The antimicrobial activity of Hwangheuk-san and synergy effect with oxacillin against methicillin-resistant Staphylococcus aureus (황흑산(黃黑散)의 메티실린 내성 황색포도상구균에 대한 항균 활성과 옥사실린과의 시너지 효과)

  • Objectives : Hwangheuk-san is a complex prescription composed of oriental traditional medicine and has been reported for antioxidant, antimicrobial and anticancer effects in the recent study. Methicillin-resistant Staphylococcus aureus (MRSA) is one of important causes of fatal infectious diseases such as septicemia, endocarditis, toxic shock syndrome, pneumonia, skin and soft tissue infections (SSTIs). S. aureus is reported as being for a variety of human diseases and its epidemiological relevance is mainly due to their ability of becoming highly resistant to common antimicrobials such as tetracycline, penicillin, cphalosporin and aminoglycoside. The objective of this study is to determine the antimicrobial effect of Hwangheuk-san ethanol extracts (HHS) and synergistic effects with antibiotics oxacillin against MRSA.Methods : The antimicrobial activity of HHS was measured by the disc diffusion method, broth microdilution method and the checkerboard dilution test, time-kill curve assay was performed to investigate synergistic effects with antibiotics oxacillin against MRSA.Results : HHS showed antimicrobial activity against MRSA with a MIC value of 125 ㎍/㎖. In the checkerboard test, the interaction of HHS with antibiotics oxacillin produced almost synergy or partial synergy against MRSA. This study showed that HHS reduced the MICs of oxacillin tested, and a remarkable antibacterial effect of HHS, with membrane permeability enhancers.Conclusions : These results suggest that HHS has the antimicrobial effect and synergistic effects with antibiotics oxacillin against MRSA. This study thus can be a valuable source for the development of a new drug with low MRSA resistance.

Comparatie Study on the Characteristics and Development Stage of Micro Manufacturers Clusters: Focused on Apparel Sewing Clusters in Changsin and Jangwi, Seoul, Korea (소공인클러스터의 특성과 발달단계에 대한 비교연구: 서울시 창신 및 장위 의류봉제 클러스터를 중심으로)

  • The support policy of Micro manufacturers clusters should reflect the regional characteristics of Micro manufacturers clusters, but only similar policies are proposed based on the normal collection of information from related companies. Since the regional characteristics of Micro manufacturers clusters are not understood, they are not reflected in the support policy. Therefore, this study analyzed the characteristics of Micro manufacturers clusters as the formation factors of localization, networking/embeddedness, and innovation synergy/collective learning which are the characteristics of clusters, and applied the development stage based on the analyzed formation factors. Since regional characteristics may be different in the same industry, Micro manufacturers clusters in Changsin and Jangwi, which are representative clusters of apparel sewing industry in Seoul, were compared and studied in terms of regional characteristics and developmental stages. As a result of analyzing the characteristics of Apparel Sewing Micro manufacturers clusters, clusters of peers were found to be higher in Changsin than in Jangwi in localization. However, in the synergy and group learning, the Changsin area was mainly centered on designers and the Jangwi area was centered on Micro manufacturers, the Changsin area was active while the Jangwi area was passive in participating in fairs and seminars, and in sample and production. As a result of examining the two formation factors, the development stage of Micro manufacturers clusters in the two regions is progressing beyond the industrial district in the case of Micro manufacturers clusters in Changsin apparel sewing, and the Micro manufacturers clusters in the Jangwi apparel sewing are analyzed to stay in the industrial district. Innovative and long-established garment sewing clusters suggest that the government's support policy needs to reflect the characteristics and development stages of the two regions for sustainable growth.

Anti-aging Effects of Cedrol and Collagen-derived Peptide (세드롤과 콜라겐 유래 펩타이드의 피부노화 개선효과)

  • Skin aging is the phenomena finally expressed on the skin surface and related to the changes in the microstructure of the skin texture. Which is resulted in wrinkle formation and uneven tone of skin and so on. In this study, the synergy effect of Cedrol and a collagen-derived peptide in type III collagen synthesis was evaluated by in vitro test. The physiological skin state of 22 female volunteers was measured after using the cosmetics for 4 weeks. Results showed that Cedrol and a collagen-derived peptide had the excellent synergy effect in type III collagen synthesis. The cosmetics improved skin microrelief, star configurations, skin gloss, skin tone, hydration and elasticity except skin lightening. In conclusion, this study proved that Cedrol and collagen-derived peptide had the synergy effect of type III collagen synthesis in the cell level and cosmetics with those was improved skin aging in human volunteer test.

Prevalence of Extended-spectrum β-Lactamase and Quinolone Resistance Genes in Escherichia coli Clinical Isolates and their Antibiotic Resistance (임상검체로부터 분리된 Escherichia coli 의 Extended-spectrum β-lactamase와 퀴놀론 내성 유전자의 출현빈도 및 항생제 내성)

  • The aim of this study was to investigate the prevalence of Extended-spectrum ${\beta}$-lactamase (ESBL) gene and quinolone resistance determinant (qnr) and the pattern of antibiotic resistance in the ESBL-producing Escherichia coli clinical isolates. The 42 ESBL-producing strains from total 274 isolates were detected using a double disk synergy test. They were isolated from various specimens, such as urine (28 strains), sputum (6 strains), pus (3 strains), wound (2 strains), blood (2 strains), and tissue (1 strain). Using the PCR with the specific primers ESBL, ESBL and qnr gene types were determined. Thirty-five strains possessed one or two ESBL genes. CTX-M-1 type was the most abundant followed by CTX-M-9 type and TEM, but SHV, CTX-M-2, and CTX-M-8 gene types were not detected. qnr gene types were detected from ten isolates in the order of qnrB4, qnrB1, and qnrS. Coexistence of ESBL and qnr genes was found. ESBL-producing isolates showed high resistance against some antibiotics, such as cefotaxmie (80.0%), levofloxacin (82.9%), and ampicillin (100%). Neither a synergy effect from the coexistence of ESBL and qnr genes on antibiotic resistance nor a correlation between the production of qnr gene and quinolone resistance were found.

A Study on Synergistic Antibacterial Activity of Cosmetic Ingredients (화장품 성분들의 세균에 대한 항균 시너지 효과 연구)

  • In this study, 1,2-hexanediol, ethylhexylglycerin, and glyceryl caprylate known as skin conditioning agents with antimicrobial activity in cosmetics were investigated for their antimicrobial activity against gram-positive and gram-negative bacteria when combining them with various ingredients in cosmetic formulations. Seven kinds of substances expected to improve the antibacterial activity in formulations were selected to blend with these three compounds.; 1,3-propanediol, 1,3-butanediol, glycerin, dipropylene glycol, niacinamide, EDTA-2Na, and ethanol. The checkerboard assay was conducted to confirm the antibacterial synergy of these seven substances and the three skin conditioning agents. Consequently, ethanol and EDTA-2Na were detected as the significant materials with synergistic effect as well as ethanol showed antibacterial synergy with all the three compounds. When seven of selected materials were mixed with glyceryl caprylate, there were synergistic or additive activity against S. aureus, gram-positive bacteria and it was suggested that the combination with 1,2-hexanediol and glyceryl caprylate was useful to control both gram-positive and gram-negative bacteria when there were four kinds of polyols. This study is meaningful in that it confirmed the antibacterial synergy of the skin conditioning agents with antimicrobial activity and the main raw materials frequently used in cosmetics, thereby contributing to prediction of the antibacterial activity of the skin conditioning agents in cosmetic formulations.

The Comparative Efficacy of Colistin Monotherapy and Combination Therapy Based on in vitro Antimicrobial Synergy in Ventilator-associated Pneumonia Caused by Multi-drug Resistant Acinetobacter baumannii (다제내성 Acinetobacter baumannii 에 의한 인공호흡기연관 페렴에서 Colistin 단독요법과 시험관 내 상승작용에 근거한 병합요법간의 효능 비교)

  • Background: Ventilator-associated pneumonia caused by multi-drug resistant Acinetobacter baumannii has been increasing and growing as a threat in intensive care units. Limited therapeutic options have forced clinicians to choose colistin with or without combination of other antibiotics. We tried to compare the effectiveness between colistin monotherapy and combination therapy based on in vitro synergistic tests. Methods: From January 2006 to December 2007 in medical ICU of a tertiary care hospital in Korea, We reviewed the medical records of patients treated with intravenous colistin due to ventilator-associated pneumonia caused by multi-drug resistant Acinetobacter baumannii. Results: A total of 41 patients were analyzed. 22 patients had been treated with colistin monotherapy and 19 patients with colistin and combination antibiotics that were found to have in vitro synergistic effects. Baseline characteristics were similar in both groups but the mean duration of colistin administration was significantly longer in the combination group (19.1${\pm}$11.2 days vs. 12.3${\pm}$6.8 days, p=0.042). There were no significant differences in outcome variables between the two groups. Conclusion: Combination treatment based on the in vitro antimicrobial synergy test did not show better outcomes compared with colistin monotherapy in VAP caused by multi-drug resistant A. baumannii.

In vitro Antimicrobial Combination Therapy in Metallo-β-lactamase Producing Pseudomonas aeruginosa (Metallo-β-lactamase 생성 Pseudomonas aeruginosa의 시험관내 항균제 병합요법에 대한 연구)

  • Metallo-${\beta}$-lactamase (MBL) can hydrolyze all ${\beta}$-lactams except monobactams and frequently coexists with various antibiotic resistance genes such as aminoglycoside resistance, sulfonamide resistance gene, etc. Therefore, the effective antibiotics against infections by these bacteria are markedly limited or can't even be found. We tried to search in-vitro antimicrobial combinations with synergistic effects for a VIM-2 type MBL producing Pseudomonas aeruginosa, isolated from clinical specimen. On the selection of antibiotic combinations with synergistic effects, we performed a one disk synergy test, modified Pestel's method, in agar without aztreonam (AZT). The bacteriostatic synergistic effects of this tests were scored as $S_1$ (by susceptibility pattern in agar without antibiotics), $S_2$ (by the change of susceptibility in agar with or without antibiotics) and $S_3$ ($S_1$ + $S_2$) and was classified into weak (1 point), moderate (2 points) and strong (3 points) by $S_3$ score. Subsequently, we carried out the time-killing curve for the antibiotic combinations with the strong synergistic bacteriostatic effect. One VIM-2 type MBL producing P. aeruginosa confirmed by the PCR showed all resistance against all ${\beta}$-lactams except AZT, aminoglycoside and ciprofloxacin. In the one disk synergy test, this isolate showed a strong bacteriostatic synergistic effect for the antibiotic combination of AZT and piperacillin-tazobactam (PIP-TZP) or AZT and amikacin (AN). On the time-killing curve after six hours of incubation, the colony forming units (CFUs/mL) of this bacteria in the medium broth with both combination antibiotics were decreased to 1/18.7, 1/17.1 of the least CFUs of each single antibiotics. The triple antibiotic combination therapy including AZT, PIP-TZP and AN was shown to be significantly synergistic after 8 hrs of exposure. In a VIM-2 MBL producing P. aeruginosa with susceptibility for AZT, the triple antibiotic combination therapy including AZT, PIP-TZP and AN may be considered as an alternative antibiotics modality against the infection by some MBL type. But the antimicrobial combination therapy for many more MBL producing isolates is essential to know as soon as possible for the selection of effective treatment against the infection by this bacteria.

A Study on the Antibacterial Activity of Combined Administration of Jakyakgamcho-tang and Antibiotics Against MRSA (MRSA에 대한 작약감초탕과 항생제 병용투여의 항균활성에 관한 연구)

  • MRSA is Staphylococcus aureus resistant to β-lactam antibiotics, and is a worldwide infectious disease. Even with the discovery of new antibiotics, resistance develops rapidly, so new alternatives are needed. Jakyakgamcho-tang (JGT) is a combination of Jakyak and Gamcho, and has been mainly used as an antispasmodic and analgesic in oriental medicine. This study was conducted to find out whether there is an effect on MRSA in relation to the anti-inflammatory effect of JGT and the antibacterial effect of Jakyak and Gamcho found in previous studies. In this study, in order to investigate the antibacterial activity of JGT and the combined effect of existing antibiotics, after extracting JGT with 70% EtoH, the disc diffusion method, minimum inhibitory concentration (MIC), drug combination effect (FICI), and time-kill analysis (Time-kill assay), metabolic inhibition, Western blot and qRT-PCR analysis were used to confirm the antibacterial activity mechanism of MRSA of JGT. As a result of the experiment, all of MRSA showed antibacterial activity in JGT's disc diffusion method, and the MIC was 250-1000 ㎍/mL. When existing antibiotics and JGT were combined with drugs, most had synergy or partial synergy. In addition, it was confirmed that the degree of bacterial growth was suppressed over time when simultaneous administration for 24 hours. JGT showed a synergistic effect when administered together with the ATPase-inhibitor DCCD, suggesting that it affected the inhibition of ATPase. As a result of observing the expression of PBP2a, and hla protein in the JGT-treated group and the untreated control group through wstern blot, it was confirmed that the protein expression of the JGT-treated group was significantly suppressed, and the expression levels of mecA, mecR1 and hla genes were also suppressed during JGT treatment. was observed by qRT-PCR. Combining the results of the experiment, it can be seen that JGT has antibacterial activity in MRSA, and when combined with existing antibiotics, the effect was increased compared to treatment with the drug alone. This suggests that JGT can be an alternative to treatment for antibiotic resistance of MRSA.

민.군 겸용기술사업의 추진-민.군 겸용기술개발과 기술이전을 중심으로

  • 경제개발 초창기부터 우리 나라의 산업발전에 일익을 담담해 온 국방과학기술은 그 특수성 때문에 민간기술과 혼합되어 시너지효과(Synergy Effect)를 유발시킴으로써 신기술의 개발은 물론 국가경쟁력 제고에 상당한 기여를 하였던 것이 사실이다. 하지만, 걸음마 단계에 불과한 민.군겸용기술사업은 현재의 여건상 소요예산확보, 대국민 홍보, 산.학.연.군 및 정부부처와의 협조체제 구축 등의 몇 가지 현안 문제점들을 안고는 있지만 사업담당자 및 관련부처 협조와 공감대 형성을 통하여 한국적인 "민.군겸용기술개발사업"구축에 지속적인 관심과 노력이 있어야 할 것이다.

  • DEA has been effectively applied to various areas which need the evaluation of relative efficiency. We propose a DEA model based on fuzzy LP in order to consider uncertain synergy effects due to M&A of existing organization. We apply the suggested approach to forecasting the efficiency of merged academic departments in a university in Korea. We expect that our approach can be utilized to effectively realign existing departments.
