  • The present study was conducted to examine the effect of recombinant bovine somatotropin(${\gamma}$ bST), which was administered to cow to promote milk production, on bST levels in milk. Fourteen cows were divided into two groups: 1) control cows received neither y bST nor vehicle, 2) treated cows were administered twice at two-week interval with 500mg ${\gamma}$ bST each cow by subcutaneous injection. Milk samples were taken on day 0 (prior to injection), day 7 (7 days after 1st injection), day 21 (7 days after 2nd injection) and day 35 (21 days after 2nd injection).

Subcutaneous progesterone versus vaginal progesterone for luteal phase support in in vitro fertilization: A retrospective analysis from daily clinical practice

  • Objective: Progesterone application for luteal phase support is a well-established concept in in vitro fertilization (IVF) treatment. Water-soluble subcutaneous progesterone injections have shown pregnancy rates equivalent to those observed in patients receiving vaginal administration in randomized controlled trials. Our study aimed to investigate whether the results from those pivotal trials could be reproduced in daily clinical practice in an unselected patient population. Methods: In this retrospective cohort study in non-standardized daily clinical practice, we compared 273 IVF cycles from 195 women undergoing IVF at our center for luteal phase support with vaginal administration of 200 mg of micronized progesterone three times daily or subcutaneous injection of 25 mg of progesterone per day. Results: Various patient characteristics including age, weight, height, number of oocytes, and body mass index were similar between both groups. We observed no significant differences in the clinical pregnancy rate (CPR) per treatment cycle between the subcutaneous (39.9%) and vaginal group (36.5%) (p=0.630). Covariate analysis showed significant correlations of the number of transferred embryos and the total dosage of stimulation medication with the CPR. However, after adjustment of the CPR for these covariates using a regression model, no significant difference was observed between the two groups (odds ratio, 0.956; 95% confidence interval, 0.512-1.786; p=0.888). Conclusion: In agreement with randomized controlled trials in study populations with strict selection criteria, our study determined that subcutaneous progesterone was equally effective as vaginally applied progesterone in daily clinical practice in an unselected patient population.

Leucocyte Promoting Factor of B. Pertussis Vaccine (백일해 예방약의 백혈구 증가 인자에 관한 연구)

  • It was well known that B. pertussis cells possess protective antigen, histamine sensitizing factor, heatstable and labile toxin, hemagglutinin, agglutinogen and the others. Previous reports involving above antigenic properties of B. pertussis have been carried out for several years. However, leucocyte promoting property was not yet investigated. In this report, the results of studies on the leucocytosis, particulary the lymphocytosis, produced in mice by injecting pertussis vaccine were presented. Especially leucocyte promoting property and histamine sensitizing property of B. pertussis vaccine treated at various temperatures were compared. The relationship between the leucocyte promoting property and histamine sensitizing property was investigated. Results were as follows. 1. Although leucocytosis was significantly rised in both 0.5ml injection and 0.1ml injection of pertussis vaccine than in control, at the higher dose (0.5ml injection) an elevation in white cell count was more significant. The leucocyte responce to pertussis vaccine was greater following 0.5 ml injection than following 0.1ml injection. 2. Lymphocytosis was significantly rised in both 0.5ml injection and 0.1ml injection of pertussis vaccine than in control. At higher dose (0.5ml injection), an elevation in lymphocyte count was more significant. 3. Order of elevation in differential leucocyte counts was lymphocyte, polymorphonuclear leucocyte and monocyte. 4. The leucocyte response to pertussis vaccine was 2 fold greater following intravenous injection than following subcutaneous injection. 5. Decrease leucocyte promoting activity and histamine sensitizing activity resulted from exposure to temperature above $56^{\circ}C$. Histamine sensitizing activity of pertussis vaccine treated at various temperatures paralleled leucocyte promoting activity.

Effects of Exogenous Bovine Somatotropin on Mammary Function of Late Lactating Crossbred Holstein Cows

  • The objective of the present study was to determine the effect of exogenous bovine somatotropin on the mammary function in late lactating crossbred Holstein cows. Twelve 87.5% late lactating Holstein cows, approximately 30 weeks postpartum, were divided into two groups of 6 animals each. Animals in the control group were given sodium bicarbonate buffer by subcutaneous injection, while animals in the treated group were given recombinant bovine somatotropin (bST) by subcutaneous injection with 500 mg of bST (14 day prolonged-release bST). After bST injection, milk yield significantly increased from the control level on day 8 to day 20 (p<0.05) with a concomitant increase in mammary blood flow (p<0.01). An increase in mammary blood flow in response to bST treatment was greater than an increase in milk production. An increased plasma concentration of IGF-I coincided with an increase in mammary blood flow in animals treated with bST. There were no significant changes in the concentration of arterial plasma glucose concentration, the arteriovenous concentration difference (A-V difference) and mammary extraction ratio while the mammary glucose uptake increased when compared to the control group. The concentration of arterial plasma triglyceride decreased throughout the experimental period in animals give bST. The plasma concentration of acetate, and the mammary uptake for acetate significantly increased (p<0.05) after bST treatment. The action of bST did not affect the plasma concentration, A-V difference and extraction ratio across the mammary gland for $\beta$-hydroxybutyrate. The concentrations of milk fat and lactose tended to increase during bST treatment. Milk protein concentration initially increased in the first few days and decreased after bST injection when compared to the pretreated period. The present results indicated that bST could affect the mammary function in late lactating cows by increase in milk yield involving changes in both extra-mammary and intra-mammary mechanisms. The exogenous bST exerted its galactopoietic action through an increase in circulating IGF-I of the late lactating Crossbred Holstein cattle.

Compression of The Trigeminal Ganglion Enhances Nociceptive Behavior Produced by Formalin in The Orofacial Area of Rats

  • The present study investigated inflammatory hypersensitivity following compression of the trigeminal ganglion in rats. Experiments were carried out on male Sprague-Dawley rats weighing 250-260 g. Under anesthesia, rats were mounted on a stereotaxic frame and injected with $8{\mu}L$ of 4% agar solution through a stainless steel injector to compress the trigeminal ganglion. In the control group, rats underwent a sham operation without agar injection. Injection sites were examined with a light micrograph after compression of the trigeminal ganglion. Air-puff thresholds (mechanical allodynia) were evaluated 3 days before surgery and 3, 7, 10, 14, 17, 21, 24, 30, and 40 days after surgery. Air-puff thresholds significantly decreased after compression of the trigeminal ganglion. Mechanical allodynia was established within 3 days and remained strong over 24 days, returning to preoperative levels approximately 40 days following compression. After subcutaneous injection of 5% formalin ($50{\mu}L$) in the compression of the trigeminal ganglion-treated rats, nociceptive scratching behavior was recorded for 9 successive 5-min internals. Injection of formalin into the vibrissa pad significantly increased the number of scratches and duration of noxious behavioral responses in sham-treated rats. Noxious behavioral responses induced by subcutaneous formalin administration were significantly potentiated in rats with trigeminal ganglion compression. These findings suggest that compression of the trigeminal ganglion enhanced formalin-induced infla-mmatory pain in the orofacial area.

Nicolau Syndrome after Intramuscular Injection: 3 Cases

  • Nicolau syndrome is a rare complication of intramuscular injection consisting of ischemic necrosis of skin, soft tissue, and muscular tissue that arises locoregionally. The characteristic pattern is pain around the injection site, developing into erythema, a livedoid dermatitis patch, and necrosis of the skin, subcutaneous fat, and muscle tissue. Three patients were injected with drugs (diclofenac sodium, ketoprofen, meperidine) for pain relief. Three patients complained of pain, and a skin lesion was observed, after which necrosis developed on their buttocks. Each patient underwent debridement and coverage. The wound healed uneventfully. We report three cases of Nicolau syndrome in the buttocks following diclofenac intramuscular injection.

  • The Present study was examined the response of mice of 5 strains to superovulation treatment. Female mice of CF#1, C57BL/6, ICR, dd and CHN strains, aged 60 days were used. Superovulation was induced by the subcutaneous injection of 5 IU PMS and followed 48 hours later by the subcutaneous injection of 5 IU HeG. All females were killed 24 hours after RCG and then ova in the oviduct were counted under a microscope. The average number of ova ovulated in CF#1, C57BL/6, ICR, dd and CHN were 21.3, 17.9, 20.7, 21.0 and 20.5, respectively.

  • We have developed an injectable thermosensitive hydrogel for local drug delivery to treat cancers clinically. We selected chitosan as a polymer matrix because of its biocompatibility and biodegradability. Glycerol 2-phosphate disodium salt hydrate (${\beta}$-GP) was used to neutralize the chitosan solution to physiological pH. The chitosan solution displayed a sol-gel phase transition in a pH-and temperature-dependent manner and formed an endothermic hydrogel after subcutaneous injection into mouse in the presence of ${\beta}$-GP. Additionally, we evaluated the biodegradation of chitosan hydrogel in mice by measuring the volume of injected chitosan hydrogel after subcutaneous injection. The injected chitosan hydrogel in mice was sected and stained with hematoxylin-eosin reagent for histological observation to confirm biodegradation of the hydrogel by the infiltrated cells. Chitosan hydrogel systems that possess biocompatibility and biodegradability could be promising thermosensitive injectable materials useful as depot systems for local anti-cancer drug delivery.

Effect of Endurance Exercise during Dexamethasone Treatment on the Attenuation of Atrophied Hindlimb Muscle Induced by Dexamethasone in Rats (Dexamethasone 치료기간중의 지구력 운동이 dexamethasone에 의해 유발된 쥐의 뒷다리근 위축경감에 미치는 영향)

  • The purpose of this study was to determine the effect of regular exercise during dexamethasone injection on the body weight, weight of hindlimb muslces, myofibrillar protein content and glutamine synthetase activity. 180-200g female Wistar rats were divided into four groups : control, exercise, dexamethasone injection (dexa), and exercise during dexamethasone injection(D+E) group. The dexa group received daily subcutaneous injection of dexamethasone at a dose of 4mg/kg body weight for 7 days. The exercise group ran on a treadmill for 60min/day(20minutes every 4 hours) at 10m/min and a 10$^{\circ}$grade. The control group received daily subcutaneous injection of normal saline at a dose of 4mg/kg body weight for 7 days. The D+E group ran on a treadmill for 60min/day(20minutes every 4 hours) at 10m/min and a 10$^{\circ}$ grade during dexamethasone injection. Body weight of the control group increased significantly from days of experiment, that of the dexa group decreased significantly from day 4 of the experiment resulting in a 82.4% decrease compared to the first day of the experiment. Body weight of the D+E group decreased significantly from day 5 of experiment resulting in a 81.77% decrease comprared to the first day of the experiment. Body weights, muscle weight and myofibrillar protein content of the plantaris and gastrocnemius decreased significantly and muscle weight of the soleus tended to decrease with dexamethasone injection. Glutamine synthetase activity of the hindlimb muscles increased significantly with the dexamethasone injection. The relative weight of the soleus was comparable to the control group and that of plantaris decreased significantly and that of gastrocnemius tended to decrease compared to that of the control in the dexa group. Body weight and muscle weight of the plantaris and gastrocnemius of the excrcise group were comparable to the control group, and the muscle weight of soleus showed a tendencey to increase. The relative weight of the soleus increased significantly and that of the plantaris and gastrocnemius were comparable to the control in the exercise group. Myofibrillar protein content of the soleus and plantaris increased significantly and there was no change of GS activity of the hindlimb muscles compared to the control in the exercise group. Body weight of the D+E group was comparable to the dexa group, muscle weight of the plantaris increased significantly and that of the soleus and gastrocnemius showed a tendency to increase. The relative weight of the hindlimb muscles increased significantly. Myofibrillar protein content of the soleus and plantaris increased significantly and that of the gastrocnemius tended to increase compared to the dexa group. Body weight and muscle weight of the plantaris and gastrocnemius of the D+E group did not recover to that of the control group. Muscle weight of the soleus recovered to that of the control group. The relative weight and of myofibrillar protein content of the hindlimb muscles recovered to that of the control group. From these results, it is suggested that regular exercise during dexamethasone injection might attenuate the muscle atrophy of the hindlimb muscles.

  • The purpose of this study was to determine the effect of regular exercise during dexamethasone injection on the body weight, weight of hindlimb muscles and adrenal gland in Young rats. 80-100g Wistar rats were divided into control, exercise, dexamethasone injection(dexa), and exercise during dexamethasone injection(D+E) group. The dexa group received daily subcutaneous injection of dexamethasone at a dose of 5mg/kg body weight for 10 days. The exercise group ran on a treadmill for 60min /day(20 minutes every 4 hour) at l0m/min and a 10$^{\circ}$ grade. The control group received daily subcutaneous injection of normal saline at a dose of 5mg /kg body weight for 10 days. The D+E group ran on a treadmill for 60min /day (20 minutes every 4 hour) at 10m/min and a 10$^{\circ}$ grade. Body weight of both control and exercise group increased significantly until 10 days, that of both dexa and D+E group decreased significantly, resulting in 79.47 and 78.75% decrease respectively compared to the first day of experiment. Body weight and muscle weight of the soleus, plantaris and gastrocnemius decreased significantly with dexamethasone injection. Relative weight of the plantaris and gastrocnemius of the dexa group decreased significantly compared to that of the control group. Body weight and muscle weight of the gastrocnemius of the exercise group increased significantly, and the muscle weight of the soleus and plantaris tended to increase. The Relative weight of the plantaris was comparable to the control group and that of the soleus and gastrocnemius tended to increase in the exercise group. Body weight and muscle weight of the soleus and plantaris of the D+E group showed a tendency to increase, and muscle weight of the gastrocnemius increased significantly compared to the dexa group. The Relative weight of the soleus and gastrocnemius tended to increase, and that of the plantaris of the D+E group increased significantly compared to the dexa group. Body weight, muscle weight and relative weight of the soleus, plantaris and gastrocnemius of the D+E group did not recover to that of the control group. Adrenal gland weight of the dexa and D+E group tended to increase, and that of the exercise group increased significantly. From these results, it can be suggested that regular exercise during dexamethasone injection might attenuate the decrease of body weight and hindlimb muscle weight induced by the dexamethasone injection.
