Fiber-optic fluoroimmunosensor for foodborn pathogens using an optical evanescent field (광섬유 소산장을 이용한 식중독균 신속검출용 형광면역센서)

  • In this study, the fiber-optic fluoro-immunosensor was designed to detect foodborne pathogens. The fabricated system is composed of the multimode optical fiber on which antibodies are immobilized. Then, a sandwich immunoassay is applied to the fabricated the fiber-optic fluoro-immunosensor. In the "sandwich" binding format, a primary or "capture" antibody is immobilized on the core surface of the multimode optical fiber and a secondary or named as "tracer" antibody is added to the bulk solution. A tracer is labeled FITC (fluorescein isothiocyanate; ${\lambda}ex$=492 nm, ${\lambda}em$= 520 nm). Different concentrations of antigens are tested in different fibers. The detection limit of the fabricated system is 5.08×103 cfu/ml for Vibrio antigen and $0.1{\mu}g/ml$, $0.05{\mu}g/ml$ in non-labeled monolayer phosphate buffered saline (NMP), non-labeled monolayer carbonate bicarbonate buffer (NMC), respectively.

  • Currently, wireless power transmission technology based on magnetic induction was employed in battery charger for smart phone application. The system consists of wireless power transmitter in base station and receiver in smart phone. Size and thickness of receiver was strictly limited in the newest smart phone. In order to achieve high efficiency of a tiny small wireless power receiver module, sub-millimeter thick electromagnetic wave shielding sheet having high permeability and Q was essential component. It was found that magnetic field from transmitter to receiver can be intensified by sufficient shielding cause to minimize leakage magnetic flux by those magnetic properties. This leads to high efficiency of wireless power transmission and protects crucial integrated circuit of main board from electromagnetic noise. The important soft magnetic materials were introduced and summarized for the current small-power wireless power charger and NFC application and mid-power home appliance and high-power automotive application in the near future.

A Meta-Analysis of Internet-Related Policy Research in Communication (인터넷 관련 규제정책 연구의 동향분석)

  • This study reviewed and ascertained policy researches relating to the Internet in the field of communication. With using a meta-analysis, this study attempts to classify 44 academic papers from 2000 to 2009 by research theme, methodology, theoretic rationale, related sub-theme, and targeted service and area. It is found that the themes of preceding researches examined herein were limited to the 4th domain in Developmental Model of Media Research. Such researches mainly relied on exploratory studies and only a few of them addressed theoretic rationales. Also, most of Internet regulation discussions focused on content regulation and general internet service. Lastly, researchers discussed laws having bearing on the Internet and social-cultural issues of the Internet.

A REVIEW OF CANDU FEEDER WALL THINNING

  • Flow Accelerated Corrosion is an active degradation mechanism of CANDU feeder. The tight bend downstream to Gray loc weld connection, close to reactor face, suffers significant wall thinning by FAC. Extensive in-service inspection of feeder wall thinning is very difficult because of the intense radiation field, complex geometry, and space restrictions. Development of a knowledge-based inspection program is important in order to guarantee that adequate wall thickness is maintained throughout the whole life of feeder. Research results and plant experiences are reviewed, and the plant inspection databases from Wolsong Units One to Four are analyzed in order to support developing such a knowledge-based inspection program. The initial thickness before wall thinning is highly non-uniform because of bending during manufacturing stage, and the thinning rate is non-uniform because of the mass transfer coefficient distributed non-uniformly depending on local hydraulics. It is obvious that the knowledge-based feeder inspection program should focus on both fastest thinning locations and thinnest locations. The feeder wall thinning rate is found to be correlated proportionately with QV of each channel. A statistical model is proposed to assess the remaining life of each feeder using the QV correlation and the measured thicknesses. W-1 feeder suffered significant thinning so that the shortest remaining life barely exceeded one year at the end of operation before replacement. W-2 feeder showed far slower thinning than W-1 feeder despite the faster coolant flow. It is believed that slower thinning in W-2 is because of higher chromium content in the carbon steel feeder material. The average Cr content of W-2 feeder is 0.051%, while that value is 0.02% for W-1 feeder. It is to be noted that FAC is reduced substantially even though the Cr content of W-2 feeder is still very low.

The first insight into the structure of the Photosystem II reaction centre complex at $6{\AA}$ resolution determined by electron crystallography

  • Electron crystallography of two-dimensional crystalsand electron cryo-microscopy is becoming an established method for determining the structure and function of a variety of membrane proteins that are providing difficult to crystallize in three dimension. In this study this technique has been used to investigate the structure of a ~160 kDa reaction centre sub-core complex of photosystem II. Photosystem II is a photosynthetic membrane protein consisting of more than 25 subunits. It uses solar energy to split water releasing molecular oxygen into the atmosphere and creates electrochemical potential across the thylakoid membrane, which is eventually utilized to generate ATP and NADPH. Images were taken using Philips CM200 field emission gun electron microscope with an acceleration voltage of 200kW at liquid nitrogen temperature. In total, 79 images recorded dat tilt angles ranging from 0 to 67 degree yielded amplitudes and phases for a three-dimensional map with an in-plant resolution of 6$\AA$ and 11.4$\AA$ in the third dimension shows at least 23 transmembrane helices resolved in a monomeric complex, of which 18 were able to be assigned to the D1, D2, CP47 , and cytochrome b559 alfa beta-subunits with their associated pigments that ae active in electron transport (Rhee, 1998, Ph.D.thesis). The D1/D2 heterodimer is located in the central position within the complex and its helical scalffold is remarkably similar to that of the reaction centres not only in purple bacteria but also in plant photosystem I (PSI) , indicating a common evoluationary origin of all types of reaction centre in photosynthetic organism known today 9RHee et al. 1998). The structural homology is now extended to the inner antenna subunit, ascribed to CP47 in our map, where the 6 transmembrane helices show a striking structural similarity to the corresponding helices of the PSI reaction centre proteins. The overall arrangement of the chlorophylls in the D1 /D2 heterodimer, and in particular the distance between the central pair, is ocnsistent with the weak exciton coupling of P680 that distinguishes this reaction centre from bacterial counterpart. The map in most progress towards high resolution structure will be presented and discussed.

Stem Rot of Convallaria keiskei Caused by Sclerotium rolfsii (Sclerotium rolfsii에 의한 은방울꽃 흰비단병)

  • Stem rot symptoms of Convallaria keiskei occurred sporadically in the herb exhibition field at Hamyang-gun Gyeongnam province in Korea. The typical symptom is water-soaking on the main stem, rotting, wilting, and blighting, which eventually leads to death of the plant. The sclerotia, which were white to brown in color, globoid in shape with size of 1~3 mm hyphal width of which was 4~9 ${\mu}m$, were formed over lesions and surface soil line. The optimum temperature for mycelial growth and sclerotia formation was $30^{\circ}C$ on PDA. The typical clamp connections were observed in the hyphae of the fungus grown on PDA. Mycological characteristics and pathogenicity to host plants indicated that the infected fungus was Sclerotium rolfsii Saccardo. This is the first report of stem rot on Convallaria keiskei caused by S. rolfsii in Korea.

  • Molecular simulation is an exceptionally useful method for predicting self-assembled structures in various macromolecular systems, enlightening the origins of many interesting molecular events such as protein folding, polymer micellization, and ordering of molten block copolymer. The length scales of those events ranges widely from sub-nanometer scale to micron-scale or to even larger, which is the main obstacle to simulate all the events in an ab initio principle. In order to detour this major obstacle in the molecular simulation approach, a molecular model can be rebuilt by sacrificing some unimportant molecular details, based on two different perspectives with respect to the resolution of model. These two perspectives are generally referred to as 'atomistic' and 'mesoscopit'. This paper reviews various simulation methods for macromolecular self-assembly in both atomistic and mesoscopic perspectives.

Ketamine-Induced Blood Pressure Lowering in the Rat (흰쥐에서 Ketamine에 의한 혈압하강)

  • Although ketamine has been used in the field of anesthetic medicine for its safety and favourable respiratory effects, the cardiovascular effects of ketamine is still controversial. To clarify the action and mechanism of ketamine upon cardiovascular system, arterial blood pressure, tension of aortic ring, left ventricular developed pressure and heart rate were measured in rats, Ketamine produced two types of effects on arterial blood pressure in anesthetized rats; monophasic effect (blood pressure lowering) and biphasic effect (initial transient blood pressure increasing following sustained lowering), The ketamine-induced lowering of aterial blood pressure showed a concentration-dependent manner, inhibited by the pretreament of $MgCl_2$ and potentiated by the pretreatment of $CaCl_2$. The ketamine-induced lowering of aterial blood pressure was suppressed by the pretreatment of nifedipine, verapamil or lidocaine. In phenylephrine-precontracted endothelium intact (+E) aortic rings, ketamine sometimes caused a small enhancement of contraction ($112.5{\pm}3.6{\%}$). However, in many experiments, ketamine produced a concentration-dependent relaxation in +E aortic rings precontracted with either phenylephrine or KCl. Ketamine-induced relaxation was significantly greater in KCl-precontracted strips than phenylephrine-precontracted strips. In phenylephrine-precontracted +E aortic rings, the ketamine-induced vasorelaxation was not suppressed by endothelium removal or by the pretreatment of a nitric oxide synthase inhibitors, L-$N^G$-nitro-arginine and a guanylate cyclase inhibitors, methylene blue, suggesting that the ketamine-induced vasorelaxation is not dependent on the endothelial function. In addition, ketamine elicited an increase in left ventricular developed pressure in perfused hearts accompanied by decrease in heart rate. These results suggest that ketamine could evoke a hypotension due to vasorelaxation and decrease in heart rate in rats. The inhibitory effect of cardiovascular system might be associated with modulation of $Ca^{2+}$ homeostasis.

Inhibitory effects of xylamine on the arterial contraction in rats (흰쥐 대동맥 수축에 대한 xylamine의 억제효과)

  • The therapeutic efficacy of xylamine in the field of psychological medicine has been recognized for years and the drug is used to treat depression and some other conditions, but little is known about its mechanism of action on vascular system. Therefore, the present study was designed to investigate the influence of xylamine on the contractile responses of isolated rat thoracic arteries to phenylephrine(PE) and potassium chloride(KCl). Xylamine produced a concentration-dependent relaxation in PE-precontracted endothelium intact(+E) rat aortic rings, but not in a KCl-precontracted aortic rings. Also, xylamine inhibited the PE-induced contraction in concentration-dependent manner, but not in the high KCl-induced contraction in +E rings. This concentration-dependent inhibition was suppressed by the removal of the endothelium (-E). The inhibitory effects of xylamine($0.3{\mu}M$) on the PE-induced contractions were suppressed by N(G)-nitro-L-arginine(L-NNA), N(omega)-nitro-L-arginine methyl ester(L-NAME), aminoguanidine, dexamethasone, methylene blue, 1H-[1,2,4]oxadiazolo [4,3-a]quinoxalin-1-one(ODQ), indomethacin, ryanodine, tetrabutylammonium(TBA), lidocaine, procaine and 0 mM extracellular $Na^+$, but not by 2-nitro-4-carboxyphenyl-n,n-diphenylcarbamate(NCDC), lithium, nifedipine, verapamil, 0 mM extracellular $Ca^{2+}$, glibenclamide and clotrimazole. These findings suggest that xylamine could act as a vasorelaxant and direct inhibitor of arterial contraction. This vasorelaxation involves an endothelial nitric oxide (NO)/cGMP (guanosine 3',5'-cyclic monophosphate) pathway or cyclooxygenase system, and an interference with $Ca^{2+}$ release, TBA-sensitive $Ca^{2+}$-activated $K^+$ channels and $Na^+$$ channels.

Application of Modified-WASP5 for Daecheong Dam Watershed Management (대청댐 유역관리를 위한 수정-WASP5 모형의 적용)

  • The objective of this study was to develop and apply a water quality simulation model for the evaluation of ungaged watershed. The Modified WASP5 consisted of three sub-models, LOAD-M, DYN-M, and EUT-M. LOAD-M, an empirical model, estimates runoff loadings using point and non-point source data of villages. Daecheong Dam watershed was selected for the research to calibrate, verify and application of Modified-WASP5. LOAD-M model was established using field data collected from all items of water quality and water quantity gaging stations of the watersheds, and was applied to the ungauged watersheds, taking the watershed properties under consideration. The result of water quality simulation using ModifiedWASP5 shows that the observed BOD data of Yongpo and Daechong Dam in 1999 were 0.8 mg/L and 1.0 mg/L, and simulated data were 0.9 mg/L and 0.9 mg/L, respectively. In case of 1999, average BOD concentrations were 0.8 mg/L and 1.0 mg/L. Simulated concentration showed 1.1 mg/L and 1.5 mg/L, respectively. Generally, the simulation results were in good agreement with the observed data. This study was focused on formulating an integrated model for evaluating ungauged watersheds. Even though simulation results varied slightly due to limited availability of data, the model developed in this study would be a useful tool for the assessment and management of ungauged watersheds.
