Relationship Among Body Mass Index, Nutrient Intake and Antioxidant Enzyme Activity of Postmenopausal Women

  • To elucidate the relationship among body mass index, nutrient intake and blood antioxidant capacity in the postmenopausal period, 60 women residing in Iksan area were recruited. Body mass index (BMI) was calculated base on height and weight, and food and nutrient intakes were estimated by 24-hour recalls of 3 non-consecutive days. Parameters of antioxidant capacity including the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) and total antioxidant capacity (TA) were measured in fasting blood samples from the subjects. The average age, height, weight and BMI of the subjects were 65 years, 151.1cm, 59.5 kg and 26.0 m/kg$^2$, respectively. The macronutrient intake rate of carbohydrate : protein : fat were 65:17.5:17.5; the mean intakes of energy and protein were 1532.7 kcal (86.3% of RDA) and 67.1 g (122.0% of RDA) respectively. The mean intakes of phosphorus, vitamin A, niacin and vitamin C were higher than Recommended Daily Allowance (RDA) for Koreans. On the other hand, calcium and riboflavin intakes were only 84.6% and 70.4% of RDA. Among the parameters of antioxidant capacity, SOD activity was significantly lower in lean subjects (BMI<20) than in the normal or overweight subjects (BMI${\geq}$20) (p<0.05). TAs of the subjects with the highest intakes of vegetables and fruits were significantly higher than those of subjects with lower intakes (p<0.05). Antioxidant capacity was compared among subjects according to 3 different nutrient intake levels according percentage of RDA for Koreans for selected nutrients with the following results: The high protein and niacin groups exhibited significantly lower TA status than those of the other intake groups (p<0.05). In conclusion, the low BMI was associated with lower SOD activity in postmenopausal women. Higher consumption of fruits and vegetables was associated with higher TA. When protein and niacin intakes were excessive, SOD activity and TA tended to be low. SOD and TA, among antioxidant indexes, seemed to be mostly influenced by other factors. Therefore, more studies on the effects of nutritional intake and the activity of antioxidant enzyme should be conducted.

추출용매에 따른 산초 (Zanthoxylum schnifolium)열매 추출물의 라디칼소거능과 항염증 효과 (Radical Scavenging and Anti-inflammation Activities from Different Extracts of Zanthoxylum schnifolium Fruits)

  • This study was presented to the physiological activities such as metal chelating capacity, superoxide dismutase-like activity, anti-inflammation, cytotoxicity from Zanthoxylum schnifolium fruit. The values for metal chelating capacity of 1 mg/mL of 70% EtOH and 70% MeOH extracts were 68% and 67%, respectively, it was shown that metal chelating capacity in a dose-dependent manner. The 100% EtOH and 100% MeOH extract exhibited strong superoxide dismutase-like activity. The highest SOD-like activity obtained from 100% EtOH was 51% at a concentration of 4 mg/mL. The all extracts of Zanthoxylum schnifolium fruit except $H_2O$ extract significantly decreased the concentration of LPS-induced NO in Raw 264.7 cells. The 100% EtOH, 100% MeOH and 70% EtOH extract represented the highest activity in the anti-inflammation properties in vitro. However, the 100% EtOH and 100% MeOH extracts has cytotoxicity in Raw 264.7 cells and it may affect cell viability. Conclude that 70% EtOH extract was most suitable for anti-inflammation. Our results revealed that the Zanthoxylum schnifolium fruit expected to physiological activities.

Effect of Red Pepper Seed on Kimchi Antioxidant Activity During Fermentation

  • In this study, the antioxidant activities of red pepper seed kimchi extracts were examined. The extracts were evaluated by various antioxidant assays that included determinations of total phenolic and flavonoid contents, DPPH radical scavenging, superoxide anion radical scavenging, nitric oxide scavenging, superoxide dismutase (SOD) activity, metal chelating activity, and reducing power. All the kimchi extracts showed strong antioxidant activities by the tested methods. The 7% red pepper seed kimchi that was fermented for 6 days possessed the highest activity according to the performed tests. Yet, the highest scavenging activity was exhibited against nitrite, by extracts from kimchi for 0 day of fermentation rather than 6 days. All the activities for the kimchi fermented for 0 day with the addition of 7% red pepper seed increased markedly with an increase in concentration. With the exception of metal chelating and SOD activities, for the antioxidant properties assayed, the red pepper seed kimchi extracts were found to be highly effective.

  • Antioxidant enzymes such as catalase (CAT), glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) are known to inhibit oxidative reactions by incativating compounds responsible for the formation of ree radicals. SOD transforms superoxide radical into hydrogen peroxide which is precursor to active free radicals. CAT reduces hydrogen peroxide to water. GSH-Px reduces hydroperoxides to corresponding alcohols. Antioxidant enzyme activities of muscle are different by animal species age, stress and exercise, muscle type and part, conditions of post mortem, storage and processing which are related to oxidative deterioration I muscle foods as well as oxidative defence in living systems. Antioxidant enzyme systems are enhanced rather than weakened in aging skeletal muscle. Red muscle contains higher antioxidant enzyme activity than white muscle. The antioxidant enzyme activities of poultry are higher in leg than in breast, and those of beef are higher in redder and more unstable muscles. It is clear that the effectiveness of the antioxidant enzyme in muscle foods seems to be influenced by meat processing operations. Both GSH-Px and CAT are inactivated by heat processing NaCl also influence the efficiency of the antioxident enzymes since its presence diminishes their catalyitc activity.

매자나무 Ethyl acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과 (Antioxidant Activity of Ethyl acetate Fraction of Berberis koreana Palibin in Caenorhabditis elegans)

  • Ethyl acetate (EA) soluble fraction of the Berberis amurensis (Berberidaceae) methanol extract showed the potent DPPH radical scavenging activity through Caenorhabditis elegans model system. The EA fraction was measured for the activity of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species (ROS) level. In addition, SOD-3 expression was conducted using a transgenic strain (CF1553) to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the EA fraction. As a result, the EA soluble fraction of B. amurensis increased SOD and catalase activity, and decreased ROS accumulation in a dose-dependent manner. Furthermore, the EA fraction-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

Effects of zinc bearing palygorskite supplementation on the growth performance, hepatic mineral content, and antioxidant status of broilers at early age

  • Objective: This study was conducted to investigate effects of zinc (Zn) bearing palygorskite (ZnPal) supplementation on growth performance, hepatic mineral content, and antioxidant status of broilers at early age. Methods: A total of 240 1-day-old Arbor Acres broiler chicks were allocated into 5 treatments with 6 replicates of 8 chicks each. Birds in 5 treatments were fed a basal diet supplemented with 0 (Control group; Analyzed Zn content: 81 mg/kg), 20, 40, 60, and 80 mg/kg Zn as ZnPal for 21 days, respectively. Blood, liver and intestinal mucosa were collected at 21 days of age. Results: Treatments did not affect growth performance of broilers during the 21-day study (p>0.05). The contents of hepatic Zn and magnesium (Mg) were linearly increased (p<0.001) by ZnPal supplementation. ZnPal inclusion linearly (p = 0.007) reduced malondialdehyde (MDA) concentration in serum. The activity of total superoxide dismutase (T-SOD) in liver increased linearly (p = 0.001) with concentration of ZnPal in diet. ZnPal inclusion linearly (p = 0.036) and quadratically (p = 0.005) increased T-SOD activity, and linearly (p = 0.012) increased copper/zinc superoxide dismutase (Cu/Zn SOD) activity in jejunal mucosa. The maximum responses of hepatic and jejunal antioxidant enzymes activities (T-SOD and Cu/Zn SOD) were found when supplementing the basal diet with 60 mg/kg Zn as ZnPal. Furthermore, ZnPal supplementation quadratically (p = 0.001) increased Cu/Zn SOD activity in ileal mucosa, and its maximum activity was observed in the diet supplemented with 20 mg/kg Zn as ZnPal. Conclusion: ZnPal supplementation did not alter growth performance of broilers. Dietary ZnPal inclusion could increase concentrations of hepatic trace minerals (Zn and Mg) and inhibit lipid peroxidation by reducing serum MDA accumulation, with the optimal dosage of Zn from ZnPal being 80 mg/kg diet (analyzed Zn content in the diet: 165 mg/kg), and 60 mg/kg Zn as ZnPal (analyzed Zn content in the diet: 148 mg/kg) was the optimum dosage for broilers to achieve maximum antioxidant enzyme activities.

항산화 활성과 Melanoma 세포에서 멜라닌조절에 대한 Oenanthe javanica 에탄올 추출액의 효과 (Effect of Oenanthe javanica Ethanolic Extracts on Antioxidant Activity and Melanogenesis in Melanoma Cells)

  • 본 연구의 목적은 melanocyte (B16F1)에서 quercetin과 kaempferol을 포함하는 미나리 에탄올 추출물(OJE)의 멜라닌 합성효과에 미치는 영향을 조사한 것이다. OJE가 세포수준에서 멜라닌 합성을 억제하는지를 조사하기 위하여 여러 농도의 OJE 존재 하에서 B16F1세포를 배양하였다. 현재 연구에서 DPPH radical scavenging, reducing power, lipid peroxidation 및 DNA oxidation에 미치는 항산화 효과는 cell free system에서 평가되었다. 더욱이 멜라닌 생성에 대한 OJE 효과는 dopaquinone (DOPA) assay 및 tyrosinase 활성으로 결정되었다. 뿐만 아니라 superoxide dismutase (SOD)-1, -2, glutathione reductase (GSH)와 같은 항산화 효소 및 tyrosinase의 단백질발현이 western blot 분석을 이용하여 평가되었다. 본 연구에서 OJE는 지질과산화 억제효과를 나타내었고 fenton 반응에 의해서 생성되는 hydroxyl radical에 의하여 유발되는 DNA 산화를 보호하였다. OJE는 50 ${\mu}g/ml$ 이상에서 멜라닌 합성을 증가시켰고 tyrosinase 활성도 50 ${\mu}g/ml$에서 검출되었다. Western blot 분석에서는 OJE가 농도에 비례하여 tyrosinase SOD-1, -2 및 GSH의 발현 수준을 증가시켰다. 이러한 발견들은 항산화 효과를 가진 OJE가 melanocyte에서 tyrosinase 활성과 melanin 생성을 조절할 수 있어 피부를 산화스트레스로부터 보호할 수 있다는 것을 암시하고 있다.

Ellagic acid plays a protective role against UV-B-induced oxidative stress by up-regulating antioxidant components in human dermal fibroblasts

  • Ellagic acid (EA), an antioxidant polyphenolic constituent of plant origin, has been reported to possess diverse pharmacological properties, including anti-inflammatory, anti-tumor and immunomodulatory activities. This work aimed to clarify the skin anti-photoaging properties of EA in human dermal fibroblasts. The skin anti-photoaging activity was evaluated by analyzing the reactive oxygen species (ROS), matrix metalloproteinase-2 (MMP-2), total glutathione (GSH) and superoxide dismutase (SOD) activity levels as well as cell viability in dermal fibroblasts under UV-B irradiation. When fibroblasts were exposed to EA prior to UV-B irradiation, EA suppressed UV-B-induced ROS and proMMP-2 elevation. However, EA restored total GSH and SOD activity levels diminished in fibroblasts under UV-B irradiation. EA had an up-regulating activity on the UV-B-reduced Nrf2 levels in fibroblasts. EA, at the concentrations used, was unable to interfere with cell viabilities in both non-irradiated and irradiated fibroblasts. In human dermal fibroblasts, EA plays a defensive role against UV-B-induced oxidative stress possibly through an Nrf2-dependent pathway, indicating that this compound has potential skin antiphotoaging properties.

Antioxidant Activities of Mulberry (Morus alba L.) Leaf Extracted with Different Concentrations of EtOH

  • Antioxidant activities of mulberry leaf extracted with different concentrations of EtOH were investigated. Total phenolic content and electron donating abilities of extract from 70% EtOH were the highest. Extracts obtained from EtOH-water mixture were shown to be significantly higher superoxide dismutase (SOD)-like activities than other treatment (p<0.05). Angiotensin-converting enzyme (ACE) inhibition was the greatest at 50% EtOH concentration (p<0.05). The extracts from 30-70% EtOH exhibited higher ferric reducing ability of plasma (FRAP) value than rest of the concentration (p<0.05). In case of nitrite scavenging activity, much higher scavenging activities were observed when the extraction was performed with EtOH or EtOH-water mixture (p<0.05). The results indicate that concentration of EtOH as extraction solvents can affect the antioxidant activity of mulberry leaf, which may provide useful information on the optimal solvent conditions for the extraction.

항산화 효소의 산화적 변형에 뽕잎 발효물이 미치는 영향 (Effects of Fermented Mulberry Leaves (Morus alba L.) on Oxidative Modification of Antioxidnat Enzymes)

  • 뽕잎 (Morus alba L.)을 노루궁뎅이 버섯균사체(Hericium erinaceum)로 발효시켜 열수 추출한 발효물(MA-HE)이 항산화 효소인 Cu,Zn-superoxide dismutase (SOD)와 ceruloplasmin (CP)의 산화적 변형에 미치는 영향을 관찰하였다. 세포질에서 중요한 항산화 활성을 나타내는 Cu,Zn-SOD와 CP는 모두 구리이온을 포함하는 단백질이다. MA-HE은 peroxyl radical에 의해 변형되는 Cu,Zn-SOD와 CP를 모두 보호하였다. MA-HE은 peroxyl radical에 의한 Cu,Zn-SOD의 단백질 fragmentation과 효소의 inactivation을 모두 효과적으로 억제하였다. 또한, MA-HE는 CP의 fragmentation과 inactivation도 효과적으로 억제하였다. MA-HE가 peroxyl radical을 소거할 수 있는 기능을 가지고 있는 지를 확인하기 위해 thiobarbituric acid를 이용한 활성 측정법으로 관찰한 결과, MA-HE는 농도가 증가함에 따라 비례적으로 peroxyl radical 소거 활성이 높아지는 것으로 관찰되었다. MA-HE는 $100{\mu}g/mL$에서 44.03% 소거활성을 나타냈다. 따라서, MA-HE는 peroxyl radical을 효과적으로 소거함으로서 세포 내의 항산화 효소를 산화적 손상으로부터 보호할 것으로 생각된다. 이와 같은 결과는 MA-HE가 세포 내에서 발생하는 활성산소종을 효과적으로 소거하여 산화적 스트레스에 의해 야기되는 세포 독성에 대한 보호 작용을 할 것으로 생각된다.
