고강도-신장플랜지성 열연강의 미세조직 및 기계적 성질 (Microstructure and Mechanical Properties of High Strength and Stretch-Flangeability Hot-Rolled Steels)

  • Research into the development of high strength (1 GPa) and superior formability, such as total elongation (10%), and stretch-flangeability (50%) in hot-rolled steel was conducted with a thermomechanically controlled hot-rolling process. To improve the overall mechanical properties simultaneously, low-carbon steel using precipitation hardening of Ti-Nb-V multimicroalloying elements was employed. And, ideal microstructural characteristics for the realization of balanced mechanical properties were determined using SEM, EBSD, and TEM analyses. The developed steel, 0.06C-2.0Mn-0.5Cr-0.2(Ti + Nb + V), consisted of ferrite as the matrix phase and second phase of granular bainite with fine carbides (20-50 nm) in both phases. The significant factor of the microstructural characteristics that affect stretch-flangeability was found to be the microstructural homogeneity. The microstructural homogeneity, manifest in such characteristics as low localization of plastic strain and internally stored energy, was identified by grain average misorientation method, analyzed by electron backscattered diffraction (EBSD) and hardness deviation between the phases. In summar, a hot-rolled steel having a composition 0.06C-2.0Mn-0.5Cr-0.2(Ti + Nb + V) demonstrated a tensile strength of 998 MPa, a total elongation of 19%, and a hole expansion ratio of 65%. The most important factors to satisfy the mechanical property were the presence of fine carbides and the microstructural homogeneity, which provided low hardness deviation between the phases.

Efficient and Cost-Reduced Glucoamylase Fed-Batch Production with Alternative Carbon Sources

  • Glucoamylase is an important industrial enzyme. Glucoamylase production by industrial Aspergillus niger strain featured with two major problems: (i) empirical substrate feeding methods deteriorating the fermentation performance; and (ii) the high raw materials cost limiting the economics of the glucoamylase product with delegated specification. In this study, we first proposed a novel three-stage varied-rate substrate feeding strategy for efficient glucoamylase production in a 5 L bioreactor using the standard feeding medium, by comparing the changing patterns of the important physiological parameters such as DO, OUR, RQ, etc., when using different substrate feeding strategies. With this strategy, the glucoamylase activity and productivity reached higher levels of 11,000 U/ml and 84.6 U/ml/h, respectively. The performance enhancement in this case was beneficial from the following results: DO and OUR could be controlled at the higher levels (30%, 43.83 mmol/l/h), while RQ was maintained at a stable/lower level of 0.60 simultaneously throughout the fed-batch phase. Based on this three-stage varied-rate substrate feeding strategy, we further evaluated the economics of using alternative carbon sources, attempting to reduce the raw materials cost. The results revealed that cornstarch hydrolysate could be considered as the best carbon source to replace the standard and expensive feeding medium. In this case, the production cost of the glucoamylase with delegated specification (5,000 U/ml) could be saved by more than 61% while the product quality be ensured simultaneously. The proposed strategy showed application potential in improving the economics of industrial glucoamylase production.

Kinetics of Kojic Acid Fermentation by Aspergillus flavus Link S44-1 Using Sucrose as a Carbon Source under Different pH Conditions

  • Kojic acid production by Aspergillus flavus strain S44-1 using sucrose as a carbon source was carried out in a 250-mL shake flask and a 2-L stirred tank fermenter. For comparison, production of kojic acid using glucose, fructose and its mixture was also carried out. Kojic acid production in shake flask fermentation was 25.8 g/L using glucose as the sole carbon source, 23.6 g/L with sucrose, and 6.4 g/L from fructose. Reduced kojic acid production (13.5 g/L) was observed when a combination of glucose and fructose was used as a carbon source. The highest production of kojic acid (40.2 g/L) was obtained from 150 g/L sucrose in a 2 L fermenter, while the lowest kojic acid production (10.3 g/L) was seen in fermentation using fructose as the sole carbon source. The experimental data from batch fermentation and resuspended cell system was analysed in order to form the basis for a kinetic model of the process. An unstructured model based on logistic and Luedeking-Piret equations was found suitable to describe the growth, substrate consumption, and efficiency of kojic acid production by A. flavus in batch fermentation using sucrose. From this model, it was found that kojic acid production by A. flavus was not a growth-associated process. Fermentation without pH control (from an initial culture pH of 3.0) showed higher kojic acid production than single-phase pH-controlled fermentation (pH 2.5, 2.75, and 3.0).

Improved Production of Medium-Chain-Length Polyhydroxyalkanoates in Glucose-Based Fed-Batch Cultivations of Metabolically Engineered Pseudomonas putida Strains

  • One of the major challenges in metabolic engineering for enhanced synthesis of value-added chemicals is to design and develop new strains that can be translated into well-controlled fermentation processes using bioreactors. The aim of this study was to assess the influence of various fed-batch strategies in the performance of metabolically engineered Pseudomonas putida strains, ${\Delta}gcd$ and ${\Delta}gcd-pgl$, for improving production of medium-chain-length polyhydroxyalkanoates (mcl-PHAs) using glucose as the only carbon source. First we developed a fed-batch process that comprised an initial phase of biomass accumulation based on an exponential feeding carbon-limited strategy. For the mcl-PHA accumulation stage, three induction techniques were tested under nitrogen limitation. The substrate-pulse feeding was more efficient than the constant-feeding approach to promote the accumulation of the desirable product. Nonetheless, the most efficient approach for maximum PHA synthesis was the application of a dissolved-oxygen-stat feeding strategy (DO-stat), where P. putida ${\Delta}gcd$ mutant strain showed a final PHA content and specific PHA productivity of 67% and $0.83g{\cdot}l^{-1}{\cdot}h^{-1}$, respectively. To our knowledge, this mcl-PHA titer is the highest value that has been ever reported using glucose as the sole carbon and energy source. Our results also highlighted the effect of different fed-batch strategies upon the extent of realization of the intended metabolic modification of the mutant strains.

동결토의 압축강도에 관한 실험적 연구 (Experimental Studies on the Compressive Strength of the Frozen Soils)

  • Upon freezing a soil swells due to phase change and its compression stress increase a lot. As the soil undergo thawing, however, it becomes a soft soil layer because the 'soil changes from a solid state to a plastic state. These changes are largely dependent on freezing temperature and repeated freezing-thawing cycle as well as the density of the soil and applied loading condition. This study was initiated to describe the effect of the freezing temperature and repeated freezing-thawing cycle on the unconfined compressive strength. Soil samples were collected at about 20 sites where soil structures were installed in Kangwon provincial area and necessary laboratory tests were conducted. The results could be used to help manage effectively the field structures and can be used as a basic data for designing and constructing new projects in the future. The results were as follows ; 1. Unconfined compressive strength decreased as the number of freezing and thawing cycle went up. But the strength increased as compression speed, water content and temperature decreased. The largest effect on the strength was observed at the first freezing and thawing cycle. 2. Compression strain went up with the increase of deformation speed, and was largely influenced by the number of the freezing-thawing cycle. 3. Secant modulus was responded sensitivefy to the material of the loading plates, increased with decrease of temperature down to - -10$^{\circ}$C, but was nearly constant below the temperature. Thixotropic ratio characteristic became large as compression strain got smaller and was significantly larger in the controlled soil than in the soil treated with freezing and thawing processes 4. Vertical compression strength of ice crystal(development direction) was 3 to 4 times larger than that of perpendicular to the crystal. The vertical compression strength was agreed well with Clausius-Clapeyrons equation when temperature were between 0 to 5C$^{\circ}$, but the strength below - 5$^{\circ}$C were different from the equation and showed a strong dependency on temperature and deformation speed. When the skew was less then 20 degrees, the vertical compression strength was gradually decreased but when the skew was higher than that, the strength became nearly constant. Almost all samples showed ductile failure. As considered above, strength reduction of the soil due to cyclic freezing-thawing prosses must be considered when trenching and cutting the soil to construct soil structures if the soil is likely subject to the processes. Especially, if a soil no freezing-thawing history, cares for the strength reduction must be given before any design or construction works begin. It is suggested that special design and construction techniques for the strength reduction be developed.

수열합성을 이용한 ZnO 마이크로 구조의 성장 및 전사 (Fabrication of Vertically Oriented ZnO Micro-crystals array embedded in Polymeric matrix for Flexible Device)

  • 최근, 유연하며 몸에 부착 가능한 소자들에 대한 관심이 늘어나고 있다. 이런 관심을 뒷받침 하여 이와 관련된 다양한 연구들이 진행되고 있는데, 기존 딱딱한 성질을 가진 소자에 사용되던 무기물 기반의 재료의 경우 유연 소자로 만들기에 여러 가지 제약이 있어 유연하게 제작할 수 있는 유기물 반도체나 탄소 나노튜브 필름 등을 이용한 소자들이 주로 연구되고 개발되어 왔다. 하지만 이런 재료들을 이용한 소자의 경우 유기물 분자와 분자 사이 또는 탄소 나노튜브와 나노튜브 사이에서 전하들이 산란되는 등 재료 자체의 한계로 인해 기존의 재료를 사용한 소자들보다 전기적 성능이 떨어지는 단점을 가지고 있다. 이런 단점들을 해결하기 위하여 이 연구에서는 수직 정렬된 반도체 결정 어레이를 투명 유연한 폴리머와 결합하는 방법을 이용, 고품질 나노/마이크로 반도체 결정을 유연한 기판으로 전사 시킬 수 있는 방법을 제시한다. 위와 같은 구조는 재료에 가해지는 힘을 완화 시켜줄 수 있으며, 이로 인해 큰 변형에도 재료의 손상이 없는 소자 제작이 가능하다. 이런 구조를 구현하기 위해 위치 및 크기가 정교하게 제어된 ZnO 나노막대 단결정을 저온에서 용액공정을 통하여 합성시킨다. 이후 성장시킨 ZnO 단결정 어레이와 polydimethylsiloxane (PDMS) 폴리머를 결합시킨 후 단단한 기판에서 기계적으로 박리시켜 ZnO/폴리머 복합체를 분리해 낸다. 추가적으로 전사된 ZnO의 결정성을 확인하기 위하여 photoluminescent 분석을 진행하였으며, ZnO/폴리머 복합체를 이용한 외부 힘에 반응하는 압력 센서를 제작하였다.

담배거세미나방에 살충효과를 나타내는 새로운 Bacillus thuringiensis 균주의 특성 (Characterization of New Bacillus thuringiensis Isolated with Bioactivities to Tobacco Cutworm, Spodoptera litura (Lepidoptera: Noctuidae))

  • 국내 토양으로부터 분리한 Bacillus thuringiensis 균주에서 담배거세미나방(Spodoptera litura)에 선택적으로 살충효과를 나타내는 새로운 균주를 선발하였다. 이 균주의 결정성 독소단백질은 위상차 현미경과 주사전자현미경으로 관찰한 결과 전형적인 이중피라미드 형태이며 H serotype으로 동정한 결과 Bacillus thuringiensis subsp. CAB 109로 동정되었다. 난방제 해충인 담배거세미나방 3령충에 대해 B. t. subsp. aizawai CAB 109균주외 분리된 다른 3균주의 생물활성을 비교 검토하였다. 살충활성 결과에서 B. t. subsp. aizawai CAB 109 균주가 $1.3{\times}10^7$ (cfu/ml)에서 100% 사망률로 다른 균주에 비해 높은 활성을 나타내었다. 담배거세미나방은 배추좀나방과 다르게 7일 이상 경과한 뒤에야 100% 사충률을 나타내었다. 한편, 담배거세미나방 2령, 3령, 4령충에 대한 살충활성의 검정에서 B. t. aizawai subsp. CAB 109 균주의 $LD_{50}$값이 각각 $9.78{\times}10^5,\;6.87{\times}10^6,\;1.83{\times}10^7$ (cfu/ml)으로 살충활성을 나타내었다. B. t.를 섭식한 담배거세미나방 유충의 사망 현상은 지효성이며 대조군보다 무게가 $6{\sim}7$배정도 적으며 다음 령기로 성장하지 못하고 죽었다. 또한 B. t. subsp. aizawai CAB 109 균주의 SDS-PAGE에서의 단백질 패턴과 trypsin을 처리한 단백질의 결과에서 기존의 B. t. subsp. aizawai와 CAB 109균주와의 차이점은 발견할 수 없었다.
