자가면역 질환과 HERV의 병인학적 관계 (The Etiological Relationship between Autoimmune Diseases and HERVs)

  • 인간의 면역계는 외부의 병원균 등의 침입에 대하여 매우 강력한 방어작용을 수행하고 있다. 하지만, 때로 면역계의 손상으로 인해 비정상적인 면역작용이 유발됨으로써 면역성 질환이 야기되기도 한다. 특히, 자가면역 질환은 그 종류가 매우 다양하며 심각한 위험을 초래하기도 한다 이들 질환에 대한 활발한 병인학적 연구가 진행되고 있으나, 아직 정확한 메커니즘을 밝히지는 못하고 있다. 자가면역성 당뇨병의 원인 유전자로 인간의 게놈에 내생하고 있는 HERV가 거론된 후, 사이토카인의 변화나 외생 레트로바기러스의 감염에 의해 HERV의 발현이 활성화되어 superantigen으로 작용함으로써 비정상적인 면역반응이 유도된다는 새로운 사실이 밝혀져 많은 주목을 끌고 있다. 이는 내생 레트로바이러스 유래의 superantigen과 인간의 질병과의 관련성을 비교적명확하게 제시함으로써, 자가면역 질환의 극복을 위한 향후 연구에 주요한 실마리를 제공하고 있다.

  • Background: Rheumatoid arthritis is an autoimmune disease characterized by a chronic inflammatory process, primarily involving the synovial membrane of peripheral j oints, where T cell activation is found. To address the superantigen stimulation in rheumatoid arthritis, T cell clonality and the expression of activation markers were analyzed. Methods: To detect TCRB V usage, inverse PCR and sequencing were done. Monoclonal antibodies were used for flow cytometric analysis of TCRBV8 or TCRBV5. As results, a restricted usage of TCRBV3 gene was detected in synovial lymphocytes from one rheumatoid arthritis patient. However, preferential usage for TCRB V8, which may be one indicator for stimulation by staphylococcal superantigen, was not obvious although general activation of T cells was found as high DR+ percentage in synovial T cells. These data show specific antigen rather than superantigen might involve the pathogenesis of rheumatoid arthritis.

식육유래의 Staphylococcus aureus으로부터 산생되는 staphylococcal superantigens의 유전자형 분석 (Gene typing of staphylococcal superantigens produced by Staphylococcus aureus isolates from meat)

  • Using the previous established multiplex PCR (mPCR), the presence of six kinds of staphylococcal superantigen (SAg) genes was investigated for nineteen Staphylococcus aureus isolates from the commercialized meat sources. As a result, only one isolate from pork among 19 S aureus isolates (5.3%) was confirmed as a potential SAg producer and harbored sec gene. The results in this study suggest that meat may not be major contagion of staphylococcal food poisoning (SFP) in Korea and that staphylococcal enterotoxin type C may be associated with the disease. Also, the mPCR method in this study can be a useful genotypic method which can overcome the typical disadvantages of conventional antibody-based methods due to antigenic homology, and furthur survey on food-borne S aureus isolates can provide the important epidemiological data for SFP in Korea.

아토피 피부염에서 황색포도상구균 분포에 관한 연구 (A Study on the Distribution of Staphylococcus aureus in Atopic Dermatitis)

  • 황색포도상구균이 정상인 피부의 경우 $2{\sim}25%$에서 검출되는 반면 아토피 피부염이 있는 소아와 어른에 있어 $78{\sim}100%$의 높은 비율로 검출되며, 황색포도상구균과 초항원으로 작용하는 독소가 아토피 피부염의 악화 및 발병기간에 중요한 역할을 하는 것으로 알려져 있다. 본 연구는 아토피 피부염 환자 피부의 황색포도상구균의 분석과 함께 아토피 피부염 환자의 연령, 성별, 발병 연령과 투병기간, 악화되는 계절 및 진물과 황색포도상구균과의 관계에 대하여 알아보고자 실시하였다. 아토피 피부염 환자 대부분(84%)에서 병변부에 S. aureus가 집락을 형성하고 있는 것으로 나타났으며, 대부분의 아토피 피부염 환자의 피부에 S. aureus가 분포하고 있음을 알 수 있었고 아토피 피부염 병변부의 진물과 S. aureus의 밀접한 관계를 확인할 수 있었다. 따라서 아토피 피부염 환자의 피부에서 황색포도상구균 및 초항원독소를 차단한다면 아토피 피부염의 개선에 많은 도움이 될 것으로 사료된다.

Regulatory T Cell Therapy for Autoimmune Disease

  • It has now been well documented in a variety of models that T regulatory T cells (Treg cells) play a pivotal role in the maintenance of self-tolerance, T cell homeostasis, tumor, allergy, autoimmunity, allograft transplantation and control of microbial infection. Recently, Treg cell are isolated and can be expanded in vitro and in vivo, and their role is the subject of intensive investigation, particularly on the possible Treg cell therapy for various immune-mediated diseases. A growing body of evidence has demonstrated that Treg cells can prevent or even cure a wide range of diseases, including tumor, allergic and autoimmune diseases, transplant rejection, graft-versus-host disease. Currently, a large body of data in the literature has been emerging and provided evidence that clear understanding of Treg cell work will present definite opportunities for successful Treg cell immunotherapy for the treatment of a broad spectrum of diseases. In this Review, I briefly discuss the biology of Treg cells, and summarize efforts to exploit Treg cell therapy for autoimmune diseases. This article also explores recent observations on pharmaceutical agents that abrogate or enhance the function of Treg cells for manipulation of Treg cells for therapeutic purpose.

Construction of Novel Bifunctional Chimeric Proteins Possessing Antitumor and Thrombolytic Activities

  • Based on their respective antitumor and thrombolytic activities, the superantigen staphylococcal enterotoxin C2 (SEC2) and staphylokinase (Sak) were chosen for the construction of the novel chimeric proteins Sak-linker-SEC2 and SEC2-linker-Sak using a linker composed of nine Ala residues. Both chimeric proteins possessed nearly the same PBMC proliferation stimulating activity and antitumor activity as SEC2 and thrombolytic activity as Sak. Neither the SEC2 or Sak component of each chimeric protein affected the activity of the other component. The results presented in this study provide a possible strategy to prevent and cure tumor thrombus.

  • 우리나라에서는 아직 마이코플라스마 폐렴이 가와사키병에 병발한 경우가 보고된 적이 없으며 마이코플라스마 폐렴의 특성상 다양한 호흡기 외의 증상이 비교적 흔하게 동반되는 점 등을 고려할 때, 적절한 치료에도 불구하고 지속되는 발열을 보이는 마이코플라스마 폐렴의 경우 반드시 비전형적 가와사키병이 감별되어야 할 것이며, 이미 언급한 바와 같이 초항원(superantigen)에 근거해 가와사키병의 병태생리를 규명하고자 하는 연구들을 지지할 수 있는 또 하나의 근거로 생각되어 본 증례를 보고하는 바이다.

포도상구균에서 분비하는 장내독소 B(SEB)에 대한 재조합 단백질을 이용한 면역특이적 난황항체 생산 (Production of Immunospecific Egg Yolk Antibody with Recombinant Staphylococcal Enterotoxin B (SEB) Protein)

  • 본 연구는 식중독은 물론, 아토피성 피부염의 원인물질로 알려져 있는 포도상구균 장내독소 B(Staphylococcal enterotoxin B; SEB)에 대한 특이 난황항체를 개발하고자 실험을 실시하였다. 우선, SEB 유전자를 클로닝한 다음, 대장균 발현시스템을 이용하여 약 30 kDa 정도의 재조합 SEB 단백질을 생산하였다. 재조합 SEB 단백질을 산란계에 2주 간격으로 3회 면역접종을 실시하고, 혈청 및 난황 내 항체가를 측정한 결과, 면역 후 4주경에 항체가가 최고치에 달하였으며, 산란계로부터 획득한 난황항체를 이용한 Western blot 결과, 재조합 SEB 단백질은 물론, 상용화 SEB 단백질과도 특이적으로 반응한다는 것을 규명하였다. 결론적으로, 식중독과 아토피성 피부염 등의 원인물질로 알려진 SEB에 특이적인 난황항체를 생산에 성공하였으며, 이러한 특이적인 난황항체는 식중독 및 아토피성 피부염의 예방 및 치료에 활용 가능할 것으로 사료된다.

감마선 조사가 Staphylococcal Enterotoxin B의 비장세포 증식률 및 Interleukin-2 분비능에 미치는 영향 (Effect of Gamma-Irradiation on the Cell Proliferating and Interleukin-2 Producing Activity of Mouse Splenocytes of Staphylococcal Enterotoxin B)

  • The purpose of this study was to investigate the cell proliferating and interleukin-2 producing activity of staphylococcal enterotoxin B by gamma-irradiation. Staphylococcal enterotoxin B was gamma-irradiated with the various doses of 0, 2, 20 and 50 kGy. SDS-PAGE analysis showed that gamma-irradiation caused the sharp decrease of the content of staphylococcal enterotoxin B and the effect was irradiating dose-dependent. Non-irradiated staphylococcal enterotoxin B increased the cell proliferation of splenocytes isolated from female Balb/c mouse, whereas 2 kGy-irradiated toxin significantly decreased the activity. 20 and 50 kGy-irradiated staphylococcal enterotoxin B was no effect. A similar effect on the interleukin-2 production of mouse splenocytes was observed with non-irradiated and irradiated staphylococcal enterotoxin B. It was considered to be due to the decrease of the antigenicity of staphylococcal enterotoxin B by gamma-irradiation. Therefore, these results suggest that gamma-irradiation can be effective for the decrease of the antigenicity of staphylococcal enterotoxin B as superantigen.

Association between specific IgE to staphylococcal enterotoxin B and the eosinophilic phenotype of asthma

  • Background/Aims: Sensitization to staphylococcal superantigens (SAgs) could contribute to asthma severity. However, its relevance with eosinophilic phenotype has not yet been clarified. This study aimed to investigate associations between serum specific IgE levels to SAg and eosinophilic airway inflammation in adult asthmatics. Methods: The serum specific IgE levels to 3 SAgs, including staphylococcal enterotoxin A (SEA) and B (SEB), and toxic shock syndrome toxin-1 (TSST-1) were measured by ImmunoCAP in 230 adult asthmatic patients and 50 healthy controls (HCs). Clinical characteristics and laboratory parameters, including serum total/free IgE, and 2 eosinophil-activation markers, eosinophil cationic protein (ECP), and eosinophil-derived neurotoxin (EDN), were analyzed according to blood eosinophil counts (BEC; 150 cells/µL) and serum specific IgE levels to 3 SAgs (0.35 kU/L). Results: Asthmatic patients showed higher serum specific IgE levels to 3 SAgs than HCs (p < 0.05 for all). The serum total/free IgE levels were significantly higher in asthmatics with positive IgE responses to 3 SAgs than those without (p < 0.05 for all). There were no significant differences in clinical parameters including age, asthma severity, comorbidities, or smoking according to IgE responses to 3 SAgs. Patients with positive IgE responses to SEB (not to SEA/TSST-1) had higher serum specific IgE levels to house dust mites and ECP/EDN as well as higher BEC with positive correlations between serum SEB-specific IgE levels and BEC/ECP/EDN (p < 0.05 for all). Conclusions: These findings suggest that serum SEB-specific IgE levels could contribute to eosinophil activation as well as IgE production in adult asthma.
