수공구조물 여유고 산정을 위한 파랑모형의 적용성 검토 (Review on Application of Wave Model for Calculation of Freeboard in Hydraulic Structure)

  • Most of dams and reservoirs were made from natural materials, such as soil, sand and gravel. This type of hydraulic structure has the danger of collapse by overflow during a flood. Freeboard is the vertical distance between the crest of the dam and the full supply level in the reservoir. It must be sufficient to prevent overtopping from over flow. Thus, freeboard determination involves engineering judgment, statistical analysis, and consideration of the damage that would result from the overtopping of a hydraulic structure. This study attempts to calculate the wave height in dam, which is needed for the determination of the freeboard of the dam. Chung-ju dam is selected as the study area. Using the empirical formulas, the wave heights in dam were calculated, and the results were compared with those by the SWAN model, which is a typical wave model. The difference between the calculated results from the empirical formulas and those by the SWAN model is considerably large. This is because empirical equations consider only fetch or fetch and wind velocity, while the SWAN model considers depth and topography data as well.

NMR Studies of Ni-binding Luteinizing Hormone Releasing Hormone

  • Luteinizing Hormone Releasing Hormone (LHRH) is composed of 10 amino acids, and is best known as a neurotransmitter. Because of the 80% homology in animals, much more concerns have focused on the substances that have similar functions or can control LHRH. Ni, Cu-LHRH complexes were synthesized. The degree of complexation was monitored by $^1H,\;^{13}C$-NMR chemical shifts, and final products were identified by ESI-Mass spectrum. Solution-state structure determination of Ni-LHRH complex was accomplished by using NMR results and NMR-based distance geometry (DG). Interproton distances from nuclear Overhauser effect spectroscopy (NOESY) were utilized for the molecular structure determination. Results were compared with previous structures obtained from energy minimization and other spectroscopic methods. Structure obtained in this study has a cyclic conformation which is similar to that of energy minimized, and exhibits a specific a-helical turn with residue numbers (2~7) out of 10 amino acids. Comparison of chemical shifts and EPR studies of Ni, Cu-LHRH complexes exhibit that Ni-LHRH complex has same binding sites with the 4-coordination mode as in Zn-LHRH complex.

  • In this paper, we have studied about the determination methode of the operating frequency, and the design technique for the self-exited electronic ballast. Because we use the saturation characteristics of the ferrite core, the variation of the B/H curve characteristics for the selected core in the most important factor for the stabilization. Although it is impossible to fix the operation frequency, the self-exited electronic ballast is widely used because of its cost and the simplicity of its structure.

Practices for Readiness of Future Specialists for Professional Self-Determination in the Information Society

  • Professional self-determination of the individual is a complex and lengthy process of finding and realizing yourself in the profession. The main goal of professional self-determination is clarified. The basic concepts of readiness for professional self-determination of future specialists in the modern information society are revealed. The following approaches to the consideration of the concept of readiness are defined: functional-psychological, personal, activity-based. Based on the components of readiness identified by the researchers, it can be assumed that the structure of professional self-determination of the future specialist contains motivational, cognitive and activity components. Self-determination is defined as a multidimensional process that can be considered from different points of view: as a series of tasks, that society sets for the emerging individual, and which the individual must solve in a certain period. As a process of step-by-step decision-making, with the help of which the individual forms a balance between his desires and inclinations, on the one hand, and the needs of society, on the other; as a process of forming an individual lifestyle, part of which is professional activity. A number of tasks of professional self-determination of a future specialist in the information society are formulated. Diagnostic practices for determining the degree of readiness of future specialists for future professional success are characterized. Practices are developed as a basis for creating an individually oriented correctional and development program to promote the formation of future specialists' focus on future professional success. Their task is to ensure control over the dynamics of this process, assess the effectiveness of this career guidance work. Practices are aimed at identifying the degree of thorough knowledge of the conditions for achieving professional success in the chosen field of activity among future specialists.

Solid-state NMR Studies of Membrane Proteins Using Phospholipid Bicelles

  • Membrane proteins in highly oriented lipid bilayer samples are useful for membrane protein structure determination. We used in the past planar lipid bilayers which were aligned and supported on the glass slide. These samples were mechanically aligned in a magnetic field. However, these stacks of glass slides with planar lipid bilayers are not well suited for use with a commercial solid-state NMR probe with a round coil. Therefore, a homebuilt solid-state NMR probe was built and used with a stack of thin glass plates wherein the RF coil was wrapped directly around the flat square sample. Recently, we began to use magnetically aligned bicelles that are suitable for the structure determination of membrane proteins by solid-state NMR spectroscopy without any effort to build a flat square coil probe. These bicelle samples are well suited for use with a commercial solidstate NMR probe with a round coil, are very easy to prepare and are very stable, so that they can be kept for more than a year. In this paper, we present the solid-state NMR spectra of optimized and magnetically oriented bicelle samples of membrane proteins.

Expression and Preparation of Periostin FAS1 Domains for NMR Structure Determination

  • Periostin, a component of extracellular matrix (ECM) protein, is produced and secreted by the fibroblasts that are involved in chronic allergic inflammation diseases and various types of human cancers. Periostin protein is composed of multiple domains including four FAS1 domains which play important roles in cell adhesion and tumor metastasis by interacting with integrins. In spite of their important biological role, the structural information of periosin FAS1 domains was not revealed yet. Recently we systemically prepared various constructs of the FAS1 domains and tried to express them in E. coli. Of them, only single FAS1-II and -IV domains were highly soluble. Circular dichroism (CD) and nuclear magnetic resonance (NMR) studies revealed that the FAS1-IV domain might be suitable for three-dimensional structure determination using NMR spectroscopy.

  • A key parameter in the design of a laterally loaded pile is the determination of its performance level. Performance level of a pile is usually expressed as the maximum head deflection and bending moment. In general, uncertainties in the performance of a pile originates from many factors such as inherent variability of soil properties, inadequate soil exploration programs, errors taking place in the determination of soil parameters, limited calculation models as well as uncertainties in loads. This makes it difficult for practicing engineers to decide for the reliability of laterally loaded piles both in cohesive and cohesionless soils. In this paper, limit state functions and consequent performance functions are obtained for single concrete piles to predict the maximum bending moment, a widely accepted design criterion along with the permissible pile head displacement. Analyses were made utilizing three dimensional finite element method and soil-structure-interaction (SSI) effects were accounted for.

  • Three typical solvent suppression methods employed for measuring the NOE data that are used for structure determination of polypeptides by modeling were discussed and compared with one another. In the experiments with several peptides composed of 10 to 65 amino acids the presaturation method was found to give severely distorted signal intensities of exchangeable protons, thus making the results based on this method much less reliable.

An inverse determination method for strain rate and temperature dependent constitutive model of elastoplastic materials

  • With the continuous increase of computational capacity, more and more complex nonlinear elastoplastic constitutive models were developed to study the mechanical behavior of elastoplastic materials. These constitutive models generally contain a large amount of physical and phenomenological parameters, which often require a large amount of computational costs to determine. In this paper, an inverse parameter determination method is proposed to identify the constitutive parameters of elastoplastic materials, with the consideration of both strain rate effect and temperature effect. To carry out an efficient design, a hybrid optimization algorithm that combines the genetic algorithm and the Nelder-Mead simplex algorithm is proposed and developed. The proposed inverse method was employed to determine the parameters for an elasto-viscoplastic constitutive model and Johnson-cook model, which demonstrates the capability of this method in considering strain rate and temperature effect, simultaneously. This hybrid optimization algorithm shows a better accuracy and efficiency than using a single algorithm. Finally, the predictability analysis using partial experimental data is completed to further demonstrate the feasibility of the proposed method.

Sample size calculations for clustered count data based on zero-inflated discrete Weibull regression models

  • In this study, we consider the sample size determination problem for clustered count data with many zeros. In general, zero-inflated Poisson and binomial models are commonly used for zero-inflated data; however, in real data the assumptions that should be satisfied when using each model might be violated. We calculate the required sample size based on a discrete Weibull regression model that can handle both underdispersed and overdispersed data types. We use the Monte Carlo simulation to compute the required sample size. With our proposed method, a unified model with a low failure risk can be used to cope with the dispersed data type and handle data with many zeros, which appear in groups or clusters sharing a common variation source. A simulation study shows that our proposed method provides accurate results, revealing that the sample size is affected by the distribution skewness, covariance structure of covariates, and amount of zeros. We apply our method to the pancreas disorder length of the stay data collected from Western Australia.
