Establishment of Artificial Screening Methods and Evaluation of Barley Germplasms for Resistance to Fusarium Head Blight (보리 붉은곰팡이병 검정법과 저항성 품종 선발)

  • Fusarium head blight (FHB) is a severe disease problem that affects the quality and yield of barley grain. The evaluation of FHB resistance is difficult because environmental conditions greatly influence FHB infection and development. The objectives of this study were to: 1) establish an efficient screening method for selecting resistant barley to FHB, 2) compare FHB severity between the cut-spike method and pot-plant method for development of mass screening, and 3) estimate FHB resistance for barley germplasms. Barley cultivars and lines were evaluated for reaction to FHB in controlled-greenhouse condition. Spikes were spray-inoculated with a suspension $(5.0\times10^5\;macroconidia\;mL^{-1})$ of Fusarium graminearum SCK-O4 strain, and then kept in a greenhouse at $18-25^{\circ}C$ with $80-100\%$ relative humidity. Inoculation were employed at 3 different heading growth stages (heading date, three days after heading, and five days after heading). The inoculation was performed in 2 consecutive days in order to avoid escapes. The inoculated plants were maintained in the greenhouse at 4 different free moisture periods (1, 3, 5, and 7 days). The percentage of FHB severity was scored from 0 to 9 according to the rate of infected kernels per spike, and three spikes were evaluated per replication with 3 replicates. There were significant differences of FHB severity depending on the different free moisture periods, but not by the inoculation at different heading stages. The optimum evaluation point of FHB severity in the greenhouse condition was on the 7th day under free moisture condition after inoculation at the heading date. Infection level in cut-spike method highly correlated with that in pot-plant method. This suggested that cut-spike method is useful in evaluating of FHB resistance in barley. Six cultivars, such as Jinkwang, Buheung, Atahualpha 92, Chevron-b, Gobernadora-d, and MNBrite-c, were selected as resistant varieties to FHB. Correlation coefficient for the FHB severity evaluated by the pot-plant method between two seasons was 0.794, indicating the stability and accuracy of the screening method.

Characteristics for Heavy Metal Pollution in Road Dust from Daebul Industrial Complex: Classification by Particle Size and Magnetic Separation (대불산업단지 도로먼지 내 중금속류 오염 특성: 입도와 자성에 따른 구분)

  • In this study, we investigated physical and chemical properties such as grain size, heavy metal pollution, magnetic properties, and their environmental impacts of road dusts (RD) collected from 14 sampling points in Daebul industrial Complex. Heavy metal concentrations in RD were in the order of Fe>Zn>Cu>Pb>Cr>Ni>As>Cd>Hg, and this pollution pattern was related to major industries and traffic activities in this area. The results of the correlation analysis between heavy metal elements and particle size in RD showed that Fe and all of analyzed heavy metals had a significant correlation with each other and metal concentrations had a significantly negative correlation (p<0.05). However, due to the input of large metal particles some heavy metal concentrations in the particle fraction of >1000 ㎛ were highest. Pollution load per unit area of this fraction was the highest among the grain size fractions. Cr, Ni, Cu, Zn, Cd, Pb levels in RD decreased and the levels of Cr, Ni, Cu, Zn, Cd, and Pb were reduced to 85 (As) -22 (Ni)% of the whole after removal of MFs fraction from RD. The mean heavy metal levels in the study area did not exceed the soil contamination guide value of Korea, indicating that heavy metal levels in RD were not a concern. However, at some sampling points, Zn concentrations were exceeded the soil contamination guide value for the 3rd areas of Korea and this result indicated that further studies of the impact of RD on the surrounding environment through re-suspension or non-point pollution, and of effective management methods are required.

A Lab-Made Wound Maker for Analysis of Cell Migration in a 96-Well Plate (세포 이동능력 분석을 위한 96-Well Plate 전용 Lab-Made Wound Maker)

  • Cell migration is a central process for recovering from wounds triggered by physical distress besides embryogenesis and cancer metastasis. Wound healing assay is widely used as a fundamental research technique for investigation of two-dimensional cell migration in vitro. The most common approach for imitating physical wound in vitro is mechanical scratching on the surface of the confluent monolayer by using sharp materials. The iron metal pin with a suspension spring for fine adjustment of the orthogonal contact surface between the scratching point and the individual bottom of multi-well plate with planar curvatures were adopted for the creative invention of a 96-well plate wound maker. While classic tips drew diverse and zigzag scratching patterns on the confluent monolayer, our wound maker displayed synchronized linear wounds in the middle of each well of a 96-well plate that was seeded with several cell lines. Given that several types of multi-well plates commercially available are compatible with our lab-made wound maker for creating uniform scratches on the confluent monolayer for the collective cell migration in wound healing assay, it is certain that the application of this wound maker to the real-time wound healing assay in high content screening (HCS) is superior than utilization of typical polypropylene pipette tips.

Flotation for Recycling of a Waste Water Filtered from Molybdenite Tailings (몰리브덴 선광광미 응집여과액 재활용을 위한 부유선별 특성)

  • Froth flotation using the residual water in the end of flotation process has been performed through controlling of pH. IEP (isoelectric point) of molybdenite and quartz in distilled water was below pH 3 and pH 2.7, respectively and the stabilized range was pH 5~10. In case of a suspension in reusing water, zeta potential of molybdenite decreased to below -10 mV or less at over pH 4 due to residual flocculants. As result of pH control, flotation efficiency in the alkaline conditions was deteriorated by flocculation, resulting from expanded polymer chain, ion bridge of the divalent metal cations ($Ca^{2+}$), and hydrophobic interactions between the nonpolar site of polymer/the hydrophobic areas of the particle surfaces. However, the weak acid conditions (pH 5.5~6) improved the efficiency of flotation as hydrogen ions neutralize polymer chains and then weakened its function. In cleans after rougher flotation, the Mo grade of 52.7% and recovery of 90.1% could be successfully obtained under the conditions of 20 g/t kerosene, 50 g/t AF65, 300 g/t $Na_2SiO_3$, pH 5.5 and 2 cleaning times. Hence, we developed a technique which can continuously supply waste water filtered from tailings into the grinding-rougher-cleaning processes.

Effect of Green Microstructure on Sintered Microstructure and Mechanical Properties of Reaction-Bonded Silicon Carbide (성형미세구조가 반응소결 탄화규소체의 소결미세구조 및 기계적 특성에 미치는 영향)

  • In the binary system of SiC and carbon, porosity and pore size distribution of green body was controlled by varying pH, by the addition of polyelectrolyte dispersants, and by using different particle size of starting powders. The preforms having different green microstructure were fabricated by slip casting from suspensions having different dispersion condition. The reaction bonding process was carried out for these preforms. The condition of reaction bonding was 1600$^{\circ}C$ and 20 min. under vacuum atmosphere. The analyses of optical and SEM were studied to investigate the effect of green microstructure on that of reaction bonded silicon carbide and subsequently the mechanical properties of sintered body was investigated. Different green microstructures were obtained from suspensions having different dispersion condition. It was found that the pore size could be remarkably reduced for a fine SiC(0.5$\mu\textrm{m}$). The bimodal microstructure was not found in the present study, which is frequently observed in the typical reaction bonded silicon carbide. It is considered that the ratio between SiC and C was responsible for the formation of bimodal microstructure. For the preform fabricated from the well dispersed suspension, the 3-point bending strength of reaction-bonded silicon carbide was 310${\pm}$40 MPa compared to the specimen fabricated from relatively agglomerated particles having lower value 260${\pm}$MPa.

  • The purpose of present study was to compare the speed of coronal leakage before and after post space preparation using Streptococcus mutans. Forty straight extracted human teeth were selected. The crowns were removed to a uniform remaining root length 14 mm. Canals were enlarged by 06 taper $Profiles^{(R)}$ to a size $\#40$ as a master apical file. And these were filled with gutta percha point and $Tubuliseal^{(R)}$ sealer, using continuous wave technique. Groupings are as follows. Group 1 - These teeth were obturated without sealer. Group 2 - These teeth were obturated and covered the surface of the root completely with sticky wax. Group 3 - These teeth were obturated. Group 4 - These teeth were obturated and prepared for post space remaining 5 mm of gutta percha. The teeth were suspended in plastic tubes. The upper chamber received the bacterial suspension everyday to simulate clinical situation. The lower chamber consisted of BHI added Andrade's indicator. All roots in the positive control group (Group 1) turned yellow within 24 h and those of negative control group (Group 2) remained red throughout the experimental period (70 days) The samples of group 3 were contaminated within an average of 27.2 days. The samples of group 4 were contaminated within an average of 15.7 days, ranging from 9 to 22 days. There was significant difference between group 3 and group 4 statistically (p < 0.05).

Experiences of Ageism and "Self-Ageism" (노인차별 경험과 자기연령주의(self-ageism))

  • This study focuses on the process of internalizing the perception of negative discrimination among elderly people who have experienced age discrimination. The grounded theory method was used to identify the age discrimination experienced by the elderly in their daily lives and to explore the consequences of such discrimination through self-ageism. According to the analysis results, the elderly respond not only to explicit discrimination but also to implicit discrimination. In this process, the stronger the pain of old age (poverty, disease, ignorance, and solitude) and the stronger the memory of failing to respond to discrimination, the higher the level of self-ageism. "Self-ageism" has internalized the negative image of the elderly, who have been discriminated against by age, resulting in disadvantages in terms of quality of life through various reject/separate/suspension actions in reality. In the presence of supportive resources such as health, institutional support, and caregivers, the elderly have room to overcome self-ageism through more active ways. However elderly people who cannot motivate themselves and they lack these resources, elderly are trapped in negative reflux caused by self-ageism and sustained a depressed and shrinking life. In this state, it is not easy to motivate themselves and make physical, cognitive, emotional and social responses. Therefore, in this analysis, we specifically noted two aspects, contextual conditions and interventional conditions, and proposed programs for synchronizing senior citizens and improving resiliency from a microscopic point of view, and argued for the need to develop systems such as supplementing welfare and health service systems related to the entire life cycle, expanding accessibility and 'age-integration' through 'Community Care', awareness improvement and anti-discrimination laws.

Fundamental Studies on the Net-Shape of the Drag Net-II On the Section Shape of the Simplified Model of the Bag Net in Four-Point Suspension (저예망의 어구형상에 관한 기초적 연구-II 단순화모형에 의한 4폭짜리 자루그물의 단면형상)

  • As a primary step in studying the relationship between the arrangement of netting and the section shape of the bag net in the four-seam trawling net, a series of experiments were performed with the simplified model of the bag net made of vinyl film. This model was suspended horizontally in circularly flowing water, with two pairs of suspending threads to four points of symetry at the border of the bag mouth in place of both wing nets. And then, the area distribution of each panel in the bag net was arranged by the portion of the length of the side panel, q to that of the lower or upper panel, p at the border of bag mouth. In the experiments, the section shape of the bag mouth photographed and the tensions on both pairs of suspending threads were measured with two loadcells in circularly flowing water. From the results that the section shape of the bag mouth in circularly flowing water is controlled by the area distribution of panels in the bag net, the author estimated an experimental equation. h/w=k sub(1) e super(k) 2 super(q/p)+C, where h is the central height of the bag mouth, and w is the lateral width of that.

Optimum Condition of Soil Dispersion for Remediating Heavy Metal-Contaminated Soils using Wet Magnetic Separation (중금속 오염 토양 정화를 위한 습식자력선별법 사용 시 최적 토양분산 조건)

  • Soil dispersion and heavy metal leaching with two heavy metal-contaminated soils were studied to derive the optimal dispersion condition in the course of developing the remedial technology using magnetic separation. The dispersion solutions of pyrophosphate, hexametaphosphate, orthophosphate and sodium dodecylsulfate (SDS) at 1 - 200 mM and the pH of solutions was adjusted to be 9 - 12 with NaOH. The clay content of suspension as an indicator of dispersion rate and the heavy metal concentration of the solution were tested at the different pHs and concentrations of the dispersion solution during the experiment. The dispersion rate increased with increasing the pH and dispersion agent concentration of the solution. The dispersion efficiency of the agents showed as follows: pyrophosphate > hexametaphosphate > SDS > orthophosphate. Arsenic leaching was sharply increased at 50 mM of phosphates and 100 mM of SDS. The adsorption of $OH^-$, phosphates and dodecysulfate on the surface of Fe- and Mn-oxides and soil organic matter and the broken edge of clay mineral might decrease the surface charge and might increase the repulsion force among soil particles. The competition between arsenic and $OH^-$, phosphates and dodecylsulfate for the adsorption site of soil particles might induce the arsenic leaching. The dispersion and heavy metal leaching data indicate that pH 11 and 10 mM pyrophosphate is the optimum dispersion solution for maximizing dispersion and minimizing heavy metal leaching.

Effect of Imipramine on the Contractility of Single Cells Isolated from Canine Detrusor (Imipramine이 배뇨근 세포의 수축성에 미치는 직접작용)

  • The objective of this study was to establish a good methodology to isolate single smooth muscle cells that are alive and respond properly to pharmacological agents. Canine urinary bladders were employed as the source of single cells, and acetylcholine, atropine and imipramine were used as indicators of pharmacological responsiveness. Imipramine, an antidepressant drug exhibited the anticholinergic and calcium antagonizing properties on rat detrusor muscle. To establish a control value for a further experiment to elucidate the mechanism of action of imipramine on detrusor muscle, we measured the concentration-response of single cells to acetylcholine in the presesnce of imipramine by length of the cells and compared the result with the response in the presence of atropine. Tiny chops of smooth muscle taken from anesthetized canine urinary bladder were incubated in collagenase solution at $36^{\circ}C$ for 17-20 minutes. The collagenase solution included collagenase 1.2 mg/ml, soybean tryspin inhibitor 0.08 mg/ml, bovine serum albumin 2% in 10 ml Krebs-Henseleit buffer solution aerated with a consistent breeze of 95/5% $O_2/CO_2$, to maintain the pH at 7.4. After washing with plain K-H solution on 450 mesh, cells were dissociated from the digested tissue for 12-15 minutes. Cell suspension was transfered in 5 ml test tubes and acetylcholine was added for the final concentration to be $10^{-14}M{\sim}10^{-9}M$. To find the optimal time to fix the cells to determine the contractile responses, 1% acrolein was added 5, 10, 20, 30, 60 and 120 seconds after the administration of ACh. The length of cells fixed by acrolein were measured by microscaler via CCTV camera on phaes-contrast microscope. The average length of 50 cells from a slide glass was taken as the value of a sample at the very concentration point. Single cells were isolated from canine detrusor. The length of untreated cells varied from 82 ${\mu}m$ to 94 ${\mu}m$. The maximal response to actylcholine $10^{-9}M$ was accomplished within 5 seconds of exposure, and the shortening was $19{\pm}3$%. Atropine reduced the contraction of the cells concentration-dependently. Imipramine which exerts a cholinergic blocking action on some smooth muscles also reduced the contraction concentration-dependently and by a similar pattern as atropine. These findings document that imipramine may exerts a cholinergic blocking activity in the single smooth muscle cells isolated from canine urinary bladder.
