Subtraction 기법을 이용한 한우 성장 단계 특이 발현 유전자 탐색 (Identification of the Differentially Expressed Genes of Hanwoo During the Growth Stage by Subtractive cDNA Hybridization)

  • 한우의 성장단계 특이발현 유전자를 탐색하기 위하여, 본 연구에서는 유전자의 발현 유무 및 발현정도의 차이를 나타내는 유전자를 분리하는데 있어 가장 강력한 수단으로 알려진 subtractive cDNA hybridization 기법을 이용하여 한우 등심조직으로부터 12개월령 및 24개월령 특이적인 subtractive cDNA library를 제작하였다. 성장단계 특이적인 유전자를 탐색하기 위하여, 6, 12 및 24개월령 cDNA를 사용하여 reverse northern blot 분석을 실시하였으며, 6개월령 cDNA probe에 대하여 특이적인 signal을 나타낸 3개의 clone은 EPV 20, Ca2+ ATPase, 및 TCTP 유전자와 유사성을 나타내었다. 12개월령 cDNA probe에 대하여 특이적인 signal을 나타낸 9개의 cDNA clone은 각각 VCP, HSP 70, aldolase A, MSSK1, GM-2 activator protein, ryanodine receptor, acidic ribosomal phosphoprotein p1, ADP/ATP translocase T1 및 UCP 2 유전자와 높은 homology를 가지고 있었다. 또한 2개의 clone이 각각 12개월령 및 24개월령 cDNA probe에 대하여 특이적인 signal을 나타내었는데, 12개월령 cDNA probe에 대해서만 signal을 나타낸 clone은 ferrochelatase 유전자와 유사하였으며, 24개월령 probe에 대해서만 signal을 나타낸 clone은 ADRP 유전자와 유사하였다. 이상에서와 같이, 본 연구에서 제작한 성장단계 특이적인 subtractive cDNA library를 분석하여 14종의 유전자를 한우 성장단계 특이 발현 후보 유전자로 선정하여 염기서열을 분석하였으며, 이외에도 성장단계에 있어 특이적으로 발현될 것으로 추정되는 cDNA 클론의 염기서열을 분석하였다.

디지털 광학기술인 3D 프린터로 제작된 임시수복용 레진의 두께별 색 평가 (Color evaluation by thickness of interim restorative resin produced by digital light processing 3D printer)

  • Purpose: The purpose of this in vitro study was to measure and compare the thickness-dependent color dimensions of digital light processing (DLP) three-dimensional (3D) printer and conventional interim restorative resin. Methods: Specimens (N=60) were fabricated using either subtractive manufacturing (S group) or DLP 3D printing (D group) material. All milled and 3D-printed specimens were allocated into three different groups (n=10) according to different thicknesses as follows: 1.0, 1.5, and 2.0 mm. Color measurements in the CIELab coordinates were made using a spectrophotometer under room light conditions (1,003 lux). The color differences (𝚫E*) between the specimen and control target data were calculated. Data were analyzed using the oneway analysis of variance (ANOVA). Post hoc comparisons were conducted using Tukey's honestly significant difference method (α=0.05 for all tests). Results: The 𝚫L*, 𝚫a*, 𝚫b*, and 𝚫E* values of interim restorative resin produced by DLP 3D printing were obtained in terms of the specimen's thickness increased compared with the increases by subtractive manufacturing. When the thickness was similar, the color difference between subtractive manufacturing and DLP 3D printing was ≥5.5, which is a value required by the dentist for remanufacturing. Conclusion: Color was influenced by the thickness of the interim restorative resin produced by DLP 3D printing.

Suppression subtractive hybridization (SSH) for isolation and characterization of genes related to testicular development in the giant tiger shrimp Penaeus monodon

  • Suppression subtractive hybridization (SSH) cDNA libraries of the giant tiger shrimp, Penaeus monodon, were constructed. In total, 178 and 187 clones from the forward and reverse SSH libraries, respectively, of P. monodon were unidirectionally sequenced. From these, 37.1% and 53.5% Expressed Sequence Tags (ESTs) significantly matched known genes (E-value < 1e-04). Three isoforms of P. monodon progestin membrane receptor component 1: PM-PGMRC1-s (1980 bp), PM-PGMRC1- m (2848 bp), and PM-PGMRC1-l (2971 bp), with an identical ORF of 573 bp corresponding to a deduced polypeptide of 190 amino acids, were successfully identified by RACE-PCR. Interestingly, PMPGMRC1 showed a greater expression level in testes of juvenile than broodstock P. monodon (P < 0.05). Dopamine administration ($10^{-6}$ mol/shrimp) resulted in up-regulation of PM-PGMRC1 in testes of juveniles at 3 hrs post treatment (P < 0.05), but had no effect on PM-Dmc1 (P > 0.05).

  • An improved single-phase full-bridge zero-voltage-switching inverter using a subtractive coupled magnetics is proposed in this paper. The proposed topology overcomes several drawbacks of the conventional ARCPI zero-voltage-switching inverter including two bulky capacitors which can cause problems such as the need for a protection circuit and voltage fluctuation of split capacitors. Also the proposed topology can reduce the number of devices required for ZVS through a simplified auxiliary circuit, thus achieving low cost and small volume and is applicable a modified unipolar PWM scheme. Detail mode analysis and design considerations are provided for optimal efficiency. In the end, the effectiveness and feasibility of the proposed topology are verified experimentally under various conditions.

  • The concept of Level Of Detail (LOD) was introduced and has been used to enhance display performance and to carry out certain engineering analysis effectively. We would like to use an adequate complexity level for each geometric model depending on specific engineering needs and purposes. Solid modeling systems are widely used in industry, and are applied to advanced applications such as virtual assembly. In addition, as the demand to share these engineering tasks through networks is emerging, the problem of building a solid model of an appropriate resolution to a given application becomes a matter of great necessity. However, current researches are mostly focused on triangular mesh models and various operators to reduce the number of triangles. So we are working on the multi-resolution of the solid model itself, rather than that of the triangular mesh model. In this paper, we propose multi-resolution representation of B-rep model by reordering and converting design features into an enclosing volume and subtractive features.

  • In this paper, a procedure is developed for identifying a number of representative solutions manageable for decision-making in a multiobjective optimization problem concerning the test intervals of the components of a safety system of a nuclear power plant. Pareto Front solutions are identified by a genetic algorithm and then clustered by subtractive clustering into "families". On the basis of the decision maker's preferences, each family is then synthetically represented by a "head of the family" solution. This is done by introducing a scoring system that ranks the solutions with respect to the different objectives: a fuzzy preference assignment is employed to this purpose. Level Diagrams are then used to represent, analyze and interpret the Pareto Fronts reduced to the head-of-the-family solutions.

3D 프린팅 : 새로운 산업혁명인가? (3D Printing : A New Industrial Revolution?)

  • 다양한 연구기관에서 4차 산업혁명을 이끌 핵심기술로 AI, IoT, Blockchain 등의 기술과 더불어 3D 프린팅 기술을 들고 있다. 3D 프린팅은 전통적인 생산방식인 subtractive manufacturing(SM)과는 대조적으로 additive manufacturing(AM)으로 그 영향력이 매우 커 혁명이라는 용어를 사용하기도 한다. 이에 본 연구에서는 3D 프린팅 산업의 특성을 경제 관점, 생산 관점, 마케팅 관점에서 전통적인 방식과 비교하여 분석하였다. 이후 비즈니스 생태계 구축 측면에서 쟁점을 분석하였다. 끝으로 향후 연구를 위한 아젠더를 제시하였다. 본 연구는 실무적으로는 3D 프린팅을 둘러싼 핵심적인 이슈 파악 및 향후 적용을 위한 단초를 제공하였다. 학문적으로는 기존의 연구 이슈에 대해 쟁점을 정리함으로써 향후 연구를 위한 틀을 제시하였다. 향후 연구에서는 이러한 논의를 토대로 실제 사례를 중심으로 실증적인 분석이 뒷받침될 필요가 있다.
