Mass Screening of Lovastatin High-yielding Mutants through Statistical Optimization of Sporulation Medium and Application of Miniaturized Fungal Cell Cultures (Lovastatin 고생산성 변이주의 신속 선별을 위해 통계적 방법을 적용한 Sporulation 배지 개발 및 Miniature 배양 방법 개발)

  • For large and rapid screening of high-yielding mutants of lovastatin produced by filamentous fungal cells of Aspergillus terreus, one of the most important stage is to test as large amounts of mutated strains as possible. For this purpose, we intended to develop a miniaturized cultivation method using $7m{\ell}$ culture tube instead of traditional $250m{\ell}$ flask (working volume $50m{\ell}$). For obtaining large amounts of conidiospores to be used as inoculums for miniaturized cultures, 4 components i.e., glucose, sucrose, yeast extract and $KH_2PO_4$ were intensively investigated, which had been observed to show positive effect on enhancement of spore production through Plackett-Burman design experimet. When optimum concentrations of these components that were determined through application of response surface method (RSM) based on central composite design (CCD) were used, maximum spore numbers amounting to $1.9\times10^{10}$ spores/plate were obtained, resulting in approximately 190 fold increase as compared to the commonly used PDA sporulation medium. Using the miniaturized cultures, intensive strain development programs were carried out for screening of lovastatin high-yielding as well as highly reproducible mutants. It was observed that, for maximum production of lovastatin, the producers should be activated through 'PaB' adaptation process during the early solid culture stage. In addition, they should be proliferated in condensed filamentous forms in miniaturized growth cultures, so that optimum amounts of highly active cells could be transferred to the production culture-tube as reproducible inoculums. Under these highly controlled fermentation conditions, compact-pelleted morphology of optimum size (less than 1 mm in diameter) was successfully induced in the miniaturized production cultures, which proved essential for maximal utilization of the producers' physiology leading to significantly enhanced production of lovastatin. As a result of continuous screening in the miniaturized cultures, lovastatin production levels of the 81% of the daughter cells derived from the high-yielding producers turned out to be in the range of 80%$\sim$120% of the lovastatin production level of the parallel flask cultures. These results demonstrate that the miniaturized cultivation method developed in this study is efficient high throughput system for large and rapid screening of highly stable and productive strains.

Role of the Inferior Thyroid Vein after Left Brachiocephalic Vein Division During Aortic Surgery

  • Background: In aortic surgery, division and ligation of the left brachiocephalic vein(LBV) may improve exposure of the aortic arch but controversy continues about the safety of this division and whether a divided vein should be reanastomosed after arch replacement was completed. The safety of LBV division and the fate of the left subclavian venous drainage after LBV division were studied. Material and Method: From November 1998 to January 2001, planned division and ligation of the LBV on the mid-line after median sternotomy was peformed in 10 patients during the aortic surgery with the consideration of local anatomy and distal aortic anastomosis. Assessment for upper extremity edema and neurologic symptoms, measurement of venous pressure in the right atrium and left internal jugular vein, and digital subtraction venography(DSV) of the left arm were made postoperatively. Result: In 10 patients there was improvement in access to the aortic arch for procedures on the ascending aorta or aortic arch. The mean age of patients was 62 years(range 24 to 70). Follow-up ranged from 3 weeks to 13 months. One patient died because of mediastinitis from methicilline-resistant staphylococcus aureus strain. All patients had edema on the left upper extremity, but resolved by the postoperative day 4. No patient had any residual edema or difficulty in using the left upper extremity during the entire follow-up period. No patient had postoperative stroke. Pressure difference between the right atrium and left internal jugular vein was peaked on the immediate postoperative period(mean peak pressure difference = 25mmHg), but gradually decreased, then plated by the postoperative day 4. In all DSV studies left subclavian vein flowed across the midline through the inferior thyroid venous plexus. Conclusion: We conclude that division of LBV is safe and reanastomosis is not necessary if inferior thyroid vein, which is developed as a main bridge connecting the left subclavian vein with right venous system, is preserved.

Fermentation Process Characteristics of Phaffia rhodozyma Mutant B76 for Astaxanthin Biosynthesis (Astaxanthin 생합성을 위한 Phaffia rhodoxyma 변이주 B76의 발효공정 특성)

  • Specific carotenoids and astaxanthin biosynthesis power of Phaffia rhodozyma mutant 876, which was obtained after NTG a and UV treatments, was higher than those of the wild type by 40% and 50%, respectively. The mutant strain did not show t the catabolite repression even at 22% (w/v) glucose concentration. The optimum C{N ratio was 2.0, and the optimum t temperature and initial pH were $22^{\circ}C$ and 6.0, respectively. 80th cell growth and astaxanthin formation decreased drastically a as the fermentation temperature was increased over $22^{\circ}C$, whereas they were comparable in the pH range between 5.0 and 7 7.0. Inoculum size did not affect the final cell density nor the carotenoids biosynthesis, and 3%(v/v) was selected as optimal. H Higher dissolved oxygen concentration facilitated astaxanthin biosynthesis, and aeration rate of 1.0 v/0/m and agitation speed of 400 rpm were selected as optimum. The final cell dens때 of 43.3 g/L and the volumetric astaxanthin and carotenoids concentrations of 110.6 mg/L and 149.4 mg/L, respectively, were obtained. The specific carotenoids concentration was 3.45 m mg{g-yeast(dry). Yx/s and Yp/s values of 0.37 and 1.08 were obtained. The result of this study will provide basic information u useful for mass production of astaxanthin from P. rhodozyma fermentation.

Inbreeding Levels and Pedigree Structure of Korean Indigenous Chicken Population (한국 토종닭 집단의 혈통구조 및 유효집단크기 추정)

  • The purpose of this study was to estimate the inbreeding level and effective population size of Korean indigenous chickens. In the study, two variables were considered to evaluate the pedigree completeness: (1) the proportion (%) of animals with complete pedigree, and (2) the proportion of animals with inbreeding coefficients greater than zero. In the proportion of animals having complete pedigree, all strains reached almost 100% completeness in 1~2 years. In the proportion of animals with inbreeding coefficients greater than zero, all strains reached almost 100% completeness in 5~6 years. We considered that the pedigree recoding system is well managed and that the inbreeding coefficient is a reliable measure. Over the past 20 years, the increase of inbreeding coefficients in Korean indigenous chicken strains has been 7.6~10.9%. The S strain showed the most rapid increase of inbreeding coefficient of 8.2% in 10 years. The reason for this rapid increase is considered to be associated with the fact that the numbers of sires and dams involved in reproduction was 115 and 91, respectively, which are lower than those of the other strains. According to average rates of increase in inbreeding coefficients (${\Delta}F$), all strains have ${\Delta}F$ values of 0.39~0.85%, which is lower than 1%, and the effective population size is above 50. The results showed that inbreeding levels were within the acceptable range and that Korean indigenous chicken population scan be regarded as safe from the threat of extinction.

  • An experimental Chung-Kook-Jang was prepared using the strain Bacillus subtilis sp. isolated by the author Samples were taken in 12 hrs interval during the fermentation and the oligopeptides were separated by the method of molecular sieving using the ion exchange resin column of Dowex-50. Only the X-16 fraction of oligopeptides was taken and the components of oligopeptides were developed in two dimensional thin layer chromatograms. The each peptide spot was eluted and each peptide was isolated. The pattern and kinds of amino acids, and N and C-terminal amino acids were studied. Fourteen different oligopeptides could be detected by the two dimensional thin layer chromatography, all of which were consisted of $4{\sim}9$ kinds of amino acids. No dipeptides and no tripeptides could be found. The N and C-terminal amino acids and the residual component amino acids of all these 14 peptides could be summarized as the follows. [P]-I. Study on Characteristics of Antibody from Milk Immunized with Some Helicobacter pylori Antigen (Helicobacter pylori 항원을 이용한 면역우유의 항체특성에 관한 연구)

  • This study has been carried out to examine the characteristics of anti-H. pylori antibodies of milk produced from cows immunized with antigen of Helicobacter pylori such as peculiarity of antigen antibody, agglutination of H. pylori strain, stability of antibody against acid, alkali and heat treatments. The molecular weight of anti-H. pylori antibody measured by SDS-PAGE were turned out as about 50 kDa in the heavy chain and about 24 kDa in the light chain. Twenty protein bands were visualized in H. pylori interacting with anti-H. pylori antibody which was made in dairy cow by immunization with H. pylori. The western blotting was peformed in order to examine the antigen peculiarity of anti-H. pylori, The results were all 7 antigen substances including serum, furified serum, whey and furified whey could be confirmed and the major antigen substances were 97, 66,34 kDa of molecular weight. As a result of agglutination response anti-H. pylori antibody in whey showed 1/10 agglutination value against H. pylori. In stability test about acid and alkali of antibody, there was $100\%$ activated at the range of $pH5\~pH10$. In stability test about heat, the antibody showed stable condition at $60^{\circ}C$ for 60 minutes and comparatively stable condition at $70^{\circ}C$, but reduced activation to $40\%$ after 60 minutes. It maintained $77\%$ activation at $80^{\circ}C$ for 4 minutes and comparatively stable at $100^{\circ}C$ for 1 minute.

  • In this study, enzymatic hydrolysis of the holocellulose from Alnus hirsuta (Spach) Rupr. (8-14 yr's) was investigated using crude cellulase preparations of Trichoderma viride Pers. ex. Fr. SANK 16374. And conducted on the optimum condition of the treated substrate for saccharification. A strain of Trichoderma viride Pers. ex. Fr. SANK 16374 was found to be highly efficient for the cellulase productivity, especially in the submerged culture process. The culture medium used in this experiment was prepared from an extract of wheat bran consisting also of $KH_2PO_410$, $(NH_4)_2$ $SO_4$ 3, $NaNO_3$ 3, and $MgSO_4$ $7H_2O$ 0.5g/l. Cellulose powder (Toyo filter paper, 60 mesh) was found to be an importent factar for inducing the cellulase formation. And the cellulase produced in the culture fluid was salted out quantitatively by the use of ammonium sulfate (Fig. 1) Reducing sugar was determined by the Dinitrosalicylic acid (DNS) method, using reagents prepared according to the method of Sumner (1925). The results obtained were summerized as follows; 1. The method of delignification were treated by the Peracetic acid (PA) method, according to the method of Toyama (1970). The yield of holocellulose were decreased in accordance with increasing concentration of Peracetic acid solution; delignification of Alnus hirsuta Rupr. with 20% Peracetic acid was satisfied for 48 hours and 40%~60% peracetic acid was satisfied for 24 hrs: 2. The substrate (holocellulose) was changed easely into fine powder with enzymatic hydrolysis and cellulase exhibits optimum activity on the reducing sugar formation from substrate at the range of 60-100 mesh. 3. The reducing sugar formation increased in accordance with increasing dry temperature on holocellulose substrate was found to be $190{\pm}5^{\circ}C$. 4. The optimal heat treated time of holocellulose substrate was found to be 45 min. for the reducing sugar formation showed the best products. The reducing sugar formation did not show statisticaly significent diflerences at 5% levels by heat treated time for 45 min. and 60 min.

  • The mechanical and rheological properties of agricultural materials are important for engineering design and analysis of their mechanical harvesting, handling, transporting and processing systems. Agricultural materials, which composed of structural members and fluids do not react in a purely elastic manner, and their response when subjected to stress and strain is a combination of elastic and viscous behavior so called viscoelastic behavior. Many researchers have conducted studies on the mechanical and rheological properties of the various agricultural products, but a few researcher has studied those properties of rice plant, and also those data are available only for foreign varieties of rice plant. This study are conducted to experimentally determine the mechanical and the rheological properties such as axial compressive strength, tensile strength, bending and shear strength, stress relaxation and creep behavior of rice stems, and grain detachment strength. The rheological models for the rice stem were developed from the test data. The shearing characteristics were examined at some different levels of portion, cross-sectional area, moisture content of rice stem and shearing angle. The results obtained from this study were summarized as follows 1. The mechanical properties of the stems of the J aponica types were greater than those of the Indica ${\times}$ Japonica hybrid in compression, tension, bendingand shearing. 2. The mean value of the compressive force was 80.5 N in the Japonica types and 55.5 N in the Indica ${\times}$ Japonica hybrid which was about 70 percent to that of the Japonica types, and then the value increased progressively at the lower portion of the stems generally. 3. The average tensile force was about 226.6 N in the Japonica types and 123.6 N in the Indica ${\times}$ Japonica hybrid which was about 55 percent to that of the Japonica types. 4. The bending moment was $0.19N{\cdot}m$ in the Japonica types and $0.13N{\cdot}m$ in the Indica ${\times}$ Japonica hybrid which was 68 percent to that of the Japonica types and the bending strength was 7.7 MPa in the Japonica types and 6.5 MPa in the Indica ${\times}$ Japonica hybrid respectively. 5. The shearing force was 141.1 N in Jinju, the Japonica type and 101.4 N in Taebaeg, the Indica ${\times}$ Japonica hybrid which was 72 percent to that of Jinju, and the shearing strength of Taebaeg was 63 percent to that of Jinju. 6. The shearing force and the shearing energy along the stem portion in Jinju increased progressively together at the lower portions, meanwhile in Taebaeg the shearing force showed the maximum value at the intermediate portion and the shearing energy was the greatest at the portion of 21 cm from the ground level, and also the shearing strength and the shearing energy per unit cross-sectional area of the stem were the greater values at the intermediate portion than at any other portions. 7. The shearing force and the shearing energy increased with increase of the cross-sectional area of the rice stem and with decrease of the shearing angie from $90^{\circ}$ to $50^{\circ}$. 8. The shearing forces showed the minimum values of 110 N at Jinju and of 60 N at Taebaeg, the shearing energy at the moisture content decreased about 15 percent point from initial moisture content showed value of 50 mJ in Jinju and of 30 mJ in Taebaeg, respectively. 9. The stress relaxation behavior could be described by the generalized Maxwell model and also the compression creep behavior by Burger's model, respectively in the rice stem. 10. With increase of loading rate, the stress relaxation intensity increased, meanwhile the relaxation time and residual stress decreased. 11. In the compression creep test, the logarithmic creep occured at the stress less than 2.0 MPa and the steady-state creep at the stress larger than 2.0 MPa. 12. The stress level had not a significant effect on the relaxation time, while the relaxation intensity and residual stress increased with increase of the stress level. 13. In the compression creep test of the rice stem, the instantaneous elastic modulus of Burger's model showed the range of 60 to 80 MPa and the viscosities of the free dashpot were very large numerical value which was well explained that the rice stem was viscoelastic material. 14. The tensile detachment forces were about 1.7 to 2.3 N in the Japonica types while about 1.0 to 1.3 N in Indica ${\times}$ Japonica hybrid corresponding to 58 percent of Japonica types, and the bending detachment forces were about 0.6 to 1.1 N corresponding to 30 to 50 percent of the tensile detachment forces, and the bending detachment of the Indica ${\times}$ Japonica hybrid was 0.1 to 0.3 N which was 7 to 21 percent of Japonica types. 15. The detachment force of the lower portion was little bigger than that of the upper portion in a penicle and was not significantly affected by the harvesting period from September 28 to October 20. 16. The tensile and bending detachment forces decreased with decrease of the moisture content from 23 to 13 percent (w.b.) by the natural drying, and the decreasing rate of detachment forces along the moisture content was the greater in the bending detachment force than the tensile detachment force.

Effect o( Restricted Feeding of Layer on the Egg Productivity in Summer of Korea (산란기 제한급사가 산란성적에 미치는 영향)

  • In order to investigate an effect of the feed restriction on the laying performance, 208 White Leghorn strain layer of 36 week-old were divided to 4 groups of 52 birds each and raised for 1 week of previous feeding and for the subsequent 3 weeks of experimental restricted feeding. The egg production, daily egg mass and feed efficiency of four groups of birds fed daily 100g, 95g, 90f and 85g of a commercial diet, respectively, which were compared with those of the original 2879 birds fed l13g of diet per day as control. During 3 weeks of experimental restricted feeding, period, daily body weight nam was decreased linearly as the degree of restriction increased in birds fed 100, 95, 90 and 85g of diet. Hen day egg production, egg weight and daily egg mass was linearly related to the diet intake level. then feed intake(x, g day$^{-1}$ ) showed a positive regression equations with the henday egg production(y, % ), egg weight(y, g egg$^{-1}$ ) and egg mass(y, g bird$^{-1}$ ) as y=38.75+0.3753$\times$(r=0.503, n=15), y=48.2+0.08868$\times$(r=0.835, n= 15) and y=15.69+0.2786$\times$(r=0.597, n=15), respectively. Feed efficiency was increased to a plateau in birds fed 95g of diet. The estimated energy utilization for egg production was reached to a plateau in birds fed 95g of diet and the highst protein utilization was shown in birds fed 90g of diet anions birds fed graded levels of diet. And the feed restriction did not affect on the egg shell contents, while protein contents of egg were shown a trend to be increased and lipids and cholesterol contents of eggs was decreased according to the diet intake lowered. The results suggested that the improved feed efficiencies of birds restricted under 16% of diet(above 95g of diet) will be due to increased energy and protein utilization for egg production and feed restriction above 16% will be aboided. In the range from 113g to 95g of diet feeding, the crude profit was increased as the feed restricted in the case of egg price 600 won kg$^{-1}$ and feed price 200 won kg$^{-1}$ .

Strain Improvement of the Genus Pleurotus by Protoplast Fusion (원형질체(原形質體) 융합(融合)에 의한 느타리버섯속(屬)의 품종개발(品種開發))

  • Somatic hybrids of Pleurotus florida ASI 2016 and Pleurotus ostreatus ASI 2018 were obtained by protoplast fusion. The 40 fusants($P1{\sim}P40$) was examined for the yield on fermented and pasteurized rice straw in a tray. The carpophore yield of them were showed as the range of $27.0{\sim}155.2$, based on parental values of 100(ASI 2018), The pilei of fusants between orange white colored P. florida and dark grey colored P. ostreatus had mixed colors in the young stage. Other breeding programmes were performed to improve new varieties with high yield and good quality. A new oyster mushroom variety, Wonhyeongneutaribeosus(P72), was developed at the Agricultural Sciences Institute, Rural Development Administration in 1990. This P.florida-ostreatus-ostreatus hybrid P72 was selected from 38 protoplast fusion products($P41{\sim}P78$) between P.florida-ostreatus recombinant P5-M 43-arg rib and P. ostreatus ASI 2-13-0 2001-19-pro orn. The yield indexes of 38 hybrids ranged $40.5{\sim}152.7$ compared with the parental values of 100(ASI 2001). Hybrid P72 was characterized by the large fruiting bundle of semispherical shape with long stipe and by the small and circular pileus, resulting in lower harvesting cost. A significant increase in carpophore production was observed in somatic hybrids of protoplasts due to heterosis. A comparision of hybrid with parents P72 was made using isozyme analysis. The esterase banding patterns could be characterized by new bands in the hybrids. Seven fusion products of four crosses between P.florida ASI 2016 and P. ostreatus ASI 2018 were analysed with respect to the distribution of progenies and segregation of gene markers by random basidiospore analysis. Segregation of alleles should yield progeny of four genotypes in a Mendelian ratio of 1 : 1 : 1 : 1 for prototrophs, auxotrophs of one parental type, auxotrophs of the other parental type, and auxotrophic recombinants, respectively. However, five fusants of them did not detect one parental, P.ostreatus, type. Basidiospores could yield progeny of 16 genotypes in the cross of one of the recombinant P5-M43-arg $rib{\times}P. ostreatus$ ASI 2-13-pro orn but the segregants of three fusants were not detected clearly. The allele ratio of loci could be expected 1 : 1 : 1 : 1 for arg, rib, pro and orn. The ratio, however, would be changed to 4 : 1 : 1 : 1 with increasing proportion of argo In almost all the fusants, prototrophic recombinants were recovered in large numbers against auxotrophic markers. Parental genotypes were recovered with the recombinant progeny amounting to $38.68{\sim}99.56%$. The analysis provides proof of heterokaryosis and strong evidence for haploidy of vegetative nuclei, a sexual cycle consisting of nuclear fusion and meiosis.
