당뇨 마우스에서 우뭇가사리(Gelidium amansii)의 식후 고혈당 완화 효과 (Gelidium amansii Extract, a Potent α-glucosidase and α-amylase Inhibitor, Alleviates Postprandial Hyperglycemia in Diabetic Mice)

  • 우뭇가사리(Gelidium amansi)는 홍조류로서 우뭇가사리과(Gelidiaceae)에 속한다. 현재까지 우뭇가사리의 항산화 및 항비만 효과 등의 기능들이 연구되었으나 식후 혈당 수치에 미치는 영향에 관한 연구는 부족한 실정이다. 이에 본 연구에서는 Gelidium amansii extract (GAE)가 탄수화물 가수분해 효소(${\alpha}-glucosidase$, ${\alpha}-amylase$)에 미치는 억제 효과 및 streptozotocin (STZ)으로 유도된 당뇨병 마우스의 식후고혈당에 미치는 완화효과를 조사하였다. 정상군과 STZ으로 유도된 당뇨병 마우스에 수용성 전분(2 g/kg body weight)을 경구투여한 후 GAE (300 mg/kg body weight) 또는 acarbose (100 mg/kg body weight)를 단독 또는 함께 투여하였다. 혈당은 꼬리채혈을 통해 0, 30, 60, 120분 간격으로 측정하였다. ${\alpha}-glucosidase$${\alpha}-amylase$에 대한 GAE의 $IC_{50}$ 값은 각각 $0.099{\pm}0.009mg/ml$$0.178{\pm}0.038mg/ml$의 결과값을 나타내어, 양성대조군인 acarbose보다 더 효과적이었다. STZ으로 유발된 당뇨병 마우스의 식후 혈당 수치는 대조군에 비해 GAE 투여 시 유의적으로 더 낮았다(p<0.05). 또한 GAE 투여는 당뇨병 마우스에서 포도당 반응에 대한 곡선하면적 감소와 관련이 있었다(p<0.05). 이러한 결과는 GAE가 ${\alpha}-glucosidase$, ${\alpha}-amylas$와 같은 탄수화물 가수분해 효소를 억제함으로써 식후 고혈당을 완화시키는 유용한 천연기능성 식품이 될 것으로 사료된다.

Hypoglycemic and antioxidant effects of Daraesoon (Actinidia arguta shoot) in animal models of diabetes mellitus

  • BACKGROUND/OBJECTIVES: The primary objective of the treatment of diabetes mellitus is the attainment of glycemic control. Hyperglycemia increases oxidative stress which contributes to the progression of diabetic complications. Thus, the purpose of this study was to investigate the hypoglycemic and antioxidant effects of Daraesoon (Actinidia arguta shoot) in animal models of diabetes mellitus. MATERIALS/METHODS: Rats with streptozotocin-induced diabetes received an oral administration of a starch solution (1 g/kg) either with or without a 70% ethanol extract of Daraesoon (400 mg/kg) or acarbose (40 mg/kg) after an overnight fast and their postprandial blood glucose levels were measured. Five-week-old C57BL/6J mice were fed either a basal or high-fat/high-sucrose (HFHS) diet with or without Daraesoon extract (0.4%) or acarbose (0.04%) for 12 weeks after 1 week of adaptation to determine the effects of the chronic consumption of Daraesoon on fasting hyperglycemia and antioxidant status. RESULTS: Compared to the control group, rats that received Daraesoon extract (400 mg/kg) or acarbose (40 mg/kg) exhibited a significant reduction in the area under the postprandial glucose response curve after the oral ingestion of starch. Additionally, the long-term consumption of Daraesoon extract or acarbose significantly decreased serum glucose and insulin levels as well as small intestinal maltase activity in HFHS-fed mice. Furthermore, the consumption of Daraesoon extract significantly reduced thiobarbituric acid reactive substances and increased glutathione levels in the livers of HFHS-fed mice compared to HFHS-fed mice that did not ingest Daraesoon. CONCLUSIONS: Daraesoon effectively suppressed postprandial hyperglycemia via the inhibition of ${\alpha}$-glucosidase in STZ-induced diabetic rats. Chronic consumption of Daraesoon alleviated fasting hyperglycemia and oxidative stress in mice fed a HFHS diet.

Alleviating Effects of Baechu Kimchi Added Ecklonia cava on Postprandial Hyperglycemia in Diabetic Mice

  • In this study, we investigated the inhibitory effects of Baechu kimchi added Ecklonia cava on the activities of ${\alpha}$-glucosidase and ${\alpha}$-amylase and its alleviating effect on the postprandial hyperglycemia in STZ-induced diabetic mice. Baechu kimchi added Ecklonia cava (BKE, 15%) was fermented at $5^{\circ}C$ for 28 days. Optimum ripened BKE was used in this study as it showedthe strongest inhibitory activities on ${\alpha}$-glucosidase and ${\alpha}$-amylaseby fermentation time among the BKEs in our previous study. The BKE was extracted with 80% methanol and the extract solution was concentrated, and then used in this study. The BKE extract showed higher inhibitory activities than Baechu kimchi extract against ${\alpha}$-glucosidase and ${\alpha}$-amylase. The $IC_{50}$ values of the BKE extract against ${\alpha}$-glucosidase and ${\alpha}$-amylase were 0.58 and 0.35 mg/mL, respectively; BKE exhibited a lower ${\alpha}$-glucosidase inhibitory activity but a higher ${\alpha}$-amylase inhibitory activity than those of acarbose. The BKE extract alleviated postprandial hyperglycemia caused by starch loading in normal and streptozotocin- induced diabetic mice. Furthermore, the BKE extract significantly lowered the incremental area under the curve in both normal and diabetic mice (P<0.05). These results indicated that the BKE extract may delay carbohydrate digestion and thus glucose absorption.

Crocin Improves Oxidative Stress by Potentiating Intrinsic Anti-Oxidant Defense Systems in Pancreatic Cells During Uncontrolled Hyperglycemia

  • Introduction: Oxidative stress (OS) during uncontrolled hyperglycemia has a pivotal role in pancreatic dysfunction. Our study aimed to demonstrate that crocin can potentiate anti-oxidant defense systems of pancreatic cells to improve oxidative stress. Methods: Male Wistar rats were divided randomly into four groups: a normal group, a normal-treated group, a diabetic group and a diabetic-treated group (n = 6 rats per group). Diabetes was induced by a single dose of streptozotocin (45 mg/kg/IV). The treated groups received crocin daily for 8 weeks (40 mg/kg/IP). At the end of the experiment, rats were sacrificed and pancreas tissue was obtained. Subsequently, the concentrations of malondialdehyde (MDA), nitrate and glutathione as well as the enzymatic activities of catalase and superoxide dismutase (SOD) were determined in all animals. Data were analyzed by two-way ANOVA with appropriate post hoc testing and a probability value of P < 0.05 was considered to represent a statistically significant difference in mean values. Results: Uncontrolled hyperglycemia weakened the anti-oxidant system by decreasing SOD and catalase enzyme activity in pancreatic tissues and induced OS by increasing the MDA content in diabetic non-treated animals. Crocin potentiated the anti-oxidant defense system by increasing the activity of both SOD and catalase, and improved OS by diminishing MDA production in pancreatic cells of rats contained in the diabetic-treated group. Conclusion: Based on our results, it is concluded that uncontrolled hyperglycemia can weaken the anti-oxidant defense system and cause the development of OS. Also, crocin can improve OS in pancreatic cells by potentiating the anti-oxidant defense system.

The Inhibitory Activity of Polygonum Multiflorum Thunberg and its Effect on Postprandial Hyperglycemia in Streptozotocin-induced Diabetic Rats

  • To keep blood glucose levels as close to normal as possible is the major goal of diabetes mellitus treatment $\alpha$-Glucosidase is the enzyme that digests die1my carbohydrate and inhibition of this enzyme may suppress postprandial hyperglycemia. The methanol extract of Polygonum multiflorum Thunberg was tested for inhibitoty activity against $\alpha$-glucosidase in vitro and in vivo. Polygonum multiflorum Thunberg extract inhibited yeast $\alpha$-glucosidase activity in a concentration-dependent manner. Polygonum multiflorum Thunberg showed an $IC_{50}$ value of 0.48 mg/mL. The ability of Polygonum multiflorum Thunberg extract to lower postprandial glucose was studied in streptozotocin-induced diabetic rats. A starch solution (1 g/kg) with and without the methanol extract of Polygonum multiflorum Thunberg extract (500 mg/kg) was administered to diabetic rats by gastric intubation after an overnight fast A single oral dose of Polygonum multiflorum Thunberg extract significantly inhibited increases in blood glucose levels at 60 and 90 min (p<0.05) and significantly decreased incremental response areas under the glycemic response curve (p<0.05). These results suggest that Polygonum multiflorum Thunberg may have an antihyperglycemic effect by inhibiting $\alpha$-glucosidase activity in the animal model of diabetes mellitus.

눈꽃 동충하초의 약물활성 (Pharmacological Activities of Paecilomyces japonica, A New Type Cordyceps sp.)

  • Cordyceps is reputed for its broad biological activities and as a tonic for replenishing vital function in Chinese traditional medicines. As an attempt to obtain fundamental data for the development of a new type Cordyceps, the effects of the fruiting bodies of cultivated fungus of Paecilomyces japonica grown on silkworm larvae on hyperglycemia induced by streptozotocin(STZ) and by epinephrine in rats and in mice as well as on immunological functions in mice were investigated. The 70% methanol extract of the fungus, when administered orally at 100 and 300 mg/kg in STZ-induced hyperglycemic rats, caused a significant decrease in blood glucose level 3hr after sample treatments. The methanol extract, when administered p.o. at the same dose levels in epinephrine-induced hyperglycemic mice, also caused a significant decrease in serum glucose levels as well as a significant reversal of the liver glycogen contents suggesting its hypoglycemic activity might be due to glycogen breakdown in the liver. Treatment of normoglycaemic mice with the methanol extract of the fungus exhibited a significant glucose tolerance up to 3hr after oral glucose load(2.0 g/kg). The methanol extract also showed immuno-stimulating activity as measured by carbon clearance in mice and a significant antifatigue effect as measured by weight loaded forced swimming performance in mice.

Hypoglycemic Effect of Jicama (Pachyrhizus erosus) Extract on Streptozotocin-Induced Diabetic Mice

  • The purpose of this research was to investigate the inhibitory effect of jicama extract on ${\alpha}$-glucosidase activity, ${\alpha}$-amylase activity, and postprandial hyperglycemia in streptozotocin (STZ)-induced diabetic mice. Jicama extract showed prominent inhibitory effects against ${\alpha}$-glucosidase and ${\alpha}$-amylase. The $IC_{50}$ values of jicama extract against ${\alpha}$-glucosidase and ${\alpha}$-amylase were $0.083{\pm}0.004$ and $0.091{\pm}0.017mg/mL$, respectively. The increase in postprandial blood glucose levels was more significantly suppressed in the jicama extract-administered group than in the control group of both STZ-induced diabetic and normal mice. Blood glucose levels of the control group increased to $383.75{\pm}11.54$ and $402.50{\pm}15.32mg/dL$ at 30 and 60 min after a meal and decreased to $349.67{\pm}11.62mg/dL$ at 120 min. However, postprandial blood glucose levels were significantly decreased, when diabetic mice were fed with jicama extract ($342.00{\pm}15.73$, $367.00{\pm}13.00$, and $329.67{\pm}12.43mg/dL$ at 30, 60, and 120 min, respectively). Furthermore, the area under the curve was significantly decreased with jicama extract administration in diabetic mice (P<0.05). Therefore, these results indicate that jicama extract may help decrease postprandial blood glucose level by inhibiting ${\alpha}$-glucosidase.

Altered Gene Expression Profiles in the Lungs of Streptozotocin-induced Diabetic Mice

  • Diabetes mellitus is a common heterogeneous metabolic disorder, characterized by deposition of extracellular matrix, oxidative stress, and vascular dysfunction, thereby leading to gradual loss of function in multiple organs. However, little attention has been paid to gene expression changes in the lung under hyperglycemic conditions. In this study, we found that diabetes inuced histological changes in the lung of streptozotocin-induced diabetic mice. Global gene expression profiling revealed a set of genes that are up- and down-regulated in the lung of diabetic mice. Among these, expression of Amigo2, Adrb2, and Zbtb16 were confirmed at the transcript level to correlate significantly with hyperglycemia in the lung. We further evaluated the effect of human umbilical cord-derived perivascular stem cells (PVCs) on these gene expression in the lung of diabetic mice. Our results show that administration of PVC-conditioned medium significantly suppressed Amig2, Adrb2, and Zbtb16 upregulation in these mice, suggesting that these genes may be useful indicators of lung injury during hyperglycemia. Furthermore, PVCs offer a promising alternative cell therapy for treating diabetic complications via regulation of gene expression.

가미오령산(加味五苓散)이 Streptozotocin으로 유발된 흰쥐의 당뇨병성(糖尿病性) 신증(腎症)에 미치는 영향 (The Effects of Gamioryung-san Extracts on Streptozotocin-induced Diabetic Nephropathy Rats)

  • Objectives : The object of this study was to observe the effects of Gamioryung-san (GOS), which consists of 22 types of herbs, on streptozotocin (STZ)-induced diabetic nephropathy rats. Methods : Three different dosages of GOS were orally administered once a day for 28 days from 3 weeks after STZ treatment. Six groups, each of 8 rats per group were used. Changes on the body weights, blood glucose levels, serum BUN and creatinine levels, urine volumes, and UAER were observed with changes on the kidney malondialdehyde contents and glutathione, dismutase and catalase contents. In addition, histopathology of kidney, pancreas, thymus and spleen were observed. The results were compared with antioxidant silymarin 100 mg/kg, of which the effects on STZ -induced diabetes and related complications are already confirmed. Results : As a result of treatment of GOS 800, 400 or 200 mg/kg for 28 days, STZ-induced decreases of body weights, hyperglycemia, atrophic changes of pancreatic islets with decreases of insulin-immunoreactive cells and decreases of glucagon -immunoreactive cells were inhibited dose-dependently. Increases in kidney weight, serum BUN and creatinine levels, urine volumes, UAER, vasodilated atrophic glomerulus and abnormal tubules were inhibited dose-dependently. Also increases of kidney MDA contents and decreases of GSH contents, SOD and CAT activities, decreases of thymus and spleen weights, and atrophic changes at histopathological observation were also inhibited. The effects of GOS 400 mg/kg showed similar effects to silymarin 100 mg/kg. Conclusions : These results suggest that 400 mg/kg of GOS retarded the STZ-induced diabetic nephropathies as similarly to silymarin 100 mg/kg, through modulations of oxidative stress and immune systems.

  • The studies were conducted to investigate the anti-diabetic activities on the hyperglycemia induced by streptozotocin in rats, Anti-fatigue, Decrease of body weight activities in mouse and anti-gastric ulcer activities in stress-induced rats by Mixed Extracts of Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex in Korea. 1. The blood glucose levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 2. The serum total cholesterol levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses (100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 3. The serum triglyceride levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 4. The swimming time levels in mouse were dose-dependently extended by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 5. The body weight levels in mouse were dose-dependently decreased by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 6. Stress-induced gastric ulcer were dose-dependently repaired by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex.
