A Study on Abdomen Ultrasonography Classified by Particular Disease Practiced in Health Promotion Center of a University Hospital (한 대학병원 종합건강진단센터에서 시행한 복부 초음파검사의 유소견 연구)

  • This study is to get preliminary data for an effectiveness evaluation of abdominal examination and improvement of it. Abnormal cases of abdominal ultrasonography are classified by sex, frequency, diagnosis and age. 4,924 examinees were included at a university hospital of health promotion center from January to December in 1999. The results are as follows. 1. According to the distribution of sex, there are more male patients(55.0%) than females patients(48.0%). For men, 40's showed the highest percentage among examinees. For women, 50's were the highest. 2. The reason that they visited the health promotion center was that 'they wanted to check their health status'. This answers were reported the highest(59.3%). 3. Patients that had abnormal cases of abdominal ultrasonography were 48.3%. Liver, kidney, gallbladder showed the highest percentage of abnormal cases in order of organs. Additionally, abnormal cases were discovered in liver cases. 4. According to the frequency of abnormal cases among examinees, the slight fatty liver were the highest regardless of sex. Men had the slight fatty liver, kidney simple cyst, liver calcification and liver simple cyst in order of abnormal cases. Women showed the slight fatty liver kidney simple cyst, kidney calcification, liver simple cyst, and blood vessel tumor in order of abnormal cases. 5. For the abnormal cases of liver by sex and age, the 50's reported the highest number of abnormal cases in men(299 patients). In addition, 60's had the highest of disease rata 47.8%. For women, 50's reported the highest number of abnormal cases(361 patients). Over 70's patients had the highest of disease rata 52.6%. For kidney, men and women showed the highest number of abnormal cases -62 vs 44 respectively. Over 70's patients had the highest percentage of disease rata-23.2% vs 14.0% respectively. For gallbladder, the number of abnormal cases were the most in men's 60's (31 patients) and in women's in the same age group (32patients). Disease showed the highest percentage in men's 60's(7.6%) and in women's 70's (14.0%). 6. According to malignant tumor, 17patients were liver cancer, 2patients stomach ca and 1pt kidney cancer. 7. The relationship between the malignant tumor and the examination motive was that 'they wanted to check their health status(41.0%)' and 'regular checkup (24.0%)'.

Morphological Characteristics and Classification of 25 Selected Clones of Aralia elata Seem (두릅나무 25개 선발 클론의 형태적 특성과 유연관계)

  • Aralia elata Seem. have a typical traditional significance among the wild herbs traditionally. Edible shoots of A. elata will augment consumer's interest due to its high value functional value, eco-friendly and pesticide-free produce. A. elata's root, fruit and bark are used as material of hypoglycemic agent and medicine for diabetes, kidney trouble, acute hepatitis, rheumation arthritis, stomach cancer and gastroenteric trouble. Flavonoid glycoside compound which is separated from A. elata's shoot shows high antioxidative activity. Also, root's identified active materials of antimicrobial was reported to be produced as food preservative and handy antimicrobial. Therefore, this research investigated quantitative morphological characteristics of leaves, spine and bud in naturally dominated and introducted A. elata in south Korea and then considered its principal compound analysis(PCA) and classification analysis(CA) among the 6 improved cultivars and 19 clones. PCA results showed that it show 76% accumulated explanation from four PC. The A. elata clones were classified into five groups; the first group of 15 clones including Yeongok, the second group of 5 clones including Yeoju, the third group of Bonghwa, Ulleung, the fourth group of Yongmunsa, Boseong and the fifth group of Singu. The object of this study will give us invaluable information about breeding by selection of A. elata in south Korea.

Correlation between HER2 Overexpression and Clinicopathological Characteristics in Gastric Cancer Patients Who Have Undergone Curative Resection

  • Purpose: At present, a human epidermal growth factor receptor 2 (HER2)-based concept of tumor biology has been established, and trastuzumab ($Herceptin^{(R)}$; Genentech/Roche, San Francisco, CA, USA), a monoclonal humanized antibody directed against HER2, is a pivotal agent for the management of HER2 positive (HER2+) metastatic breast cancer. It is also known that HER2 has a predictive value in gastric cancer; however, its association with the prognosis of this disease remains uncertain. The purpose of this study was to evaluate both the relationship between HER2 overexpression in the tumors of gastric cancer patients, and the prognosis of these patients who have had curative resection. Materials and Methods: A total of 139 consecutive patients with gastric cancer who underwent surgery at the Kosin University Gospel Hospital between October 2011 and March 2012 were included in this retrospective study. All tumor samples were examined for HER2 expression by immunohistochemistry. A retrospective review of the medical records was conducted to determine the correlation between the presence of HER2 overexpression and clinicopathological factors. Results: The HER2+ rate was 15.1%. HER2 overexpression was associated with histological grade (P=0.044) and Lauren classification (P=0.036). There was no significant difference in the 2-year overall survival between HER2+ and HER2- patients (P=0.396). Multivariate analysis showed that HER2 was not an independent prognostic factor. Conclusions: HER2 overexpression in tumors was associated with histological grade and Lauren classification in gastric cancer patients with curative resection. However, HER2 was not an independent prognostic factor for gastric cancer in our study.

A study in Mack-Gyung(脈經) on taking pulse of relative weight to five Viscera attachment system in Nan-Gyung(難經) (난경(難經)에서 안맥(按脈)의 경중(輕重)에 따른 오장배속방법(五臟配屬方法)에 대(對)한 맥경(脈經)과의 비교(比較) 연구(硏究))

  • In Nan-Gyung, showed that could know the lung condition taking pulse with the weight of three beans, the heart condition taking pulse with the weight of six beans, the spleen condition taking pulse with the weight of nine beans, the liver condition taking pulse with the weight of twelve beans, the kidney condition pressing to bone(骨). This theory is first suggested in Nan-Gyung(難經). In those case, the weight of three, six, nine, twelve beans and pressing to bone don't mean not the real weight but the relative weight(輕重) of taking pulse(按脈). In other words, those represent Boo Jung Chin(浮中沈), which are the conception of the upper, the meddle, the lower part(上中下). So, we could take pulse of the heart and the lung condition in Boo(浮), the spleen condition in Jung(中), and the liver and the kidney condition in Chim(沈). The heart and the lung pulse showed in the Boo(float level) must be seen with Boo-Mack(부맥 : float pulse), the liver and the kidney pulse showed in Chim (sinklevel) must be seen also with Chin-Mack(沈脈 : sink pulse). The result of the method of taking pulse of viscera with relative weight focused on the as pect of mornal pulse(平脈) and disease pulse(病脈) of five viscera in Mack-Gyung publeshed later than Nan-Gyung and special works which made a comprehensive survey the result is as follow. 1. In normal pulse of five viscera, the heart and the lunk pulse were shown with Boo-Mack(浮脈:float pulse) as the central figure, the liver's and the kidney's pulse were shown centering around Chim-Mack(沈脈: sink pulse) and the spleen's pulse was shown with Wan-Mack(緩脈) which is vital force of stomach(胃氣) and seen in only middle part. 2. In disease pulse of five viscera, frequently, the heart and the lung pulse was shown as Chim-Mack(sink pulse), the liver and the kidney pulse was seen as Boo-Mack (float pulse). 3. In the case of normal pulse. the method of taking pulse with relative weight in Nan-Gyung agree with the normal pulse of five viscera in Mack-Gyung. But in the case of disease pulse, they didn't correspond with the other. 4. So the method of taking pulse with relative weight in Nan-Gyung is not the exam pulse which ca be used in the clinical diagnosis but one of the feeling pulse way to bring in the conception of location of the visceras. 5. From now on, the method of taking pulse rdlated to relative weight need to be looked into minutely compared with later physician's theory than Mack-Gyung.

Analysis of Immunomodulating Gene Expression by cDNA Microarray in $\beta$-Glucan-treated Murine Macrophage

  • ${\beta}$-(1,3)-D-Glucans have been known to exhibit antitumor and antimicrobial activities. The presence of dectin-1,${\alpha}$, ${\beta}$-glucan receptor of dendritic cell, on macrophage has been controvertial. RT-PCR analysis led to the detection of dectin-1${\alpha}$ and ${\beta}$ in murine macrophage Raw264.7 cell line. Among the various organs of mouse, dectin-1${\alpha}$ and ${\beta}$ were detected in the thymus, lung, spleen, stomach and intestine. To analyze gene expression modulated by ${\beta}$-glucan treated murine Raw264.7 macrophage, total mRNA was applied to cDNA microarray to interrogate the expression of 7,000 known genes. cDNA chip analysis showed that ${\beta}$-glucan of P. osteatus increased gene expressions of immunomodulating genes, membrane antigenic proteins, chemokine ligands, complements, cytokines, various kinases, lectin associated genes and oncogenes in Raw 264.7 cell line. When treated with ${\beta}$-glucan of P. osteatus and LPS, induction of gene expression of TNF-${\alpha}$ and IFN-R1 was confirmed by RT-PCR analysis. Induction of TNF-R type II expression was confirmed by FACS analysis. IL-6 expression was abolished by EDTA in ${\beta}$-glucan and LPS treated Raw264.7 cell line, indicating that ${\beta}$-glucan binds to dectin-l in a Ca$\^$++/ -dependent manner. To increase antitumor efficacy of ${\beta}$-glucan, ginsenoside Rh2 (GRh2) was co-treated with ${\beta}$-glucan in vivo and in vitro tests. IC$\sub$50/ values of GRh2 were 20 and 25 $\mu\textrm{g}$/$m\ell$ in SNU-1 and B16 melanoma F10 cell line, respectively. Co-treatment with ${\beta}$-glucan and GRh2 showed synergistic antitumor activity with cisplatin and mitomycin C both in vitro and in vivo. Single or co-treatment with ${\beta}$-glucan and GRh2 increased tumor bearing mouse life span. Co-treatment with ${\beta}$-glucan and GRh2 showed more increased life span with mitomycin C than that with cisplatin. Antitumor activities were 67% and 72 % by co-injection with ${\beta}$-glucan and GRh2 in the absence or presence of mitomycin C, respectively.

Comparison of Digestive Organ and Body Composition among the Cultured, Wild and Released Fish, 1-Year Black Sea Bream (Acanthopagrus schlegeli) (양식, 방류 및 자연산 감성돔 1년어의 소화기관 및 체성분 비교)

  • In order to evaluate adaptation to the environment in released fish, digestive organ and body composition were compared among 1-year old cultured (CUL), released (REL) and wild (WIL) Black Sea Bream, Acanthopagrus schlegeli. Hepatosomatic index (HSI) of the REL was significantly lower than those of the CUL and WIL fish, but intestine weight index (IWI) of REL was significantly higher than those of the CUL and WIL fish. There were no significant differences in stomach weight index (SWI) and condition facto. (CF) among those different fish groups. The number of pyloric caeca of the REL and WIL fish were 4, but the $30{\%}$ of CUL fish was 3 counts. The moisture content of whole body in the WIL fish was significantly higher than the CUL and REL fish, but significant lowest in the crude lipid of whole body. The content of moisture and crude protein of dorsal muscle were no significant differences among the CUL, REL and WIL fish, but crude lipid content of WIL fish was significantly lower than the CUL and REL fish. Moisture content of CUL fish in the liver was significantly lower than the REL and WIL fish, and crude lipid was significant lowest in the WIL fish. Amino acid content of dorsal muscle in the WIL fish was highest in the total amino acid, EAA and E/A ratio, and CUL fish was highest EAA and E/A ratio in liver. EPA content in dorsal muscle of WIL fish was lower than CUL and REL fish, and the REL fish was highest in DHA content of the fatty acid. EPA content in liver of WIL fish was lower than CUL and REL fish, and DHA content in REL and WIL fish was highest and lowest. Crude protein content of CUL and WIL fish in the scale was significantly higher than REL fish, but there were no significant differences in contents of crude lipid and ash. Ca and P of scale were formed most of mineral and content of P in CUL fish was significantly higher than those of REL and WIL fish. There were no significant differences in Mg, K, Na and S of scale, but CUL fish was significantly lower than REL fish in C1. The results suggest that difference of digestive organs and body composition concluded it from differences of inhabitation environment and feed formulations.

Mechanism underlying Chios gum mastic-induced apoptosis on SCC25 human tongue squamous cell carcinoma cell line

  • Chios gum mastic (CGM) is a resin produced from the stem and leaves of Pistiacia lentiscus L var chia, a plant which grows only on Chios Island in Greece. CGM has been used for many centuries as a dietary supplement and folk medicine for stomach and duodenal ulcers in many Mediterranean countries and is known also to induce cell cycle arrest and apoptosis in some cancer cells. In this study, we further investigated the induction and mechanisms underlying the apoptotic response to CGM treatment in the SCC25 human tongue squamous cell carcinoma cell line. The viability of SCC25 cells, human normal keratinocytes (HaCaT cells) and human gingival fibroblasts (HGF-1 cells), and the growth inhibition of SCC25 cells were assessed by MTT assay and clonogenic assay, respectively. Staining with Hoechst and hemacolor dyes and TUNEL assays were employed to detect SCC25 cells undergoing apoptosis. SCC25 cells were treated with CGM, and this was followed by western blotting, immunocytochemistry, confocal microscopy, FACScan flow cytometry, MMP activity and proteasome activity analyses. CGM treatment of SCC25 cells was found to result in a time- and dosedependent decrease in cell viability, a dose-dependent inhibition of cell growth, and apoptotic cell death. Interestingly, CGM showed a remarkable level of cytotoxicity in SCC25 cells but not in normal cells. Tested SCC25 cells also showed several lines of apoptotic manifestation. Taken together, our present findings demonstrate that CGM strongly inhibits cell proliferation by modulating the expression of G1 cell cycle-related proteins and induces apoptosis via the proteasome, mitochondria and caspase cascades in SCC25 cells.

Osteonectin Interacts with Human Nebulin C-terminus in Skeletal Muscle

  • Nebulin is a giant actin binding protein (600-900 kDa) which is specific to skeletal muscle. This protein is known to regulate thin filaments length in sarcomere as a molecular template. The C-terminus of nebulin is located in the Z-disc of muscle sarcomere and is bound to other proteins such like myopalladin, titin, archvillin, and desmin. The N-terminus of nebulin binds to tropomodulin at the pointed ends of the thin filaments. In recent research, nebulin not only found in brain but also expressed in heart, stomach, and liver. So, the roles of nebulin in non-muscle tissue have been studied. However, lack of information or studies on nebulin binding proteins and nebulin function in brain are available so far. Therefore, the current study have investigated a novel binding partner of Nebulin C-terminus by using yeast two-hybrid screening with human brain cDNA library. Nebulin C-terminus, containing simple repeats, serine rich and SH3 domain, interacts with osteonectin C-terminal region. The specific interaction of nebulin and osteonectin were confirmed in vitro by using GST pull-down assay and reconfirmed in vivo by using transfected COS-7 cells with EGFP-tagged nebulin and DsRed-tagged osteonectin. Consequently, this study identified SH3 domain in nebulin C-terminus specifically binds to extracellular Ca-binding (EeC domain in osteonectin. Also, nebulin C-terminus fusion protein colocalized with osteonectin EC domain fusion protein in transfected COS-7 cells. The current study found the interaction between nebulin and osteonectin in human brain for the first time and suggested the nebulin in brain may be associated with osteonectin, as a regulator of cell cycle progression and mitosis.

  • Gastric cancer is the most common disease which is the highest genetic rate, chemotherapy is the only known way to treat, especially advanced gastric cancer. Although, it is the remedy, the harmful side-effects are an obstacle of the sustaining treatment and negative effect on immunologic ability of patients prologed. Consequently, it is surely necessary that effective control for sequele of anti-cancer treatment, and remedy which can improve the quality of patients life. 1, the writer, medicate patients, diagnose as gastric cancer in 4th stage, with Sohap-hyangwon and Chungwoolhyadamjen who complained of pain in sequele of anti-cancer treatment. The size of the metastastic liver tumor is decreasing in patients and the immunologic abilities are maintaining stable. This clinical cases are shown that the oriental medical care in long-term can improve the quality of life of the patients who has sequele of anti-cancer treatment.

The Effects of Electrolyzed Reduced Water on Blood and Organ Tissues of Mice (전해환원수 음용이 마우스의 혈액과 장기조직에 미치는 영향)

  • The main purpose of this study is to investigate the effects of electrolyzed reduced water (ERW) on blood components and electrolytes, as well as elected tissues on mice. The mice were supplied either tap water (control group) and ERW (experimental group) for two months. There were no significant different between two groups anatomically and physiologically. In the blood electrolyte study, the experimental group had less Na and BUN compared to the control group. In the blood component study, the experimental group had less neutrophiles and the control group had more lymphocytes. In histological study, no tissue changes were noticed in various organs, including the stomach, small intestine, heart, and liver tissues. In conclusion, ERW has no noticable side affects on blood and organ tissues, and might be safe to a living body.
