed (> 90 %) and was obtained in a large range of initial pH from 4 to 8 with, and for a dye concentration lower than $235mg\;L^{-1}$. After addition of the coagulant, the medium had to be mixed for 30 min at 60 rpm, then allowed to settle for 40 min. The effects of water conductivity in the range $0.035-2.42mS\;cm^{-1}$ and dye concentration up to $380mg\;L^{-1}$ were also followed and discussed.

Microbial and Physicochemical Monitoring of Granular Sludge During Start-up of Thermophilic UASB Reactor

  • Mesophilically-grown granular sludge seeded in thermophilic UASB reactor was monitored to better understand the start-up process of the reactor. The reactor was fed with a synthetic wastewater containing glucose. As COD loading rate increased stepwise, methane production rate increased. Maximum values of COD removal efficiency (95%) and methane production rate (5.3 l/day) were achieved by approximately day-80 and remained constant afterward. However, physicochemical and microbial properties of granules kept changing even after day-80. Specific methanogenic activity (SMA) was initially negligible, and increased continuously until day-153 and remained constant afterward, showing the maximum value of $1.51{\pm}0.13\;g\;CH_4-COD/g$ VSS/day. Deteriorated settling ability of granules recovered the initial value by day-98 and was maintained afterward, as determined by sludge volume index. Initially reduced granule size increased until day-126, reaching a plateau of 1.1 mm. Combined use of fluorescence in situ hybridization and confocal laser scanning microscopy (CLSM) allowed to localize families of Methanosaetaceae and Merhanosarcinaceae in granules with time Quantitative analyses of CLSM images of granule sections showed abundance patterns of the methanogens and numerical dominance of Methanosaeta spp. throughout the start-up period. The trend of SMA agreed well with abundance patterns of the methanogens.

Population Dynamics of Phage-Host System of Microlunatus phosphovorus Indigenous in Activated Sludge

  • Monitoring of the phage-host system of Microlunatus phosphovorus indigenous in activated sludge was attempted. A laboratory-scale activated sludge process was operated for 5 weeks with synthetic wastewater. The phage-host system population in the process was monitored by plaque assay and FISH methods at every 3 days. During the process operation, the phage-host system populations were more or less steady, except for 1 week in the middle of the operation. In that period, initially M. phosphovorus decreased significantly and its lytic bacteriophages increased, and then M. phosphovorus increased back to its original level while its lytic bacteriophages decreased. This observation suggests that lytic bacteriophages should be considered as one of the biological factors affecting the bacterial population dynamics in activated sludge processes.

  • Granular activated carbon(GAC) adsorption is one of the best available technology to remove synthetic organic chemicals(SOCs) from water supplies and wastewater. In order to satisfy enviromental criteria and reduce GAC treatment cost, optimal study of reverse stratified tapered adsorber(RSTA) has been conducted. The RSTA was found to provide on increase in breakthrough time when compared to a conventional cylindrical adsorber(CA). Through the RSTA optimal experiment, optimal mean bed velocity was decided 19.10cm/min and optimal angle was decided RSTA($3.0^{\circ}$). Adsorption efficiency was increased with increasing activated carbon doses and the number of activated carbon layers.

Removal of Manganese(II) from Aqueous Solution Using Manganese Coated Media (망간코팅 여재를 이용한 수용액상의 망간 제거연구)

  • This study investigated the applicability of manganese coated media such as manganese coated sand (MCS), manganese coated sericite (MCSe) and manganese coated starfish material calcined at $550^{\circ}C$ (MCSf) to remove Mn(II) in synthetic wastewater. Manganese coated media prepared at different pH was applied in the treatment of soluble Mn(II) in batch and column experiments at various Mn(II) concentrations. The amount of Mn coated on three different media was approximately 800~1100 mg/kg. From the stability test, negligible dissolution of Mn was observed above pH 3.0. In batch test, more than 40% of Mn(II) was removed by all sand media at various manganese concentrations. In order to see the effect of additional oxidant for the removal of Mn(II), 4 mg/L of hypochlorite was added in Mn(II) solution during column experiment. Breakthrough of Mn(II) was greatly retarded in the presence of hypochlorite in all column reactors packed with different media. Among the manganese coated media, MCSf prepared at pH 4 indicated the highest removal capacity. The removal efficiency of Mn(II) was also increased in the multi-layer system (0.5 g of MCS, MCSe, and MCSf each).

  • This study was carried out to investigate the variation of organic, nitrogen and phosphorus in $(AO)_2$ SBBR process according to the variation of operating cycle at the high TOC concentration. The operation time in anoxic (anaerobic) time to oxic time was 1:1. Three lab-scale SBBRS were fed with synthetic wastewater based on glucose as carbon source, The variation of total TOC removal was similar each other irrespective of operation time, however, the TOC concentrations in SBBRs showed a little difference according to the operating condition. In SBBR, complete nitrification was not occurred at all reactors, however, R3 showed a higher nitrification than R2. And in SBBR, the variation of operating time more affected at phosphorus removal than nitrogen removal. R2 which had the shortest time at the 1st aeration time showed the lowest phosphorus release and uptake efficacy.
