Analysis of the Effect of Temperature on the Pesticide Efficacy and Simulation of the Change in the Amount of Pesticide Use (온도가 농약효과에 미치는 영향분석 및 농약사용량 예측 모의실험)

  • Pest population density models are very important to monitor the initial occurrence and to understand the continuous fluctuation pattern of pest in pest management. This is one of the major issues in agriculture because these predictions make pesticides more effective and environmental impact of pesticides less. In this study, we combined and predicted the mortality change of pest caused by pesticides with temperature change and population dynamic model. Sensitive strain of two-spotted spider mite (Tetranychus urticae Koch) with kidney bean leaf as host was exposed to mixed acaricide, Acrinathrin-Spiromesifen and organotin acaricide, Azocyclotin, at 20, 25, 30, and $35^{\circ}C$, respectively. There was significant difference in mortality of T. urticae among pesticides and temperatures. We used DYMEX to simulate population density of T. urticae and predicted that the initial management time and number of chemical control would be changed in the future with climate change. There would be implications for strategies for pest management and selection process of pesticide in the future corresponding climate change.

Expression of CP4 5-Enol-Pyruvylshikimate-3- Phosphate Synthase Transgene in Inbred Line of Korean Domestic Maize (Zea may L.) (국내 옥수수 순계주에서 CP4 5-Enol- Pyruvylshikimate-3- Phosphate Synthase 유전자의 발현)

  • This study was conducted to develop herbicide-resistance domestic maize plants by introducing the CP4 5-enol-pyruvylshikimate-3-phosphate synthase (CP4 EPSPS) gene using Agrobacterium tumefaciens-mediated immature embryo transformation. Immature embryos of five genotypes (HW1, KL103, HW3, HW4, HW7) were co-cultivated with strains Agrobacterium tumefaciens (strain C58C1) containing the binary vector (pCAMBIA2300) carrying Ubiquitin promoter-CP4 EPSPS gene and Cauliflower mosaic virus 35S (CaMV35S) promoter-nptll gene conferring resistance to paromomycin as a selective agent. The presence and expression of CP4 EPSPS transgene were confirmed by PCR, RT-PCR and Northern blot analysis, respectively. Also, the resistance to glyphosate in the transgenic maize ($T_1$) was analyzed by shikimate accumulation assay. The frequency (%) of paromomycin-resistance callus was 0.37, 0.03, 2.20, 2.37, and 0.81% in pure lines HW1, KL103, HW3, HW4 and HW7, respectively. EPSP transgene sequences were amplified in putative transgenic plants that regenerated from paromomycin-resistance calli of two inbred lines (HW3, HW4). Of them, RT-PCR and Northern blot analyses revealed that the transgene was only expressed in two transgenic events (M266, M104) of HW4 inbred line, and a mild glyphosate resistance of transgenic event (M266) was confirmed by the lower shikimate accumulation in leaf segments. These results demonstrate that transgenic maize with herbicide-resistance traits in Korean genotype can be genetically obtained.

Variation of Ergosterol Content in Lentinula edodes Culture (표고 균사 배양체내 에르고스테롤 함량의 변이)

  • Ergosterol is an indicator chemical of fungi and involves in fungal cell growth as a major component in fungal cell membranes. Thus, this chemical can be used to estimate live fungal biomass within various solid substrates. Ergosterol content in liquid culture of Lentinula edodes had a linear relationship (r=0.98) with the hyphal mass of the fungus. This chemical content differed depending on the fungal strains, age of culture and water content levels of sawdust substrate. Ergosterol content was 0.13% in the 10 weeks old liquid culture while it was 0.10% in 20 weeks old one. The chemical content in the sawdust cultures of the fungus varied 0.015% to 0.042% depending upon strains and water content levels within sawdust substrate. Ergosterol content in the culture of a L. edodes strain, Sanrim 4, was higher by $20{\mu}g$ to $140{\mu}g$/g dry substrate than those of strains, Mok-H and Sanrim 6. And the chemical contents in the sawdust cultures with 125% or 175% water, 297 to 425 ${\mu}g$/g dry substrate, were higher than those with 75% or 225% water, 148 to $286{\mu}g$/g dry substrate. We conclude that ergosterol analysis can estimate the fungal biomass within solid substrate such as logs and sawdust.

  • The damage and fracture of machine or structure are caused by the crack happened from the defect existed at the inside of material. The properties of crack propagation and growth characteristic must be considered because there are many cases at which these cracks are densely existed. Therefore, this study investigates the fracture property due to the position of crack and hole inside the standard compact tension (C. T.) specimen. When the concentrated load is applied eccentrically at the standard C. T. specimen, the fracture mechanical behavior due to the existence or non-existence and the position of hole near crack is investigated. As the result of analysis study, model 3 (in case of the distance of 2mm on the horizontal direction between the end part and hole as the specimen model existed with one hole near the crack) has the maximum deformation, stress and deformation energy of the most values among three models. As the distance between the crack and hole inside the specimen becomes nearer, the maximum stress becomes higher in cases of three models. Apart from the number of holes, it is seen that the maximum stress becomes higher near the crack when the hole exists near the crack inside the specimen. If the hole inside the machine or the mechanical structure is punctured by using the result of this study, it is thought that the occurred breakage or breakdown can be prevented by reducing the fracture stress happened at the specimen.

Enhanced Production of Astaxanthin by Archaea Chaperonin in Escherichia coli (대장균에서 고세균 샤페론을 이용한 아스타잔틴 생산능 향상을 위한 연구)

  • The aim of this study is to increase production of carotenoids in recombinant Escherichia coli by Archaea chaperonin. The carotenoids are a widely distributed class of structurally and functionally diverse yellow, orange, and red natural pigments. These pigments are synthesized in bacteria, algae, fungi, and plants, and have been widely used as a feed supplement from poultry rearing to aquaculture. Carotenoids also exhibit diverse biological properties, such as strong antioxidant and antitumor activities, and enhancement of immune responses. In the microbial world, carotenoids are present in both anoxygenic and oxygenic photosynthetic bacteria and algae and in many fungi. We have previously reported cloning and functional analysis of the carotenoid biosynthesis genes from Paracoccus haeundaensis. The carotenogenic gene cluster involved in astaxanthin production contained seven carotenogenic genes (crtE, crtB, crtI, crtY, crtZ, crtW and crtX genes) and recombinant Escherichia coli harboring seven carotenogenic genes from Paracoccus haeundaensis produced 400 μg/g dry cell weight (DCW) of astaxanthin. In order to increase production of astaxanthin, we have co-expressed chaperone genes (ApCpnA and ApCpnB) in recombinant Escherichia coli harboring the astaxanthin biosynthesis genes. This engineered Escherichia coli strain containing both chaperone gene and astaxanthin biosynthesis gene cluster produced 890 μg/g DCW of astaxanthin, resulting 2-fold increased production of astaxanthin.

  • The goal of this study is to increase production of astaxanthin in recombinant Escherichia coli by engineered isoprenoid pathway. We have previously reported structural and functional analysis of the astaxanthin biosynthesis genes from a marine bacterium, Paracoccus haeundaensis. The carotenoid biosynthesis gene cluster involved in astaxanthin production contained six carotenogenic genes (crtW, crtZ, crtY, crtI, crtB, and crtE genes) and recombinant E. coli harboring six carotenogenic genes from P. haeundaensis produced 400 ${\mu}g$/g dry cell weight (DCW) of astaxanthin. In order to increase production of astaxanthin in recombinant E. coli, we have cloned 4-hydroxy-3-methylbut-2-enyl diphosphate reductase (lytB), farnesyl diphosphate (FPP) synthase (ispA), and isopentenyl (IPP) diphossphate isomerase (idi) in the isoprenoid pathway from E. coli and coexpressed these genes in recombinant E. coli harboring the astaxanthin biosynthesis genes. This engineered E. coli strain containing both isoprenoid pathway gene and astaxanthin biosynthesis gene cluster produced 1,200 ${\mu}g$/g DCW of astaxanthin, resulting 3-fold increased production of astaxanthin.

Association of A/T Rich Microsatellites with Responses to Artificial Selection for Larval Developmental Duration in the Silkworm Bombyx mori

  • Simple sequence repeats (SSRs) and interSSR (ISSR) marker systems were used in this study to reveal genetic changes induced by artificial selection for short/long larval duration in the tropical strain Nistari of the silkworm Bombyx mori. Artificial selection separated longer larval duration (LLD) ($29.428{\pm}0.723days$) and shorter larval duration (SLD) ($22.573{\pm}0.839days$) lines from a base, inbred population of Nistari (larval span of $23.143{\pm}0.35days$). SSR polymorphism was observed between the LLD and SLD lines at one microsatellite locus, Bmsat106 ($CA_7$) and at two loci of 1074 bp and 823 bp generated with the ISSR primer UBC873. Each of these loci was present only in the LLD line. The loci segregated in the third generation of selection and were fixed in opposite directions. In the $F_2$ generation of the $LLD{\times}SLD$ lines, the alleles of Bmsat106 and $UBC873_{1074bp}$ segregated in a 1:1 ratio and the loci were present only in the LLD individuals. $UBC873_{823bp}$ was homozygous. Single factor ANOVA showed a significant association between the segregating loci and longer larval duration. Together, the two alleles contributed to an 18% increase in larval duration. The nucleotide sequences of the $UBC873_{1074bp}$ and $UBC873_{823bp}$ loci had 67% A/T content and consisted of direct, reverse, complementary and palindromic repeats. The repeats appeared to be "nested" (59%) in larger repeats or as clustered elements adjacent to other repeats. Of 203 microsatellites identified, dinucleotides (67.8%) predominated and were rich in A/T and T/A motifs. The sequences of the $UBC873_{1074bp}$ and $UBC873_{823bp}$ loci showed similarity (E = 0.0) to contigs located in Scaffold 010774 and Scaffold 000139, respectively, of the B. mori genome. BLASTN analysis of the $UBC873_{1074bp}$ sequence showed significant homology of (nt.) 45-122 with upstream region of three exons from Bombyx. The complete sequence of this locus showed ~49% nucleotide conservation with transposon 412 of Drosophila melanogaster and the Ikirara insertions of Anopheles gambiae. The A + T richness and lack of coding potential of these small loci, and their absence in the SLD line, reflect the active process of genetic change associated with the switch to short larval duration as an adaptation to the tropics.

Anti-inflammatory Activity of the Galactomannans Produced by a New Fungal Strain Trichoderma erinaceum DG-312 (Trichoderma erinaceum DG-312 신규 곰팡이 균주로부터 생산된 galactomannan의 항염증 활성)

  • The crude galactomannans (GMs) were obtained from the culture of a newly isolated fungus Trichoderma erinaceum DG-312 and their anti-inflammatory activity was investigated in mice. The maximum concentrations of mycelial biomass and GMs reached 9.44 g/l, 2.72 g/l at day 3 in a 5-l stirred-tank bioreactor, respectively. The results of Sepharose CL-6B gel chromatography and compositional analysis revealed that the crude GMs contain heterogeneous polysaccharides consisting of $74.9\%$ mannose and $24.1\%$ galactose. The GMs was shown to possess a significant anti-inflammatory activity against acetic acid-induced inflammatory mouse model in a dose-dependent manner, when mice were treated with 100 and 200 mg GMs/kg body weight. The inhibition in vascular permeability $(60.6\%)$ and in writhing response $(62.5\%)$ evidenced an anti-inflammatory activity of the GMs. The marked anti-inflammatory and writhing-lowering properties of the GMs suggest its potential therapeutic use.

Two-dimensional gel Electrophoresis of Helicobacter pylori for Proteomic Analysis

  • Two-dimensional gel electrophoresis (2-DE) is an essential tool of proteomics to analyse the entire set of proteins of an organism and its variation between organisms. Helicobacter pylori was tried to identify differences between strains. As the first step, whole H. pylori was lysed using high concentration urea contained lysis buffer [9.5 M Urea, 4% CHAPS, 35 mM Tris, 65 mM DTT, 0.01% SDS and 0.5% Ampholite (Bio-Rad, pH 3-10)]. The extract ($10\;{\mu}g$) was rehydrated to commercially available immobilised pH gradient (IPG) strips, then the proteins were separated according to their charges as the first dimensional separation. The IPG strips were placed on Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis (SDS-PAGE) to separate according to molecular mass of the proteins as the second dimension. The separated protein spots were visualised by silver staining in order to compare different expression of proteins between strains. Approximately 120 spots were identified in each mini-protein electrophoresised gel, furthermore about 65 to 75 spots were regarded as identical proteins in terms of pI value and molecular weight between strains used. In addition, distinct differences were found between strains, such as 219-1, Y7 and Y14, CH150. Two representative strains were examined using strips which had pH range from 4 to 7. This strips showed a number of isoforms which were considered large spots on pH range 3-10. Furthermore, the rest of spots on pH 4-7 IPG strips appeared very distinctive compared to broad range IPG strips. 2-DE seems to be an excellent tool for analysing and identifying variations between H. pylori strains.

Pulsed Field Gel Electrophoresis Profile of Erythromycin-Clindamycin Resistant Streptococcus pyogenes Isolated in Korea (국내분리 Erythromycin-Clindamycin 내성 Streptococcus pyogenes에 대한 Pulsed Field Gel Electrophoresis 양상 분석)

  • Ninety two strains of Streptococcus pyogenes were isolated from patients with pharyngitis, scarlet fever, skin infection, and invasive streptococcal infections in Seoul, Korea from January to December, 1998. All isolates were epidemiologically characterized by T protein serotype, and serum opacity factor (OF) detection to phenotypes. To analyze the genetic relationship, fifty two isolates including 32 erythromycin-clindamycin (Em-Cm) resistant strains, 20 antimicrobial susceptible strains were attempted to the pulsed-field gel electrophoresis (PFGE). T protein serotype showed 16 kinds in distribution including T12 and T4. Among the total isolates, 40 strains (43.5%) belonged to the T12 serotype and twenty strains (21.7%) to T4 serotype. On the other hand, when infection aspect of S. pyogenes isolates were analysed by T serotype distribution, T12 type was predominant for pharyngitidis which contributed to 21 strains (53%) and for skin infection isolates which contributed to 11 strains (28%), respectively. In case of T4 type, it was the most predominant pharyngitidis isolates which contributed to 8 strains (40%). In T serotype distribution of Em-Cm resistant strains, 27 strains (84%) of the thirty two showed T12 serotype. In minimum inhibitory concentration (MIC) values of Em-Cm resistance isolates, thirty two isolates showed resistant to erythromycin 27 strains (84%), had high MIC of >$128\;{\mu}g/ml$. And also to clindamycin, twenty two strains (69%) had high MIC of >$128\;{\mu}g/ml$. When OF detection of Em-Cm resistance of S. pyogenes isolates were analyzed by T serotype distribution, T12 serotype isolates revealed that all of the isolates except one strain were OF negative. In PFGE profile analysis to Em-Cm resistance isolates, of the twenty seven, Em-Cm resistance of T12 serotype isolates, 26 strains showed identical PFGE profile and all of these isolates revealed that OF negative. Eighty four percent of Em-Cm resistance S. pyogenes isolates had identical phenotype and PFGE profile. These results strongly suggested that the Em-Cm resistant S. pyogenes isolates from Seoul area showed close genetic correlation and PFGE could be available tool for molecular epidemiology.
