The study investigated the effect of carrier composition (ionic strength and pH) on the retention of various proteins in flow field-flow fractionation (Flow FFF) as well as the conformational change of Bovine Serum Albumin (BSA) with urea concentration, storage time and temperature. The study found that the retention of protein in Flow FFF increased with the ionic strength of the carrier liquid. Most proteins were well solubilized at pH = 7-8. The hydrodynamic diameters obtained from Flow FFF retention data agree well with theoretical values. The retention increased and the peak shape became distorted at extreme pH conditions of the carrier solution. The selected carrier composition for comparison between the literature value of proteins was 0.05 M tris buffer solution with a pH of 8. Storing BSA at 4 ±2℃ over a period of three months resulted in slow dimerization. Also, in case of the storage of BSA at 37 ±5℃ for one week, the retention of both BSA monomer and dimer increased with the urea concentration. Finally, the structural composition of specific enzymes: malonyl-CoA decarboxylase (MCDC) and malonyl-CoA synthesis (MCS) was determined by using Flow FFF at specific carrier solutions. The molecular weight of the natural MCDC was determined to be 208 kDa, which means it is a homotetramer, while that of the MCS was determined to be 47 kDa, which means it is a monomer.
This study investigated how ozone treatment can successfully inactivate pathogenic bacteria in both artificial seawater and effluents discharged from the fishery quarantine station in Pyeongtaek Port, Korea. Vibrio sp. and Streptococcus sp. were initially inoculated into the artificial seawater. All microbes were almost completely inactivated within 10 min and 30 min by injecting 6.4 mg/min and 2.0 mg/min of ozone, respectively. It was discovered that the water storing Pleuronichthys, Pelteobagrus, and Cyprinus imported from China contained the indicator bacteria, Vibrio sp., Enterococcus sp., total coliforms, and heterotrophic microorganisms. Compared to the control, three indicator bacteria were detected at two to six times higher concentrations. The water samples displayed a diverse microbial community, comprising the following four phyla: Bacteroidetes, Proteobacteria, Firmicutes, and Actinobacteria. Almost all indicator bacteria were inactivated in 5 min at 2.0 mg/min of ozonation; comparatively, 92.9%-98.2% of the less heterotrophic microorganisms were deactivated within the same time period. By increasing the dosage to 6.4 mg/min, 100% deactivation was achieved after 10 min. Despite the almost complete inactivation of most indicator bacteria at high doses after 10 min, several bacterial strains belonging to the Proteobacteria have still been found to be resistant under the given operational conditions.
Background: Stroke patients usually experience damage to multiple functions and a long rehabilitation period. Hence, there is a large volume of patient clinical information. It thus takes a long time for clinicians to identify the patient's information and essential pieces of information may be overlooked. To solve this, we stored the essential clinical information of stroke patients in a blockchain and implemented the blockchain technology using the Java programming language. Methods: We created a mini blockchain to store the medical information of patients using the Java programming language. Results: After generating a unique pair of public/private keys for identity verification, a patient's identity is verified by applying the Elliptic Curve Digital Signature Algorithm based on the generated keys. When the identity verification is complete, new medical data are stored in the transaction list and the generated transaction is verified. When verification is completed normally, the block hash value is derived using the transaction value and the hash value of the previous block. The hash value of the previous block is then stored in the generated block to interconnect the blocks. Conclusion: We demonstrated that blockchain can be used to store and deliver the patient information of stroke patients. It may be difficult to directly implement the code that we developed in the medical field, but it can serve as a starting point for the creation of a blockchain system to be used in the field.
To suppress mold generation of yujacheong, Penicillium chrysogenum LB31 was cultured, and spores were harvested and put into yujacheong. Antioxidant activity, useful ingredients, mold size and incidence were investigated while storing yujacheong for 30 days, after sterilization with different methods (nontreatment, ozone gas emission, heating after ozone gas emission and heating). The results showed that the content of narirutin, naringin, hesperidin, or neohesperidin, which are functional components of yuzu, increased as the storage period increased in all the treatment units. In addition, mold generation was not observed until the 15th day in the heat treatment group after ozone gas emission. As the treatment group emitted ozone gas. molds of 34.8 and 112 mm2 in size were observed on the 30th day. These results suggested that ozone sterilization can prevent microbial contamination, further extending the shelf life of yuzacheong and maintaining a fresh state.
이동 통신 기술의 급격한 발전으로, 이동 컴퓨팅 환경에서 데이터 서비스에 대한 수요가 점차 증가하고 있다. 이동 컴퓨팅 환경에서의 여러 단점을 극복하고자 캐쉬가 등장하게 되었다. 이동 호스트 내에 캐쉬가 존재하면, 통신 대역폭의 절약 및 질의에 대한 빠른 응답을 가져올 수 있지만, 이동 호스트가 서버 데이터와의 캐쉬 일관성(consistency)을 유지시켜야 한다는 문제가 생기게 된다. 본 논문에서는 기존의 기법들이 가지는 문제점들을 보완하기 위해 데이터의 주기성을 고려한 캐쉬 일관성 유지 기법을 제안한다. 제안한 기법은 데이터를 주기적, 비주기적 데이터로 분류하고 주기적 데이터에 만료시간을 삽입한다. 또한 무효화 메시지를 저장해서 단절 후에 선별적으로 캐쉬 데이터가 나누어질 수 있도록 해서, 캐쉬 내의 데이터가 접속 단절 후에 일방적으로 버려지는 것을 막을 수 있게 된다. 이렇게 함으로서 서버와 이동 호스트간의 통신에 사용하는 전체 대역폭의 낭비와 서버접속에 필요한 이동 호스트의 전력의 낭비를 줄일 수 있다. 마지막으로 기존의 기법과의 실험을 통해서 제안한 기법의 효율성을 알아보고자 한다.

Purpose: The aim of this study was to evaluate the rat periodontal ligament cell viability under $0^{\circ}C/2$ MPa condition up to one week using in vivo 3-(4,5-dimethylthiazol-2-y1)-2,5-diphenyltetrazolium bromide (MTT) assay. Materials and Methods: As soon as 110 upper molar teeth of rats were extracted, they were stored in Hartman's solution under $0^{\circ}C/2$ MPa condition for 1, 2, 3, 4, and 7 days each. All specimens were treated with in vivo MTT assay and the value of optical density was measured by ELISA reader. These values were statistically analyzed by one-way ANOVA. Result: There was no statistical difference on MTT value between immediate and 1 day storage group. There were statistically significant differences between 1 day and 2 days tsorage, 2 and 3 days storage groups, respectively. Teeth of 34, and 7 days storage groups showed significantly lower MTT valuesc ompared with shorter period storage groups. Conclusion: When the MTT values were substituted in standard curve, 1 day storage group at $0^{\circ}C/2$ MPa condition showed 68% cell viability when compared with immediate group. It dropped to 13% at 2 days, and to less than 5% at 3 days or more.
Pulse diagnosis is the one of the most important diagnostic process to traditional medical doctors. Although the pulse diagnosis position, Gwan is apart from Chon or Cheok by 10$\sim$20mm at most, traditional medical doctors applies different indent pressures and even they states different pulse images are felt at Chon, Gwan and Cheok. One the other hand, the education on pulse diagnosis behavior includes tantalizing problem caused by no tool for communication between trainer and trainee. On account of this situation, we tried to develop a system which can measure the hold-down pressure during a pulse diagnosis and compare the hold-down pressure profile of trainer and that of trainee. This system can be divided into three parts - pulse pressure sensing part, signal acquisition part and data storing part. A correction curve was generated by the relation between output voltages and standard weights. Using this system, 3 channel hold-down pressure profile of a oriental medical doctor were recorded three times. In the profile, three period were observed and all period included two process for searching the depth of pulsation and for classifying the pulse feeling into one or more of 28 pulse types. The maximum value of pulse profile was 1.3$kg{\cdot}f$ which was more than reported by previous chinese groups and the mean values of three channel ranged from 240$g{\cdot}f$ to 430$g{\cdot}f$. In frequency domain, each channel has some dominant frequency components - about 10Hz, 35Hz and 75Hz. In further study, we want to collect more profiles from lage number of oriental medicine doctors and hope to develop a measuring system which can measure the hold-down pressure on subject's skin directly.
Background: In order to prepare for the regulation about the limit of manure production, the status of manure management and the amount of nutrients in the compost discharged from Hanwoo breeding farm shall be known. This study aimed to find the practical amount of nutrients (volatile solids, VS; total nitrogen, T-N; total phosphorus, T-P) in manure, and compost samples collected from 40 Hanwoo breeding farms and the loss of the nutrients was calculated during the composting period, which supports to develop nutrient loading coefficients (NLCs) for each nutrient. Results: Although the addition of bedding materials for composting caused the increase of the VS amount before composting, the comparison of VS, N, and P amounts in between manure and compost showed the lower VS by 4 % as well as T-N and T-P amounts by 69 and 40 %, respectively, of which values were corresponded with the NLCs of 0.96, 0.31, and 0.60 for VS, N, and P, respectively, based on the questionnaire, and sample analyses. Considering with the environmental impacts including land application from Hanwoo manure, P loss should be zero before and after composting. In this regard, nitrogen loss of 50 % occurs and VS was increased by 30 %. In addition, feasible cases for the calculations based on the notification from Ministry of Environment were compared with this study. Conclusions: The development of NLCs from Hanwoo manure in this study implies that the loss of nutrients in manure occurs during the composting or storing period. The mass balances of N and P from livestock manure to land application may be overestimated over the practical values. It is necessary to build up the database about each livestock category other than Hanwoo.
본 연구에서는 동결채소의 저장 중 품질 변화를 측정하여 동결채소의 장기저장 가능성과 이용 가능성을 모색해 보고자 하였다. 수확직후 여러 가지 채소는 선행실험을 통한 최적 조건에서 blanching 처리한 후 $-40^{\circ}C$에서 24시간 동안 급속동결시켜 $-20^{\circ}C$에서 저장하면서 분석하였다. 동결시킨 취나물, 콩나물, 얼갈이배추, 애호박 및 대파는 저장 기간 12개월 동안 색도의 증가와 감소가 크게 나타나지 않았다. 취나물, 애호박 및 대파의 경도는 저장기간 동안 비슷한 수준을 유지하였고, 얼갈이배추는 약간 증가하는 경향을 나타내었다. 반면에 콩나물의 자엽부와 배축부는 저장 6개월째에 경도가 큰 폭으로 증가하는 경향을 나타내었다. 저장기간 동안 취나물의 총균수는 $10^3$ CFU/g 수준을 콩나물, 얼갈이배추, 애호박 및 대파는 $10^2$ CFU/g 수준을 유지하였으며, 대장균군은 모든 시료에서 검출되지 않았다. 관능검사는 외관, 색, 이취, 조직감 그리고 맛 등의 전반적인 품질 요소들을 종합하여 전체기호도를 조사한 결과 5점을 marketable의 한계점으로 두었을 때 저장기간에 따른 동결채소의 상품성에는 큰 문제가 없는 것으로 나타났다. 따라서 동결 취나물, 콩나물, 얼갈이배추, 애호박 및 대파는 동결저장 12개월까지 저장 안전성이 있는 것으로 판단된다.
본 연구에서는 조선후기 기록물의 편찬과 관리의 측면을 "승정원일기"와 "일성록", 의궤(儀軌)의 편찬과, 사고 및 외규장각의 설치, 실록청의궤와 실록형지안의 작성을 중심 소재로 하여 연구를 진행하였다. 조선후기에는 고문서, 문집, 지리지 등 다양한 국가기록물을 편찬하였지만 본 연구에서는 주로 시기적으로 연속해서 간행된 거질의 편찬물을 중심으로 연구를 진행하였다. 연구의 결과 조선후기에 왕실이 중심이 되어 철저하게 기록물을 편찬, 보관, 관리했음을 다시금 확인할 수가 있었다. 국왕의 일거수일투족을 기록에 담은 것은 무엇보다 정치의 공개성과 투명성을 강조하기 위함이었을 것이다. 당대에 그치지 않고 영원히 기록물이 보존되는 전통을 확립하기 위하여 험준한 산간 지역에 사고를 설치하고, 강화도와 같은 국방상 안전하다고 판단한 지역에 외사고를 설치하기도 하였다. 왕명을 받은 사관이 정기적으로 사고에 가서 서책을 점검하는 작업을 게을리 하지 않았으며, 기록물 점검의 전 과정을 실록형지안으로 작성하기도 하였다. 기록물에 대한 영구적인 보존과 관리를 위해서는 왕명을 받은 사관이 포쇄 작업을 정기적으로 수행하도록 하였다. 이처럼 철저하게 기록물을 보존하고 관리한 결과 현재에도 거의 완전한 형태로 옛 문헌을 접할 수가 있다. 선조들이 남겨준 뛰어난 기록문화와 그 기록물을 철저히 보존한 정신을 계승하는 것이 현재에도 절실히 요청되고 있다.
