Combined effect of recombinant human bone morphogenetic protein-2 and low level laser irradiation on bisphosphonate-treated osteoblasts

  • Objectives: The purpose of this study was to evaluate the synergic effect of recombinant human bone morphogenetic protein-2 (rhBMP-2) and low-level laser therapy (LLLT) on bisphosphonate-treated osteoblasts. Materials and Methods: Human fetal osteoblast cells (hFOB 1.19) were cultured with $100{\mu}M$ alendronate. Low-level Ga-Al-As laser alone or with 100 ng/mL rhBMP-2 was then applied. Cell viability was measured with MTT assay. The expression levels of receptor activator of nuclear factor kappa-B ligand (RANKL), macrophage colony-stimulating factor (M-CSF), and osteoprotegerin (OPG) were analyzed for osteoblastic activity inducing osteoclastic activity. Collagen type and transforming growth factor beta-1 were also evaluated for bone matrix formation. Results: The results showed that rhBMP-2 and LLLT had a synergic effect on alendronate-treated osteoblasts for enhancing osteoblastic activity and bone matrix formation. Between rhBMP-2 and LLLT, rhBMP-2 exhibited a greater effect, but did not show a significant difference. Conclusion: rhBMP-2 and LLLT have synergic effects on bisphosphonate-treated osteoblasts through enhancement of osteoblastic activity and bone formation activity.

Synergism among Endo-xylanase, $\beta$-Xylosidase, and Acetyl Xylan Esterase from Bacillus stearothermophilus

  • Synergic effects among endo-xylanase, $\beta$-xylosidase, and acetyl xylan esterase of Bacillus stearothermophilus in the hydrolysis of xylan were studied by using birchwood, oat spelt, and acetylated xylan as substrates. Synergism between endo-xylanase and $\beta$-xylosidase was observed on all three substrates tested, indicating that $\beta$-xylosidase enhanced the production of xylose by relieving the end-product inhibition upon endo-xylanase conferred by xylooligomers. Endo-xylanase and $\beta$-xylosidase also showed synergism with acetyl xylan esterase in the hydrolysis of birchwood and acetylated xylan, while no synergic effect was detected in oat spelt xylan hydrolysis. Thus, the hydrolysis of xylan containing acetic acid side chains required the action of acetyl xylan esterase, which eliminated the steric hindrance of the side chains, leading to the better hydrolysis by endo-xylanase and $\beta$-xylosidase , and the acetyl xylan esterase activity was also enhanced by endo-xylanase and $\beta$-xylosidase for the latter enzymes provided acetyl xylan esterase with shorter xylan oligomers, the better substrate for the enzyme.

Prestress and excitation force identification in a prestressed concrete box-girder bridge

  • Prestress force identification (PFI) is crucial to maintain the safety of prestressed concrete bridges. A synergic identification method has been proposed recently by the authors that can determine the prestress force (PF) and the excitation force simultaneously in prestressed concrete beams with good accuracy. In this paper, the ability of this method in the application with prestressed concrete box-girder bridges is demonstrated. A reasonable assumption is made to capture the similarity of the dynamic behavior of the prestressed concrete box-girder bridge and a beam under a certain loading scenario, and the feasibility of this method for application in a prestressed box-girder bridge is affirmed. A comprehensive laboratory test program is conducted, and the effects of PF, excitation, measuring time and uncertainties are studied. Results show that the proposed method can predict the PF and the excitation force in a prestressed concrete box-girder accurately and has a great robustness against uncertainties.

Synergic Effect of GamiSamgieum (SGMX) and Lipitor on Hyperlipidemia in Animal Model

  • Objectives: To investigate the possibility of GamiSamgieum (SGMX) as a combination therapy with statins on hyperlipidemia using an animal model. Methods: Forty eight ICR mice (male) were divided into six groups of eight mice each: naive, induced, Lipitor 5 mg/kg, Lipitor 5 mg/kg plus SGMX 100 mg/kg, Lipitor 10 mg/kg, and Lipitor 10 mg/kg plus SGMX mg/kg treatment group. Hyperlipidemia was induced by feeding a purified high fat diet for all groups (except naive) along with treatment of drugs for 6 weeks, and then biological parameters were examined on the last experiential day. Results: Lipitor treatment lowered total cholesterol and increased HDL-cholesterol compared to the induced group with no statistical significance. However, co-treatment of SGMX with Lipitor revealed synergic effects on total cholesterol and HDL-cholesterol significantly (P < 0.05) in both. SGMX co-treatment also significantly protected liver tissues from the oxidative stress in liver tissues (P < 0.05) and augmented inhibitory effect of Lipitor against fat accumulation in the body. Conclusion: These results indicate the possibility of that SGMX can be used for patients having hyperlipidemia as a combination therapy with statin drugs.

순기화중탕(順氣和中湯)과 Doxorubicin의 병용이 Colon-26의 항암효과에 미치는 영향 (The Anti-tumor Effect of Soonkiwhajungtang with Doxorubicin in Colon-26)

  • In order to evaluate the anti-tumor and synergic effect of Soonkiwhajungtang with doxorubicin, the inhibitory concentration(IC), IC50 and IC90 of single use of doxorubicin and Soonkiwhajungtang with their concomitant treatment against Colon-26(Murine Rectum Carcinoma) was observed using MTT(Microculture Tetrazolium test) assay. In addition, their anti-tumor effects were also observed in the xenograft nude mice models against 3LL cell lines. Soonkiwhajungtang may only mimic direct anti-tumor effects against 3LL cell lines, but signs of worsening induced by implantation of tumor cell lines generally decreased, while the total WBC and lymphocyte numbers increased. Therefore, experimentation suggests that Soonkiwhajungtang extracts reduced the critical toxicity of doxorubicin, and that Soonkiwhajungtang extracts have favorable synergic effects when combined with doxorubicin.

  • Objectives & Methods : In order to evaluate the in vitro synergic effects or Eunkyo-san which is a traditional poly-herbal formula that has been used in the treatment of respiratory diseases in oriental medicine, and quinolone antibiotics, rufloxacin (RUFX) and iprofloxacin(CPFX), experimentation was designed to determine minimal inhibitory concentration(MIC), $MIC_{50}\;and\;MIC_{90}$ of single use of quinolones and concomitant treatment with Eunkyo-san against 5 strains of aerobic gram negative bacteria, Escherichia coli, Klebsiella peumoniae, Hemophilus influenzae, Citrobacter freundii and Pseudomonas aeruginosae. Result : In the case of aerobic gram negative bacteria, the MIC, $MIC_{50}\;and\;MIC_{90}$ against Klebsiella peumoniae and Citrobacter freundii significantly decreased in concomitant-treated groups with Eunkyo-san compared to those of single-treated groups of RUFX and CPFX, respectively. However, no significant changes were demonstrated against Echerichia coli, Hemophilus influenzae and Pseudomonas aeruginosae. Conclusion : According to these results, concomitant use of Eunkyo-san against some strains of aerobic gram-negative bacteria dramatically increases in vitro antibacterial activity of RUFX and CPFX, and the increase and selectivity of antibacterial activities against these strains is attributable to Eunkyo-san, and not RUFX or CPFX activity.

국내허브(석창포)와 에센셜 오일 항균효과 (A Study on Synergy Effect of Antimicrobial Activity on Aroma essential oil)

  • 석창포와 에센셜오일 Geranium(Pelargonum aspeum), lemongrass(Cymbpogen), Cypress(Cupressus Sempervirens), Chamomile Roman(Anthemis nobilis), Tea tree(malaleuca lterniflia), Bergamot(citrus aurant bergamia), lavender(Lavendula officinalis), Sandal wood(Santalum album)을 혼합하여 나타나는 항균 시너지 효과를 Streptococcus aureus, Pseudomonas aeruginosa, Candida albicans에 대하여 검토 해본 결과 다음과 같은 사실이 확인되었다. 1. 석창포는 Streptococcus aureus, Pseudomonas aeruginosa, Candida albicans에 대하여 뚜렷한 항균 효과를 나타냈다. 2. 아로마오일, Geranium(Pelargonum aspeum) lemongrass(Cymbpogen), Cypress(Cupressus Sempervirens), Chamomile Roman(Anthemis nobilis), Tea tree(malaleuca lterniflia), Bergamot(citrus aurant bergamia).lavender(Lavendula officinalis), Sandal wood(Santalum album)등은 Streptococcus aureus, Pseudomonas aeruginosa, Candida albicans에 대하여 강력한 항균 효과를 나타냈다. 3. 석창포와 아로마오일을 혼합 시에도 Streptococcus aureus, Pseudomonas aeruginosa, Candida albicans에 대하여 항균 시너지 효과를 나타냈다. 이로써 석창포와 아로마오일 모두 항균력을 가지고 있음이 확인되었고, 혼합시 또한 석창포와 아로마 오일의 대부분 상승효과가 있으며 이는 화장품에 폭넓게 응용할 수 있으리라 사료된다.

LPS로 활성화된 대식세포에서 현부리경탕(玄附理經湯)의 시험관내 항염효과 및 Piroxicam과 병용효과 (In Vitro Anti-inflammatory Effects of Hyunbulikyung-tang Aqueous Extracts on LPS activated Raw 264.7 Cells, and Synergic Effects with Piroxicam)

  • Objectives: To observe the in vitro anti-inflammatory effects of Hyunbulikyung-tang aqueous extracts (HBLKT) and the possible synergic combination effects with a nonsteroidal anti-inflammatory drug, piroxicam. Methods: Anti-inflammatory effects of HBLKT (yield=16.17%) were observed on LPS activated raw 264.7 cells based on $ED_{50}$ to cell viability, NO, $PGE_2$, $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 productions as compared with piroxicam, in the present study. In addition, the combination effects of HBLKT with piroxicam were observed after treatment of HBLKT 1/4 $ED_{50}$ + piroxicam 1/4 $ED_{50}$, 1/8 $ED_{50}$, 1/16 $ED_{50}$, 1/32 $ED_{50}$ and 1/64 $ED_{50}$ concentrations, respectively. Results: Significant (p<0.01 or p<0.05) increases of cell viabilities and decreases of NO, $PGE_2$, $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 cytokine releases were detected in HBLKT 1/4 $ED_{50}$ + piroxicam 1/4 $ED_{50}$, 1/8 $ED_{50}$, 1/16 $ED_{50}$ and 1/32 $ED_{50}$ concentration co-treatment as compared with each of single 1/4 $ED_{50}$ concentration of piroxicam and HBLKT treatments, respectively. Although significant (p<0.01 or p<0.05) increases of cell viabilities and decreases of NO, $PGE_2$, $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 cytokine releases were also demonstrated in piroxicam 1/64 $ED_{50}$ + HBLKT 1/4 $ED_{50}$ co-treatment as compared with LPS control, no significant changes were detected as compared with each of single 1/4 $ED_{50}$ concentration of piroxicam and HBLKT treatments, in this experiment. Conclusions: Hyunbulikyung-tang showed cell protective and anti-inflammatory effects against LPS activated raw 264.7 cells. It, therefore, expected that HBLKT will be showed favorable effects to relieve dysmenorrhea related to over expressed inflammatory cytokines, and it also expected that the clinical dosages of piroxicam can be reduced as 1/8 levels as combination with HBLKT.

  • This research is based on the survey of strategic alliance of the foodservice industry conducted by interviewing their customers. We found out that cooperating and information of benefits influenced revisiting followed by service quality and customer satisfaction. As a result of research, we noticed that for breaking through the depression and having synergic effects among the enterprises, providing high service and quality to customer is a key to success. It must be used as an essential part of marketing strategic plans for maintaining and increasing customers, guiding partner selection among the enterprises.

MCF-7에서 Cisplatin과 타우린의 병용처리로 인한 항암효과 및 관련 기전 (Anticancer Effects and Mechanisms of Co-Treatment of Cisplatin with Taurine in MCF-7 Cells)

  • The objective of this study is to evaluate the synergic effects of combined treatment with taurine and cisplatin in human breast cancer, MCF-7 cells. For this study, MCF-7 cells were treated with taurine (5, 10, and 20 mM) and cisplatin (0.5 ${\mu}M$) for 48 and 72 hrs. Co-treatment of cisplatin with taurine decreased cell proliferation more compared with cisplatin alone. Reduced cell proliferation was caused by apoptosis. Therefore we investigated the apoptotic cells. After treatment of cisplatin and taurine, apoptotic cells were slightly increased. Apoptosis-related proteins, cleaved caspases and cytochrome c were increased. The present study suggests that combination treatment of cisplatin with taurine enhance anticancer activity of cisplatin in MCF-7 cells.
