  • Here, we have cloned and sequenced Streptococcus parauberis pgm gene, encoding the enzyme phosphoglucomutase (PGM), which is known to be in association with virulence in other streptococcal species. The PGM of S. parauberis is the most closely related to that of S. iniae based on their amino acid sequences.

국내산 봉독의 어류병원성 세균에 대한 항균활성 (Antimicrobial activity of honeybee venom against fish pathogenic bacteria)

  • 본 연구에서는 양식 넙치의 주요 어병세균인 E. tarda, V. ichthyoenteri와 S. iniae에 대한 국내산 꿀벌로부터 분리한 봉독의 항균활성을 검색하였다. 그 결과 봉독은 폐사 납치로부터 분리한 그람음성균인 E. tarda, V. ichthyoenteri와 양성균인 S. iniae 모두에서 강한 항균 활성을 나타내는 것으로 확인되었다. 봉독은 E. tarda와 V. ichthyoenteri에 대하여 최소발육저지 농도는 각각 17.6 ${\mu}g$/ml, 1.76 ${\mu}g$/ml 였으며, 최소살균 농도는 34.6 ${\mu}g$/ml, 6.8 ${\mu}g$/ml, 항균력 지속시간은 5시간, 6시간으로 측정되었다. 또한 그람 양성균인 S. iniae에 대한 최소발육저지농도는 3.49 ${\mu}g$/ml, 최소살균농도는 11 ${\mu}g$/ml였으며, 항균력지속시간은 7시간으로 봉독은 매우 강한 증식 및 살균 억제력을 보였다. 뿐만 아니라, 다양한 pH 조건하에서도 향균활성이 유지되는 것으로 확인되었다.

  • Streptococcal diseases are known as serious problems in fresh water and marine fish culture industry worldwide. Recently, the importance of the infection of Streptococcus parauberis has been increased among the streptococcicosis since severe outbreaks in cultured olive flounder (Paralichthys olivaceus) were recorded in Korea. The aims of the present study were to investigate immunomodulatory differences between S. parauberis and S. iniae. In this study, S. parauberis isolated from diseased olive flounder was investigated on the characteristics of morphological and immune responses. Immune response of the fish to the pathogen was characterized by bactericidal activity in the serum, phagocytic activity and reactive oxygen intermediaters (ROIs) production of olive flounder. The capsules of both bacteria, S. parauberis and S. iniae seem to be associated with the ability to resist killing activity of normal sera. Also, capsulated strains could survive in the phagocytes and induce to lower ROIs production.

Effects of Dietary Hizikia fusiformis on Growth and Immune Responses in Juvenile Olive Flounder (Paralichthys olivaceus)

  • An eight week feeding trial was conducted to investigate the effects of dietary supplementation of hizikia (Hizikia fusiformis) on growth performance, immune responses and resistance of juvenile olive flounder (Paralichthys olivaceus) to Streptococcus iniae. Four experimental diets (designated as Hiz 0, Hiz 2, Hiz 4 and Hiz 6) were formulated to be isonitrogenous (50% crude protein) and isocaloric (17.2 MJ/kg DM). Hizikia powder was added at 0%, 2%, 4% and 6% in diets Hiz 0, Hiz 2, Hiz 4 and Hiz 6, respectively. Three replicates of fish groups (15 fish/tank) were fed one of the experimental diets. At the end of feeding trial, no significant differences were observed in final body weight, specific growth rate, protein efficiency ratio, feed utilization and feed intake among fish groups fed the experimental diets. However, there was clear trend that the growth performances of fish were improved by the increment of dietary hizikia showing a positive growth effects. Mean phagocytes activated with nitro-blue-tetrazolium were significantly increased with the increment of dietary hizikia. The cumulative mortality was significantly (p<0.05) lower in the fish groups fed Hiz 6 diet (no mortality) than that in the other fish groups for 15 days of S. iniae challenge test. The findings of this study suggest that a dietary supplementation of hizikia could enhance the nonspecific immune response and improve the resistance of juvenile olive flounder to S. iniae.

국내 양식 넙치에서 발생하는 연쇄구균병에 관하여 (About Streptococcosis of Cultured Olive Flounder, Paralichthys olivaceus in South Korea)

  • 본 내용은 최근 국내 양식 넙치에서 발생하는 연쇄구균병에 관하여 정리한 내용으로, 주요 균종은 Streptococcus iniae, S. parauberis 그리고 Lactococcus garvieae이다. 최근 발생경향은 지금까지 보고된 내용과 2002년에서 2017년 사이에 국립수산과학원 균주은행의 기탁균주에 대한 정보를 기초자료로 검토하였다. S. iniae와 L. garvieae는 계속 검출빈도가 줄어들었고, 균주은행 기탁균주의 경우에도 두 균주 각각 2012년과 2002년 이후로 기탁 건수가 없었다. 최근 양식 넙치에서 우점적으로 분리되는 연쇄구균병의 원인세균은 S. parauberis이다. 또한 육안적 증상과 병리조직학적 변화를 함께 비교·검토하였는데, S. iniae 감염에서는 안구백탁, 출혈, 장염 그리고 복수저류가, S. parauberis 감염에서는 체색흑화, 심외막염이 주요증상이다. L. garvieae 감염은 연쇄구균병에서 나타나는 증상이 공통적으로 나타나지만, 독성은 다른 두 종의 증상과 비교하여 약한 것으로 판단되었다. 병리조직학적 증상은 급성인지 만성인지에 따라 내부 장기별로 변성진행에 차이를 나타내지만 육안적으로 관찰되는 주요증상이 조직학적 소견에 반영된다.

  • 세균성 어류질병의 치료 및 예방을 위하여 어류와 인체에 안전한 수산용 probiotic의 개발이 요구되고 있다. 본 연구에서는 남해안 일대에서 양식되고 있는 참굴 (Crassostrea gigas), 바지락 (Ruditapes philippinarum), 피조개 (Scapharca broughtonii), 새조개 (Fulvia mutica)의 가식 부위로부터 17균주의 candidate probiotic bacteria (CPB)를 분리하였다. 나아가 다양한 연쇄상구균 (Lactococcus garvieae, L. piscium, Streptococcus sp., S. iniae, and S. parauberis)에 대하여 강한 생장 억제력을 보이는 균주(CPB-St)를 선별하여 어류의 연쇄구균증 관리를 위한 probiotic 균주로서의 개발 가능성을 알아보았다. CPB-St 균주를 double layer test를 통하여 다양한 연쇄상구균들에 대한 생장 억제 정도를 알아보았으며, 혼합 배양에서의 Streptococcus sp.에 대한 생장 억제 능력을 확인하였고, CPB-St의 어류에 대한 안전성 및 장내 생존 유무를 평가하였다. CPB-St는 대부분의 연쇄상구균에 대하여 18~26 mm의 생장 억제대를 형성함으로써 높은 생장 억제 능력을 보였다. CPB-St와의 혼합 배양에서 Streptococcus sp.는 6시간째부터 생장 억제 현상이 관찰되기 시작하여 12시간 전후로 8~55배 정도의 감소를 나타내었다. CPB-St의 어류에 대한 병원성 유 무를 알아본 결과, 2주일 동안 CPB-St로 인한 어류 폐사는 관찰되지 않았다. CPB-St의 1회 강제 경구 투여 후, 위와 장에서 CPB-St의 생존을 확인한 결과 투여 24시간 후에는 CPB-St가 모두 배출되는 것으로 나타났다. CPB-St를 probiotic bacteria로 개발하기 위해서는 어류의 장내 정착 가능성과 먹이를 통한 효과의 정확한 검증이 추후 이루어져야 할 것이다.

  • Two consecutive studies were conducted to evaluate the effects of dietary supplementation with fermented garlic powder (FGP) or fermented garlic fluid (FGF) on growth performance, immune responses, and disease resistance of olive flounder Paralichthys olivaceus. In experiment I, olive flounder (BW: 65 g) were fed four experimental diets formulated to contain 0%, 0.5%, 1%, and 1.5% FGP (designated as FGP-0, FGP-0.5, FGP-1, and FGP-1.5, respectively). After the 10-weeks feeding trial, feed intake was significantly lower in fish fed the FGP-0.5 and FGP-1.0 diets, as compared to those fed the control diet. Fish fed the FGP-0 and FGP-0.5 diets showed significantly lower survival, as compared to the other treatments. Dietary supplementation with FGP resulted in higher non-specific immune responses than the FGP-0 group. Plasma cholesterol and triglyceride levels decreased as dietary FGP level increased. In experiment II, olive flounder (BW: 65 g) were fed four experimental diets for 10 weeks. The diets were prepared with a commercial expanded pellet to have 0%, 0.25%, 0.5%, and 1% FGF (designated as FGF-0, FGF-0.25, FGF-0.5, and FGF-1, respectively) by adsorption. At the end of the second feeding trial, feed intake was significantly lower in fish fed the FGF-0 diet, as compared to other treatments. Fish fed the FGF-0.25 and FGF-0.5 diets exhibited significantly lower cholesterol levels, as compared to other treatments. Lysozyme activity significantly increased with increases in dietary FGF. Cumulative mortality in a challenge test with Streptococcus iniae was significantly lower in the fish groups fed FGF-supplemented diets than in fish fed the control diet. The results of this study indicated that dietary supplementation with FGP or FGF can enhance the non-specific immune responses and disease resistance of olive flounder against S. iniae.

손바닥선인장 줄기 가수분해 분획물의 어류 병원성 세균에 대한 항균활성 (Antimicrobial activity of hydrolysis compound of cactus stem (Opuntia ficus-indica var. saboten Makino) against fish pathogenic bacteria)

  • 손바닥선인장(Opuntia ficus-indica var. saboten Makino) 줄기 가수분해 분획물(MBT-01108)의 어병세균에 대한 in vitro 항균활성을 조사하였다. 실험에 사용한 어병세균은 Edwardsiella tarda, Listonella anguillarum, Vibrio sp., Lactococcus garvieae, L. raffinose, Streptococcus parauberis, S. iniae 등 총 18종이었다. 또한, oxytetracycline을 포함한 시판 항균제 disc를 MBT-01108의 항균활성에 대한 양성 대조군으로 하여 항균활성을 비교 조사하였다. 그 결과, MBT-01108은 실험에 사용한 어병세균들에 대하여 우수한 항균활성을 나타내었다.

식물 정유 주요 성분의 어병 세균에 대한 항균활성 (Antibacterial Effects of Major Compounds in Essential Oil against Fish Disease Bacteria)

  • In this study, the antibacterial activity of major compounds obtained from domestic plant essential oils was investigated against four species of fish pathogenic bacteria, namely, Edwardsiella tarda, Vibrio harveyi, Streptococcus iniae, and Streptococcus parauberis. We observed that α-terpineol and eucalyptol showed high antibacterial activity against S. iniae, whereas α-terpineol and β-linalool showed high antibacterial activity against S. parauberis. The antibacterial activity of eucalyptol and β-linalool was significantly high against E. tarda, and α-terpineol and β-linalool exerted antibacterial activity against V. harveyi. All compounds showing significant antibacterial activity also exhibited high solubility in water (≥1,000 mg/L). In contrast, compounds such as α-limonene and α-pinene with low water solubility showed significantly low antibacterial activity against all bacteria. These findings can be utilized further to estimate the antibacterial activity of compounds isolated from plant essential oils for the prevention of fish disease.

Fluctuation of Temperature Induces Pathogenicity of Streptococcus iniae and Changes of Immunology Related Genes of Korean Rockfish, Sebastes schlegeli

  • This study was designed to examine the immune response in Korean rockfish during water temperature fluctuation and to elucidate the factors contributing to streptococcal pathogenesis in cultured Korean rockfish, S. schlegeli. We investigated cumulative mortality against Streptococcus iniae (FP5228 strain) infection in the exposed Korean rockfish (39.7±5.8 g) to environmentally relevant temperature (Control, 23℃; High temperature, 28℃ and 23℃ and 28℃ with 12 hours interval exchange, 23↔28℃) for 48 hours. Also, the expression of the mRNA related to the immune response genes (heat shock protein 70, interleukin1β, lysozyme g-type and thioredoxin-like 1) were measured in spleen and head kidney by real-time PCR analysis in the exposed fish to thermal stress. In this study, the combined stress with bacterial challenge in fishes exposed to thermal stress lowered the survival rate than that of control (23℃). The cumulative mortality in the group of control, 28℃ and 23↔28℃ was 24%, 24% and 40% (P<0.05), respectively. Also, thermal stress modulated the mRNA level of immune related genes; heat shock protein 70, interleukin-1β, lysozyme g-type and thioredoxin-like 1 in Korean rockfish. The present study indicates that a high and sudden water temperature change affect immune responses and reduce the disease resistance in Korean rockfish.
