MyoD, expressed in skeletal muscle lineages of vertebrate embryo, is one of muscle-specific basic helix-loop-helix (bHLH) transcription factors, which plays a key role in the determination and differentiation of all skeletal muscle lineages. In this study, a cDNA of grass carp MyoD was cloned and characterized from total RNA of grass carp embryos by RT-PCR. The full-length cDNA of grass carp MyoD is 1597 bp. The cDNA sequence analysis reveals an open reading frame of 825 bp coding for a protein of 275 amino acids, which includes a bHLH domain composed of basic domain (1-84th amino acids) and HLH domain (98-142th amino acids), without signal peptide. Then the MyoD cDNA of grass carp was cloned to yeast expression vector pPICZ$\alpha$A and transformed into P. pastoris GS115 strain, the recombinant MyoD protein with a molecular weight of about 31KD was obtained after inducing for 2d with 0.5% methanol in pH 8.0 BMGY medium, and the maximum yield was about 250 mg/L in shaking-flask fermentation. The results were expected to benefit for further studies on the crystal structure and physiological function of fish MyoD.
The proximate composition and mineral contents of 18 subtropical fish species caught off the coast of Jeju Island, Korea were determined. The mean moisture content was highest (78.0%) in the eight species of lean fish, lowest (72.6%) in the four species of medium-fat fish, and intermediate (75.4%) in the six species of low-fat fish. The lipid content was in the order of medium-fat fish > low-fat fish > lean fish. The protein content ranged from 17.7% to 18.9%. The mean macro-mineral contents in the 18 species were 290.2 mg/100 g edible portion K, 146.6 mg P, 113.5 mg Na, 67.2 mg Ca and 38.3 mg Mg. The mean value of micro-mineral contents were 2.1 mg Fe, 1.7 mg Cu and 1.5 mg Zn. These results suggest that subtropical fish from Jeju Island are good sources of protein, lipid and minerals.
Lipid levels and fatty acid compositions of 20 species of subtropical fish caught off the coast of Jeju Island, Korea were determined. The fish were categorized according to total lipid (TL) content as follows; lean fish ($TL=1.19{\pm}0.32%$, n=10 species), low-fat fish ($TL=3.02{\pm}0.44%$, n=6 species) and medium-fat fish ($TL=6.87{\pm}1.23%$, n=4 species). The prominent fatty acids were saturated fatty acids (SFAs), at 16:0 and 18:0 monounsaturated fatty acids (MUFAs), at 18:1n-9 and 16:1n-7, and polyunsaturated fatty acids (PUFAs), at 22:6n-3, 20:5n-3 and/or 20:4n-6. Across all species, the percentages of PUFAs and SFAs were $38.0{\pm}7.89%$ and $34.3{\pm}3.08%$, respectively, with no significant difference between both groups (P<0.05), but showed higher proportions than that of MUFA group ($27.8{\pm}7.23%$) (P<0.05). The proportion of n-3 PUFAs was $27.1{\pm}8.68%$ across all fish species, similar to levels in other fishes from Korea. These results suggest that subtropical fish from Jeju Island are a good source of n-3 PUFAs for humans.

Annual mean temperature and salinity were $18.8^{\circ}C$, 33.7psu (2012) and $18.8^{\circ}C$, 33.8psu (2013) which shows stable water mass. nutrient concentrations represent that dissolved inorganic nitrogen (DIN) ranged from 0.030 to 0.217mg/L, dissolved inorganic phosphorus (DIP) from 0.002 to 0.010mg/L, and silicate ($SiO_2$) from 0.096 to 0.292mg/L. Chlorophyll a concentrations of phytoplankton ranged from 0.905 to $1.125{\mu}g/L$ were appeared the highest in summer and the lowest in winter. Suspended Solid(SS) concentrations ranged from 1.200 to 2.100mg/L (mean, 1.634mg/L) were appeared the highest in autumn and the lowest in spring. During the survey period, a total of 83 species (48 families and 12 orders) of fishes were identified, Perciformes and Scorpaeniformes accounted for 63.9% of the total number. The number of species was higher in summer than in winter, showing a peak in summer at 57 species and a low in winter at 47 species. The number of individuals and the biomass peaked in summer at 694 individuals and 151.9kg, and were the lowest in autumn at 466 individuals and 106.5kg. The diversity index, evenness index and richness index were appeared 2.92~3.14, 0.75~0.83 and 7.06~8.56 at each stations. the dominance index were appeared highest in spring and summer, were appeared the lowest in winter and autumn. The Filefish (Stephanolepis cirrhifer), Bluestriped angelfish (Chaetodontoplus septentrionalis), Marbled rockfish (Sebastiscus marmoratus), Rabbit fish (Siganus fuscescens), Blackfin sweeper (Pempheris japonica) were dominant species, which occurred abundantly during the entire survey period. The subtropical fishes were identified total of 41 species (49.4%) and the number of species was higher in summer at 29 (50.9%) species and a low in winter at 20 (42.6%) species. The dominant species of subtropical fishes were appeared Bluestriped angelfish, Blackfin sweeper, Rabbit fish, Scarbreast tuskfish (Choerodon azurio), Ballonfish (Diodon holocanthus), Flagfish (Goniistius zonatus), Blue-spotted boxfish (Ostracion immaculatus) in study sites. The subtropical fishes of species, individual and biomass showed significant correlation with environmental factors (Chlorophyll a, DIN, DIP, $SiO_2$) in almost all sampling sites.
The age and growth of rabbit fish, Siganus fuscescens were investigated from samples monthly collected in the coast of Jeju island, Korea from February to December 2012. Ages were determined from annuli in otoliths, and annuli were formed in one or two months immediately after spawning once a year. Also, main spawning period was estimated between July and August, thus rings were considered as annual marks. The von Bertalanffy growth equations estimated from a non-linear regression method were $L_t=37.24(1-e^{-0.33(t+1.24)})$ ($R^2=0.92$) for female and $L_t=31.44(1-e^{-0.45(t+1.23)})$ ($R^2=0.90$) for male, and the growth between female and male was different.
Ciguatera is the commonest syndrome of marine poisoning that is caused by the ingestion of ciguatoxins that accumulate in certain tropical and subtropical finfish. It is endemic throughout the subtropical and tropical regions of the Indo-Pacific and Caribbean and has been rarely reported in Korea. With the expansion of travel, tourism, and the import of fish from the tropics, ciguatera poisoning now affects a diverse population. We report a case of ciguatera in a 70-year-old woman, presenting with general paralysis and coma, which developed after consuming codfish and mackerel in Korea.
