대형 와 모사를 통한 레이놀즈 수 증가에 따른 혼합 탱크 내의 유동 구조의 연구 (The study of Flow Structure in a Mixing Tank for Different Reynolds Numbers Using LES)

  • The stirred tank reactor is one of the most commonly used devices in industry for achieving mixing and reaction. Here we report on results obtained from the large eddy simulations of flow inside the tank performed using a spectral multi-domain technique. The computations were driven by specifying the impeller-induced flow at the blade tip radius. Stereoscopic PlY measurements (Hill et al. $^{(1)}$) along with the theoretical model of the impeller-induced flow (Yoon et al. $^{(2)}$) were used in defining the impeller-induced flow as superposition of circumferential, jet and tip vortex pair components. Large eddy simulation of flow in a stirred tank was carried out for the three different Reynolds numbers of 4000, 16000 and 64000. The effect of different Reynolds numbers is well observed in both instantaneous and time averaged flow fields. The instantaneous and mean vortex structures are identified by plotting an isosurfaces of swirling strength for all Reynolds numbers. The Reynolds number dependency of the non-dimensional eddy viscosity, resolved scale and subgrid scale dissipations is clearly shown in this study.

혼합탱크 내의 임펠라 형태에 따른 유동 특성에 관한 수치해석 (Numerical Study on Flow Patterns in a Stirred Tank with Impeller Types)

  • The present study is concerned with the flow patterns induced by various impellers in a rectangular tank. Impellers are FBT (Flat blade turbine), PBT (Pitched blade turbine), Shroud turbine, Rushton turbine, and Helical ribbon turbine types. The solutions of flows in moving reference frames require the use of 'moving' cell zone. The moving zone approaches are based on MRF (Multiple reference frame), which is a steady-state approximation and sliding method, which is an unsteady-state approximation. Numerical results using two moving zone approaches we compared with experiments by Ranade & Joshi, which have done extensive LDA measurements of the flow generated by a standard six-bladed Rushton turbine in a cylindrical baffled vessel. In this paper, we simulated the flow patterns with above-mentioned moving zone approaches and impellers. Turbulence model used is RNG $k-{\epsilon}$ model. Sliding-mesh method is more effective than MRF for simulating the rectangular tank with inlet and outlet. RNG $k-{\epsilon}$ model strongly underestimates the velocity of experimental data and velocity by Chen & Kim's model, but it seems to be correctly predicted in overall distribution.

Cultivation of Transgenic Nicotiana tabacum Suspension Cells in Bioreacters for the Production of mGM-CSF

  • Transgenic Nicotiana tabacum cells were cultivated for the production of murine granulocyte macrophage-colony stimulating factor (mGM-CSF) in both a stirred tank bioreactor and an airlift bioreactor with draft tube. Cell growth and mGM-CSF production in the airlift bioreactor were found to be better than those achieved in the stirred tank bioreactor. In the airlift bioreactor, 9.0g/L of cells and 2.2ng/mL of mGM-CSF were obtained (11.0g/L and 2.4ng/mL, respectively in shake flasks). Although the lag period was prolonged and mGM-CSF production was lowered by 33% in the stirred thank bioreactor as compared to the control culture, the maximum cell density was increased up to 12.0g/L due to better mixing by agitation at the higher cell density.

Nonlinear Observer flay Applications of Fermentation Process in Stirred Tank Bioreactor

  • This paper proposed a modified observer based on Busawon's high gain observer using an appropriate time depended function, which can be chosen to make each estimated state converge faster to its real value. The stability of the modified observer is proved by using Lyapunov function. The modified nonlinear observer is applied to estimate the states in stirred tank bioreactor: out-put substrate concentration, output biomass concentration and the specific growth rate of the process. The convergences of the modified observer and Busawon's observer are compared trough simulation results. As the results, the modified observer converges faster to its real value than the well-known Busawon's observer.

  • The effects of fermentor types on biopolymer production with Cordyceps militaris has been studied in this articles. Since the shear stresses of stirred tank ferementor and bubble column fermentor were different, the mycelium growth and biopolymer production were different. The mycerial growth was high with the bubble column fermentor for the early stage but biopolymer production was lower than that with stirred tank fermentor. The production of biopolymer was closely related to shear stress to the cells and biopolymer production was increased with increased shear stress.

  • This paper proposes a nonlinear adaptive controller based on back-stepping method for tracking reference substrate concentration by manipulating dilution rate in a continuous baker´s yeast cultivating process in stirred tank bioreactor. Control law is obtained from Lyapunov control function to ensure asymptotical stability of the system. The Haldane model for the specific growth rate depending on only substrate concentration is used in this paper. Due to the uncertainty of specific growth rate, it has been modified as a function including the unknown parameter with known bounded values. The substrate concentration in the bioreactor and feed line are measured. The deviation from the reference is observed when the external disturbance such as the change of the feed is introduced to the system ...

당밀을 이용한 곤충병원성 곰팡이 Beauveria bassiana 726의 포자생산 (Spore Production of Entomopathogenic Fungus, Beauveria bassiana 726, Using Molasses)

  • To optimize the culture conditions for Beauveria bassiana 726, the effects of culture medium, pH, and temperature on mycelium and spore production were investigated. The optimum temperature and pH for the cultivation of B. bassiana 726 were 28 $^{\circ}C$ and 5.0, respectively. The optimized medium was composed of 1.0~2.0% total sugar from molasses, 0.5% corn steep liquor and 0.05% KH$_2$PO$_4$. In the cultivation of B. bassiana 726 with the optimum medium, the specific growth rate and substrate utilization were well-fitted with the proposed kinetic model in the shake flask and stirred tank reactor. When the fed-batch cultivation using carbon suorce, nitrogen source, and mineral salt as a feeding medium was compared with batch cultivation in stirred tank reactor, mycelium (12.7 g/L) and spore production (5.4$\times$$10^8/mL$) were enhanced up to 110% and 85%, respectively.

글리세롤 기반의 바이오에탄올 생산을 위한 연속생산반응기의 성능 비교 (Performance Comparison of Continuous Reactors for Bioethanol Production Based on Glycerol)

  • Ethanol production using glycerol as a carbon source was performed by Enterobacter aerogenes immobilized on calcium alginate beads. To improve the ethanol production, the optimal conditions such as loading amount of immobilized cells and glycerol concentration were investigated. The optimal loading amount of immobilized cells and glycerol concentration were 10 mL of calcium alginate bead and 10 g/L, respectively. Consequently, glycerol consumption rate, ethanol concentration and yield were 0.32 g/$L{\cdot}h$, 3.38 g/L and 0.43 g/g on the batch production, respectively. Continuous production of ethanol was successfully achieved using two types of immobilized cell reactors (continuous stirred tank reactor and packed bed reactor) from 10 g/L of glycerol. In the continuous stirred tank reactor, glycerol consumption, ethanol concentration, specific productivity and yield were 9.8 g, 4.67 g/L, 1.17 g/$L{\cdot}h$, 0.48 g/g, respectively. The concentration of produced ethanol was 38-44% higher comparison to batch fermentation, and continuous stirred tank reactor showed better performance than packed bed reactor.
