Antimicrobial Effects of Ethanol Extract of Yongdamgosam-hwan against Streptococcus mutans (용담고삼환(龍膽苦參丸) 에탄올 추출물의 Streptococcus mutans에 대한 항균활성에 관한 연구)

  • Objectives : Yongdamgosam-hwan(YGH) has been used as a traditional medicine from old times for antiinflammatory effects. Streptococcus mutans(S. mutans) is known as a prime bacteria responsible for causing caries by forming a biofilm referred to as dental plaque on the tooth surface. But antimicrobial activity of YGH with dental disease is not sufficiently understood. This study was designed to investigate the effects of YGH ethanol extract on antimicrobial effect against Streptococcus mutans.Methods : The antimicrobial effect of YGH ethanol extract was assessed by the paper disk diffusion method and optical density method to determine minimum inhibition concentration(MIC), also observed by fractional inhibitory concentration index(FICI) and time-kill assay to figure out the synergic effect on the combination of YGH ethanol extract with antibiotics.Results : The YGH ethanol extract 500 μg was 7.5-8.5 mm diameter of clear zone of inhibition against Streptococcus mutans in a concentration-dependent manner and MIC was 250 μg/mL. The administration of the ethanol extract in combination with gentamicin and streptomycin induced a reduction of ≥4-8-fold in all tested bacteria. Furthermore, time-kill study was found that a combination of YGH ethanol extract with oxacillin and streptomycin produced a more rapid decrease in the concentration of bacteria CFU/mL than the YGH ethanol extract or antibiotics alone.Conclusions : As a result, the YGH ethanol extract has good antimicrobial effects. And the results suggest that YGH could be employed as a natural antibacterial agent in dental care products.

Effects of Lignum sappan Extract on the Growth of Streptococcus mutans KCTC 3065 (소목 분획물이 Streptococcus mutans KCTC 3065의 생장에 미치는 효과)

  • This study was to study the antibacterial effect of Lignum sappan extract on S. mutans growth. The antibacterial activities of Lignum sappan's crude extract was measured 17.3 mm at 20 mg/ml concentration. The growth of S. mutans in control medium was the highest at 8hr, while the media of Lignum sappan extract added-medium (2 mg/ml) showed maximum growth at 16hr. The pH values of the control media was 5.08 at 8hr, but the media supplemented with Lignum sappan extract was 6.69 at 8hr. The amounts of total carbohydrate of the control media was 0.81 mg/ml at 8hr, but the media supplemented with Lignum sappan extract was 2.06 mg/ml at 8hr. In the protein change of culture medium, the control culture broth and the cultures supplemented with Lignum sappan extract was 8.39 mg/ml and 12.3 mg/ml at 8hr, respectively. The S. mutans polysaccharide contents of the control media and the media supplemented with Lignum sappan extract was 300 mg/100ml and 60 mg/100ml at 8hr, respectively.

Isolation and Identification of Anticariotic Compound from Sophora flavescens Ait. (고삼으로부터 항우식활성 물질의 분리)

  • The purpose of this study is to investigate anticariotic activity of the ethyl acetate soluble extract of Sophora flavescens Ait. for the prevention of dental caries and glucosyltransferase activity caused by Streptococcus mutans. The fraction 5-4-3 showed strong growth inhibition activity against Streptococcus mutans (MIC, 3.13 $\mu\textrm{g}$/ml). The glucosyltransferase activity of the active fraction 5-4-3 inhibited the formation of glucan and showed 77% of the antiproliferative effect at 100 $\mu\textrm{g}$/ml (P<0.05). Two flavanones, (2S)-2'-methoxy kurarinone (1) and (+)-kurarinone (2), were isolated from the active fraction 5-4-3 of the ethyl acetate soluble extract of S. flavescens Ait. Their structures were elucidated using spectroscopic methods.

Inhibitory Effects of Galla Chinensis Extract on Cariogenic Properties of Streptococcus mutans (오배자 Galla Chinensis 추출물이 Streptococcus mutans의 우식활성 억제에 미치는 영향)

  • Streptococcus mutans (S. mutans) is one of the most important bacteria in the formation of dental plaque and dental caries. S. mutans adheres to an acquired pellicle formed on the tooth surface, and aggregates with many oral bacteria, and initiates plaque formation by synthesizing glucan from sucrose, which is catalyzed by glucosyltransferases. S. mutans metabolizes the dietary sugar to the organic acids. The organic acids demineralize tooth surface and result in dental caries. Galla Chinensis have been traditionally used for stopping bleeding of gingiva, removing edema and halitosis, drainage, fixing the teeth and as an antiphlogistic agent. In previous reports, antibacterial effects of Galla Chinensis have been investigated whereas anti-cariogenic effects is still not examined enough. Therefore we tested effects of ethanol extracts of Galla Chinensis on the cariogenic properties such as the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. In the result, ethanol extracts of Galla Chinensis showed the inhibition of S. mutans growth and organic acids production over 0.031 mg/ml concentrations. The adhesion of S. mutans to Saliva-coated Hydroxyapatite beads S-HAs has decreased with the increase of concentration of ethanol extracts of Galla Chinensis. And it seems to have adhesion inhibitory effect in concentration of over 0.25 mg/ml. It gives us the result that Galla Chinensis have anti-caries effects. But ethanol extract of Galla Chinensis didn't have inhibitory effect on insoluble glucan synthesis. Preliminary phytochemical analysis of the ethanol extract of Galla Chinensis showed strong phenolic compounds, medium steroids & terpenoids and glycosides, and weak organic acids and peptides. These results suggest that the ethanol extracts of Galla Chinensis may have anti-cariogenic properties, which may be able to be related with strong phenolic compounds.

Analysis of Acid Stress Response in Streptococcus mutans KCTC 3065 (산에 대한 Streptococcus mutans KCTC 3065의 스트레스 반응에 관한 연구)

  • Dental caries is initiated by the acid accumulated in dental plaque. Streptococcus mutans, one of a major causal agents of dental caries, is component of the dental plaque and produces various organic acids such as lactic acid as the end-product of glycolysis. As a consequence, we investigated the acid stress response of S. mutans KCTC 3065 in this study. The addition of lactic acid to the growth media had a concentration-dependent effect on the growth of S. mutans. S. mutans exhibited higher maximum culture OD compared with the more acidic growth pH values. At treatment of centration of 20 mmol/L lactic acid in the mid-log phage, cell growth was reduced to 40% relative to the control. The following results were obtained with the treatment of cells with a concentration of 20 mmol/L lactic acid in the mid-log phage for 2hrs: Analysis of fatty acids extracted from cells showed that growth at a concentration of 20 mmol/L lactic acid resulted in changes in $C_{14:0}$, $C_{16:0}$, $C_{18:0}$ and $C_{18:1}$ fatty acids. Protein profiles investigated by SDS-PAGE showed that approximately 70, 60, 45, 40 and 23 kDa proteins were highly expressed in S. mutans KCTC 3065.

Neutral Electrolyzed Water for Prevention of Dental Caries (기존 구강청결제를 대체할 수 있는 치아우식 예방을 위한 전기분해수)

  • Gargle solution has typically been used for the prevention of oral infectious disease such as dental caries and periodontitis. However, the use of most gargle solutions is controversial in application for children because some gargle solutions have harmful side effects. Electrolyzed water is generated by passed an electric current and has antimicrobial activity. The purpose of this study was to investigate and compare the efficacy of electrolyzed water in various conditions for eliminating cariogenic bacteria. Electrolyzed water was generated by a platinum electrode in the presence of sodium chloride at various concentrations. Streptococcus mutans and Streptococcus sobrinus were cultivated into a brain heart infusion broth. After harvesting planktonic bacteria, the pellets were treated with the electrolyzed water and commercial gargle solutions and plated on a mitis-salivarius agar plate. Also, the anti-biofilm activity of the electrolyzed water and commercial gargle solutions was investigated after biofilm formation of S. mutans and S. sobrinus. The bacteria in the biofilm were plated onto a mitis-salivarius agar plate. The plates were incubated, and the colony forming unit was measured. The electrolyzed water containing sodium chloride showed significant antibacterial activity against S. mutans and S. sobrinus as well as some gargle solutions. Furthermore, the electrolyzed water had more disruptive effect on the biofilm of S. mutans and S. sobrinus and killed more bacteria in the biofilm than commercial gargle solutions. The results demonstrate that electrolyzed water may be a useful gargle solution for prevention of dental caries.

Effect of Diet Supplements on the Production of Anti-Dental Caries Hen's Eggs by Immunization of Streptococcus mutans (Streptococcus mutans 균의 면역과 사료첨가제가 충치 예방용 계란의 생산에 미치는 영향)

  • To increase IgY in egg yolks, hens were fed a feed supplemented with kelp meal $4\%$ cinnamon $0.3\%$ and mint $2\%$ respectively, and immunized 5 times with Streptococcus mutans(S. mutans) at 2 week intervals. Groups fed experimental feeds without immunization showed higher laying rate than the control group, without supplementary feed and immunization. After the immunization, the laying rates had been decreasing due to the stress of immunization. The laying rate was recovered after the termination of immunization. Egg weight was not affected by the immunization but diets. Feed intake was dependent on the laying rate. Total IgY concentration in eggs laid from hens fed feeds containing supplementary feeds was higher than that of control. Especially, total IgY was increased up to $7.9\%$ in eggs laid from hens fed feeds supplemented with $4\%$ of kelp meal. Anti-S. mutans IgY was detected at 4 weeks after first immunization. Activity of anti-S. mutans IgY was sustained at 5 week after the final immunization. As the average concentration of specific IgY during the experimental period showed that eggs from hens fed the feed containing $4\%$ of kelp meal increased the specific IgY by $8.5\%$ kelp meal supplement improved specific IgY production by immunization.

Screening of Leaves of Higher Plants for Antibacterial Action

  • The methanol and benzene extracts of the leaves of 55 higher plants in Korea were tested for their antibacterial activity against three Gram positive bacteria, Streptococcus mutans, Staphylococcus aureus and Bacillus subtilis, and one Gram negative bacterium Escherichia coli. Among them, the methanol extract of the leaves of Liriodendron tulipifera showed remarkably potent antibacterial activity against both Gram positive and negative bacteria.

Acid Stress-Induced Proteins of the Streptococcus mutans Isolated from Korean Children with Caries (한국인 우식아동으로부터 분리한 Streptococcus mutans의 내산성 단백질의 발현)

  • 한국인 아동의 우식치아로부터 S. mutans를 분리하고, acid stress하에서 분리한 S. mutans의 내산성 능력과 관련된 단백질을 규명하고자 하였다. S. mutans의 16S rDNA 유전자 핵산염기서열을 바탕으로 설계된 프라이머 쌍을 이용하여 중합효소연쇄반응을 실시하고 16S rDNA sequencing을 실시한 결과, 한국인 아동으로부터 분리 된 S. mutans K7은 기존에 보고된 표준균주와 99.9% 일치하는 결과를 나타내 S. mutans로 판명되었다. 또한 2D gel electrophoresis 를 수행한 결과, acid stress에 관여하는 것으로 추정되는 단백질들을 확인 할 수 있었다.
