To determine the level of heterosis, higher cocoon shell weight multivoltine congenic lines (Con. L) and bivoltine syngenic lines (Syn. L) of silkworm were used for crosses. First filial generations $(F_1s)$ expressed heterobeltiotic genetic interaction at significant magnitude (p < 0.01) for single cocoon shell weight (SCSW). The other linked characters viz., single cocoon weight (SCW) and yield by weight per 10, 000 larvae were also significantly higher (p < 0.01) than the better parental lines. All the hybrids showed significant improvement for these aforesaid characters over standard heterosis (Standard check). The reeling parameters viz., filament length, raw silk, neatness, cohesionstrokes etc, also showed improvement among the hybrids than check in congenial environment. Overall results suggested that the cross between congenic and syngenic lines provide better heterosis with good quality silk than conventional hybrids and may be used for commercial exploitation.
Pedunculagin is an ellagitannin purified from Alnus hirsuta var. microphylla. Betulaceae. The effects of pedunculagin on immune system have been characterized to induce enhancement of NK (natural killer) cell cytotoxicities against tumor cells. Here, we report the evaluation of the effects of pedunculagin on the growth of murine Bl6-F10 melanoma in vivo. After the intradermal inoculation of Bl6-F10 melanoma, Bl6-F10 tumors grew progressively in immunocompetent syngenic C57BL/6 mice. The mice treated with pedunculagin(10 mg/kg, every 48 hrs) resulted in a significant improvement in survival. Inhibitory effects of pedunculagin on lung metastasis in C57BL/6 mice were also detected. Summarizing treatment with pedunculagin has a significant antitumor effect upon Bl6-F10 murine melanoma.
In the present study, we investigated the antitumor effects of Ginsenoside Rh2 and $\beta$-glucan using an experimental metastatic mouse model intravenously injected with B 16 melanoma F10 cells. Oral administration to various concentration of $\beta$-glucan (50mg/kg, l00mg/kg and 200mg/kg) dose-dependently reduced the lung-metastatic potential of metastatic BI6 melanoma F10 cells in syngenic mice. At same dose, Ginsenoside Rh2(50mg/kg) has more antitumor effect than $\beta$-glucan(50mg/kg). (omitted)

Dendritic cells (DCs) are known to professional antigen presenting cell (APC). Due to the role as an effective activator of cytotoxic T Lymphocytes by expressing MHC, adhesion and co-stimulatory molecules, DCs are now widely recognized to play an important role in the immune responses to tumors.We investigated the effect of cultured DCs in murine melanoma pulmonary metastasis model. To follow the metastasis protocol, syngenic melanoma cells were inoculated intra-venously into the mouse (B16F10 into the C57BL/6)8 days prior to the first DC injection (1$\times$106 DCs/ mouse, i.p.) and the autologous tumor cell lysate pulsed-DCs were injected as a therapeutic module twice in two weeks. Amylase isozyme based three multivoltine viz., N+p, Np, N+ $p^{cho}$ and two bivoltine-D6+p, D6p syngenic lines (Syn. L) were developed from germplasm (GP) stocks Nistari (N) and D6 respectively. haemolymph isozyme pattern at pH 7.0 and 8.5 depicted a total 11 number (Am $y_{1 to 6}$ at pH 7.0 and Am $y^{l to 5}$ at pH 8.5) of native proteins (NP) of various sizes are amylase isozyme expressers. Among eleven NPs, two NPs of 770 kDa (Am $y^{6}$ at pH 7.0) and 376 kDa (Am $y^3$ at pH 8.5) are $\alpha$-amylase expressers and remaining NPs of 370, 364, 350, 329 and 274 kDa at pH 7.0 and 206, 292, 416, 725 kDa at pH 8.5 are $\beta$-amylase expressers. Accordingly, digestive juice amylase isozyme pattern at aforesaid pH also depicted a total number of 10 NPs (Am $y^{1 to 5}$) at each pH 7.0 and 8.5 are amylase expressers of which NP of 387 kDa (Am $y^4$ at pH 7.0) and 780 kDa (Am $y^{5}$ at pH 8.5) are a-amylase expresser. Remaining NPs of 338,297 & 216 kDa at pH 7.0 and 370, 341, 329 &302 kDa at pH 8.5 are $\beta$-amylase expresser. Recurrent backcross lines (RBL) viz., N+pRBL and NpRBL were developed through introgression of high shell weight character (a multigenic trait) to be used further for congenic line (Con. L) development and to understand any association with introgressed character. Isozyme pattern in haemolymph of RBLs depicted only one $\alpha$-amylase of 770 kDa at pH 7.0 and 376 kDa at pH 8.0 with three and four respective $\beta$-amylase bands but in bivoltine lines numbers of $\beta$-amylase bands vary between 1 to 2 at aforesaid pH. Variability was also observed in digestive juice of multivolitine and its RBLs but bivoltine lines express null activity at both pH except appearance of one very week $\alpha$-amylase band D6+p at pH 8.5. Overall study suggests that not a single NP at both pH is common for expression of any band of amylase isozyme i.e., a totally different set of proteins are the amylase isozyme expresser at specific pH and no molecular factor of amylase is associated in developed RBLs which showed improvement on survival, single cocoon shell weight (SCSW) and single filament length over receptor parents.s.s.s.

Manifestation of heterotic genetic interaction was studied in different hybrids made between multivoltine recurrent backcross (RBL)/congenic lines (Con. L) during unfavourable season when temperature and relative humidity are > $30^{\circ}C$ and 86%, respectively. A few number of silkworm race or strain or breed like Nistari (N + p or Np) can sustain the temperature above 3$0^{\circ}C$ and RH above 86%. The present heterosis study screened a hybrid i.e., CB$_{5}$Lm5RBL1M$_{6}$DPC-LmE$^1$RBL and its reciprocal provided heterobeltiotic effect on survival by number and pupation rate at a magnitude of 20% (p < 0.01) and yield by weight of 10% (p < 0.01). Beside all the hybrids expressed heterosis over check - Nistari (N + p) with better quality silk. Therefore, aforesaid hybrid may be useful for utilization at commercial level during adverse seasons of West Bengal.gal.
Purpose : Selective introduction of genes conferring chemosensitivity into proliferating tumor cells may be used to treat cancer. We first investigated the bystander effect of retrovirus-mediated gene transfer of herpes simplex virus thymidine kinase(HSV-TK) gene to murine neuroblstoma cell line(neuro-2a) in vitro and in vivo. Second, we examined the mechanism and its enhancement of the bystander effect in murine neuroblastoma. Methods : To investigate the bystander effect, we studied tumor growth and survival time after HSV-TK/ganciclovir(GCV) treatment in a syngenic A/J mouse neuroblastoma model by mixing various ratios of HSV-TK-expressing neuro-2a cells with wild type neuro-2a cells followed by GCV treatment. To investigate the mechanism of the bystander effect in murine neuroblastoma, immunohistochemistry using connexin 43, CD4 and CD8-specific monoclonal antibodies was analyzed. We studied whether IL-2-secreting neuro-2a cells(neuro-2a/IL-2) would potentiate the bystander effect. Results : A strong bystander effect was observed in vitro and in vivo. The bystander effect in murine neuroblastoma was dependent on the immune response rather than connexin-mediated gap junction. Neuro-2a/IL-2 treatment enhanced the bystander effect in the HSV-TK/GCV system in murine neuroblastoma model. Conclusion : We conclude that the bystander effect in murine neuroblastoma depends on immune response and is enhanced by neuro-2a/IL-2.

We have investigated the antimetastatic and antitumor effects of Ginsenoside Rh2 and ${\beta}-glucan$ unsing an experimental metastatic mouse model intravenously injected with B 16 melanoma F 10 cells. Animal groups are divided into six groups according to the dosage of drug administration and the kind of drugs. The groups are control, ${\beta}-glucan$ with 50, 100 and 200 mg/kg, Geinsenoside Rh2 50 mg/kg, and ${\beta}-glucan$ 50 mg/kg + Ginsenoside Rh2 50 mg/kg. Oral administration of various concentration of ${\beta}-glucan$( 50, 100, and 200 mg/kg) were reduced the lung- metastatics induced by metastatic B16 melanoma F 10 cells injection with a dose dependent manner in the syngenic mice. At same dosage group, Ginsenoside Rh2 (50 mg/kg) has more antimetastatic effect than the ${\beta}-glucan$(50 mg/kg). The highest antimetastatic effects was observed in the ${\beta}-glucan$ 50 mg/kg + Ginsenoside Rh2 50 mg/kg group and has a similar tendency in the anti-tumor effects, including decrease of the average tumor weight and increase of the average survival rate. There are no differences of the average tumor weights were apparent in the ${\beta}-glucan$ groups, however there were little decrease of the average tumor weight in Ginsenoside 50 mg/kg group and ${\beta}-glucan$ 50 mg/kg + Ginsenoside Rh2 50 mg/kg group than that of the control group. The rate of average survival rate in the ${\beta}-glucan$ 50 mg/kg + Ginsenoside Rh2 50 mg/kg group, ${\beta}-glucan$ 200 mg/kg, ${\beta}-glucan$ 100 mg/kg and ${\beta}-glucan$ 50 mg/kg, and Ginsenoside 50 mg/kg groups were highly in order. These data suggest that antimetastatic and antitumor effect of combination of Ginsenodide Rh2 and ${\beta}-glucan$ be the highest in this study.
The present study was carried out to sex effectively mouse and rabbit embryos using rat H-Y antiserum and to improve the rate of rat producing H-Y antiserum with H-Y antibody. The H-Y antiserum was prepared from inbred SD strain female rat immunized by intrasplenic injection and subsequent intraperitioneal booster of testis cell of syngenic male newborn rat. the titer of antiserum was identified by in vitro cytotoxicity test of mouse embryos. The rabbit embryos exposed to the H-Y antiserum was classified to the developed (H-Y negative) or delayed (H-Y positive) group. The H-Y negative rabbit embryos were transferred to recipients and sex of offspring was examined. 1. When mouse embryos were exposed to the rat H-Y antisera, the ratio of embryos developed vs delayed was various. The H-Y antisera where the ratio of embryos developed vs delayed showed the range of 40~60% were recognized as having titer of H-Y antibody. 2. When the subsequent intraperitioneal boosters were followed after priming of intrasplenic injection of H-Y antigen, the rate of rat producting the H-Y antiserum with H-U antibody was 13, 27, 70 and 73% in control, 1st B, 2nd B and 3rd B, respectively. The rate in 2nd B and 3rd B was significantly(P<0.05) higher than that in control and 1st B. 3. When the rabbit morulae were exposed to the rat H-Y antiserum with H-Y antibody, the ratio of morulae developed versus delayed was 42:58% and it was close to the natural sex ratio 50:50%. It was confirmed that the rat H-Y antiserum was cross reactive to the rabbit morulae. 4. When the H-Y negative rabbit embryos were transferred to the recipients, the pregnancy rate was 50% and 83% of the newborns were females. In conclusion, the rat H-Y antiserum with high titer of H-Y antibody was able to be obtained from the female rat immunized by the intrasplenic injection followed by the second intrapent oneal booster of testis cells at a week interval. When the rabbit embryos negative to the rat H-Y antiserum were transferred, 83% of the newborns were females.
Purpose : Bronchopulmonary dysplasia (BPD) is characterized by arrested vascular and alveolar growth in the premature lung. Considering the consequences of arrested lung growth, the idea of administering bone marrow cells to enhance the inborn repair mechanism is promising as this may reduce the morbidity and mortality of BPD. We followed enhanced green fluorescent protein (EGFP)-labeled bone marrow cells (BMC) injected intraperitoneally into non-EGFP mice in order to determine their fate after transplantation. Methods : An angiogenesis inhibitor, SU1498, was injected subcutaneously on day 3 in non-EGFP C57BL/6 newborn mice to create a model of arrested alveolar development. On the following day, $1{\times}10^6$ BMCs isolated from major histocompatibility complex (MHC)- matched syngenic EGFP mice were injected intraperitoneally to non-EGFP BPD mice. Morphometric analysis, immunostaining, and confocal microscopy were performed to determine the fate of EGFP-positive stem cells in the injured lung. Results : SU1498 injection reduced alveolar surface area and mean alveolar volume in newborn mice. BMC injection resulted in recovery of lung structure comparable to controls. EGFP-positive BMCs were identified in the lungs of the recipient mice after intraperitoneal injection. The injected EGFP cells were co-stained with endothelial and epithelial cells of the developing lung as determined by confocal microscopy. Conclusion : Our results illustrated that EGFP-positive BMCs engrafted and trans-differentiated into epithelial and endothelial cells after intraperitoneal injection in a mouse model of arrested alveolar development.
