Ultrastructural Alterations of Rabbit Liver after Overdose of Nitrate (질산염과잉투여(窒酸鹽過剩投與)에 의(依)한 간장(肝臟)의 변화(變化)에 관한 전자현미경적연구(電子顯微鏡的硏究))

  • In order to know the morphological changes of liver in nitrate poisoning, the ultrastructural studies were carried out on the rabbit liver after potassium nitrate was administered orally at lethal dose, in single treatment, as acute case and at two different levels. 1.0 and 0.5g/kg of body weight daily for 43 and 60 days as chronic case, respectively, The results were summarized as followings: 1. In the hepatic cells of acute case, mitochondria were swollen, disappearance of cristae and variable in shape. Dilatation of rough endoplasmic reticulum and vacuoles containing degenerated cell organells were observed. Glyogen particles were decreased in number. Degenerated Kupffer cells were often seen in acute case. 2. In the hepatic cells of chronic case, there were increase of smooth endoplasmic reticulum, marked enlargement of rough endcplasmic reticulum, detachment of membrane bound ribosome and some rough endoplasmic reticulum changed into smooth endoplasmic reticulum. Secondary lysosome, abundant glycogen paricles and myelin-figure structures were also observed in the cytoplasm of the hepatic cells. The endothelial cells were proliferated in the area of the necrotic cells.

  • This research was undertaken to determine the effects of the cyclophosphamide (CP) on the epididymal corpus of the male rat in terms of ultrastructural alteration and protein analysis by SDS-PAGE at different groups; control group, 1 week group, 3 weeks group and 5 weeks group were treated with saline (control group) or CP at doses of 20mg/kg/week, 1 time a week, respectively. In the cytoplasm of the principal cells on the epididymal corpus, the mitochondria were significantly swollen or disrupted. The lumens of rough endoplasmic reticulum (rER) were also dilated and the number of secretory vesicles and lysosomes were increased respectively. CP caused changes in protein concentrations in the corpus of epididymis after CP treatment. Total proteins of 31 to 36 species were expressed in the corpus fluid. Then the more CP was increased, the more concentration of proteins caused to decrease, synthesize or increase in epididymal corpus. In contrast to the control group, in particular 88KD and the other 8 proteins in the corpus fluid, were decreased or disappeared respectively, whereas acid phosphatase and the other 9 proteins in the corpus fluid, were increased or synthesized respectively. The other proteins are not showed distinctive difference. It is suggested that treatment with CP alters the specific cell organelles and proteins in segment of the epididymal corpus.

Ultrastructure of the Epiphytic Sooty Mold Capnodium on Walnut Leaves

  • Cellular aspects of sooty mold on walnut leaves were investigated by using light and electron microscopy. A black coating developed on the adaxial leaf surface of a walnut tree. No infestations were found on the abaxial leaf surface with peltate glandular trichomes. Light microscopy showed that fungal complexes from the leaf surface were composed of brown conidia and hyphae. Conidia, with longitudinal and transverse septa, were variable in length ranging from 10 to $30{\mu}m$, and commonly found in clusters, forming microsclerotia. Neither epidermal penetration nor hyphal entrance to host tissues was observed. Based on their morphological characteristics, the fungal complexes were assumed to be Capnodium species. An electron-dense melanized layer was present on the cell wall of multi-celled conidia. Concentric bodies in the fungal cytoplasm had an electron-translucent core surrounded by an electron-dense margin with a fibrillar sheath. Chloroplasts without starch granules in the palisade mesophyll cells of sooty leaves had electron-dense stromata and swollen plastoglobuli. These results suggest that the epiphytic growth of fungal complexes can be attributed to the melanized layer and concentric bodies against a water-deficient environment on the leaf surface. Ultrastructural characteristics of the sooty leaves indicate typical features of dark-adapted and non-photosynthetic shade leaves.

  • Silver nanoparticles (AgNPs) can be used in the areas such as integrate circuit, cell electrode and antimicrobial deodorant. In this study, AgNPs have been prepared by using $AgNO_3$ aqueous solution in the carboxymethyl cellulose (CMC) hydrogel. CMC powders were dissolved in deionized water, and then irradiated by a gamma-ray with a radiation dose of 50 kGy to make CMC hydrogel. CMC hydrogels were dipped into $1.0{\times}10^{-2}M$ $AgNO_3$ solution for 1 hour. After that, the swollen hydrogels were irradiated by gamma-ray for the formation of AgNPs. The characteristics of silver nanoparticles in the CMC hydrogels were monitored by UV-Vis and the morphological study and dispersed coefficient of particles were investigated by FE-SEM/EDX. It was observed that the sodium salt in the CMC is crucial to the formation of silver nanoparticle. Finally, antibacterial tests indiacted that the hydrogel containing silver nanoparticle has antibacterial activity.

Biocontrol of Citrus Canker Disease Caused by Xanthomonas citri subsp. citri Using an Endophytic Bacillus thuringiensis

  • Citrus canker is a devastating disease of citrus caused by Xanthomonas citri subsp. citri (Xcc). A total of 134 endophytic bacteria were isolated from various gymnospermic and angiospermic plants. They were screened for their antagonistic activities against three wild-type and six streptomycin-resistant Xcc strains. TbL-22 and TbL-26, both later identified as Bacillus thuringiensis, inhibited all the wild and resistant Xcc strains. TbL-22 exerted the highest antagonistic activity against XccW3 and XccM6 with inhibition zones of $20.64{\pm}0.69$ and $19.91{\pm}0.87mm$, respectively. Similarly ethyl acetate extract of TbL-22 showed highest inhibition zones $15.31{\pm}2.08$ and $19.37{\pm}3.17mm$ against XccW3 and XccM6, respectively. TbL-22 reduced canker incidence on infected leaves by 64.05% relative to positive controls. Scanning electron microscopy revealed that the cell membranes of Xcc treated with ethyl acetate extract of TbL-22 were ruptured, lysed, and swollen. B. thuringiensis TbL-22 can effectively and sustainably controls streptomycin-resistant citrus canker.

Histological study on the skin structure in two mudskippers, Periophthalmus waltoni and Boleophthalmus dussumieri in relation to their terrestrial life

  • Microscopic structure of skin in two amphibious mudskipper fish; Boleophthalmus dussumieri Valenciennes, 1837 (B. dussumeri) and Periophthalmus waltoni Koumans, 1941 (P. waltoni) were investigated in relation to their lifestyle. The general structure of skin is the same among the two species. Epidermis in B. dussumeri was thicker significantly than P. waltoni. The dermal bulges were only well developed in the skin of B. dussumeri. Mucous cells were absent in the epidermis of P. waltoni but present in B. dussumeri. Both B. dussumeri and P. waltoni have well-developed swollen middle cells as a shared epidermal feature. The thickness of the middle cell layer of the epidermis in B. dussumeri was significantly greater than in P. waltoni. Capillaries in the dorsal and ventral parts of the body are more closely distributed to the epidermal surface in P. waltoni than in B. dussumeri. The diffusion distance in the dorsal epidermis of P. waltoni was less than that in the ventral epidermis of B. dussumeri. A comparative examination of the skin of mudskipper species suggests that, due to the more terrestrial lifestyle adopted by P. waltoni, the skin contributes more to respiration.

ELECTRON MICROSCOPIC STUDY ON THE HISTOPATHOLOGICAL CHANGES OF THE PERIODONTAL TISSUE OF PRESSURE SIDE FOLLOWING EXPERIMENTAL TOOTH MOVEMENT IN GUINEA PIGS (Guinea pig에서 실험적 치아이동에 따른 압박측 치아주위 조직의 변화에 관한 전자현미경적 연구)

  • The hyalinized zone in compressed periodontal ligament seems to be an unavoidable aspect during certain phases of orthodontically produced tooth movement. And these hyalinized zones leads to a standstill of tooth movement. But when hyalinized zones disappears after a certain period of time and tooth movement is established. In the basic aspect of clinical science of orthodontics, it is very important, to study about the process involved and to establish whether a difference of periodontal response by the amount of the applied experimental force exists. The 35 Guineapigs were divided into the control group (5 animals) and the experimental group (Group I-Group VI 30 animals). The experimental tooth movement of Guineapig's maxillary incisors were carried out by rendering continuous force, 5g (Group I, Group II) 35g (Group III, Group IV), 100g (Group V, Group VI) respectively. 15 animals (Group I, Group III, Group V) were sacrificed one week after this experiment. Another 15 animals (Group II, Group IV, Group VI) were sacrificed one week after the removal of active appliences. At the end of the experimental periods, specimens containing tooth and adjacent periodontal structure were obtained and processed for light and electron microscopy. The results were as follows: 1. In Group I, cellular changes of pressured zones showed swollen mitochondria, dilation of rough surfaced endoplasmic reticulum (RER), vesicles and pyknosis of nucleus. 2. In Group III and Group V, the hyalinized tissues showed cell necrosis accompaning ruptures of cytoplasmic membrane and perinuclear envelope, large cytoplasmic vacuoles and many necrotic cell debris. 3. In Group IV and Group VI, hyalinized tissue were eliminated and the primitive mesenchymal cells and blood capillaries were proliferated. 4. In group V, the destruction of the collagenous fibers were observed, while in group I and group III were not observed. 5. In Croup IV and Group VI, the hyalinized zones were still remained partly.

  • This study was carried out to investigate the effect of the red light on embryo tissue of barley during germination. The solubility of starch in endosperm of germinated barley was different between dark and red treatments at the 3rd day of germination, but was increased by the red light from the 4th day of germination. Blue value of the starch in the germinated barley decreased rapidly from 0.42 at the 1st day to 0.13 at the 6th day in the dark, and same tendency was found in the red light, but blue value was lower in the red light than in the dark. Aleurone cell wall was swollen much faster in the red light than in the dark during germination. The cell wall was broken down more greatly in the red light than in the dark at the 5th day of germination.

Outbreak of carprine arthritis-encephalitis in dairy goat flocks (유산양에서 관절염.뇌염 발생)

  • This is a case report on the occurrence of caprine arthritis-encephalitis (CAE) disease among dairy goats in a local farm located in Yeongdong-gun, Chungbuk. Previously, it was reported that the farm experienced intermittent deaths numbering 15 of the 97 goats raised for 5 months. Most of the goats less than 6 months of age were suffering from ataxia and posterior paresis, body tremor and abnormal head posterior. Affected animals frequently had stunted growth and had a rough coat. Goats more than 6 months of age were affected with an insidious, chronic arthritis characterized by articular swelling ('big knee') of the carpal, hock, and stifle joints. Necropsy revealed severely swollen mesenteric lymph nodes, under- flow of 2-3ml synovial fluid in the articular space and fibrous proliferation of synovial membrane. Histopathological examination showed perivascular accumulations of mononuclear inflammatory cells in the white matter of the brain, proliferative synovitis characterized by villous hypertrophy, synovial cell hyperplasia and infiltration by mononuclear inflammatory cells. Pulmonary lesions consists of patchy interstitial pneumonia with hyperplasia of lymphoid tissues and an extensive mononuclear inflammatory cell infiltration into the alveolar septa. Confirmation by nested PCR involves amplification of a 296 bp (lst PCR) and 184 bp (2nd PCR) fragments corresponding to the gag region of the CAE virus. This is the first time CAE has been reported in a local farm in Korea and emphasizes the importances of developing preventive measures against CAE.

Studies on Ultrastructure of Rat's Liver Cell and Fetal Liver Cell Treated by Actinomycin D (Actinomycin D가 흰쥐의 모체 및 태아 간세포에 미치는 영향에 관한 전자현미경적 연구)

  • This study was made to investigate the ultrastructural changes of the hepatocyte of the maternal liver, and fetal liver by Actinomycin D in Wistar rats at the stage of pregnancy. Peritoneal injection of Actinomycin D to rats carried out gestation day 7 to 9 at the level of $15{\mu}g(11.5{\mu}g/100g$ body wt.), $20{\mu}g(15.8{\mu}g/100g$ body wt.) on each day. Treated animals with saline only were used for controls. Animals were sacrificed on day 15 of gestation. On electron microscopic examination, the hepatocytes of maternal liver given Actinomycin D $15{\mu}g$/ml had evidence of serious cellular damage, for example, hypertrophy of rough endoplasmic reticulum, loss in nucleolar osmiophilia, swelling of Golgi apparatus and change of mitochondrial structure. Maternal liver given Actinomycin D $20{\mu}g/ml$ shown similar changes to that of the $15{\mu}g/ml$ treated animals. But mitochondria of this group were not changed than that of $15{\mu}g/ml$ treated group. In the hepatocytes of fetal liver, changes were more pronounced. The drug produced alteration in nuclei and cytoplasm. The rough endoplasmic reticulum was swollen and there were ribosomes detachement. In addition, damages of mitochondria, Golgi apparatus were detected.
