Mammal microsomal glutathione transferase 1 (MGST1) can conjugate many toxic or carcinogenic substances and depress oxidative stress. In this study, Chicken MGST1 and its variants were cloned for the first time and were composed of 956 or 944 nucleotides. The 12 nt deletion in the exon 2 did not alter the GT-AG rule and the ORFs for the two MGST1 variants were the same, which both comprised 465 nucletides and encoded a peptide with 155 amino acids. It was found that the two different splice variants identified using RT-PCR expressed in all three organs investigated of Dwarf Brown Chicken, namely liver, spleen and shell gland. Moreover, the expression level of MGST1 mRNA in the liver of Dwarf Brown chickens was the highest (p<0.01), and there were no significant differences between the spleen and the shell gland. These results provide a base for studying the biological function of Chicken MGST1.
The purpose of this study is to compare the results between conventional and accelerated rehabilitation program following ACL reconstruction using bone-patellar tendon-bone. Conventional rehabilitation focused on protecting the new ligament by blocking terminal extension and avoiding active quadriceps function in the terminal degrees of extension. But there is current trend toward early postoperative mobilization and intensive. so called 'accelerated', rehabilitation stressing hyperextension of the knee. The results of intraarticular ACL reconstruction with conventional and accelerated rehabilitation were prospectively compared for one year postoperatively in a series of 27 patients. Range of motion and thigh circumference were checked preoperatively, and weekly up to 8 weeks, 3 months. 6 months, and 1 year postoperatively. Stress radiologic test, KT-1000 arthrometer. Cybex II dynamometer were checked in preoperatively, and 3 months, 6 months, and 1 year postoperatively. There were no differences of objective stability and restoration of muscle power. But the accelerated group had a low incidence of extension loss. excellent range of motion, and less difference of thigh circumference. We concluded that accelerated rehabilitation program is recommendable due to superiority in terms of range of motion, especially less extension loss without increasing laxity of knee joint.
A Bridge Navigational Watch Alarm System (hereafter 'BNWAS') is to monitor and detect if an officer of watch(hereafter 'OOW') keeps a sharp lookout on the bridge. The careless lookout of an OOW could lead to marine accidents. For this reason on June 5th, 2009, IMO decided that a ship is equipped with a BNWAS. However, an existing BNWAS gives the OOW a lot of inconvenience and stress in its operation. It requires that the OOW should press reset buttons to confirm their alert watch on the bridge at every three to twelve minute. Many OOWs have complained that at some circumstances they cannot focus on their bridge activities including watch-keeping due to a lots of resetting inputs of BNWAS. Accordingly, IMO has allowed the use of a motion sensor as a resetting device. The motion sensor detects the movements of human body on the bridge and subsequently sends reset signals directly to BNWAS automatically. As a result, OOWs can work uninterrupted. However, some of classification societies and flag authorities have a slightly different stance on the use of motion sensor as a resetting method for BNWAS. The reason is that the motion sensor may trigger false reset signals caused by the motion of objects on the bridge, especially a slight movement such as toss and turn of human body which can extend the period of careless watch. As a basic study to minimize the false reset signals, this paper proposes a simple configuration of BNWAS, which consists of only three motion sensors associated with 'AND' and 'OR' logic gates. Additionally, several considerations are also proposed for the implementation of motion sensors. This study found that the proposed configuration which consists of three motion sensors is better than an existing one by reducing false reset signals caused by a slight movement of human body in one's sleep. The proposed configuration in this paper filters false reset signals and is simple to be implemented on existing vessels. In addition, it can be easily installed just by a basic electrical knowledge.
Heme oxygenase-1 (HO-1), an inducible enzyme with broad tissue expression, is wel1-regulated in response to hematopoietic stress and preserves vascular homeostasis. We investigated the involvement of HO-1 in HL-60 cell differentiation. Dimethyl sulfoxide (DMSO) completely decreased HO-1 expression in a time-dependent manner, but clearly induced HL-60 cell differentiation, as evidenced by a marked increase in CD11b expression. Interestingly, zinc protoporphyrin (ZnPP), a strong inhibitor of HO-1, induced HL-60 cell differentiation. In contrast, treatment with cobalt protoporphyrin (CoPP), an activator of HO-1, decreased CD11b expression. Additionally, ZnPP down-regulated HO-1 protein expression in HL-60 cells, whereas CoPP induced upregulation. These results suggest that HO-1 might have a negative function in DMSO-induced HL-60 cell differentiation. This study provides the first evidence that HO-1 plays an important role in DMSO-induced HL-60 cell differentiation.
Vascular smooth muscle cells (VSMCs) are a unique cell type that has unusual plasticity controlled by environmental stimuli. As an abnormal increase of VSMC proliferation is associated with various vascular diseases, tight regulation of VSMC phenotypes is essential for maintaining vascular homeostasis. Hypoxia is one environmental stress that stimulates VSMC proliferation. Emerging evidence has indicated that microRNAs (miRNAs) are critical regulators in the hypoxic responses of VSMCs. Therefore, we previously investigated miRNAs modulated by hypoxia in VSMCs and found that miR-1260b is one of the most upregulated miRNAs under hypoxia. However, the mechanism that underlies the regulation of VSMCs via miR-1260b in response to hypoxia has not been explored. Here we demonstrated that hypoxia-induced miR-1260b promotes VSMC proliferation. We also identified growth differentiation factor 11 (GDF11), a member of the TGF-β superfamily, as a novel target of miR-1260b. miR-1260b directly targets the 3'UTR of GDF11. Downregulation of GDF11 inhibited Smad signaling and consequently enhanced the proliferation of VSMCs. Our findings suggest that miR-1260b-mediated GDF11-Smad-dependent signaling is an essential regulatory mechanism in the proliferation of VSMCs, and this axis is modulated by hypoxia to promote abnormal VSMC proliferation. Therefore, our study unveils a novel function of miR-1260b in the pathological proliferation of VSMCs under hypoxia.

The purpose of this study was to investigate the anticancer effects of Caesalpiniae Lignum (Somok) on HepG2 cells, a human liver cancer cell line. To study the cytotoxic effect of Caesalpiniae Lignum methanol extract (CL-MeOH) on HepG2 cells, the cells were treated with various concentrations of CL-MeOH and then cell viability was determined by XTT reduction method and trypan blue exclusion assay. CL-MeOH reduced proliferation of HepG2 cells in a dose-dependent manner. To confirm the induction of apoptosis, HepG2 cells were treated with various concentrations of CL-MeOH. The activation of caspase 3 and the cleavage of poly ADP-ribose polymerase (PARP), a substrate for caspase-3 and a typical sign of apoptosis, was examined by western blot analysis. CL-MeOH decreased procaspase 3 level in a dose-dependent manner and induced the clevage of PARP at concentration> $200{\mu}/ml$. Mitogen-activated protein (MAP) kinase signaling cascades are multi-functional signaling networks that influence cell growth, differentiation, apoptosis, and cellular responses to stress. CL-MeOH-induced MAPK activation was examined by Western blot for phosphorylated ERK, p38 and JNK. CL-MeOH significantly increased p38 phosphorylation and JNK phosphorylation in a dose-dependent manner. Inhibition of p38 function using the selective inhibitor SB20358O results in inhibition of apoptosis by CL-MeOH. These results suggest that CL-MeOH-induced apoptosis is MAP kinase-dependent apoptoric pathway. These results suggest that CL-MeOH is potentially useful as a chemotherapeutic agent in human liver cancer.

Especially, from the Jang-Fu physiological point of view, the memory is closly related with the heart and kidney In oriental medicine, some experiments on animal and literature studies on the subject of memory promotion have done. But because of difference in memory mechanism between man and animal, it is not in reason to apply the result of experiment on animal to human. Therefore I have methodological study of memory promotion to set up the concept of oriental medicine and experimental theory about this and can obtain such conclusion. 1. The oriental medical therapy for memory promotion is following. nourishing the heart and blood, regulating the function of spleen, relieving the mental stress, reinforcing the heart and kidney, invigorating and enriching the blood. 2. The insufficient intelligence in a child is considered to not be full and in an old man, it is considered to decline by degrees. 3. It is needed to molecular biological study of neurotransmitter after the using of oriental medical therapy. 4. It is possible to study using the genetic mutation or observing the collateral of brain nerve cell.

Recently, epidural morphine has been administrated to decrease patients' systemic stress responses such as: suffers, endocrine responses and impairment of pulmonary function, etc. Epidural morphine provided excellent analgesic effect, but incomplete sensory blockade as compared to epidural local anesthetics, which has sympathetic blockade effect and tachyphylaxis. Therefore, the authors surmised that low dose bupivacaine on low dose epidural morphine improved postoperative pain with greater sensory analgesia than epidural morphine alone. The effect of low dose bupivacaine on epidural morphine analgesia for postoperative pain was evaluated in seventy patients. They were physical status I-III by ASA classification. Patients were randomly divided into 2 groups and they were administrated morphine 2.5 mg only (group I), morphine 2.5 mg plus 0.125% bupivacaine (group II) through epidural catheter 1 hour before the end of the operation. During postoperative second days, their analgesic effects were evaluated by visual analogue scale (0-10). Side effects were also evaluated. The results were as follows, 1) On the day of the operation, VAS score showed significant differences between two groups (morphine group $3.20{\pm}0.16$, morphine plus bupivacaine group $2.77{\pm}0.08$; p < 0.05). 2) On the postoperative and second day, there were no statistical differences between the groups according to VAS score. 3) The incidence of pruritus, nausea, and vomiting were no differences in both groups. 4) None of the patients showed objective sedation or a low respiratory rate (< 10 bpm). We concluded that epidural administration of low dose bupivacaine on the epidural morphine analgesia was an effective method to decrease postoperative pain with little change in frequencies of side effects compared to epidural morphine alone.

This study proposes medical gowns for interns and residents that address the functions of utility and activity. Based on the questionnaires, the prototype focused on the pockets for practical use and movement adaptability. Functional pockets were made by increasing pocket sizes and separating pockets with sections according to use. An inside pocket and a loop for an identification tag was placed on the chest pocket and the side seam pocket were oblique lined. To resolve horizontal stress on the back of the gown,6 cm action pleats were added at both ends of the bladebone. The elbow area was tucked up to improve the workplace activities. To reduce hamper on the gown hemline, slits were added along the gown side seam. The front button was changed into an inside button closure. The cuffs, belt, and back waist belt were removed due to occupational cases of inconvenience. The length of the sleeves was adjusted with belts to improve the movement of the arms. In accordance with design preference, the gown was designed to be knee-length with wrist length sleeves. Blue colored (2.5PB 3/12: by Munsell) material was inserted to reduce the contamination on the neck, cuffs, front closure, and hemline area. The planning cloth of the experimental gown was designed to improve activity function, cleaning, and comfort from light materials. The field test (with 30 doctors at university hospitals and the objective assessment with 9 subject groups)were done by the valuation between the existing gown and the prototype. The results showed that the prototype had to be modified by decreasing the pocket size, expanding the usage of the penholder, and simplify front pockets in order to maintain unity. The loop for the identification tag, inside pocket, and the blue colored material had to be removed. The front closure of the gown had to return to its original state.

Platycodon grandiflorum, commonly known as Doraji in Korea, has a wide range of pharmacologic properties, such as reducing adiposity and hyperlipidemia, and antiatherosclerotic effects. However, the mechanisms underlying these effects remain unclear. In order to profile proteins from the nodal segment, callus, root and shoot, high throughput proteome approach was executed in the present study. Two dimensional gels stained with CBB, a total of 84 differential expressed proteins were confirmed out of 839 protein spots using image analysis by Progenesis SameSpot software. Out of total differential expressed spots, 58 differential expressed protein spots (${\geq}$ 2-fold) were analyzed using MASCOT search engine according to the similarity of sequences with previously characterized proteins along with the UniProt database. Out of 58 differential expressed protein, 32 protein spots were up-regulated such as ribulose-1,5-bisphosphate carboxylase, endoplasmic oxidoreductin-1, heat stress transcription factor A3, RNA pseudourine synthase 4, cysteine proteinase, GntR family transcriptional regulator, E3 xyloglucan 6-xylosyltransferase, while 26 differential protein spots were down-regulated such as L-ascorbate oxidase precursor, late embryogenesis abundant protein D-34, putative SCO1 protein, oxygen-evolving enhancer protein 3. However, frequency distribution of identified proteins using iProClass databases, and assignment by function based on gene ontology revealed that the identified proteins from the explants were mainly associated with the nucleic acid binding (17%), transferase activity (14%) and ion binding (12%). In that way, the exclusive protein profile may provide insight clues for better understanding the characteristics of proteins and metabolic activity in various explants of the economically important medicinal plant Platycodon grandiflorum.
