Effect of Route of Trihalomethanes (THM) Administration on Renal Toxicity in Male Rat

  • Single non-lethal doses of chloroform $(CHCL_3)$ dichlorobromomethane $(CHCL_2Br)$, dibromochloromethane $(CHCIBr_2)$, or bromoform $(CHBr_3)$ were administered to male rats. Routes of exposure including single intraperitional (ip) and subcutaneous (sc) injection were used in order to permit comparison of severity of THM effects and renal toxicity was assessed at varied times following treatment. On an equimolar basis, sc administration of $CHBr_3$ (either 12 or 3 mmoles/kg) is more effective at increasing KW/BW than ip $CHCI_3$ treatment. Plasma urea nitrogen (BUN) following ip THM injections are markedly increased with all four THM at 24 hours post treatment. BUN response to $CHCL_2Br$ and $CHCIBr_3$-effected BUN levels have essentially returned to those of vehicle control. THM sc treatment results in a BUN response similar to that seen following ip treatment, with only the time course being different. With the exception of $CHCL_3$, sc and ip-treatments appear to be equally effective in evoking absolute BUN elevations. These results suggest that THM administration induce renal toxicity dependent upon the route or exposure.

  • Intraperitoneal administration of ginseng butanol fraction(GBF) to chronic morphinization in male Sprague-Dawley rats inhibited the development of tolerance to the analgesic effect and hyperthermic action of morphine. Rats were rendered tolerant to morphine by subcutaneous multiple morphine injections for a period of 8 days. The development of tolerance was evidenced by the decreased analgesic response to morphine and inhibition of tolerance by the greater analgesic response. Concomitant administration of morphine with GBF blocked the tolerance to the hyperthermic effect of morphine as evidenced by elevation of body temperature by morphine. Dopamine receptor sensitivity was enhanced in morphine tolerant rats as measured by apomorphine induced in spontaneous motor activity. GBF administration also blocked dopamine receptor supersensitivity induced by chronic morphinization.

  • Secure fixation is essential for continous epidural catheterization on a long-term-basis. Adhesive tape or surgical knots were commonly used for those patients, but the surgical knot method has a tendency to cause strangulation of catheter. Another invasive technique, subcutaneous tissue tunnelling is more safe than other methods but requires sophisticated technique and time. We employed a simple device using a blood transfusion set for patients who have epidural catheters placed safely un their backs. In 120 patients treated for postoperative and chronic pain by means of this technique, the results were as follows: 1) Five of 120 patients (4.2%) developed backache and pruritus, but there were no instances of respiratory depression, local infection and headache. 2) Nine of 120 patients (7.5%) failed booster-injections, but two cases were due to be non-technical errors. 3) The duration of fixation was 1~3 days in most cases (85%), the longest being for 21 days.

Ameliorative Effect of Yacon Containing Herbal Mixture against Benign Prostatic Hyperplasia Symptoms (야콘을 포함한 수종 천연물 처방의 전립선비대 증상 개선효과)

  • We evaluated the benign prostatic hyperplasia (BPH) with Yacon pill, the preparation composed of Smallanthus sonchifolius, Torilis japonica and Acorus gramineus. A total of 45 rats were divided into five groups. One group was used as a control and the other groups received subcutaneous injections of testosterone for 4 weeks to induce BPH. Yacon pill extract (200 or 400 mg/kg) was administered daily for 4 weeks to two groups by oral gavage concurrently with testosterone. The rats euthanized, the prostate and body weights were recorded, and tissues were subjected to hormone assay. In addition, we investigated proliferating cell nuclear antigen (PCNA) expression in the prostate using immunoblotting. Rats with BPH showed significantly increased prostate weights, increased dihydrotestosterone (DHT) levels in the serum and increased PCNA expression in the prostate; however, Yacon pill extracts treated rats showed significant reduction of prostate weights, DHT levels and PCNA expression compared with the BPH group. Conclusively, Yacon pill showed the possibility as ameliorable agents of BPH symptoms.

Bilateral symmetrical lipoma of the buccal fat pad as an incidental finding in a woman with weight gain after tamoxifen: a case report

  • Although lipoma is a common benign tumor, it occurs relatively infrequently in the oral and maxillofacial areas, and only 31 cases of lipoma in the buccal fat pad have been reported. Herein, we present an extremely rare case of symmetric lipomas in both buccal fat pads. These masses were incidentally discovered during a facelift procedure in a 50-year-old woman with a 4-year history of tamoxifen use. during which she had gained 10 kg. The patient stated that cheek protrusion had developed concomitantly with weight gain and was exacerbated by an injection lipolysis procedure she had received 1 year previously. This case underscores the importance of paying careful attention to the patient's medication use and surgical history when evaluating suspected cases of lipoma, and sheds light on tamoxifen use and subcutaneous injections of phosphatidylcholine and deoxycholate as potential risk factors for lipoma development.

Distribution of the Mast Cells in the Parenchymal Organs of the Cattle, Horses, Pigs, Dogs, and Rompun-induced Mast Cell Degranulation in the Dog (우(牛), 마(馬), 돈(豚) 및 견(犬)의 실질장기내(實質臟器內) Mast Cell 분포(分布)와 Rompun을 투여(投與)한 견(犬)의 Mast Cell 탈과립소견(脫顆粒所見))

  • This paper dealt with the distribution of normal mast cells in the spleen, liver and lung on cattle, horses, pigs and dogs, and also degranulation of mast cells in the dogs infected with Rompun (2% Xylazine HCl). The results observed are summarized as follows. Normal mast cells were distributed in spleen, liver and lung on cattle, horse, pig and dog. Mast cells were observed in both red pulp and surroundings of white pulp of the spleen in horse, in the white pulp of the spleen in cattle, in the trabeculae of the spleen in pigs, and in white pulp and red pulp of the spleen in dogs, respectively. Mast cells were observed in the portal triad of the liver in cattle and horses, in both portal triad and interlobular connective tissues of the liver in pigs, and not only the portal triad but also walls of the sinusoids and the central veins in dogs. A large number of mast cells were observed in the interlobular septa and peribronchioles of lung on all the species in this experiment. The mast cells are more numerous in the lungs than other organs. Author considers that numbers of normal mast cells distributed in the tissue is related to the dosage of Rompun in animal. The degranulation of mast cells were observed in the subcutaneous tissues of dog intramuscularly injected with Rompun(0.5ml/times) for 4 or 5 times and subcutaneously injected with Rompun(0.3ml/times) for 4 times. In dog intradermally injected with 0.1ml of Rompun, mast cells were decreased in number at 30 minutes and markedly decreased in number at 2 hours, but more or less increased in number at 3 hours after injection. In addition, the granules of the mast cells were decreased in number at 30 minutes and marked degranulation of the mast cells were recognized at 2 hours after injections, but normal mast cells begun to appear in subcutaneous tissue with the lapse of time from 3 hours after injection. There was also observed local infiltration of neutrophils in subcutaneous tissues of dogs intradermally injected with 0.1ml of Rompun at 30 minutes. At 2 hours after injection, numerous neutrophils and a small number of eosinophils were observed in the site of injection. Conclusionally, Rompun was regarded as a factor which causes the degranulateon of mast cell and the authors considered that histamine released from the mast cells by Rompun might cause relaxation of skeletal muscle.

Development of Polyclonal Antibodies to Abdominal and Subcutaneous Adipocyte for Producing Fat-reduced High Quality Pork (저지방 고품질 돈육 생산을 위한 돼지 복강 및 피하지방 항체 개발)

  • The aim of the present study was to develop polyclonal antibodies to regional inedible adipocytes of pigs and investigate the effect of these antibodies on adipocytes in vitro. As antigens, abdominal and subcutaneous adipocyte PMPs from pigs were injected into sheep 3 times per 3 wk intervals for passive immunization, and non-immunized serum, antisera against abodominal (AAb) or subcutaneous adipocyte PMPs (SAb) were collected before and after the injections. Titers of the antisera obtained from sheep and their cross-reactivities with the heart, kidney, liver, lung, muscle, and spleen of pig were determined by ELISA. Isolation and culture of abdominal and subcutaneous adipocytes from pigs were performed to analyze LDH concentration. At a 1:1,000 dilution, little antibody reactivity was observed for non-immunized serum whereas both AAb and SAb had relatively strong reactivity up to a dilution of 1:16,000. These findings may indicate that strong antibodies against adipocyte PMPs can be developed using an immunological approach. Extremely low reactivity of AAb and SAb was detected with the PMPs of the organs. Both antisera most strongly reacted with each adipocyte PMPs and showed statistically (p<0.05) higher cross-reactivities compared with the non-immunized serum. In conclusion, these results may indicate that the present polyclonal antibodies against regional inedible adipocyte PMPs are well developed and are safe against cross-reactivities with the organs of pigs. Further studies on the in vivo nutritional safety and fat reduction of these antibodies in pigs will be required fat-reduced high quality pork production.

Effects of Exposure Period on the Developmental Toxicity of 2-Bromopropane in Sprague-Dawley Rats

  • Recently we reported that 2-bromopropane (2-BP) has maternal toxicity, embryotoxicity, and teratogenicity in Sprague-Dawley rats. The aims of this study are to examine the potential effects of 2-BP administration on pregnant dams and embryo-fetal development, and to investigate the effects of metabolic activation induced by phenobarbital (PB) on developmental toxicities of 2-BP. Pregnant rats received 1000 mg/kg/day subcutaneous 2-BP injections on gestational days (GD) 6 through 10 (Group II and Group IIII) or 11 through 15 (Group IV). Pregnant rats in Group III received an intraperitoneal PB injection once daily at 80 mg/kg/day on GD 3 through 5 for induction of the liver metabolic enzyme system. Control rats received vehicle injections only on GD 6 through 15. All dams underwent caesarean sections on GD 20 and their fetuses were examined for external, visceral, and skeletal abnormalities. Significant adverse effects on pregnant dams and embryo-fetal development were observed in all the treatment groups, and the maternal and embryo-fetal effects of 2-BP observed in Group II were higher than those seen in Group IV. Conversely, maternal and embryo-fetal developmental toxicities observed in Group III were comparable to those seen in Group II. These results suggest that the potential effects of 2-BP on pregnant dams and embryo-fetal development are more likely in the first half of organogenesis (days $6{\sim}10$ of pregnancy) than in the second half and that the metabolic activation induced by PB pre-treatment did not modify the developmental toxic effects of 2-BP in rats.

Comparison of follitropin ${\beta}$ administered by a pen device with follitropin ${\beta}$ administered by a conventional syringe in patients undergoing IVF-ET

  • Objective: To compare the effectiveness and convenience of a pen device for the self-administration of follitropin ${\beta}$ with a conventional syringe delivering follitropin ${\beta}$ solution in patients undergoing IVF-ET. Methods: GnRH agonist long protocol was used for controlled ovarian stimulation (COS) in all subjects. A total of 100 patients were randomized into the pen device group or the conventional syringe group on the first day of COS. Local tolerance reactions were assessed within 5 minutes, at 1 hour and at 3 hours after each injection. On the day of hCG injection, patients were asked to rate their overall pain and convenience experienced with self-injection on a visual anlaogue scale (VAS). Results: There were no differences in patients' characteristics between the two groups. The duration of COS was significantly shorter in the pen device group than in the conventional syringe group. Patients included in the pen device group needed a significantly smaller amount of follitropin ${\beta}$. However, no differences between the two groups were found in IVF results and pregnancy outcome. The incidence of local pain within 5 minutes, at 1 hour and at 3 hours after the injection was significantly lower in the pen device group. VAS scores indicated that injections using the pen device were significantly less painful and more convenient. Conclusion: The pen device for self-administration of follitropin ${\beta}$ is less painful, safer and more convenient for the patients, and can be more effective because of the shorter duration and smaller dose of follitropin ${\beta}$ when compared with the conventional syringe.

Subcutaneous Administration of Highly Purified-FSH(HP-FSH) versus Intramuscular Administration of FSH in Superovulation for IVF-ET (체외수정시술을 위한 과배란유도시 Highly Purified Follicle Stimulating Hormone (HP-FSH) 피하주사와 Follicle Stimulating Hormone 근육주사의 비교연구)

  • The early studies demonstrated that the relative amount of FSH was important for stimulating normal ovarian activity and demonstrated the existence of a threshold level for FSH, above which follicular growth was activated. It was found that only a modest increase in circulating FSH level above the threshold (between 10 and 30%) was required to stimulate folliculogenesis. In addition, FSH is primary responsible for initiating estradiol production through the activation of the aromatase enzyme system in granulosa cells, follicular secretion and growth. LH on the other hand, plays a supportive role in ovarian steroidogenesis, stimulating the ovarian thecal cells to produce androgen, the precursor for estradiol synthesis. But there is now an increasing number of reports in the literature demonstrating an adverse effect of LH on fertility and miscarriage in infertile and fertile women. So HP-FSH is the drug of a highly purified FSH preparation which has a higher specific activity and far fewer impurities than FSH. This study was performed to evaluate the efficacy and safety of HP-FSH administered (SC; subcutaneous) versus FSH(IM; intramuscular) for ovulation induction. 20 candidates patients for ovulation induction were participated. All patients underwent pituitary desensitizing with a long gonadotropin-releasing hormone (GnRH) agonist protocol and ovulation induction was started with HP-FSH SC (10 patients; group I) or FSH IM (10 patients; group II). After ovulation, outcome of ovulation induction and local reaction of injection site were compared. There were no difference of outcome of ovulation in two groups except pregnancy rate/embryo transfer. Group I had a higher pregnancy rate/ embryo transfer than Group II (44.4% Vs 28.6%). Pain, redness, tenderness, bruising and itching when the injection received on the first 5 days of treated (50 SC and 50 IM injections) were assessed. There were no significant difference (P>0.05) in the incidence of tenderness, bruising and itching between the IM and SC injection. But IM injection (FSH) had a tendency of higher above incidence. The number of reports of pain, redness were significantly increased in IM injection group (P<0.05). These results indicate that SC administration of HP-FSH has been shown to be as effect for superovulation as traditional gonadotropins, with an improved safety profile due to the removal of extaneous proteins.
