Inhibition of Lipoxygenase Activity by the Extract of Various Processed Garlic - Inhibitory Effect of Garlic Extracts on Soybean Lipoxygenase Activity - (마늘의 가공 조리방법에 따른 Lipoxygenase활성도 저해효과 -마늘 추출액이 Lipoxygenase 활성도 저해에 미치는 영향-)

  • The bioactivity of garlic extract was evaluated, based on the inhibition of soybean lipoxygenase. While the inhibition of lipoxygenase by the chloroform extract (1$_{50}$ value after 10min precirculation, 55mg/$m\ell$) of garlic homogenate shows the property as irreversible inhibitors, the aqueous extract (1$_{50}$ value, 65mg/$m\ell$) appeared to contain mainly reversible inhibitors. In the related study, diallyldislufide and dimethyldisulfide inhibited the enzyme with 1$_{50}$ value of 1.3mM and 18mM, respectively. These disulfides demonstrated both irreversible and reversible patterns of inhibition. In addition, synthetic alliin(allylcysteine sulfoxide) was found to inhibit the enzyme at high concentration (approximately 22%, at 10mM), and its decomposition products showed the irreversible property in the inhibition, in contrast to S-ethyl cysteine sulfoxide which expressed no significant inhibition. Thus, it is suggested that the garlic macerate contains both irreversible and reversible sulfur inhibitors.itors.

Study of Effective Cryoprotectants on the Cryopreservation of Porcine Mesenechymal Stem Cells (돼지 중간엽 줄기세포 동결에 있어서 동결보호제에 따른 특성 연구)

  • The objective of this study was to investigate the effective cryoprotectants for the cryopreservation of porcine mesenechymal stem cells (pMSCs). In order to understand the effectiveness of various cryoprotectants on pMSCs, we studied the most commonly used cryoprotectants; dimethyl sulfoxide (DMSO), ethylene glycol (EG), DMSO and EG. pMSCs were isolated from bone marrow matrix of piglet (2 month) and characterized by alkaline phopshatase (AP) activity, colony forming, and differentiation to adipocyte. In slow cooling cryopreservation, the pMSCs were exposed to cell medium containing Dulbecco's Modified Eagle's Medium (DMEM) supplemented with 10% DMSO, 1.5M EG and 5% DMSO/0.75M EG, respectively, and freezed to $-1^{\circ}C$/min from $25^{\circ}C$ up to $-80^{\circ}C$ in a cryo-container. The proportion of viable cells and the growing rates in fresh pMSCs were significantly (P<0.05) higher than those of other groups, but did not differ between the cryopreserved groups. The expression of Sox-2 and Nanog gene was increased by extending culture time in cryopreserved groups. The expression of Bax gene in cryopreserved groups was similar with fresh pMSCs. Moreover, the gene expression of adipocyte-specific marker as well as chondrogenic/osteogenic factors in cryopreserved groups was similarly to fresh pMSCs. Taken together, our results suggested that all these cryoprotectants of 10% DMSO, 1.5M EG and 5% DMSO/0.75M EG could be used for cryopreservation of the pMSCs.

  • This study examined the possibility on the dietary value and cryopreservation of the marine microalga, Isochrysis galbana. Four cryoprotectants, dimethyl sulfoxide (DMSO), ethylene glycol (EG), glycerol (Gly) and 1.2-propanedial (PD) were tested at the concentrations of 1.0, 1.5, 2.0, 2.5 and 3.0 M respectively. The highest survival rates were obtained with 1.5 and 2.0 M of four cryoprotectants yielding a survival rate of 80%. Cell concentration of $30\;{\times}\;10^4\;cell/ml$ at the initial point of the experiment was increased to $365\;{\times}\;10^4\;cell/ml$ in 1.5 M of dimethyl sulfoxide, $298\;{\times}\;10^4\;cell/ml$ in 1.5 M of ethylene glycol, $512\;{\times}\;10^4\;cell/ml$ in 1.5 M of glycerol, and $385\;{\times}\;10^4\;cell/ml$ in 1.5 M of 1.2-propanedial after five days, respectively. In dietary value experiment, survival rate and growth were not significantly different.

Resveratrol Ameliorates High-fat-induced Metabolic Complications by Changing the Expression of Inflammasome Markers and Macrophage M1 and M2 Markers in Obese Mice (라스베라트롤 투여가 고지방식이 비만쥐의 지방조직에서의 inflammasome과 대식세포 마커에 미치는 영향)

  • The purpose of this study was to investigate the effects of resveratrol supplementation on inflammasome, inflammation, and macrophage markers in subcutaneous adipose tissue of high-fat-diet-induced obese mice. C57BL/6 mice were randomly assigned to three groups: normal diet control (NC; n=10), high-fat diet control (HC; n=10), or high fat with resveratrol (HRE; n=10) group. The mice were fed a high-fat diet (60% of calories from fat) or normal diet (18% of calories from fat). Resveratrol dissolved in a 0.1ml solution of dimethyl sulfoxide was supplemented orally at 25 mg/kg body weight. After 15 weeks, the body weight was significantly higher in the high-fat diet group than in the normal diet group. The inflammasome markers NLRP3, ASC, and caspase1 were significantly lower in the HRE group than in the HC group. The levels of an inflammation marker, IL-18, were also significantly lower in the HRE group than in the NC and HC groups. The levels of macrophage markers F480 and CD86 were significantly lower in the HRE group than in the HC group. The levels of the M2 macrophage marker CD206 were significantly decreased in the HC and HRE groups. Resveratrol had a positive effect on ameliorating the complications of high fat diet-induced obesity by reducing inflammasome and M1 macrophage gene expressions. However, resveratrol supplementation did not reduce inflammation gene expression.

  • Some photopolymer, poly(vinyl cinnamoyl acetate)(PVCiA) was synthesized by esterification of polymer(vinyl alcohol)(PVA) with monochloroacetic acid, followed by reaction poly(vinyl monochloroacetate)(PVAhA) and potassium cinnamate. When esterification of PVA with monochloroacetic acid was reacted in the dimethyl sulfoxide(DMSO), in the synthesis of PVChA, it is very good yield and the successive cinnamoyl acetoxyl esterification of PVCiA can be successfully synthesized. But PVCiA is low photosensitive polymer if net added photosensitizing dyes. Here, we synthesized photosensitizing dyes.

  • In this study eco-friendly wax remover for printing table of flat screen was investigated. Seven organic solvents, diethylene glycol monomethyl ester, triethylene glycol monomethyl ester, triethylene glycol monobutyl ester, tripropylene glycol monomethyl ester, dimethyl sulfoxide(DMSO), trichloro ethylene(TCE), tolune, were used as resin wax remover. The resin wax solubility with solvents was investigated. DMSO was found to be the most suitable eco-friendly resin wax remover.
