Review of two immunosuppressants: tacrolimus and cyclosporine

  • Immunosuppressants are vital in organ transplantation including facial transplantation (FT) but are associated with persistent side effects. This review article was prepared to compare the two most used immunosuppressants, cyclosporine and tacrolimus, in terms of mechanism of action, efficacy, and safety and to assess recent trials to mitigate their side effects. PubMed and Google Scholar queries were conducted using combinations of the following search terms: "transplantation immunosuppressant," "cyclosporine," "tacrolimus," "calcineurin inhibitor (CNI)," "efficacy," "safety," "induction therapy," "maintenance therapy," and "conversion therapy." Both immunosuppressants inhibit calcineurin and effectively down-regulate cytokines. Tacrolimus may be more advantageous since it lowers the likelihood of acute rejection, has the ability to reverse allograft rejection following cyclosporine treatment, and has the potential to reinnervate nerves. Meanwhile, graft survival rates seem to be comparable for the CNIs. To avoid nephrotoxicity, various immunosuppressants other than CNIs have been studied. Despite averting nephrotoxicity, these medications show increases in acute rejection or other types of adverse effects compared to CNIs. FT has been a topic of interest for oral and maxillofacial surgeons, and the postoperative usage of immunosuppressants is crucial for the long-term prognosis of FT. As contemporary transplantation regimens incorporate novel medications along with CNIs, further research is required.

  • This study analyzed the current operational status, business continuity, and business continuity strategies of 11 Soho fashion shopping malls through a longitudinal case study (2019/2020 and 2022/2023). The aim was to determine how Soho fashion shopping malls are continuing their business having been successfully established in the fiercely competitive current online fashion market. Regarding changes in shopping mall operation status, all cases displayed changes in items, distribution channels, marketing channels, core values, and profit structures. After establishing the businesses, a crisis arose due to the COVID-19 pandemic, leading to difficulties in creating new customers. Moreover, there was excessive business expansion, resulting in saturation of the online platform store market. This meant companies were experiencing reduced sales (5) or difficulties in maintaining their business (3). However, there were differences in the direction of overcoming the crisis and continuing or converting the business. These revealed that to continue the business after successful establishment of an online fashion shopping mall, it is essential to have the expertise of entrepreneurs, work performance, and marketing ability, as well as differentiation capabilities such as product power and management strategy and core competitiveness that provides a relative advantage over other shopping malls. Customized government support at the level of the start-up ecosystem is also essential for the survival and business continuity of a SOHO online fashion shopping mall with poor human and material resources.

Studies on the Number of Sperm Penetrated Oocytes and Survival Rate of Frozen-thawed Epididymal Dog Sperm (개 정소상체 정자의 난자내 침입율과 동결융해 후의 생존성에 관한 연구)

  • This study was carried out to investigate the general characteristics such as concentration sperm motility and abnormality of sperm on the whole epididymal semen(EWS), RSP-S(removed seminal plasma by saline) and RSP-T(removed seminal plasma by tris-buffer) semen and survival rates after freezing on motility of whole and RSP-S and RSP-T semen and extender containing 2~8% glycerol, and ability of frozen-thawed sperm to penetrate homologous oocytes. 1. The concentration, motility and abnormality of epididymal WES, RSP-S and RSP-T sperm were 4.25 $\pm$ 0.25($\times$10$^{6}$ Cells/$m\ell$), 3.85$\pm$0.20($\times$10$^{6}$ Cells/$m\ell$), 4.05 $\pm$ 0.28($\times$10$^{6}$ Cells/$m\ell$), 50.55 $\pm$ 2.75%, 67.25 $\pm$ 2.55%, 78.75 $\pm$ 3.55 and 9.45 $\pm$ 2.25%, 37.75 $\pm$ 2.10%, 24.25 $\pm$ 1.55%, respectively. 2. The survival rates of slow and rapid frozen epididymal RSP-S and RSP-T sperm were 35.00 $\pm$ 2.35%, 45.50 $\pm$ 2.15% and 16.50 $\pm$ 3.55%, 22.55 $\pm$ 3.95%, respectively. The survival rate of epididymal WES and RSP-T sperm after freezing following dilution with tris-buffer containing 2~8% glycerol were 9.25 $\pm$ 1.55%~17.50 $\pm$ 2.50%. 3. The percentage of capacitated and acrosome-reacted sperm prier to culture for fresh and frozen -thawed epididymal RSP-T semen were 45.25 $\pm$ 5.75%, 7.06 $\pm$ 0.25%, 48.20 $\pm$ 6.80% and 13.00 $\pm$ 2.35%, 3.55 $\pm$ 0.85%, 15.50 $\pm$ 1.90%, respectively. The penetration rate the number of sperm per penetrated for fresh and frozen-thawed epididymal RSP-T sperm were 39.25 $\pm$ 4.72%, 34.24 $\pm$ 3.93% and 1.30 $\pm$ 0.33, 1.10 $\pm$ 0.50., respectively.

Analysis of the Weediness Potential in Vitamin A Enforced Rice (비타민 A 강화 벼의 잡초화 가능성 분석)

  • The purposes of this study were to analyze the weediness potential associated with 'Vitamin A enforced rice' and to evaluate the relationships between growth characteristics and environmental adaption related to survival ability in the natural environment. 'Vitamin A enforced rice', 'Nagdong', donor variety of 'Vitamin A enforced rice' and three other varieties were used for this study. The 'Vitamin A enforced rice' was experimented according to ratio of ripened grain, shattering of grain, germination traits, germination ratio of low temperature, emergence ratio after wintering, ratio of viviparity, and growth ratio after ratooning ability. 'Vitamin A enforced rice' was not significant between 'Nagdong' and other varieties in seven characters. Germination ratio was similar of Vitamin A enforced rice' and 'Nagdong', but average germination ability of was different at low temperature. Also, shattering of grain and germination ratio of alternating temperature were significant between 'Nagdong' and other varieties. These results suggested that the relationships between growth characteristics and environmental adaption in 'Vitamin A enforced rice' could be applied to the stability of weediness potential and biosafety guide in GM rice.

A Review of the Neurocognitive Mechanisms of Number Sense (수 감각의 인지신경학적 기반에 관한 연구 개관)

  • Human and animals are born with an intuitive ability to determine approximate numerosity. This ability is termed approximate number sense (hereafter, number sense). Evolutionarily, number sense is thought to be an essential ability for hunting, gathering and survival. According to previous research, children with mathematical learning disability have impaired number sense. On the other hand, individuals with more accurate number sense have higher mathematical achievement. These results support the hypothesis that number sense provides a basis for the development of mathematical cognition. Recently, researchers have been examining whether number sense training can lead to enhancement in mathematical achievement and changes in brain activity in relation to mathematical problem solving. Numerosity which basically represents discontinuous quantity is expected to be closely related to continuous quantity such as representations of space and time. A theory of magnitude (ATOM) states that processing of number, space and time is based on a common magnitude system in the posterior parietal cortex, especially the intraparietal sulcus. The present paper introduces current literature and future directions for the study of the common magnitude system.

Biofilm Formation Characteristics of Major Foodborne Pathogens on Polyethylene and Stainless Steel Surfaces

  • This research was investigated the effects of temperature and time against the formation of biofilms by foodborne pathogens on surfaces of polyethylene and stainless steel. After preliminary experiments with 32 strains from 6 species of foodborne pathogens (Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Salmonella Typhimurium), one strain from each species with the highest biofilm formation efficiency was selected. All foodborne pathogens showed a tendency toward an increased ability for biofilm formation with increasing temperature, but there was no consistency between the two materials and between foodborne pathogens. At all tested temperatures, the biofilm formation ability of E. coli and P. aeruginosa on the polyethylene surface was higher than that on the stainless steel surface with significant differences. The foodborne pathogens all formed biofilms immediately upon inoculation, and biofilm formation by E. coli, P. aeruginosa, and S. Typhimurium increased on both the polyethylene and stainless steel surfaces at 1 h after inoculation compared to at 0 h. At 7 days after biofilm formation, the other strains except S. aureus showed no difference in survival rates on polyethylene and stainless steel. The ability of these 6 foodborne pathogens to form biofilms showed different trends depending on the type of bacteria and the instrument material, i.e., polyethylene and stainless steel.

Control of histamine-forming bacteria by probiotic lactic acid bacteria isolated from fish intestine (생선 내장으로부터 분리된 프로바이오틱 유산균에 의한 히스타민 생산균의 제어)

  • In this study, we examined in vitro the potential probiotic properties of lactic acid bacteria (LAB) obtained from the fish intestine and their ability to degrade histamine through the production of diamine oxidase (DAO) enzymes and bacteriocin. Among 97 LAB strains isolated from the intestine of croaker, flatfish, pollack, and rockfish, CIL08, CIL16, FIL20, FIL31, PIL45, PIL49, PIL52, and RIL60 isolates exhibited excellent survival rates under simulated gastrointestinal tract conditions, high adhesion ability to HT-29 epithelial cells, and resistance to the antibiotics such as amoxicillin, ampicillin, erythromycin, penicillin G, streptomycin, tetracycline, or vancomycin. In addition, these strains did not produce histamine in decarboxylating broth containing histidine. In particular, 4 strains (CIL08, FIL20, PIL52, and RIL60) that may produce DAO were significantly able to degrade histamine. The bacteriocins produced by FIL20, FIL31, and PIL52 LAB inhibited the growth and histamine production of Enterococcus aerogenes CIH05, Serratia marcescens CIH09, Enterococcus faecalis FIH11, Pediococcus halophilus FIH15, Lactobacillus sakei PIH16, Enterococcus faecium PIH19, Leuconostoc mesenteroides RIH25, or Aeromonas hydrophilia RIH28. Histamine-producing strains isolated from fish intestine were found to reduce histamine accumulation during co-culture with CIL08, FIL20, PIL52, and RIL60 LAB showing histamine degradation or bacteriocin production ability. The probiotic strains preventing histamine formation were identified as Pediococcus pentosaceus CIL08, Lactobacillus plantarum FIL20, Lactobacillus paracasei FIL31, Lactobacillus sakei PIL52, and Leuconostoc mesenteroides RIL60 with high similarity based on 16S rRNA gene sequencing.

Effects of Electrostimulation on In Vitro Development Ability of Single 4-cell Blastomeres and Oocyte Activation in Porcine (돼지에 있어서 4-세포기 분할구의 체외발생능과 난모세포의 활성화에 미치는 전기자극의 효과)

  • The objective of the present experiments were to determine whether micromanipulative and electro-stimulation conditions for blastomere survival overlapped those for oocyte activation in porcine. Eggs selected for in vitro development potential of blastomeres isolated from 4-cell embryos and oocyte activation by electrostimulation were equilibrated for 5~10 min, in 0.3M sucrose solution containing 7.5$\mu\textrm{g}$/ml cytochalasin B, and then electrostimulated for 30$\mu$sec using one pulse of 100, 120, 150 or 180 volts DC with electrodes 0.2mm apart. Single blastomeres were inserted into empty zona pellucida prior to electrostimulaticn. Then they were cultured in 20${mu}ell$ drops of fresh BECM to observe their developmental ability in vitro in a humidified incubat or at 38.5$^{\circ}C$. The results obtained from these experiments are as follows : 1. When one pulse of 100, 120, 150 or 180 volts DC for 30$\mu$sec were applied to porcine oocytes having the slit formed on zona pellucida for activation, activation rates were 65.1, 66.7, 70.7 and 91.7%, respectively. Higher activation rate was observed in 180V. 2. Infact oocytes incubated for 30 min, in 0.3M sucrose solution after electrostimulation were significantally different from control group with increasing of voltages(p<0.05). When voltages used for electrostimulation were increased, activation rates of oocytes were improved in all treatment groups. 3. When zona punctured-oocytes were only electrostimulated, or incubated in 0.3M sucrose solution for 30 min. after electrostimulation at 180 volt DC, activation rates were 90.5 and 95.5%, respectively. And activation rates of zona punctured-oocytes were significantly different from the groups for which zona pellucida was not punctured(P<0.05). 4. When single blastomeres form 4-cell transferred into empty zona pellucida were incubated for 0, 15 and 30 min. in 0.3M sucrose solution after electrostimulation using one pulse of 180 volt DC for 30 $\mu$sec, developmental rates of electrostimulated-single blastomeres to blastocyst were 72.5, 59.0 and 51.2%, respectively, and the ratio of control group developed to blastocyst were 80.0%. 5. The average cell number in electrostimulated-blastomeres developed to blastocyst were 7.9~10.8, and reduced than the cell number in diploid control ; Also cell number decreased with increasing of voltages. The results of these experiments indicate that the optimal condition for achieving in vitro developmental ability of single 4-cell blastomeres and oocyte activatin is 1 pulse, duration 30 $\mu$sec. in 180 volt, and incubation of blastomeres and oocytes in 0.3M sucrose solution after electrostimulation was not significantally different from another treatment groups. The results also show that this condition is suitable for nuclear transplantation using porcine eggs.

  • This paper suggested a plan to increase the start-up survival rate by identifying independent variables that significantly affect start-up success through analysis of the founder's internal and external factors. As a result of this study, it was found that internal factors (financial ability, risk sensitivity) and external factors (start-up environment, start-up support policy) had a direct positive (+) effect on start-up success. Meanwhile, self-efficacy for business models partially mediated the relationship between factors(financing ability, risk sensitivity, start-up environment and start-up support policies) and start-up success. Self-efficacy for business models fully mediated the relationship between factors(start-up expertise and challenge spirit) and start-up success. Finally, trust in business models partially mediated the relationship between factors(start-up expertise, risk sensitivity, and start-up support policies) and start-up success. And the trust in business models fully mediated the factors(the spirit of challenge, start-up environment) and start-up success.

Survival Ability of Porcine Oocytes Frozen-Thawed by Open Pulled Straw Method (Open Pulled Straw 방법에 의해 동결-융해된 돼지난자의 생존능력)

  • Vitrification of oocytes has been applied recently fur pigs, but remains elusive. The purpose of this study is to investigate the effects of vitrification in open pulled straws (OPS) on in vitro survival of porcine oocytes. When immature follicular oocytes frozen-thawed were cultured for in vitro maturation, maturation rates to metaphase-II stage were higher in oocytes with (25%) than without (15%) cumulus cells. After In vitro fertilization of oocytes frozen-thawed, the maturation rates were also significantly (P<0.05) higher in oocytes with (41%) that than without (17%) cumulus cells. However, the penetration rates were higher in oocytes without (19%) that than with (9%) cumulus. In another experiment, porcine oocytes matured in vitro were frozen and thawed for in vitro fertilization. The penetration rates were higher than in oocytes without (35%) that than with (26%) cumulus cells. However, the proportions of oocytes dead after in vitro fertilization were significantly (P<0.05) higher in oocytes with that than without cumulus cells. On the other hand, the rates of penetration and dead oocytes at 6 h after in vitro fertilization were not significant differences between oocytes with and without cumulus cells. However, the proportions of dead oocytes with (18%) and without (16%) cumulus cells were higher than in oocytes of control group (0%). These finding indicated the possible broader application for OPS, as they demonstrated that the maturation and fertilization in vitro by frozen-thawed oocytes may be accompained by cumulus cells and culture periods according to the requirements of the survival ability after freezing of mature and immature oocytes in pigs.
