FBG센서가 내장된 스마트 텐던을 이용한 7연 강연선의 인발 하중전이 특성 (Load Transfer Characteristics of the 7-wire strand using FBG Sensor Embedded Smart Tendon)

  • 최근 도심 구조물의 규모가 급격히 대형화됨에 따라 인접구조물이 존재하는 상태에서 대규모의 기초구조물의 설치를 위한 넓고 깊은 범위의 굴착이 빈번히 시행되고 있어 안전관리의 중요성이 크게 부각되고 있다. 시공 및 사용 중에 토류구조물의 안전성을 확보하기 위해서는 쏘일앵커, 쏘일네일 및 락앵커 등의 지반보강재에 작용하는 프리스트레스 하중(prestress force) 및 변형을 지속적이고 효과적으로 관측할 수 있는 방법이 필요하다. 그러나 현재 현장에서 주로 사용되는 전기저항식 로드셀과 스트레인게이지, 바이브레이팅 타입의 변형율계를 이용한 변형 및 장력의 측정은 센서 자체의 자기열화 특성 때문에 장기적인 모니터링에는 효과적이지 않을뿐더러 시험체 내부에서 다수의 측정점을 측정하기 위해서는 많은 스트레인게이지를 설치하기 위해 리드선의 공간이 크게 필요한 단점이 있다. FBG(Fiber Bragg Grating)센서는 스트레인게이지와 비교해서 매우 작은 직경을 가지며 전자기파에 의한 노이즈가 없고 내구성이 커서 장기적인 모니터링이 필요한 구조물에 매우 유용하게 사용될 수 있다. 본 연구에서는 7연 강연선의 센터 킹케이블에 FBG 센서를 내장한 스마트 텐던을 이용하여 토류구조물의 보강에 활용되고 있는 앵커와 주변 그라우트면의 하중전이 특성에 대한 일련의 실내실험 결과를 기술하였다. 연구결과 스마트 텐던은 기존의 스트레인 게이지 변형율계로는 마땅히 측정할 수 없던 7연 강연선과 그라우트면의 변형률을 매우 효과적으로 측정할 수 있을 뿐 아니라 하나의 선으로 여러 위치의 변형을 효과적으로 측정할 수 있어 지반보강재의 장력모니터링 뿐만 아니라 그라우트로 부착된 부분의 하중전이 특성을 효과적으로 파악할 수 있는 것으로 나타났다.

Microbacterium sp. A-210이 생성하는 Levan fructotransferase의 정제 및 생물학적 특성에 관한 연구 (Purification and Biological Characterization of Wild-type and Mutants of a Levan Fructotransferase from Microbacterium sp. AL-210)

  • DFA (Difructose anhydride)는 특유의 구조적인 안정성 때문에 당뇨병 환자를 위한 당원으로써 적합하다는 연구가 보고 되어 있다. DFA에는 4가지 type이 있는데 inulin에 의한 DFA I DFA III DFAV가 있고 levan에 의한 DFA IV가 있는 것으로 알려져 있다. 특히 DFA IV는 당뇨병 환자를 위한 당원 뿐 만 아니라 rat을 이용한 연구에서 칼슘의 흡수를 도와 준다는 보고가 있었다. 이러한 DFAIV를 생성하는 데 쓰이는 Microbacterium sp. AL-210에서 유래한 LFTase (Levan fructotransferase)의 wild-type과 mutants (D63A, D195N, N85S)의 구조적 특성을 밝히기 위해 정제하였다. LFTase의 wild-type과 mutants들을 대량 발현시킨 후 흡착 크로마토그래피, 이온교환 크로마토그래피 그리고 젤 여과 크로마토그래피를 이용하여 고순도로 분리 정제하였으며 이를 SDS-PAGE를 통하여 확인하였다. 분리 정제된 단백질을 JNET 이차 구조 예측 프로그램, solubility 측정, CD (원 편광 이색성 분광편광계), fluorescence spectroscopy (형광분석법), DSC (시차주사열량계)를 이용하여 분석하였다. 또한 다중 정렬과 2차 구조 예측 프로그램을 이용하여 wild-type의 2차 구조를 분석하였다. Solubility 측정에서 가장 적합한 온도는 $55^{\circ}C$, 최상의 pH는 7.5로 나타났다. CD 분석에서 wild-type과 비교한 결과 다른 mutant에 비해 N85S의 $\alpha$-helix가 많이 감소한 것과 $\beta$ strand와 random coil이 증가한 것을 확인하였다. 또한 DSC 분석을 통해 wild-type이 다른 mutants에 비해 안정적인 구조를 지닌 것을 확인하였다. 형광분석에서 N85S가 wild-type과 가장 유사하게 나타났으며 D63A와 D195N은 wild-type에 비해 높은 강도를 나타내었다. 또한 wild-type의 sequence를 Exo-inulinase from Aspegillus awamori, a plant fructan 1-exohydrolase from Cichorium intybus 그리고 invertase from Thermotogo maritime (Tm)의 sequence와 다중 정렬한 결과 Exo-inulinase와 높은 identity를 보였다.

모형실험 및 수치해석을 이용한 앵커형 네일의 적용성 검토 (Investigations of Application of Anchor Type Nail Using Experiments and Numerical Analysis)

  • 본 논문에서는 지반 내 보강재를 삽입하여 원지반 전체의 전단강도와 활동저항력을 증가시켜주는 네일링에 프리스트레스를 가할 수 있는 PC강연선을 결합한 앵커형 네일에 대한 연구를 수행하였다. 앵커형 네일의 거동특성을 확인하기 위하여 무보강, 네일링, 앵커형 네일 각각에 대하여 실내모형실험과 수치해석을 수행하여 하중-변위, 벽체변위 등을 분석하였다. 실내모형실험결과 프리텐션을 가한 앵커형 네일의 경우 일반네일과 비교할 때 하중증가에 따른 수평변위량이 크게 감소되는 것이 확인되었으며, 특히 벽체상부의 변위 억제력이 커짐을 알 수 있었다. 이를 확인하기 위한 수치해석결과도 실내모형실험과 유사한 해석결과를 나타내었다.

Minimal Medial-row Tie with Suture-bridge Technique for Medium to Large Rotator Cuff Tears

  • Background: The purpose of this study was to evaluate the postoperative magnetic resonance imaging (MRI) results of minimal-tying (one medial-row tie among 4 medial-row sutures) on the medial-row in double-row suture-bridge configuration ($2{\times}2$ anchor with $4{\times}4$ suture stands). Methods: From 2011 March to 2012 July, 79 patients underwent arthroscopic rotator cuff repair using $2{\times}2$ anchor double-row configuration. The mean age was 61.3 years (range, 31-81 years). Two double-loaded suture anchors were used for medial-row. Four medial-row stitches were made with only one medial-row knot-tying (the most anterior suture). Lateral-row was secured using the conventional suture-bridge anchor technique; all 4 strands were used for each anchor. Repair integrity was evaluated with MRI at mean 6.2 months postoperatively. Retear and the pattern of retear, change of fatty infiltration, and muscle atrophy of supraspinatus were evaluated using pre- and postoperative MRI. Results: Repaired tendon integrity was 38 for type I, 30 for type II, 6 for type III, 4 for type IV, and 1 for type V, according to Sugaya classification. Considering type IV/V as retear, the rate was 6.3% (5 out of 79 patients). Medial cuff failure was observed in 4 patients. Fatty atrophy of supraspinatus was significantly improved postoperatively according to Goutallier grading (p=0.01). The level of muscle atrophy of supraspinatus was not changed significantly after surgery. Conclusions: Minimal tying technique with suture configuration of four-by-four strand double-row suture-bridge yielded a lower retear rate (6.3%) in medium to large rotator cuff tears.

  • Cable-In-Conduit Conductor(CICC) is widely accepted as an advanced superconductor configuration for large scale applications such as tokamak fusion reactors, MAGLEV (MAGnetic LEVitation), and SMES (Superconducting Magnetic Energy Storage). The stability of CICC cooled with supercritical helium can be very high if it is operated below a certain limiting current. This limiting current can be determined by Stekly type heat balance equation. The stability characteristic of CICC for AC operation is more complicated than that of DC because there are additional instability sources which are associated with local flux change. Ramp-rate limitation is a phenomenon discovered during US-DPC (United States-Demonstration Poloidal Coil) program, which showed apparent quench current degradation associated with high dB/dt. This paper describes recent experimental investigation results on the ramp-rate limitation and discusses current imbalance, induced current, current redistribution due to local quench of the strand in the cable.

Plant RNA Virus-Host Interaction: Potato virus X as a model system

  • Potato virus X (PVX), the type member of Potexvirus genus, is a flexuous rod-shaped virus containing a single-stranded (+) RNA. Infection by PVX produces genomic plus- and minus-strand RNAs and two major subgenomic RNAs (sgRNAs). To understand the mechanism for PVX replication, we are studying the cis- and/or trans-acting elements required for RNA replication. Previous studies have shown that the conserved sequences located upstream of two major sgRNAs, as well as elements in the 5' non-translated region (NTR) affect accumulation of genomic and sg RNAs. Complementarity between sequences at the 5' NTR and those located upstream of two major sgRNAs and the binding of host protein(s) to the 5' NTR have shown to be important for PVX RNA replication. The 5 NTR of PVX contains single-stranded AC-rich sequence and stem-loop structure. The potential role(s) of these cis-elements on virus replication, assembly, and their interaction with viral and host protein(s) during virus infection will be discussed based on the data obtained by in vitro binding, in vitro assembly, gel shift mobility assay, host gene expression profiling using various mutants at these regions.

Construction of a Genetic Linkage Map of Shiitake Mushroom Lentinula Edodes Strain L-54

  • From fruiting bodies of L. edodes strain L-54, single-spore isolates (SSIs) were collected. Two parental types of L-54 were regenerated via monokaryotization. By means of random-amplified polymorphic DNA (RAPD), DNA samples from L-54, its two parental types, and 32 SSIs were amplified with arbitrary primers. Dedikaryotization was demonstrated, and 91 RAPD-based molecular markers were generated. RAPD markers that were segregated at a 1:1 ratio were used to construct a linkage map of L. edodes. This RAPD-linkage map greatly enhanced the mapping of other inheritable and stable markers [such as those that are linked to a phenotype (the mating type), a known gene (priA) and a sequenced DNA fragment (MAT)] with the aid of mating tests, bulked-segregant analysis, and PCR-single-strand conformational polymorphism. These markers comprised a genetic map of L. edodes with 14 linkage groups and a total length of 622.4 cM.

Glycation of Copper, Zinc-Superoxide Dismutase and its Effect on the Thiol-Metal Catalyzed Oxidation Mediated DNA Damage

  • The nonenzymatic glycation of copper, zinc-superoxide dismutase (Cu,Zn-SOD) led to inactivation and fragmentation of the enzyme. The glycated Cu,zn-SOD was isolated by boronate affinity chromatography. The formation of 8-hydroxy-2'-deoxyguanosine (8-OH-dG) in calf thymus DNA and the generation of strand breaks in pBhiescript plasmid DNA by a metal-catalyzed oxidation (MCO) system composed of $Fe^{3+}$, $O_2$, and glutathione (GSH) as an electron donor was enhanced more effectively by the glycated CU,Zn-SOD than by the nonglycated enzyme. The capacity of glycated Cu,Zn-SOD to enhance damage to DNA was inhibited by diethylenetriaminepentaacetic acid (DETAPAC), azide, mannitol, and catalase. These results indicated that incubation of glycated CU,Zn-SOD with GSH-MCO may result in a release of $Cu^{2+}$ from the enzyme. The released $Cu^{2+}$ then likely participated in a Fenton-type reaction to produce hydroxyl radicals, which may cause the enhancement of DNA damage.

세그멘탈 합성보의 휨 실험 (Flexural Tests on Post-Tensioned Segmental Composite Beam)

  • The interior portion of Gerber's beam are constructed with post-tensioned segmental composite beams in this study. A precast concrete member which is larger than the limits of domestic transportation regulation in weight, length, and volume is divided into three parts, transported separately, and erected with a composite member by post-tensioning in site. Static flexural loading tests are performed on Gerber's type frames which are consisted with 2.5m overhangs and 5m interior beams composited from three pieces. The connection of overhang to interior composite beam and beam to beam, and flexural performance of interior portion of Gerber's beam are examined thoroughly. All of the tests are ended with a compression failure of the interior composite beams over the design strength of homogeneous beams. The brittle connection failures or tensile failures with the failure of lower strand was not observed in any test frames.

The Production of mutant protein by a transcription-based mechanism and in vivo technique for determining transcriptional mutagenesis

  • When an elongating RNA polymerase encounters DNA damage on the template strand of a transcribed gene it can either be arrested by or be transcribed through the lesion. Lesions that arrest RNA polymerases are thought to be subject to transcription-coupled repair, whereas that damage that is bypassed can cause miscoding, resulting in mutations in the transcript (transcriptional mutagenesis). We have developed a technique using a plasmid-based luciferase reporter assay to determine the extent to which a particular type of DNA base modification is capable of causing transcriptional mutagenesis in vivo. The system uses Escherichia coli strains with different DNA repair backgrounds and is designed to detect phenotypic changes caused by transcriptional mutageneis under nongrowth conditions. In addition, this method is capable of indicating the extent to which a particular DNA repair enzyme (or pathway) suppresses the occurrence of transcriptional mutagenesis. Thus, this technique provides a tool with which the effects of various genes on non-replication-dependent pathways resulting in the generation of mutant proteins can be gauged.
