Integrated Bioinformatics Approach Reveals Crosstalk Between Tumor Stroma and Peripheral Blood Mononuclear Cells in Breast Cancer

  • Breast cancer is now the leading cause of cancer death in women worldwide. Cancer progression is driven not only by cancer cell intrinsic alterations and interactions with tumor microenvironment, but also by systemic effects. Integration of multiple profiling data may provide insights into the underlying molecular mechanisms of complex systemic processes. We performed a bioinformatic analysis of two public available microarray datasets for breast tumor stroma and peripheral blood mononuclear cells, featuring integrated transcriptomics data, protein-protein interactions (PPIs) and protein subcellular localization, to identify genes and biological pathways that contribute to dialogue between tumor stroma and the peripheral circulation. Genes of the integrin family as well as CXCR4 proved to be hub nodes of the crosstalk network and may play an important role in response to stroma-derived chemoattractants. This study pointed to potential for development of therapeutic strategies that target systemic signals travelling through the circulation and interdict tumor cell recruitment.

Tumor Stroma as a Therapeutic Target for Pancreatic Ductal Adenocarcinoma

  • Pancreatic ductal adenocarcinoma (PDAC) has a poor prognosis owing to its desmoplastic stroma. Therefore, therapeutic strategies targeting this tumor stroma should be developed. In this study, we describe the heterogeneity of cancer-associated fibroblasts (CAFs) and their diverse roles in the progression, immune evasion, and resistance to treatment of PDAC. We subclassified the spatial distribution and functional activity of CAFs to highlight their effects on prognosis and drug delivery. Extracellular matrix components such as collagen and hyaluronan are described for their roles in tumor behavior and treatment outcomes, implying their potential as therapeutic targets. We also discussed the roles of extracellular matrix (ECM) including matrix metalloproteinases and tissue inhibitors in PDAC progression. Finally, we explored the role of the adaptive and innate immune systems in shaping the PDAC microenvironment and potential therapeutic strategies, with a focus on immune cell subsets, cytokines, and immunosuppressive mechanisms. These insights provide a comprehensive understanding of PDAC and pave the way for the development of prognostic markers and therapeutic interventions.

  • Since the blastocyst is broken and spreads out on a flat plastic culture dish (two dimensional culture) during in vitro development, it has been difficult to study the implantation process. It also has been difficult to analyse the interactions between endometrial epithelial and stromal cells because of the lack of a long-term in vitro model which can stimulate in vivo characteristics, as these cells eventually fail to proliferate or cease to express differentiated functions. Recently nontransformed cell lines, CUE-P and CUS-V2, derived from rat endometrial epithelium and stroma were reported. In this study, morphology of CUE-P and CUS-V2 was examined and oxytocin gene expression by CUE-P cells was demonstrated by RT-PCR. The CUE-P cells have a cuboidal morphology and CUS-V2 cells resemble fibroblast and exhibit a spindle-like morphology. In RT-PCR, same size of PCR products of oxytocin gene at hypothalamus, uterus and CUE-P cells were demonstrated. These results showed three dimensional culture system could be made by using the new cell lines.

Diepoxybutane에 의해 유도된 애기장대 엽록체 돌연변이체의 분석 (Analysis of Chloroplast Mutants of Arabidopsis Induced by Diepoxybutane)

  • 본 연구에서는 DEB를 애기장대 종자에 처리하여 엽록체 발달에 관련된 돌연변이체들을 분리하였다. 이들은 각각 그 특징에 따라 iml, gev, yev로 명명하였으며 iml은 잎에 얼룩무늬를 나타내는 돌연변이체이며, gev는 엽맥에만 녹색을 나타내고 이외의 잎에서는 연한녹색을 나타내며, yev는 엽맥은 연한녹색을 나타내고 엽맥 이외의 잎에서는 녹색을 나타내었다. 주사 전자현미경을 이용하여 엽록체 상세구조를 관찰하여 본 결과 야생형 엽록체의 그라나 티라코이드의 모양은 규칙적인 배열하는 그라나가 쌓여 있었으나, iml, gev, yev 돌연변이체 에서는 그라나 티라코이드의 모양이 불규칙하며 그라나의 수도 불규칙하며 스트로마 티라코이드의 연결도 야생형과 상이함을 보였다. 그리고 이들을 유전분석한 결과 야생종과 돌연변이체의 F$_2$에서 3:1의 분리비를 나타내어, 이들 돌연변이들은 핵내에 존재하는 유전자에서 돌연변이가 일어났음을 알 수 있었고, 단일열성으로 유전됨이 밝혀졌다.

  • The present study was conducted to investigate the structural changes in rat cornea. Sixty eyes from one-day-old uneyed rats, fourty eyes from 4-weeks-old rats, and foully eyes from 10-weeks-old adult rats were used. With the increase of age, the epithelial layer was thickened by the addition of new successive cellular layers. Then, the new-born rat's epithelial cells formed a pentagonal shape, and the quality of decidual cells showed a high electron-density, although the boundary between cells was distinctive. The newly produced cells showed a low electron-density so that there was the distinctive difference between light and darkness. In Bowman's layer, collagen fibrils demonstrate a regularly arranged structure along with the age. In stroma's layer, the density of keratocytes was decreased and thereby Progressively flattened during the development. The collagenous layer of the adult rats was more distinctive than that of the new-born rats in a form of parallel alignment running vertically and horizontally.

교배육종에 의한 밀리타리스 동충하초 '도원홍초'의 품종특성 (Varietal characteristics of cross-bred Cordyceps militaris 'Dowonhongcho')

  • 밀리타리스 동충하초는 다양한 생리적 및 약리적 가치를 가지고 있는 약용버섯으로 인정받으며 많은 연구가 있어왔다. 교배를 위해 자낭포자를 분리한 후 균사생장, 균사밀도, 자실체 및 자낭각 형성 여부 등에 대한 검토를 하였고, 그 중에서 우수한 특성의 균주를 이용하여 교배와 선발과정을 거쳐 신품종 '도원홍초'를 육성하였다. 신품종 '도원홍초'는 SDAY배지 및 $10{\sim}25^{\circ}C$ 온도조건에서 균사 생장이 양호하였다. 신품종의 자실체는 곤봉형이고 밝은 주황색을 띠었으며 코디세핀 함량은 0.34% 였고 자좌의 평균길이는 6.1 cm였다. '예당 3호'와 비교할 때, 새로운 품종 '도원홍초'의 수량은 9% 증수되었고 자실체의 경도가 높은 특징을 보였다. 균사생장 적온은 $22{\sim}25^{\circ}C$였고, 버섯 발생 적온은 $18{\sim}22^{\circ}C$였으며 접종에서부터 자실체 발생까지의 기간은 45일이 소요되었다. 신품종 '도원홍초'는 동충하초 인공재배 및 산업적 생산에 기여할 것으로 판단된다.

A case of canine bilateral ovary granulosa cell tumor and mammary complex carcinoma

  • An 11-year-old poodle bitch was presented for investigation of multicentric mammary masses. Abdominal sonography and radiography demonstrated abnormal enlargement of uterus and ovaries. Blood analysis revealed high progesterone concentration. The ovariohysterectomy and mastectomy were performed. Histopathologically, the mammary masses revealed complex carcinoma-tubulopapillary carcinoma with papillary pattern and tubule pattern. In the uterus, cystic endometrial hyperplasia was observed. Scattered inflammatory cells were observed in the endometrial stroma and mucinous material was protruded from endometrial surface. Also, in the ovaries, bilateral ovary granulosa cell tumor was detected. The bitch made a complete recovery following the ovariohysterectomy and mastectomy. This case was a very rare multiple tumor occurrence with bilateral ovary granulosa cell tumor and mammary complex carcinoma. High progesterone concentration was characterized clinically in the bitch.

체외 배양된 자궁내막세포에서의 DOC-1 유전자의 발현 조절 (In Vitro Regulation of DOC-1 Gene Expression in Uterine Endometrial Cells)

  • 포배의 착상은 구조 및 기능적으로 준비된 자궁내막의 체계적 반응에 의하여 진행되는데, 자궁 내막에서 엄격히 통제된 급격한 세포의 증식과 분화가 진행된다. 구강 상피세포암의 억제자로 알려진 Doc-1이 자궁에서 스테로이드 호르몬에 의하여 발현되며 세포 증식을 조절할 것으로 추정되어 왔으나, 그 정확한 발현 변화에 대한 이해가 불충분하였다. 따라서 본 연구에서는 체외에서 탈락막 반응 모델 확립을 통하여 세포의 증식과 분화 진행과정에서 Doc-1의 발현 조절기작을 알아보고자 하였다. 생쥐 자궁내막을 이용한 탈락막 반응은 기질세포를 순수하게 분리하여 배양하면서 프로게스테론과 에스트로겐을 함유한 유도 배양액으로 유도하였다. 탈락막 유도 과정에서 배양 24시간까지 세포의 증식이 유의하게 증가하였고 그 후에는 감소하였다. 한편, 탈락막 세포로의 분화는 분화 유도 48시간에 거의 모든 세포에서 진행되었다. 또한 Doc-1 단백질의 발현이 분화 유도 시간과 비례적으로 증가하였고 탈락막 세포에 위치하였다. 이러한 결과를 바탕으로 착상이 진행되는 자궁내막에서 Doc-1의 발현이 스테로이드 호르몬과 탈락막 분화와 연계되어 조절되고, Doc-1 단백질이 탈락막 세포의 세포분열 억제를 유도하는 것을 제안할 수 있다.

FAP Inhibitors as Novel Small Molecules for Cancer Imaging using Radionuclide

  • Tumors are encircled by various non-cancerous cell types in the extracellular matrix, including fibroblasts, endothelial cells, immune cells, and cytokines. Fibroblasts are the most critical cells in the tumor stroma and play an important role in tumor development, which has been highlighted in some epithelial cancers. Many studies have shown a tight connection between cancerous cells and fibroblasts in the last decade. Regulatory factors secreted into the tumor environment by special fibroblast cells, cancer-associated fibroblasts (CAFs), play an important role in tumor and vessel development, metastasis, and therapy resistance. This review addresses the development of FAP inhibitors, emphasizing the first, second, and latest generations. First-generation inhibitors exhibit low selectivity and chemical stability, encouraging researchers to develop new scaffolds based on preclinical and clinical data. Second-generation enzymes such as UAMC-1110 demonstrated enhanced FAP binding and better selectivity. Targeted treatment and diagnostic imaging have become possible by further developing radionuclide-labeled fibroblast activation protein inhibitors (FAPIs). Although all three FAPIs (01, 02, and 04) showed excellent preclinical and clinical findings. The final optimization of these FAPI scaffolds resulted in FAPI-46 with the highest tumor-to-background ratio and better binding affinity.
