  • Corynebacterium glutamicum, an important industrial strain, has a relatively slower reproduction rate. To acquire a growth-boosted C. glutamicum, a descendant strain was isolated from a continuous culture after 600 generations. The isolated descendant C. glutamicum, JH41 strain, was able to double 58% faster (td=1.15 h) than the parental type strain (PT, td=1.82 h). To understand the factors boosting reproduction, the transcriptomes of JH41 and PT strains were compared. The mRNAs involved in respiration and TCA cycle were upregulated. The intracellular ATP of the JH41 strain was 50% greater than the PT strain. The upregulation of NCgl1610 operon (a putative dyp-type heme peroxidase, a putative copper chaperone, and a putative copper importer) that presumed to role in the assembly and redox control of cytochrome c oxidase was found in the JH41 transcriptome. Plasmid-driven expression of the operon enabled the PT strain to double 19% faster (td=1.82 h) than its control (td=2.17 h) with 14% greater activity of cytochrome c oxidase and 27% greater intracellular ATP under the oxidative stress conditions. Upregulations of genes those might enhance translation fitness were also found in the JH41 transcriptome. Plasmid-driven expressions of NCgl0171 (encoding a cold-shock protein) and NCgl2435 (encoding a putative peptidyl-tRNA hydrolase) enabled the PT to double 22% and 32% faster than its control, respectively (empty vector: td=1.93 h, CspA: td=1.58 h, and Pth: td=1.44 h). Based on the results, the factors boosting growth rate in C. gluctamicum were further discussed in the viewpoints of cellular energy state, oxidative stress management, and translation.

  • Fillet welding is widely used in the assembly of ships and offshore structures. The T-joint configuration is frequently reported to experience fatigue damage when a marine structure meets extreme loads such as storm loads. Fatigue damage is affected by the magnitude of residual stresses on the weld. Recently, many shipping registers and design guides have required that the fatigue strength assessment procedure of seagoing structures under wave-induced random loading and storm loading be compensated based on the effect of residual stresses. We propose a computational procedure to analyze the residual stresses in a T-joint. Residual stresses are measured by the X-ray diffraction (XRD) method, and a 3-D finite element analysis (FEA) is performed to obtain the residual stress profile in the T-joint. The proposed finite element model is validated by comparing experiments with computational results, and the characteristics of the residual stresses in the T-joint are discussed.

  • The present study investigated the litter performance of multiparous sows had different feeding frequencies during summer season. A total of 60 crossbred multiparous sows was allotted to one of two treatments based on body weight (BW) according to a completely randomized design. Two different feeding frequencies based on NRC (National Research Council) were applied as conventional feeding (T1; 2 kg per meal at 06:00, 11:00, and 17:00) and night feeding (T2; 1.5 kg per meal at 06:00, 11:00, 17:00, and 2 kg at 22:00). Sows in T2 treatment had lower (p < 0.05) BW changes during lactation. The backfat thickness change of sows was decreased in T2 treatment during lactation (p < 0.05). The daily feed intake differed significantly between T1 and T2 with increased feed intake of the T2 group at 5.47 kg/d to 5.14 kg/d in the T1 group (p < 0.05). There was a significant difference in total weight of weaned piglets between T1 (70.50 kg) and T2 (74.34 kg). A greater total litter weight gain was observed in sows in T2 treatment. Night feeding is suggested for lactating sows with significant beneficial effects on litter growth during summer season.

  • The present study was performed to assess the activity of the botulinum toxin A on breast cancer cells. The T47D cell line was exposed to diverse concentrations of the botulinum toxin A and cell viability and apoptosis were estimated using MTT and propidium iodine/annexin V methods, respectively. Botulinum toxin A exerted greater cytotoxic activity in T47D cells in comparison with MCF10A normal cells; this appeared to be via apoptotic processes caspase-3 and -7. In conclusion, botulinum toxin A induces caspase-3 and -7 dependent apoptotic processes in the T47D breast cancer cell line.

  • This paper presents application of neutron diffraction technique to the measurement of residual stresses in the T-type 20 m thick welded stainless steel plates(100$\times$50 $mm^2$ and 50$\times$50 $mm^2$). The High Resolution Powder Diffractormeter of the Korea Atomic Research Institute was utilized in the measurement. The power of nuclear reactor was 24 MWt and the measured reflection in the 220 plane (2$\theta$)was $92.66^{\circ}$. Poisson ratio of 0.265 and elastic constant of 211 GPa were applied to the calculation of stresses and strains. Three directional components such as normal, transverse, and longitudinal stresses were measured. The results showed that three components were tensile and became compressive along the y axis in the zone away from the welded center. The compressive stresses became tensile in the zone away from the center line of x axis. This may be due to the balance effect caused by the net stress to keep the specimen shape flat.

  • Premature cracking of the epoxy coatings applied on ship's ballast tanks(BT) can lead to damage of ship's hulls. To avoid this, it's important to have clear understanding of the underlying mechanism and primary factors of the coating crack. In this study, the efforts were made to clarify the integrated effects of main factors, i.e., initial coating shrinkage, thermally induced strain, steel-structural strain and the intrinsic coating flexibility at the initial and after aging, to the early cracking phenomena of epoxy coating in the ship's ballast tank. The coating crack is caused by combination of thermal stress, structural stress, and internal stresses which is closely related to chemical structures of the coatings. On the other hand, thermal stresses and dimensional stabilities would rarely play a major role in coating crack for ballast tank coatings with rather large flexibility. Crack resistance of the coatings at early stages can be estimated roughly by measuring internal stress, FT-IR and $T_g$ value of the coatings. A new screening test method was also proposed in this study, which can be possibly related to the long-term resistance of epoxy-based paints to cracking.

  • Background: Panax notoginseng saponins (PNS) are bioactive substances extracted from P. notoginseng that are widely used to treat cardiovascular and cerebrovascular diseases and interstitial diseases. PNS have the functions of scavenging free radicals, anti-inflammation, improving blood supply for tissue and so on. Objectives: The aim of this study was to investigate the effects of PNS on the oxidative stress of immune cells induced by porcine circovirus 2 (PCV2) infection in vitro and in vivo. Methods: Using an oxidative stress model of PCV2 infection in a porcine lung cell line (3D4/2 cells) and mice, the levels of nitric oxide (NO), reactive oxygen species (ROS), total glutathione (T-GSH), reduced glutathione (GSH), and oxidized glutathione (GSSG) and the activities of xanthine oxidase (XOD), myeloperoxidase (MPO) and inducible nitric oxide synthetase (iNOS) were determined to evaluate the regulatory effects of PNS on oxidative stress. Results: PNS treatment significantly reduced the levels of NO and ROS, the content of GSSG and the activities of XOD, MPO, and iNOS (p < 0.05), while significantly increasing GSH and the ratio of GSH/GSSG in infected 3D4/2 cells (p < 0.05).Similarly, in the in vivo study, PNS treatment significantly decreased the level of ROS in spleen lymphocytes of infected mice (p < 0.05), increased the levels of GSH and T-GSH (p < 0.05), significantly decreased the GSSG level (p < 0.05), and decreased the activities of XOD, MPO, and iNOS. Conclusions: PNS could regulate the oxidative stress of immune cells induced by PCV2 infection in vitro and in vivo.

  • A simple approximation method for the stress intensity factor at the tip of the axial semielliptical cracks on the cylindrical vessel is developed. The approximation methods, incorporated in VINTIN (Vessel INTegrity analysis-INner flaws), utilizes the influence coefficients to calculate the stress intensity factor at the crack tip. This method has been compared with other solution methods including 3-D finite element analysis for internal pressure, cooldown, and pressurized thermal shock loading conditions. For these, 3-D finite-element analyses are performed to obtain the stress intensity factors for various surface cracks with t/R=0.1. The approximation solutions are within $\pm2.5%$ of the those of finite element analysis using symmetric model of one-forth of a vessel under pressure loading, and 1-3% higher under pressurized thermal shock condition. The analysis results confirm that the approximation method provides sufficiently accurate stress intensity factor values for the axial semi-elliptical flaws on the surface of the reactor pressure vessel.

  • In recent, the design of diaphragm which is representative disturbed region in PSC box girder bridge have been performed according to the empirical method or beam theory. But, these methods couldn't be described the behavior of the end diaphragm, and placed reinforcements accurately. As the compressive stress transferred by the web concentrated on the lower parts of diaphragm, it was demonstrated that the basic assumption of 2-D strut-and-tie model for the diaphragm that the compressive stress acts on the upper parts of the diaphragm is wrong. Meanwhile, in this research, after analyzing the variables of end diaphragm, the 2-D strut-and-tie models appropriate to each cases are proposed. And, the problems of 2-D strut-and-tie model were analyzed, so 3-D strut-and-tie model is proposed as well. There is no codes which include the demonstration of safety of 3-D strut-and-tie model. Hence, for nodes, the stresses at the elements which included the singular node in strut-and-tie model were investigated using the finite element analysis. And, the stress states of strut has one direction, so effective stresses were considered at the stage, dimensioning of the model. From the results, 3-D strut-and-tie model could predict the behavior of end diaphragm accurately, and design of reinforcement could be performed economically.
