In this study, we characterized the physical form of allergenic Cry j 1 in the urban atmosphere. Through an immunofluorescence antibody method, we showed that allergenic Cry j 1 exists as fine particles (${\leq}1.1{\mu}m$). To determine Cry j 1 concentrations and its particle size distribution, we used the ELISA method to confirm that most Cry j 1 exists as fine particles in the urban atmosphere and is found at high concentrations on fine day next to rainy day. Furthermore, we evaluated Cry j 1 denaturation by using the Biacore J system based on the surface plasmon resonence (SPR) principle and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). We showed that the dissociation constant ($K_D$) of Cry j 1 that has been exposed to urban polluted air is lower ($1.76{\times}10^{-14}$ M) than that of Cry j 1 ($1.32{\times}10^{-9}-3.37{\times}10^{-9}$ M) of original pollen grains that has not been exposed to air pollutants. Cry j 1 turns into low molecular weight proteins by reacting with various acidic solutions. In sum, we showed that allergenic Cry j 1 exists as fine particles that can deposit in the lower respiratory tract. This finding clarifies the relationship between Japanese cedar pollinosis and air pollutants.
Label-free optical methods for the monitoring of interactions between biological molecules have become increasingly popular within the last decade. A rising number of publications have demonstrated the benefits of direct biomolecular interaction analysis(BIA) for biology and biochemistry, such as antigen-antibody Interactions, receptor-ligand interactions, protein-DNA, DNA- intercalator, and DNA-DNA interactions. This article gives an overview of the historical development, principle and application of label-free optical biosensor to examine the functional characteristics of biospecific interaction, such as kinetics, affinity, and binding position of biomolecular between an immobilized species at the transducer surface and its dissolved binding partner.
In this study, a simple procedure is described for patterning biotin on a glass substrate and then selectively immobilizing proteins of interest onto the biotin-patterned surface. Microcontact printing (CP) was used to generate the micropattern of biotin and to demonstrate the selective immobilization of proteins by using enhanced green fluorescent protein (EGFP) as a model protein, of which the C-terminus was fused to a core streptavidin (cSA) gene of Streptomyces avidinii. Confocal fluorescence microscopy was used to visualize the pattern of the immobilized protein (EGFP-cSA), and surface plasmon resonance was used to characterize biological activity of the immobilized EGFP-cSA. The results suggest that this strategy, which consists of a combination of $\mu$CP and cSA-fused proteins. is an effective way for fabricating biologically active substrates that are suitable for a wide variety of applications. one such being the use in protein-protein assays.
A bimetallic chip made of gold and silver was investigated in intensity interrogation mode to confirm enhancement of the SPR sensor resolution. Both reflectance curves of the bimetallic chip and the conventional gold chip was acquired and compared. The line width of the reflectance curve of the bimetallic chip was narrower than that of the conventional Au chip, resulting in steeper tangential slope. The reflectance was monitored at the angle related to the steepest tangential slope. The change in reflectance of the bimetallic chip was larger than that of the Au chip. The critical angle was analyzed by differentiating the reflectance with respect to incident angle twice. Acquiring the critical angle regarding to the sample informs the refractive index of the sample. Using various concentration of Bovine Serum Albumin, we confirmed that refractive index was linearly related to variation of reflectance of the bimetallic chip.
본 논문에서는 금속의 표면 플라즈몬 공명으로 인한 금속-유전체 경계면에서의 국소적 전자기장의 강화 효과를 이용하여 표면 플라즈몬을 유발하는 금 박막을 유리 기판위에 증착하고, 프리즘 커플러를 이용한 소산장의 공명 흡수현상을 이차원 영상 으로 얻었다. 특히 DNA/단백질 칩 등 향후 가능한 다채널 시스템에의 응용을 고려하여11-MUA, 11-MUOH 등 자기조립 단분자막(SAM)을 크롬 마스크와 리토그래피, 그리고 Shadow mask와 광 산화반응을 이용하여 금 표면 위에 패터닝 하였다. 텅스텐-할로겐 램프와 중심파장이 ${\lambda}_0=633$ nm의 대역통과 필터를 사용하여 이 평행광을 프리즘 커플러에 입사시켜 반사되어 나오는 반사광의 이차원 영상을 얻었다. 이와는 별도로 ${\lambda}_0=633$ nm의 레이저를 이용하여 단분자막이 코팅되어 있을 때와 없을 때의 공명각의 변화를 관찰하였다. 얻어진 이차원 영상의 위치에 따른 화소 값의 변화를 단분자 막의 두께의 변화에 따라 보정하고, 알려진 매질의 SPR 특성을 Fresnel 방정식에 따라 이론적으로 계산하면 다채널 표면 영상으로부터 항원-항체 등 단백질의 결합 정도를 정량적으로 측정할 수 있다.
Surface Plasmon Resonance(SPR) sensor system can be applicable for detecting of many biospecific interactions. In this study, the feasibility of the experimental $SPREETA^{TM}$ evaluation kit to analyze human IgG, Pseudomonas aeruginosa, was investigated. The sensor prepared for detecting of anti-human IgG has response on $0.1{\mu}{\ell}$ of the anti-human IgG solution. SPREETA was able to detect P. aeruginosa solution in the range above $10^8\;CFU/mL$ with the chitosan/ alginate multilayers.

최근 의료, 보건, 헬스케어 분야에 대한 관심이 증가함에 따라 질병의 조기 진단 연구가 각광 받고 있다. 특히 표면증강 라만 산란 (Surface Enhancement Raman scattering)은 고분자 검출을 위해 가장 유용한 물리 화학적 기법으로 SERS를 활용한 특정물질 검출 기술 개발에 대한 연구가 많이 이루어지고 있다. 나노구조의 국부적 표면 플라즈몬의 공명조건 (Surface Plasmon Resonance, SPR)으로 유도된 전자기장은 우수한 SERS 신호를 나타낸다. 따라서 표면 플라즈몬 공명 효과는 귀금속 나노입자의 종류, 크기 및 형태, 기판의 형상 및 구조 등에 의해서 달라지게 되므로 이들을 조절하여 보다 민감한 SERS 신호를 얻을 수 있다. 본 연구에서는 고감도 SERS-Active 기판을 제작하기 위해 SERS 기판 표면의 나노구조를 최적화 하였다. SERS 기판 표면을 제어하기 위해 공정파워, 공정압력, 기판의 온도 등의 증착공정 변수에 변화를 주어 표면의 나노구조를 형성하였다. 이를 분석하기 위해 SEM 분석을 통해 피라미드형 실리콘 기판 표면의 Au 나노구조 금속 박막을 확인하였고, XRD를 이용하여 결정성 및 결정크기를 확인하였다. Rhodamine 6G를 이용한 라만 분석을 통해 SERS 신호의 강도를 알 수 있었다. 금속 나노구조의 형태, 온도 제어를 통해 SERS 신호강도가 우수한 나노구조 기판을 제조 할 수 있었다.
Cryptomeria japonica pollen is the most common pollen, which are scattering during each spring season in Japan. Japanese cedar (Cryptomeria japonica) pollinosis is one of seasonal allergic rhinitis that mainly occurs in Japan. In addition, long range transportation of Yellow Sand from the East Asian continent was also found during the pollen scattering seasons in Japan. Therefore, the interaction or impact between pollen and Yellow Sand should be concerned. In this study, our objective was to investigate the airborne behaviour of Cryptomeria japonica pollen grains and its size-segregated allergenic (Cry j 1) particles as the airborne tracer of Cryptomeria japonica pollen during the Yellow Sand events. Airborne Cryptomeria japonica pollen grains and its size-segregated allergenic particles were collected at roadside of urban residential zones of Saitama city during the pollination periods from February to March in two year investigation of 2009 and 2010. The overlap of Yellow Sand events and dispersal peak of pollen grains was observed. According to the Meteorological data, we found that the peaks of airborne pollen grains appeared under higher wind speed and temperature than the previous day. It was thought that Yellow Sand events and airborne pollen counts were related to wind speed. From the investigation of the airborne behavior of the size-segregated allergen particles by determining Cry j 1 with Surface Plasmon Resonance (SPR), the higher concentrations of the allergenic Cry j 1 were detected in particle size equal to or less than $1.1{\mu}m$($PM_{1.1}$) than other particle sizes during Yellow Sand events, especially in the rainy day. We conclude that rainwater trapping Yellow Sand is one of the important factors that affect the release of allergenic pollen species of Cry j 1. Therefore, it is very important to clarify the relationships between Cryptomeria japonica pollen allergenic species and chemical contents of the Yellow Sand particles in further studies.
본 연구에서는 7 개의 아미노산으로 이루어진 헵타펩타이드의 Lgr5 binding에 따른 인체 모낭 구성 세포의 활성에 대한 영향을 확인하였다. 표면 플라즈몬 공명(surface plasmon resonance, SPR) 시스템을 이용하여 헵타펩타이드가 Lgr5에 결합하는 것을 확인하였다. 인체 모유두세포(human hair follicle dermal papilla cell, HHFDPC)에 헵타펩타이드를 처리한 결과, 농도 의존적인 세포 증식이 나타났으며 β-catenin의 세포 내핵 이동 및 하위 유전자인 LEF1, Cyclin-D1, c-Myc의 발현 증가가 관찰되었다. 그리고 세포 증식 기전 관련 인자인 Akt와 ERK의 인산화 수준이 증가되었으며, 성장인자인 hepatocyte growth factor (HGF), keratinocyte growth factor (KGF), vascular endothelial growth factor (VEGF) 발현이 유도되었다. 또한 인체 모모세포(human hair germinal matrix cell, HHGMC)의 분화 관련 전사 인자와 인체 외모근초세포(human hair outer root sheath cell, HHORSC)의 분화 표지 인자들도 헵타펩타이드 처리 시 높은 발현율을 보였다. 추가적으로 우리는 헵타펩타이드의 인체 모낭줄기세포(human hair follicle stem cell, HHFSC) 분화에 대한 영향을 조사하였다. 그 결과, HHFSC 표지인자들의 mRNA와 단백질 수준이 감소하였고 반면에 분화 표지인자들은 증가하였다. 상기의 결과들은 헵타펩타이드가 인체 모낭 구성 세포에서 Wnt/β-catenin 경로를 촉진시켜 증식 또는 분화를 유도할 수 있음을 보여준다. 이를 토대로 종합해 볼 때, 본 연구의 헵타펩타이드는 모발 성장을 유도하고 탈모 개선에 도움을 줄 수 있는 기능성 원료로 사용될 수 있을 것으로 보인다.
