Isolation and Characterization of Salt Tolerant Mutations in Budding Yeast Saccharomyces cerevisiae

  • In order to study the mechanism for the adaptation to salt stress, we mutagenized budding yeast Saccharomyces cerevisiae with Ethylmethane sulfonate, and isolated salt-tolerant mutants. Among the salt-tolerant mutants, two strains exhibit additional temperature sensitive phenotype. Here, we report that these two salt-tolerant mutants are specific to {TEX}$Na^{+}${/TEX} rather than general osmotic stress. These mutant strains may contain mutations in the genes involved in {TEX}$Na^{+}${/TEX} home-ostasis.

  • We investigated the degradation of $In_2O_3-Ga_2O_3$-ZnO (IGZO) thin-film transistors (TFTs), which is promising device for driving circuits of nextgeneration displays. We performed the electronic stress test by applying gate and drain voltage. We discussed the degradation mechanism by thermal analysis and device simulation.

  • The heat transfer and thermal stress analysis was performed on the ceramic core of the matrix-type recuperator. The efficiency was calculated as 34% to 65%. Triple-pass arrangement provided higher preheated air temperatures, lower thermal stresses and the increase of pressure drops. Higher temperature gradients and maximum peak stresses appeared on the corners of the ceramic core. The effect of boundary conditions was significant and the use of spring-load sealing mechanism could release thermal stresses.

  • Neuronal hyperexcitability followed by high level of intracellular calcium and oxidative stress play critical roles in neuronal cell death in stroke and neurotrauma. Hence, KR-31378, a novel benzopyran derivative was designed as a new therapeutic strategy for neuroprotection possessing both anti-oxidant and potassium channel modulating activities. In the present study, we tested for its neuroprotective efficacy against oxidative stress-induced cell death in primary cortical cultures and further investigated its neuroprotective mechanism. Korean Red Ginseng and Korean black ginseng extracts, JP5 and BG1, prevent hepatic oxidative stress and inflammation induced by environmental heat stress

  • Background: Continuous exposure to high temperatures can lead to heat stress. This stress response alters the expression of multiple genes and can contribute to the onset of various diseases. In particular, heat stress induces oxidative stress by increasing the production of reactive oxygen species. The liver is an essential organ that plays a variety of roles, such as detoxification and protein synthesis. Therefore, it is important to protect the liver from oxidative stress caused by heat stress. Korean ginseng has a variety of beneficial biological properties, and our previous studies showed that it provides an effective defense against heat stress. Methods: We investigated the ability of Korean Red Ginseng and Korean black ginseng extracts (JP5 and BG1) to protect against heat stress using a rat model. We then confirmed the active ingredients and mechanism of action using a cell-based model. Results: Heat stress significantly increased gene and protein expression of oxidative stress-related factors such as catalase and SOD2, but treatment with JP5 (Korean Red Ginseng extract) and BG1 (Korean black ginseng extract) abolished this response in both liver tissue and HepG2 cells. In addition, JP5 and BG1 inhibited the expression of inflammatory proteins such as p-NF-κB and tumor necrosis factor alpha-α. In particular, JP5 and BG1 decreased the expression of components of the NLRP3 inflammasome, a key inflammatory signaling factor. Thus, JP5 and BG1 inhibited both oxidative stress and inflammation. Conclusions: JP5 and BG1 protect against oxidative stress and inflammation induced by heat stress and help maintain liver function by preventing liver damage.

  • The author analyzed and compared the relationship mechanism among measured variables effecting to posttraumatic stress and posttraumatic stress, psychological wellbeing, and depression in the case of 360 policemen and 360 firefighters working in Korean police stations and fire stations by the structural equation modeling. The results showed that firefighters' posttraumatic stress deteriorated psychological wellbeing and depression more than policemen's posttraumatic stress did. In conclusion, systematic prevention and management program for posttraumatic stress and mental health of public servants are needed.

INVESTIGATION OF A STRESS FIELD EVALUATED BY ELASTIC-PLASTIC ANALYSIS IN DISCONTINUOUS COMPOSITES

  • A closed form solution of a composite mechanics system is performed for the investigation of elastic-plastic behavior in order to predict fiber stresses, fiber/matrix interfacial shear stresses, and matrix yielding behavior in short fiber reinforced metal matrix composites. The model is based on a theoretical development that considers the stress concentration between fiber ends and the propagation of matrix plasticity and is compared with the results of a conventional shear lag model as well as a modified shear lag model. For the region of matrix plasticity, slip mechanisms between the fiber and matrix which normally occur at the interface are taken into account for the derivation. Results of predicted stresses for the small-scale yielding as well as the large-scale yielding in the matrix are compared with other theories. The effects of fiber aspect ratio are also evaluated for the internal elastic-plastic stress field. It is found that the incorporation of strong fibers results in substantial improvements in composite strength relative to the fiber/matrix interfacial shear stresses, but can produce earlier matrix yielding because of intensified stress concentration effects. It is also found that the present model can be applied to investigate the stress transfer mechanism between the elastic fiber and the elastic-plastic matrix, such as in short fiber reinforced metal matrix composites.

Computational Identification and Comparative Genomic Analysis of Soybean Oxidative Stress-Related Genes

  • Reactive oxygen and nitrogen species (ROS and RNS, respectively) are messengers that carry signals to alter the redox state in order to activate plant responses and other physiological processes, such as differentiation, aging, senescence, and pathogen defense. Quite a large number of genes are involved in this signaling and lead to oxidative stress in plants. Although the role of ROS/RNS during stress conditions is well documented, a comprehensive list of genes and comparative study of these genes has not yet been completed. Accordingly, the in silico identification of oxidative stress-related genes was performed for soybeans and Arabidopsis. These genes were also studied in relation to multiple domain prediction. The presence of domains like dehydogenase and ATPase suggests that these genes are involved in various metabolic processes, as well as the transportation of ions under optimal environmental conditions. In addition to a sequence analysis, a phylogenetic analysis was also performed to identify orthologous pairs among the soybean and Arabidopsis oxidative stress-related genes based on neighbor joining. This study was also conducted with the objective of further understanding the complex molecular signaling mechanism in plants under various stress conditions.

Involvement of Caenohabditis elegans MAPK Signaling Pathways in Oxidative Stress Response Induced by Silver Nanoparticles Exposure

  • In the present study, toxicity of silver nanoparticles (AgNPs) was investigated in the nematode, Caenohabditis elegans focusing on the upstream signaling pathway responsible for regulating oxidative stress, such as mitogen-activated protein kinase (MAPK) cascades. Formation of reactive oxygen species (ROS) was observed in AgNPs exposed C.elegans, suggesting oxidative stress as an important mechanism in the toxicity of AgNPs towards C. elegans. Expression of genes in MAPK signaling pathways increased by AgNPs exposure in less than 2-fold compared to the control in wildtype C.elegans, however, those were increased dramatically in sod-3 (gk235) mutant after 48 h exposure of AgNPs (i.e. 4-fold for jnk-1 and mpk-2; 6-fold for nsy-1, sek-1, and pmk-1, and 10-fold for jkk-1). These results on the expression of oxidative stress response genes suggest that sod-3 gene expression appears to be dependent on p38 MAPK activation. The high expressions of the pmk-1 gene 48 h exposure to AgNPs in the sod-3 (gk235) mutant can also be interpreted as compensatory mechanisms in the absence of important stress response genes. Overall results suggest that MAPK-based integrated stress signaling network seems to be involved in defense to AgNPs exposure in C.elegans.

Mechanical Properties of Mica/Epoxy Composite Materials used in Genrator Stator Windings (발전기 고정자 권선에 사용되는 마이카/에폭시 복합재료의 기계적 특성에 관한 연구)

  • Experiments on mechanical fatigue were conducted using the specimens which were cut from hydrogen cooled generator(rated 22kV and 50OMW) stator windings. We have investigated the aged mechanism of mica/epoxy insulation systems under air or hydrogen by both the tensile and compressive loadings. The fracture of generator stator windings is generally affected by mechanical stress. Thus, the tensile strength test were conducted. In this case, the maximum strength and strain are quite different between sound and aged specimens. It is observed that low bonded interface parts of tapes generally have lower strength than those of normal tapes which causes stress. In order to estimate the effects of cyclic load by the electromagnetic forces while the generator starts/stops, the mechanical fatigue test was also conducted. It is confirmed that the equation of expected life depends on stress amplitude and number of cycles. Though the stress amplitude and number of cycles are very tiny, the tensile fatigue of aged specimens under hydrogen atmosphere is bigger than those under air. In the case of hydrogen atmosphere, the tensile stress gives bigger effect than the compressive one.
