Comparison of Physicochemical Properties of Starch Acetates Prepared by Conventional, Preheat Treatment and Extrusion Process (습식법과 예열처리법 및 Extrusion 공정에 의해 제조한 초산전분의 이화학적 성질비교)

  • Starch acetates were prepared by conventional method, preheat treatment, and extrusion process through acetylation of corn starch with acetic anhydride and their physicochemical properties were investigated. The optimal conditions of the acetylation of starch by conventional method(CSA) was found that starch concentration was 30%, reaction temperature $35^{\circ}C$ and pH 8.5. With increasing the molar ratio of acetic anhydride to anhydrous glucose unit from 0.03 to 0.20, DS(Degree of substitution) value of corn starch acetate prepared at the optimum condition was increased from 0.019 to 0.080, while the acetylation efficiency was decreased from 31.6% to 20.5%. In the case of the preheated (gelatinized), then acetylated starch(PSA), DS value was increased from 0.027 to 0.04 at the fixed molar ratio of the acetic anhydride with increasing preheating temperature from $60^{\circ}C\;to\;90^{\circ}C$. The DS was low as 0.02 in the case of starch acetate prepared by extrusion process(WESA). The CSA and PSA showed lowering gelatinization temperature and enthalpy than raw corn starch with increasing DS. All of starch acetates showed the increased degree of transparency, the decreased lightness and the increased yellowness as compared to the raw corn starch. WESA showed lower apparent viscosity and more close to the characteristic of the Newtonian fluid than CSA and PSA. Intrinsic viscosity was reduced in CSA and WESA, although PSA has a slightly higher one than raw corn starch. The rate of retrogradation of the gels was retarded in all starch acetates.

Effect of Sequence Variation in Bovine Mitochondrial DNA D-Ioop Region on ~ilk Production for Hanwoo (한우 산유량에 미치는 Mitochondrial DNA D-loop영역의 염기서열 변이효과)

  • This study was performed to analyze the sequence variations of mtDNA D-loop and their effects on milk in Hanwoo(Korean cattle). The resulting sequences were compared with previously published sequences for other cattle breeds (GenBank JOI394). The Polymerase Chain Reaction was performed to amplify a total of 964 bp between nucleotide 15758 and 383 within D-loop region of mtDNA using specific primers. Twenty polymorphic sites by nucleotide substitution were found in mtDNA D-loop region of Hanwoo. The frequencies of positions at 8, 169, 16042, 16051, 16057, 16093, 16119, 16122, 16209, 16255 and 16302 nt with high levels of sequence polymorphism were 0.150, 0.950, 0.085, 0.138, 0.106, 0.085, 0.138, 0.212, 0.085, 0.148 and 0.180, respectively. The substitution effect at 16119(p<0.1) and 16185(p< 0.05) nt was found significant on milk production. Polymorphism of mtDNA sequence in D-Ioop region could be useful for the analysis of cytoplasmic genetic variation and associations with the other economically important traits and maternal lineage analysis in Hanwoo.

  • In the realm of cement manufacturing, concerted efforts are underway to mitigate the emission of greenhouse gases. A significant portion, approximately 60%, of these emissions during the cement clinker sintering process is attributed to the decarbonation of limestone, which serves as a fundamental ingredient in cement production. Prompted by these environmental concerns, there is an active pursuit of alternative technologies and admixtures for cement that can substitute for limestone. Concurrently, initiatives are being explored to harness technology within the cement industry for the capture of carbon dioxide from industrial emissions, facilitating its conversion into carbonate minerals via chemical processes. Parallel to these technological advances, economic growth has precipitated a surge in construction activities, culminating in a steady escalation of construction waste, notably waste concrete. This study is anchored in the innovative production of calcium silicate cement clinkers, utilizing finely powdered waste concrete, followed by a thorough analysis of their mineral phases. Through X-ray diffraction(XRD) analysis, it was observed that increasing the substitution level of waste concrete powder and the molar ratio of SiO2 to (CaO+SiO2) leads to a decrease in Belite and γ-Belite, whereas minerals associated with carbonation, such as wollastonite and rankinite, exhibited an upsurge. Furthermore, the formation of gehlenite in cement clinkers, especially at higher substitution levels of waste concrete powder and the aforementioned molar ratio, is attributed to a synthetic reaction with Al2O3 present in the waste concrete powder. Analysis of free-CaO content revealed a decrement with increasing substitution rate of waste concrete powder and the molar ratio of SiO2/(CaO+SiO2). The outcomes of this study substantiate the viability of fabricating calcium silicate cement clinkers employing waste concrete powder.

  • The distributions of G to A substitution ($G^{-75}{\rightarrow}A$) mutation in the human apolipoprotein A1 (APOAI) gene promoter region and glutathione S-tran-sferase Mu1 (GSTM1) gene deletion were examined in subjects with Korean population. The $G^{-75}{\rightarrow}A$ mutation of APOA1 was genotyped by the polymerase chain reaction (PCR) and subsequent digestion of the PCR product using either Mspl or Mval (n=206). The observed numbers of GG, GA and AA genotypes were 132, 63 and 11, respectively. The allele frequencies of G and A were 0.794 and 0.206, respectively. The GSTM1 gene deletion was simply examined by the PCR amplification (n=106). The observed numbers of null type ($GSTM1^*0/GSTM1^*0$) and positive type were 55 and 51, respectively. The allele frequency of $GSTM1^*0$ was 0.720.

  • We have developed four chicken hybridomas secreting monoclonal antibodies to induce a protective immune response against the chicken disease avian coccidiosis, caused by the intestinal parasite Eimeria acervulina. Huwever, since the amount of antibodies secreted from these hybridomas is too low or sometimes they lost their ability to produce antibodies, the hybridoma method is not satisfactory in the production of large amounts of chicken monoclonal antibodies. To bypass these problems, we applied the antibody engineering technology using polymerase chain reaction. We cloned and determined the sequences of variable domains of the four chicken monoclonal antibodies, namely, 2-1, 5D11, 13C8 and 8C3. The sequences comparison to germline sequences skewed that the gene con version mechanism might contribute to developing diversification of heavy and λ-light chains in chicken antibodies. Several pseudogene families regarded as donors in gene conversion were identified at each framework region and the complementarily determining region of λ-light chains. In addition, as expected, numerous changes of nucleotide sequences such as nucleotide substitution, insertion and deletion were found predominantly in complementarity determining regions, which are likely to be somatic hypermutations as a result of affinity maturation in antibody-producing cells.

  • Cryptand resins have been synthesized from 1-aza-18-crown-6 macrocyclic ligand attached to styrene (2th petroleum in 4th class hazardous materials) divinylbenzene copolymer with crosslinkage of 1%, 2%, 10%, and 18% by substitution reaction. The synthesis of these resins was confirmed by the content of chlorine, element analysis, surface area (BET), and IR-spectroscopy. The effects of pH, time and crosslinkage on adsorption of metal ion from water fire extinguishing agentby synthetic resin adsorbent were investigated. Metal ions showed a great adsorption over pH 3 and adsorption equilibriumof metal ions was about two hours. The adsorption selectivity determined in water was in the increasing order of sodium ($Na^{1+}$) > zinc ($Zn^{2+}$) > chromium ($Cr^{3+}$) ion. The adsorption was in the order of 1%, 2%, 10%, and 18% crosslinkage resin.

  • The hard coatings on the polycarbonate plate were performed with the object of substitution the glass in the car to the polycarbonate plate. In this research, (3-glycidoxypropyl)trimethoxysilane (GPTMS), (3-aminopropyl)triethoxysilane (APS), and diethylenetriamine (DETA) were used to prepare the coatings by a sol-gel process. The optimum conditions and formulation to achiere the excellent physical properties of the coating were determined. In the case of using GPTMS and DETA, the smooth coating which had the 2 H class in pencil hardness was formed. In the case of using GPTMS and APS, the coating which had the 2 H class in pencil hardness was also formed. And the coatings with GPTMS and DETA showed an excellent abrasion resistant.

  • ;The electronic state calculations for various types of ceramic materials have beell performed by the use of $DV-X\;{\alpha}$ cluster method. The molecular orbital levels and wave functions for model clusters have been computed to study the electronic properties ami chemical bonding of the ceramics. For ${\beta}-sialon(Si_{6-z}Al_zO_zN_{8-z})$ which is a high temperature structural material based on ${\beta}-Si_3N_4$, we have made model cluster calculations to estimate the strength of chemical bonding between atoms by the Mulliken population analysis. It is found that the covalent bonding between Si and N atoms is very strong in pure ${\beta}-Si_3N_4$, but the covalency around solute atom is considerably weakened when Si atom is substituted by AI. This tendency is enhanced by an additional substitution of oxygen atom for N. The result calculated can well explain the experimental data of changes in mechanical properties such as the reductions of Young's modulus and Vickers hardness with increment of z-value in ${\beta}-sialon$. Various model clusters for transition metal oxides which show many interesting physical and chemical properties have also been calculated. High-valent perovskite-type iron oxides EMFe0_3E(M=Ca and Sr) possess very interesting magnetic and chemical properties. In these oxides, iron exists as $Fe^{4+}$ state, but the experimental measurement of Mossba~er effect suggests that disproportionation $2Fe^{4+}=Fe^{3+}+Fe^{5+}$ takes place for $CaFe0_3$ at low temperatures. The model cluster calculations for these compounds indicated the existence of considerably strong covalent bonding of Fe-O. The calculations of hyperfine interaction at iron neucleus show very good agreement with the experimental Mossbauer measurements. The result calculated also implies that the disproportionation reaction is strongly possible by assuming the quenching of breathing phonon mode at low temperatures.tures.

Microstructure Evolution and Dielectric Characteristics of CaCu3Ti4O12 Ceramics with Sn-Substitution

  • The doping effect of Sn on the microstructure evolution and dielectric properties was studied in $CaCu_3Ti_{4-x}Sn_xO_{12}$ polycrystals. Samples were produced by a conventional solid-state reaction method. Sintering was carried out at $1115^{\circ}C$ for 2-16 h in air. The dielectric constant and loss were examined at room temperature over a frequency range between $10^2$ and $10^6$ Hz. The microstructure was found to evolve into three stages. Addition of $SnO_2$ led to an increase in density and advanced formation of abnormal grains. The formation of coarse grains with a reduced thickness of the boundary brought about an enhanced dielectric constant and a lower dielectric loss below ~1 kHz. EDS data showed the Cu-rich phase along the grain boundary, which should contribute to the improved dielectric constant according to the internal barrier layer capacitor model. After all, $SnO_2$ was an effective dopant to elevate the dielectric characteristics of $CaCu_3Ti_{4-x}Sn_xO_{12}$ polycrystals as a promoter for abnormal grain growth.

Preparation and Physicochemical Characteristics of Octenyl Succinated Rice Starches Based on Amylose Content (아밀로오스 함량에 따른 옥테닐호박산 쌀전분의 제조 및 이화학적 특성)

  • The purpose of this study was to investigate the preparation and physicochemical characteristics of n-octenylsuccinylated (OSAn) rice starches with different rice amylose contents. Amylose contents of Jinsumi flour and Milyang 261 flour were 15.42 and 20.31%, respectively. After processing by alkali treatment, amylose contents of Jinsumi rice starch and Milyang 261 rice starch were 34.21% and 39.32%, respectively. After OSAn treatment, the degree of substitution and reaction efficiency of Jinsumi starch were higher than those of Milyang 261. The stability of the modified Jinsumi emulsion was higher than that of the Milyang 261 emulsion. Viscosity of the Jinsumi emulsion was higher than that of the Milyang 261 emulsion. When the emulsions were spray dried, modified Jinsumi rice starch showed excellent coating efficiency compared to that of modified Milyang 261 rice starch. Therefore, Jinsumi was more suitable than Milyang 261 to apply for encapsulation as wall materials.
