The purpose of this study is to define whether Asterina pectinifera Lectin (APL) is effective on the cytokine production. Isolated mRNA from hPBMC (human peripheral blood mononuclear cells) stimulated with APL for various reaction times (1 to 96 hours) was detected by RT-PCR. The intensity of band for IL-1 and $IFN{\gamma}$ mRNA was markedly increased at l hour, and IL-2 mRNA was strongly expressed at 4 hours. The mRNA band of APL-induced IL-2 and $IFN{\gamma}$ was weaker than that of IL-1, IL-6 and $TNF{\alpha}$. The mRNA expression of 4 cytokines (IL-1, IL-2, $IFN{\gamma}$ and $TNF{\alpha}$) was detected up to 48 hours, and that of IL-6 was detected until 72 hours. ELISA was used to look protein secretion of the cytokine gene with IL-1, IL-2 and TNF$\alpha$expressed strongly in RT-PCR. The highest protein secretion was at 4 hours with IL-1, at 8 hours with IL-2 and at 4 hours with $TNF{\alpha}$. These results suggest that APL can induce the production of some cytokines and the immune response from PBMC was done within the first few hours of stimulation with APL.
It is well known that a long-term exposure to a loud noise environment affects performance, since it distracts attention, and also is able to evoke stress accompanied by negative emotional states. The purpose of this study was to analyze dynamics of subjective and physiological variables during long-lasting (30 min) exposure to intensive white noise (85 dB[A]). Physiological signals on 23 college students were recorded by BIOPAC, Grass Neurodata systems and AcqKnowledge 3.5 software. Autonomic variables, namely skin conductance level (SCL), non-specific SCR number (N-SCR), inter-beat intervals in ECG (RR intervals), heart rate variability index (HF/LF ratio of HRV), respiration rate (RESP), and skin temperature (SKT) were analyzed on 5 min epoch basis. Psychological assessment (subjective rating of stress level) was also repeated on every 5 min basis. Regression and correlation analyses were employed to trace the time course of the dynamics of the subjective and autonomic physiological variables and their relationship. Results showed that intense noise evokes subjective stress with associated autonomic nervous system responses. However, it was shown that physiological variables endure specific changes in the process of exposure to loud white noise. Discussed are probable psychophysiological mechanisms mediating reactivity to long-term auditory stimulation of high intensity.
The purpose of this study was to investigate the effect of the body temperature peripheral circulation with vibration shoes in healthy 10 adults. The magnetic vibration device with non-electric power was mounted in the midsole of the vibration shoes. The experiment was divided into two groups: vibration shoes and no vibration shoes. Subjects were randomly selected and measured body surface temperature by digital infrared thermal imaging (DITI) and non-invasive capillaries change by nailfold microscope (NFM). After walking in a treadmill for 15 minutes at 4.0 km/h speed wearing normal shoes or vibration shoes, DITI and NFM were measured. The walking with vibration shoes showed the body surface temperature shift from the proximal to the distal. In addition, the diameter of the nailfold capillary in the vibration shoes group was thicker and clearer due to the increased blood flow than that of the no vibration shoes group. The vibration shoes are easy to apply to anyone who can walk because it can give vibration stimulation by walking without additional time, cost, and effort in daily life. Further studies are needed to explain the physiological effects of vibration frequency and intensity on the long-term perspective of target subjects resulting from vascular dysfunction.
Spinal dysraphism often causes neurological impairment from direct involvement of lesions or from cord tethering. The conus medullaris and lumbosacral roots are most vulnerable. Surgical intervention such as untethering surgery is indicated to minimize or prevent further neurological deficits. Because untethering surgery itself imposes risk of neural injury, intraoperative neurophysiological monitoring (IONM) is indicated to help surgeons to be guided during surgery and to improve functional outcome. Monitoring of electromyography (EMG), motor evoked potential, and bulbocavernosus reflex (BCR) is essential modalities in IONM for untethering. Sensory evoked potential can be also employed to further interpretation. In specific, free-running EMG and triggered EMG is of most utility to identify lumbosacral roots within the field of surgery and filum terminale or non-functioning cord can be also confirmed by absence of responses at higher intensity of stimulation. The sacral nervous system should be vigilantly monitored as pathophysiology of tethered cord syndrome affects the sacral function most and earliest. BCR monitoring can be readily applicable for sacral monitoring and has been shown to be useful for prediction of postoperative sacral dysfunction. Further research is guaranteed because current IONM methodology in spinal dysraphism is still deficient of quantitative and objective evaluation and fails to directly measure the sacral autonomic nervous system.
This study was undertaken to assess the effect of ginseng administration on T lymphocyte induced local xenogenic graft-versus-host(GVM) reactions which were induced with thymocyte, spleen cell and lymph node cell of ICR mice. Mice received daily 10mg of 70% alcohol ginseng extract oral1y for 100days and control mice remained untreated for the same period of time. The cells from donor mice were injected intradermally into the closely shaven abdominal skin of Sprague-Dawley rats for GVH tests. The thymocyte from control(ginseng-untreated) mice showed a negative local GVH reaction, whereas thymocyte from experimental(ginseng-treated) mice showed a positive reaction with the rate of 17.4%. When spleen cells were injected, the incidence of positive local GVH reaction was 66.7% among ginseng-treated mice, as opposed to incidence of 45.5% of positive local GVH reaction among control mice. The incidence of positive local GVH reaction of the lymph node cells when injected into a recipient was 71.4% among ginseng-treated mice as compared with that of 18.9% among control mice. The relationship between spleen cell inoculum and intensity of the local GVH reaction was assessed in ginseng-untreated mice. The intensity of GVH reaction clearly appears to be dose related. In ginseng-treated mice, a minimum of $1{\times}10^7$ spleen cell was required for production of positive local GVH reaction with almost linear relationship up to an inoculum of $5{\times}10^8$ cells. In control mice, however, a minimum of $1{\times}10^8$ spleen cells was required for positive GVH reaction. These results strongly suggest that the ginseng administration augments significantly the local xenogenic GVH reaction which was used to assess T lymphocyte function and immunocompetence of mice and in addition to this, these results appear to support previous suggestions that the local GVH reaction consitutes a qualitative test of the functional activity of T lymphocytes. These results may be the first to induce local GVH reaction, employing rats as recipient and mice as donor. This study was also desingned to investigate some of the effects of ginseng extract on lymphocyte-macrophage interactions. This was accomplished by in vitro quantification of 1) migratory inhibitory factor(MIF) synthetic capacity of splenic lymphocytes in mice previously primed with ginseng 2) MIF responsiveness of mouse peritoneal macrophages or chicken peripheral leucocytes under the presence of ginseng extract 3) migration ability of chicken peripheral leucocytes by direct stimulation of ginseng extract or ginseng saponin and 4) immunosuppressive effects of immunosuppressants such as cyclophosphamide, cyclosporin A or dexamethasone. Mice divided equally into the ginseng and the saline groups, which received intraperitoneally daily 0.2ml of ginseng absolute alcohol-extract(5mg/ml) and same amount of saline for 15 days, respectively. The cellular immune responsiveness of these mice was assayed 15 days after ginseng pretreatment. Splenic lymphocytes of mice treated with ginseng, when stimulated with sensitized specific-antigen such as sheep red blood cells or toxoplasmin, or with polyclonal activator concanavalin A, produced significantly more MIF than those of control saline group. MIF responsiveness of normal mouse macrophages was significantly augmented when assayed under the presence of ginseng extract (1mg/ml). The migratory ability of normal chicken leucocytes in the absence of MIF was significantly decreased by the stimulation of ginseng extract alone. MIF response was significantly decreased by immunosuppressants and this impaired response was not restored by ginseng pretreatment. This study was additionally performed to evaluate the effect of ginseng on the expulsion of adult Trichinella spiralis in mice. ICR mice were infected experimentally by esophageal incubation of 300 T. spiralis infective muscle larvae prepared by acid-pepsin digestion of infected mice. and received oral administration of 70% alcohol ginseng extract(10mg/mouse/day) for the indicated days plus 4 days before infection. At various times after infection, the number of adult T. spiralis worms in small intestines was determined. Interestingly, ginseng-treatment was accompanied by accelerated expulson of T. spiralis. These results led to the conclusion that Panax ginseng caused some enhancing effect on GVH reaction, macrophage migration inhibition reaction and expulsion of T. spiralis. In addition these results suggested that the mechanisms responsible for this enhancement of ginseng may be chiefly or partially due to nonspecific stimulation of cell-mediated immune response.
MFDS(Ministry of Food and Drug Safety : 식품의약품안전처)에서 고시한 고주파 자극기 기준 규격을 준수한 고주파 출력 장치를 설계 및 개발하였다.
The purpose of this study is to prove the analgesic effects of apitoxin and its mechanism via jaw-opening reflex(JOR) and measuring expression of mRNA in Phospholipase and Tryptophan hydroxylase(TPH) using RT-PCR. The experiments were carried out on Sprague-Dawley rats(300-400g) and mastocytoma(P-185 HTR) for JOR and RT-PCR, respectively. Rats anesthetized with thiopental sodium (80mg/kg) were used in the Tooth Pulp stimulation induced JOR. The amplitude of a digastric electromyogram (dEMG) was recorded during the stimulation at an intensity of 1.5 times the threshold for JOR. Apitoxin used in this experiment was diluted with normal saline by 1:1000. Apitoxin was injected intravenously into the test group while normal saline to the control group. However, it was injected directly into the cell of mastocytoma. We referred to base sequence registered in Genbank in designing primers for RT-PCR. The results were as follows; (1)Compared with control group, analgesic effect started to show right after Sprague-Dawely rats were treated with apitoxin($71.50{\pm}8.08$) and lasted for 50 minutes. (2)As a result of the experiment of RT-PCR, we witnessed significant changes in the degree of expression of phospholipase or rate-limiting enzyme of biosynthesis of prostaglandins with $10{\mu}g/ml$ apitoxin.($31.74{\pm}18.98%$, P<0.05) (3)As a result of the experiment of RT-PCR, we witnessed significant changes in the degree of expression of TPH or rate-limiting enzyme in biosynthesis of serotonin with $10{\mu}g/ml$ apitoxin.($131.37{\pm}16.87%$, P<0.05). These results suggest that $10{\mu}g/ml$ apitoxin have the most analgesic effects. This study showed that apitoxin has analgesic effects and held good for 50 minutes. The injection of apitoxin has brought out changes in the degree of expression of phospholipase and TPH. These results strongly suggest that analgesic mechanism by apitoxin is closely related to prostaglandins and serotonin.
This study is aimed to find out whether there is difference in the physiological change of a human body according to the illumination and color of interior space or not and to specify the effect of the condition of illumination and color, respectively on the attention. In order to do so, White and Green were selected for colors and 4,000k, 5,000k, and 6000k were done for color temperature, and then attention was identified. Examining the results, the more color temperature increased, the more attention improved (P < 0.05), and in the case of EEG, ${\alpha}$ wave decreased while performing the task of attention (P < 0.01), and ${\beta}$ wave decreased more in Green than White in color condition, and it increased more in 4,000k than 5,000k and 6,000k (p < 0.05) in color temperature condition. To sum up, color condition didn't contribute to the attention much, in the case of color temperature, when it is 6,000k, it is judged that it helped to improve attention. It is considered that relaxation contributed to improving attention, as ${\beta}$ wave and sympathetic nerve decreased in 6,000k (p < 0.05). It is judged that the relaxation of tensions which happened due to a beta wave and the reduction of sympathetic nervous system activity in 6,000k, a condition of high color temperature, contributed to the improvement of concentration. In further researches, it is intended that a test will be conducted for the subjects of different ages, and the correlation between color temperature and color stimulation and the influence of them on human body would be observed in subdivided, various test conditions through various color temperature and color stimulation.
