Hypoadrenocorticism in a 1-Year-Old Korean Shorthair Cat

  • A one-year-old spayed female Korean Shorthair cat presented to Kangwon National University Veterinary Hospital with vomiting, weight loss, lethargy, loss of appetite, and polyuria that lasted for more than two weeks. The body condition score, blood pressure, heart rate, and body temperature were abnormally low, and the physical examination findings were consistent with moderate dehydration. Hematological and biochemical tests demonstrated mild azotemia and a low Na:K ratio. Additional abdominal ultrasound imaging revealed reduced size of both adrenal glands. The adrenocorticotropic hormone (ACTH) stimulation test showed decreased post-ACTH cortisol and aldosterone levels and increased endogenous ACTH levels, confirming a diagnosis of primary hypoadrenocorticism. The cat was treated with subcutaneous injections of desoxycorticosterone pivalate (DOCP) and oral prednisolone supplementation, and subsequent electrolyte analysis showed a normal Na:K ratio. Clinical symptoms were also improved in response to treatment. Hypoadrenocorticism in cats is a very rare disease, but it should not be excluded as a potential diagnosis in favor of kidney diseases or other conditions, especially when the Na:K ratio is low. In addition, the prognosis for the disease and the response to DOCP treatment should be further evaluated in cats.

Synergistic Effects of Cisplatin-epigel and Interstitial KTP Laser Treatment on a Xenografted Squamous Cell Carcinoma

  • Background and Objectives Cisplatin is an important chemotherapy drug for the treatment of head and neck cancer. Interstitial laser treatment (ILT) has cosmetic utility and is very important for maintaining the function of the head and neck after cancer treatment. This study examined the synergistic effects of locally injected cisplatin-epigel and high fever induced by an interstitial potassium titanyl phosphate (KTP) laser treatment on a xenografted human Heinz squamous cell carcinoma. Materials and Methods SNU-1041 (107 cells/0.1 ml) cells were xenografted into the back of nude mice by subcutaneous injection. The ILT group (n = 10) was treated with a KTP laser (1 J/mm3) through a cylindrical diffuser tip inserted into the tumor, monitoring the temperature at 43-45℃. In the combined treatment group (n = 10), local hyperthermia was induced by intratumoral injection of 100-200 ㎍ of cisplatin into a collagen-based gel carrier (cisplatin-epigel), which was released slowly four hours before ILT. After four weeks of follow-up, the treated tumors were evaluated for tumor remission and volume change. Results Eight (80%) of the combined group showed complete tumor remission at the four-week follow-up, whereas only three (30%) of the ILT group showed remission (30%) (p < 0.01). Conclusion The current study has shown the synergistic effects of a local cisplatin injection and high fever from ILT on a xenografted human Heinz squamous cell carcinoma.

Herba Ephedrae and Rhizoma Amorphophalli modulates visceral obesity in micro-CT of high fat induced obese male mice (고지방식이 수컷 마우스 비만모델에서 micro-CT를 이용한 마황(麻黃)과 마우(魔芋)의 복부비만 조절효과)

  • Objectives : We investigated the effects of Herba Ephedrae and Rhizoma Amorphophalli on high fat diet induced obese male mice. Methods : 8 weeks old, high fat diet induced obese male mice were divided into 5 groups: C57BL/6 normal control, obese vehicle control, GGEx55 (Herba Ephedrae), GGEx61 (Rhizoma Amorphophalli), GGEx62 (Herba Ephedrae + Rhizoma Amorphophalli). After mice were treated with GGEx for 8 weeks, we measured body weight gain, food intake, feeding efficiency ratio, rectal temperature, fat weight, plasma leptin and lipid levels. We also took micro-computerized axial tomography (micro-CT) on the mice. Results : 1. GGEx55 and GGEx62 groups significantly decreased body weight gain and feeding efficiency ratio compared with vehicle control. But they significantly increased rectal temperature. 2. Plasma total cholesterol and LDL-cholesterol concentrations were significantly increased by GGEx55 groups, whereas were significantly decreased by GGEx62 groups compared with vehicle control. 3. GGEx55 and GGEx62 groups significantly decreased total, subcutaneous and visceral fat as well as fat areas in micro-CT analysis of abdomen compared with vehicle control. 4. Plasma GOT and GPT concentrations were significantly increased by GGEx55 groups compared with vehicle control. Conclusions : These results demonstrate that GGEx55 and GGEx62 effectively reduces body weight gain, feeding efficiency ratio in high fat diet induced obese mice, leading to the modulation of obesity. In addition, GGEx55 and GGEx62 decreases visceral adipose tissue mass and improves plasma lipids, suggesting that GGEx55 and GGEx62 may act as a therapeutic agent for obesity.

  • Through the study on judgment of Body form and settle Energy flow(立形定氣) before diagnose the patients, the results are as follows. The observation of the body form is to determine prosperity and deficiency of each internal organ. It is necessary to distinguish Body form loss(形脫) and Body form fullness(形充). Fat man(肥人), Thin man(瘦人), Creamy man(膏人), Muscular man(肉人), Small Fat man(脂人) are discriminated by fat distribution, fat content, and muscle mass. The observation of the body form means the observation of structure disorder, color change, develop part at body, head and face. The observation of the body form that is to determine prosperity and deficiency of each internal organ is from the limited knowledge of the anatomy. The observation of face color is considered by blood perfusion, blood oxygenation and accumulation of carotinoid, bilirubin and change of melanin in the facial skin. The prosperity and the deficiency of energy flow is considered by symptom combined with growth (<40 years) and aging (>40 years). The prosperity of energy flow includes the anger, anxious emotion and the deficiency of energy flow includes the fear, depressive emotion. The breathing type is expiratory exhalation like asthma patients in the prosperity of energy flow. The deficiency of energy flow is weakness to overcome the disease. The prosperity and the deficiency of energy flow are considered by body metabolic ratios (Basal metabolic Rate: BMR, Resting metabolic rate: RMR, Physical activity ratios: PASs). Development of subcutaneous fat is good in the person of prosperous energy flow. The person of prosperous energy flow is hard to overcome to heat weather than cold weather. The person of deficiency of energy flow has tendencies of low blood pressure, insufficiency of blood flow in the peripheral and being shocked. The person of deficiency of energy flow has tendencies of chronic fatigue syndrome or automatic nerve disorder. If the patient who has deficiency of energy flow has severe weight loss should be checked for the presence of disease. The observation of small and large of bone is to check the development and disorder of bone growth and aging. The observation of thickness and weakness of muscle is to check the development of muscle, particularly biceps, gastrocnemius, and rectus abdominal muscle. The observation of thickness and weakness of skin is to check the ability of regulating body temperature by sweating.

Studies on the Nutritional Status and Energy Balance of Korean Women Divers (한국 해녀의 영양섭취 상태 및 에너지 균형에 관한 연구)

  • This study was attempted to investigate Korean women divers' nutritional status and the effect of their putting on rubber diving suits on the energy balance. For these investigations, measurements of nutrients intake, energy intake and energy consumption during diving work were performed. This study was conducted in summer and winter of 1981. Nutrition survey was carried out by interviewing each individual and the energy consumption during diving work was calculated from the extra oxygen consumption over the resting value and the change in mean body temperature in sea water. The results obtained were summarized as follows : 1) In summer, the divers' dietary intake were 2,454 Kcal for energy, 69g of protein, 16g of fat 578mg of calcium, 11mg of iron, 526 Retinol Equivalent of vitamin A, 1.1mg of thiamin, 1.3mg of riboflavin, 19mg of niacin and 50mg of ascorbic acid. These results showed that the intakes of energy, thiamin, riboflavin, niacin and ascorbic acid were higher than the Korean Recommended Dietary Allowances (RDA). Whereas in winter, the nutrients intake except iron, riboflavin, and vitamin A were higher than the RDA. 2) The energy consumption, measured by oxygen consumption and changes in body heat content, used in diving work were 260 Kcal per day in summer and 370 Kcal per day in winter. These values were about 600-700 Kcal per day lower than 1960s' 1,000 Kcal per day. The divers' energy intake were 2,454 Kcal per day in summer and 2,487 Kcal per day in winter. These data were about 550 Kcal per day lower than 1960s' intake. Since divers have worn the rubber diving suits, the energy consumption decreased as compared with the energy consumed while wearing cotton suits. And this seemed to be resulted in decreasing the energy intake. This result also showed that the balance between energy expenditure and energy intake were kept. 3) The mean subcutaneous fat thickness of divers and nondivers were 8.85 and 9.03 mm respectively. These values were as high as four times as those of 1960s'. The total body fat contents wers 25.8 percent in both diver and nondiver groups and showed an increase as high as twice as compared with 1960s' values.

Effect of Thyroid Hormones on the Redox Balance of Broiler Chickens

  • In the present study, two trials were conducted to evaluate the effects of hyper- and hypothyroid status on the redox balance of broiler chickens. In Trial 1, 3 groups of broiler chickens were randomly subjected to one of the three treatments: subcutaneous administration of triiodothyronine (T3, $150{\mu}g/kg$ BW), methimazole (MMI, 150 mg/kg BW), or saline. The blood, liver and heart were sampled at 3 h after injection. In Trial 2, three groups of 20 broiler chickens were randomly fed with one of the three diets: control, dietary supplementation of T3 (1.5 mg/kg diet) or MMI (1 g/kg diet) for 7 days. In trial 1, the plasma concentrations of T3 and T3 to thyronine ratio (T3/T4) were significantly increased by T3 injection. Plasma levels of thiobarbituric acid reacting substances (TBARS) tended to be increased (p = 0.067) by both T3 and MMI treatments while the ferric reduced/antioxidant capacity (FRAP) was increased only by MMI treatment. Acute T3 treatment had no significant effect on the activities of superoxide dismutase (SOD) and the concentrations of FRAP and TBARS in either liver or heart tissue. In contrast, the hepatic activities of SOD were decreased (p<0.05) while the cardiac levels of FRAP were significantly increased (p<0.0001) by MMI treatment. In chronic treatments, the rectal temperature of chickens was significantly decreased (p<0.05) by MMI treatment. The circulating T3 levels were significantly increased (p<0.05) by long-term T3 treatment, and showed a trend to decrease in MMI treatment. The plasma concentrations of TBARS were significantly (p<0.05) increased by MMI treatment. All the redox parameters measured in either liver or heart were not significantly altered by either long-term T3 or MMI treatment except that the hepatic SOD activities were significantly augmented by T3 treatment. The result showed that neither acute nor long-term elevation of circulating T3 levels induced lipid peroxidation in broiler chickens. The enhanced enzymatic antioxidant system (SOD in cardiac tissue) may be involved in the protection of the bird to increased oxidative challenge. The responses of redox balance to changed thyroid state seem to be tissue specific.

Study of Bio-absorbability and Bio-compatibility of Poly-L-lactic-acid Implant in Dogs (개에서 Poly-L-lactic-acid 이식물의 생분해성과 생체적합성에 관한 연구)

  • Bioabsorbable devices have been utilized and experimented in many aspects of orthopaedic surgery. Depending upon their constituent polymers, these materials can be tailored to provide sufficient rigidity to allow bone healing, retain mechanical strength for certain period of time, and then eventually begin to undergo degradation. The objective of this study was to estimate extent in which Poly-L-latic acid (PLLA) implants had bioabsorbability and biocompatibility with bone and soft tissue in dogs and also to develop bioabsorbable, biocompatible materials with the appropriate strength and degradation characteristics to allow for regular clinical use for treating orthopedic problems in humans as well as animals. Eighteen dogs were used as experimental animals and were inserted two types of PLLA implants. PLLA rods were inserted into subcutaneous tissue of back or the abdomen wall. And the rods were tested for material properties including viscosity, molecular weight, melting point, melting temperature, crystallinity, flexural strength, and flexural modulus over time. PLLA screws were inserted through cortical bone into bone marrow in the femur of the dogs and stainless steel screw was inserted in the same femur. Radiographs were taken after surgery to observe locations of screw. Histological variations including cortical bone response, muscular response, bone marrow response were analyzed over the time for 62weeks. The physical properties of PLLA rods had delicate balances between mechanical, thermal and viscoelastic factors. PLLA screws did not induce any harmful effects and clinical complications on bone and soft tissue for degradation period. These results suggest that PLLA implants could be suitable for clinical use.

Evaluation of 99mTc-MAG3-2-nitroimidazole for hypoxic tumor imaging

  • 2-Nitroimidazole derivatives have been reported to accumulate in hypoxic tissue. We prepared a novel $^{99m}Tc-MAG_3$-2-nitroimidazole and evaluated the feasibility for hypoxia imaging agent. $Bz-MAG_3$-2-nitroimidazole was synthesized by direct coupling of $Bz-MAG_3$ and 2-nitroimidazole using dicyclohexylcarbodiimide. $Bz-MAG_3$-2-nitroimidazole was labeled with $^{99m}Tc$ in the presence of tartaric acid and $SnCl_2-2H_2O$ at $100^{\circ}C$ for 30 min. And the reaction mixture was purified by $C_{18}$ Sep-pak cartridge. The labeling efficiency and the radiochemical purity were checked by ITLC-SG/acetonitrile. The tumor was grown in balb/c mice for 8~13 days after the subcutaneous injection of tumor cells, CT-26 (murine colon adenocarcinoma cell). Biodistribution study and tumor autoradiography were performed in the xenografted mice after i.v injection of 74 kBq/0.1 mL and 19 MBq/0.1 mL of $^{99m}Tc-MAG_3$-2-nitroimidazole, respectively. In vivo images of $^{99m}Tc-MAG_3$-2-nitroimidazole in tumor bearing mice were obtained 1.5 hr post injection. The labeling efficiency was $45{\pm}20%$ and the radiochemical purity after purification was over 95%. Paper electrophoresis confirmed negative charge of $^{99m}Tc-MAG_3$-2-nitroimidazole. $^{99m}Tc-MAG_3$-2-nitroimidazole was very stable at room temperature and its protein binding was 53%. The $^{99m}Tc-MAG_3$-2-nitroimidazole exhibited high uptake in the liver, stomach and intestine. In biodistribution study using tumor bearing mice, the uptakes (% ID/g) of the tumor were $0.5{\pm}0.1$, $0.4{\pm}0.0$, $0.2{\pm}0.1$ and $0.1{\pm}0.1$ at 5, 15, 30 min and 4 hrs. Tumor/muscle ratio were $1.4{\pm}0.1$, $2.2{\pm}0.83$, $3.0{\pm}0.9$, and 3.7 (n=2) for 5, 15, 30 min and 4 hrs. The uptake in hypoxic area was found higher than in non-hypoxic area of tumor tissue by autoradiography. In vivo images showed the relatively faint uptake to the hypoxic tumor region. $^{99m}Tc-MAG_3$-2-nitroimidazole was successfully synthesized and found feasible for imaging hypoxia.
