Dietary Administration of Probiotics, Bacillus sp. IS-2, Enhance the Innate Immune Response and Disease Resistance of Paralichthys olivaceus against Streptococcus iniae (Probiotics를 양식넙치에 투여시 Streptococcus iniae에 대한 면역반응 및 병저항성)

  • The strains were added to the feed in the concentration of $10^3$, $10^5$, and $10^7$ CFU/kg and 2% of fishes were given the feed twice a day (8 AM and 5 PM) for 12 weeks. In result of the nonspecific immune response study to examine Respiratory burst activity, Lysozyme activity and Phagocytosis activity every two weeks until the end of the study, all test samples showed greater activities than control samples and improved immune activity with Bacillus sp. IS-2. The mortality test performed by artificial infection using Streptococcus iniae, a pathogenic bacterium, after the completion of this study also showed over 55% greater survival rate in all test samples. In result of performing PCR using the universal primer to verify that the probiotic stays in the intestines of the fishes, all test samples showed PCR product of 1,465 bp. Based on the above findings, it was concluded that Bacillus sp. IS-2 in the feed improved farmed flatfish's immune system and resistance against diseases as the probiotics. Also, the physiological indicators discovered by this study would be useful for identifying the mechanisms of probiotics.

  • Some physical and physiological properties of di-D-fructofuranose dianhydrideIII (DFAIII), as a new sweetener, were investigated via in vitro experiments. The disaccharide was prepared by decomposing inulin with inulin fructotransferase (depolymerizing) from Arthrobacter sp. A-6. DFAIII had more excellent heat and acid stability than sucrose. This was one of the most desirable properties especially for the oligomer types of sweetener. DFAIII showed the least pH drop in the Streptococcus mutans culture, compared with the other saccharides examined. This indicates that the sugar will be fairly effective for preventing dental caries. The saccharide also had a selective Bifidus growth-promoting effect in PYF medium. Whereas, E. coli did not show growth promotion in the DFAIII-containing medium. In the co-culture of Bifidus longum and E. coli in the BL medium, Bifidus longum had a selective growth while the growth of E. coli appeared rather to be inhibited.

  • The study was performed to investigated the excellent microbial anticaries substance which is more effective than the chlorohexidine in the dental caries treatment. For the screening of alkaliphilic microorganism, more than 1200 bacterial strains were isolated from sea soil sample. A typ-ical strain which produced the most excellent antimicrobial substance was selected. The strain was identified novel alkalophilic Bacillus sp. through the results of morphological, biochemical and chemotaxonomical characteristics and 16S rDNA sequencing and designated as Bacillus alkalophilshaggy JY-827.

  • A bacteriocin-producing organism, Enterococcus sp. T7, was isolated from human fecal samples. Bacteriocin T7, named tentatively as the bacteriocin, was produced by Enterococcus sp. T7 and it inhibited some strains of Lactobacillus. Staphylococcus, Enterococcus, and Streptococcus, but not all the lactococci and gram-negative bacteria tested. Bacteriocin T7 inhibited the growth of Listeria monocytogenes Scott A, but the degree of inhibition was less than those for other sensitive gram-positive vacteria. Bacteriocin T7 in MRS broth started to produce at the middle of the exponential growth phase and the inhibitory activity reached its maximum level during the stationary growth phase. Bacteriocin T7 was stable against heat treatments, pH variations (pH 2-10), and exposure to organic solvents. The molecular weight of bacteriocin T7 was estimated to be 6.500 Da by SDS-PAGe. All these facts, including physico-chemical stabilities, small molecular size, and inhibition of Kisteria monocytogenes, indicate that bacteriocin T7 is likely to be a member of the class IIa bacteriocins.

  • Microorganisms causing bovine mastitis, swine diarrhea and swine pneumonia were isolated from farms in Chungbuk and Kyunggi Province and the isolates were identified using microbial identification system(MIS). The most common isolates from bovine mastitis were Staphylococcus sp.,Streptococcus sp., and Corynebacterium sp., those from swine diarrhea were Escherichia coli, Salmonella sp., and Clostridium perfringens, those from swine pneumonia were Bordetella bronchiseptica and Pasteurella multocida. Examination of 16 antibiotics against these pathogens revealed that the incidence of antibiotic-resistant microoganisms were very high and that many of these isolates had multiple resistance to various commercially available antibiotics such as penicillin, ampicillin, erythromycin, streptomycin, norfloxacin, and tetracycline. These results suggest that the use of antibiotics in a farm should be controlled in order to decrease the number of antibiotic-resistant microorganisms were very high and that many of these isolates had multiple resistance to various commercially available antibiotics such as penicillin, ampicillin, erythromycin, streptomycin, norfloxacin, and tetracycline. Epidemiological Study on the Bovine Mastitis in Chonnam Area (乳牛의 乳房炎에 관한 疫學的 연구 -전남지역을 중심으로-)

  • Epidemiological study on the bovine mastitis was carried out on 1,493 raw milk samples from 7,406 dairy cows of dairyfarms in Chonnam province. And, it were classified by R. B. V. test during the period from July, 1984 to June, 1986. Of these positive samples, the mastitis causative agents were isolated, and examined for susceptibility to 14 antibiotics by disc method. The results are summarized as follows 1. Incidence rate of bovine mastitis from raw milk examined was observed in 1,393 dairy cows (18.8%) among 7,406 dairy cows of dairy farms. These outbreaks according to years were increased to 17.1% in 1984, 19.7% in 1985, and 20.5% in 1986. And, outbreaks of bovine mastitis according to seasons were high observed during the period from August to October. 2. Isolated rate of mastitis causative agents from raw milk was observed in the order of Staphylococcus sp. (50.2-51.7%), Streptococcus sp. (35.6-48.3%), Bacillus sp. (2.2-6.7%), Pseudomonas sp. (0.3-5.7%), and others (0.6%). 3. Generally, antibiotics susceptibility of isolated mastitis agents from raw milk was observed high susceptible in CP, CL, NB, PC, SM, NM, EM, GM etc.

Assessment of the ozonation against pathogenic bacteria in the effluent of the quarantine station

  • This study investigated how ozone treatment can successfully inactivate pathogenic bacteria in both artificial seawater and effluents discharged from the fishery quarantine station in Pyeongtaek Port, Korea. Vibrio sp. and Streptococcus sp. were initially inoculated into the artificial seawater. All microbes were almost completely inactivated within 10 min and 30 min by injecting 6.4 mg/min and 2.0 mg/min of ozone, respectively. It was discovered that the water storing Pleuronichthys, Pelteobagrus, and Cyprinus imported from China contained the indicator bacteria, Vibrio sp., Enterococcus sp., total coliforms, and heterotrophic microorganisms. Compared to the control, three indicator bacteria were detected at two to six times higher concentrations. The water samples displayed a diverse microbial community, comprising the following four phyla: Bacteroidetes, Proteobacteria, Firmicutes, and Actinobacteria. Almost all indicator bacteria were inactivated in 5 min at 2.0 mg/min of ozonation; comparatively, 92.9%-98.2% of the less heterotrophic microorganisms were deactivated within the same time period. By increasing the dosage to 6.4 mg/min, 100% deactivation was achieved after 10 min. Despite the almost complete inactivation of most indicator bacteria at high doses after 10 min, several bacterial strains belonging to the Proteobacteria have still been found to be resistant under the given operational conditions.

RPS5A Promoter-Driven Cas9 Produces Heritable Virus-Induced Genome Editing in Nicotiana attenuata

  • The virus-induced genome editing (VIGE) system aims to induce targeted mutations in seeds without requiring any tissue culture. Here, we show that tobacco rattle virus (TRV) harboring guide RNA (gRNA) edits germ cells in a wild tobacco, Nicotiana attenuata, that expresses Streptococcus pyogenes Cas9 (SpCas9). We first generated N. attenuata transgenic plants expressing SpCas9 under the control of 35S promoter and infected rosette leaves with TRV carrying gRNA. Gene-edited seeds were not found in the progeny of the infected N. attenuata. Next, the N. attenuata ribosomal protein S5 A (RPS5A) promoter fused to SpCas9 was employed to induce the heritable gene editing with TRV. The RPS5A promoter-driven SpCas9 successfully produced monoallelic mutations at three target genes in N. attenuata seeds with TRV-delivered guide RNA. These monoallelic mutations were found in 2%-6% seeds among M1 progenies. This editing method provides an alternative way to increase the heritable editing efficacy of VIGE.

Purification and Characterization of Bacteriocin Produced by Enterococcus sp. (Enterococcus sp.가 생산하는 Bacteriocin의 정제 및 특성에 관한 연구)

  • We isolated microorganism secreting antimicrobial substance from tomato and identified as Enterococcus faecium. This substance was completely inactivated by pretense treatment and retained activity after catalase treatment. This result indicated that the antimicrobial activity of this substance was due to proteinaceous substance known as bacteriocin. The bacteriocin inhibited growth of Gram positive bacteria, such as Listeria monocytogenes, Leuconostoc mesenteroides, Lactobacillus plantarum, Streptococcus agalactiae, Streptococcus pyrogenes, and Gram negative bacteria, such as Pseudomonas aeruginosa. Purification of the bacteriocin was achieved by ethanol precipitation, ion exchange chromatography on CM Sepharose CL-6B, and gel filtration on Sephacryl S-100 HR. After these purification steps, the specific activity of the bacteriocin was increased 35.8 fold compared with culture broth. Purified bacteriocin was shown single band on SDS-PAGE and molecular weight was estimated 51 kDa. The residual activity of this bacteriocin was 3.3% at 10$0^{\circ}C$ for 60 min, and this bacteriocin was stable at pH 2~7.

  • Purpose: Hematopoietic cell transplantation (HCT) recipients are vulnerable to invasive infection by Haemophilus influenzae type b (Hib) and Streptococcus pneumoniae (Sp). This study was performed to evaluate immune responses after Hib and Sp vaccination in Korean pediatric HCT recipients. Methods: Patients were prospectively enrolled at Samsung Medical Center during 2009-2011. ELISA tests to detect anti-PRP IgG antibody and antibodies to Sp serotypes 4, 6B, 9V, 14, 18C, 19F, and 23F were performed at the Center for Vaccine Evaluation and Study, Ewha Medical Research Institute. Results: Ten patients (two allogeneic, eight autologous recipients) with median age 5.4 years (range 2.7-12.2 years) were enrolled. Before Hib vaccination, 60% of patients' anti-PRP IgG titers were below $0.15{\mu}g/mL$. After vaccination, 100% of patients' anti-PRP IgG titers increased above $0.15{\mu}g/mL$ (cut-off value for detection) and $1.0{\mu}g/mL$ (cut-off value for seroprotection). For pneumococcus, in 2-5 year-old patients, pre-vaccination geometric mean concentrations (GMCs) of IgG for six serotypes (4, 6B, 9V, 14, 18C, and 23F) were below $0.35{\mu}g/mL$ and at 5 months post-vaccination GMCs of IgG for all seven serotypes increased to above $0.35{\mu}g/mL$. In patients older than 5 years, pre-vaccination GMCs of IgG for four serotypes (4, 9V, 14, and 23F) were below $0.35{\mu}g/mL$ and at 3 months post-vaccination GMCs of IgG for all seven serotypes increased to above $0.35{\mu}g/mL$. Conclusion: Most HCT recipients had low or no protective antibodies to Hib and Sp before vaccination, but showed good immune responses to protective levels after vaccination.
