Lipid Composition and Protein Pattern of Prunus Tomentosa Thunberg Seed (앵두(Prunus Tomentosa Thunberg)씨의 지방질 조성 및 단백질 패턴)

  • Prunus tomentosa Thunberg seed was investigated to evaluate its possibility for use as food resources of fats and proteins. The seed contained 40.38% of crude fat and 26.59% of crude protein. The lipid fractions obtained by silicic acid column chromatography were mainly composed of 95.49% of neutral lipids, whereas compound lipids were only 4.51%. Among the neutral lipid components by thin-layer chromatography, triglycerides were 89.86%, sterols, monoglycerides, sterol esters, free fatty acids and diglycerides were 4.14%, 2.98%, 1.77%, 1.07%, and 0.18%, respectively. Oleic acid (65.06-66.05%) and linoleic acid (26.56-28.40%) were the main fatty acids in the total lipid, neutral lipid and triglyceride fractions. In the glycolipid and phospholipid fractions, predominant fatty acids were oleic acid (40.55-51.46%), linoleic acid (20.26-30.89%) and palmitic acid (17.64-21.43%). The extractability of salt soluble protein of seed was 60%, and recovery rate of main protein fraction separated by Sephadex G-200 was about 46.5%. The electrophoretic analysis showed 7 bands in seed protein.

A Comparative Study on the Lipid Components of Barley and Malt -I. Composition of Neutral Lipids- (보리와 맥아(麥芽)의 지방질(脂肪質) 성분(成分)에 관한 비교(比較) 연구 -제(第)1보(報) : 중성지질(中性脂質)의 조성(組成)-)

  • Neutral lipids of the total lipid extracted from 4 representative varieties of barley grown in Korea and their corresponding malt were studied. Total lipids of barley and malt were solvent extracted with chloroform:methanol:water (1.0 : 1.0 : 0.9, v/v), The total lipids were fractionated into neutral and polar lipids by silicic acid column chromatography, and neutral lipids fraction was separated by thin layer chromatography and quantitated by TLC scanner. The fatty acid compositions was determined by gas liquid chromatography. The average content of total lipid in the 4 barleys and their malts were 3.3 and 2.9%, and average of neutral lipids content in the barley and malt lipids were 73.8 and 68.5%, respectively. Among the neutral lipids, triglycerides and free fatty acids were the major components, and triglycerides content decreased and free fatty acids content increased during malting. Sterol esters, free sterols, 1,3-and 1,2-diglycerides were the minor components of the neutral lipids, and contents of those components showed increasing tendency during malting. The major fatty acid composition of the total lipids were linoleic, palmitic and oleic acids, and in general, the malts had lower amounts of unsautrated fatty acids and high amounts of saturated fatty acids. Fatty acid composition of neutral lipids was of almost the same pattern as that of the total lipids.

Effect of Different Application of P, K, and Ca on Lipid Content and Composition of Perilla (인산(燐酸), 가리(加里), 석회(石灰) 시용(施用)이 들깨의 유지함량(油脂含量)과 그 조성(組成)에 미치는 영향(影響))

  • This study was carried out to investigate the effects of P, K and Ca on Perilla lipid content and composition. The results were summarized as follows; Seed yield of Perilla was more increased with P application than K and Ca. K application resulted in increased lipid content of Perilla seed, while P and Ca had no effect on lipid content. Triglyceride content in Perilla lipid was increased with application of P, particulary up to 92.1% with P+K+Ca application. Main fatty acids in Perilla lipid were palmitic, oleic, linoleic, and linolenic acid that is the most predominating fatty acid. P and Ca applicatin without K decreased the proportion of linolenic acid, and therefore degree of unsaturation when P and Ca were respectively applicated was 1.80 and 1.84, but when together with K they were respectively increased to 2.06 and 2.11. 4-Desmethyl sterols which is a predominating fraction in unsaponifiables of seed lipid were identified as campesterol, stigmasterol, and sitosterol. Of the 4-desmethylsterols, sitosterol showed the highest proportion at all pots with different kinds of fertilizers.

Effect of Glucosylceramides and Sterols Isolated from Agaricus Blazei Extract on Improvement of Skin Cell (신령버섯에서 분리된 Glucosylceramide 및 Sterol의 피부 세포 개선 효과)

  • Agaricus blazei Murill (Almond mushroom) has many beneficial effects, such as anti-cancer, immuneenhancement, and anti-obesity. Also, its skin benefits have been reported for antioxidant, anti-inflammatory, and whitening. In order to elucidate these effects, many studies have been conducted. In this study, we reconfirmed the skin efficacy of the extract of the mushrooms mushrooms. The Agaricus blazei extract showed inhibition of melanin synthesis, enhancement of collagen synthesis, and up-regulation of gene expression (hyaluronan syntahase-2, 3 and aquaporin-3) at 100 ㎍/mL. and identified the ingredients from the extract. We further investigated them to find an applicability as cosmetic ingredients. The ingredients were confirmed comparison of their spectroscopic data with literature values. They were identified as being ergosterol (1), 5-dihydroergosterol (2), cerevisterol (3), cerebroside B (4), cerebroside D (5), adenosine (6), and benzoic acid (7). Among these compounds, we evaluated skin efficacy for two cerebrosides and three ergosterol derivatives that have not been reported its efficacy. As a result, 5-dihydroergosterol (2) inhibited melanogenesis in B16F10 and promoted collagen biosynthesis in human dermal fibroblast. In addition, cerevisterol (3), cerebroside B (4), and cerebroside D (5) inhibited NO production in RAW 264.7 cell. In particular, cerebroside D (5) increased the expression of hyaluronan synthase-2 and aquaporin-3 genes in HaCaT. These results suggest that Agaricus blazei extract and isolated compounds can be used as cosmetic ingredients.

The Sterol Components of Undaria Pinnatifida and the Incorporation of $^14C-1-acetate$ into Them (미역의 스테롤 조성과 $^14C-1$-식초산염의 스테롤 전환에 관하여)

  • The present study was carried out to knew the sterol components of U. pinnatifida and their incorporation abilities of $^14C-1-acetate$ injected into it. The results obtained are as follows: 1. The total lipids are classified as hydrocarbon $1.6\%$, pigment and sterol ester $2.5\%$, triglyceri do $3.3\%$, free fatty acid $2.2\%$, free sterol $3.8\%$, chlorophyll $18.8\%$, and polar lipids $ 67.3\%$ 2. The sterol mixture from U. pinnatifida are omposed of cholesterol $3.5\%$, 24-methylene-cholesterol $11.2\%$, fucosterol $85.3\%$. 3. The radioactivities of the lipids classes from U. pinnatifida injected with $^14C-1-acetate$ are distributed 4,648 dpm/ug in total lipid, 2,754 dpm/mg in polar lipids, 373 dpm/mg in chlorophyll, 22,481 dpm/mg in free sterol, 6,520 dpm/mg in free fatty acid, 789 dpm/mg in sterol ester and 358 dpm/mg in hydrocarbon respectively. 4. The specific radioactivities of the sterols are 115 dpm/mg in cholesterol, 147,821 dpm/mg in 24-methylenecholesterol, 20, 887 dpm/mg in fucosterol.

Distribution of Lipids in Panax ginseng Root (인삼근의 지질 분포)

  • Diversities of lipid compositions according to the morphological differences of the Panax ginseng root were studied by means of column, thin layer, gas-liquid chromate-graphies and histochemical stainings. Purified lipids from various parts were 1.08-2.23% of dry weight, of which 64.2-73.5% were neutral lipids, 15.4-17.4% were glycolipids and 10.4-19.2% were phospholipids. Especially the contents of neutral lipids were highest in cortex, suggesting to be the presence of lipid ducts only in cortex. Triglycerides, sterol esters and hydrocarbons were abundant in the neutral lipid fractions. Twelve components were identified in the periderm and cortex, but unidentified II, IV and V components were not present in the medulla. The major components of glycolipid freactions were sterol glycoside, digalactosyl diglyceride and esterified sterol glucoside. Phosphatidyl glycerol, phosphatidyl choline and phosphatidyl ethanolamine were major components of phospholipid fractions, And phosphatidyl choline was extreamly much in the periderm and medulla, but phosphatidyl glycerol was largest in quantity in the cortex. Eighteen kinds of fatty acids were identified in the neutral lipid, glycolipid and phospholipid fractions. Linoleic, palmitic, oleic and linolnic acids were the main components of fatty acids. The contents of saturated fatty acids, unsaturated fatty acids and essential fatty acids of each three fractions were different one another regardless of the Periderm, cortex and medulla.

  • For surveying of constituents concerning the flavor of the dried tomato fruits, nonvolatile components such as minerals, sugars, free amino acids, nucleotides and lipids were analyzed and determined. Potassium, calcium and phosphorus were the major conatituents of ashes, but trace amounts of cadmium and lead were determined. Glucose and fructose were the major constituents in sugars, and the latter slightly showed higher value than the former. The content of total free amino acids was 8322.5mg%, and that of the major components such as asparagine, aspartic acid, urea. glutamic acid, ${\beta}-alanine$ and ${\gamma}-aminobuthyric$acid showed 58.01% to the total amount. The nucleotides were composed of CMP, UMP, GMP and IMP, and CMP among them showed the highest value as about 58.07% to the total amount. On the other hand, IMP showed the lowest one. The major components of the total fatty acids from the saponifiable fraction in the lipids were $C_{18:2}$, $C_{18:1}$ and $C_{16:0}$ and those of the total sterols from unsaponifiable one were stigmasterol and ${\beta}-sitosterol$.

The Effects of Metathesis for Concentrating the Tocopherols from Soybean and Rice-bran Scum Oils (대두유 및 미강유의 찌꺼기유로부터 토코페롤의 농축을 위한 메타세시스 반응효과)

  • The effects of metathesis for concentrating the tocopherols from soybean and rice-bran scum oils were studied by using the batch reactor under helium atmosphere. The contents of tocopherols in the scum oils decreased consticuously when heated under air atmosphere or when kept in hexane solution above 5 days even at room temperature. The sterols in the scum oils were removed by the mixed solvent method. Metathesis of the sterol-removed scum oils in hexane was performed over Re2O7/Al2O3 and WO3/Al2O3 catalysts, and the concentrate was obtained by distillation in vacuum at $190^{\circ}C$. The effect of metathesis was evaluated as relative ratio of ${\alpha}$-tocopherol in the concentrate to that in scum oil. The maximum ratio for both scum oils was obtained on 12.8%(w/w)$Re_2O_7/Al2O_3$ catalyst which formed effectively the active sites for metathesis by the reaction between the added tetramethyltin and $Re_2O_7$ on the surface of the catalyst. The optimum amount of the catalyst was 0.5g pre l0g scum oil, and the optimum reaction temperature was $25^{\circ}C$ for both scum oils. The metathesis was more effective in rice-bran scum oil than in soybean scum oil. These facts indicated that the tocopherols in the scum oils can be highly concentrated by applying metathesis.

Lipids and Fatty Acid Composition of Garlic (Allium sativum Linnaeus) (마늘의 지방질(脂肪質) 및 지방산(脂肪酸) 조성에 관한 연구)

  • Lipids extracted from two garlic samples (Allium sativum Linnaeus) were studied. Total lipid content of the samples ranged from 310 to 342mg/100g of fresh garlic. These lipids were shown to consist of $36.4{\sim}43.5%$ neutral lipids, $20.3{\sim}24.3%$ glycolipids, and $36.2{\sim}39.3%$ phospholipids. Among the neutral lipids, triglycerides were predominant $(80.5{\sim}83.6%)$ with smaller amounts of free sterols, free fatty acids, and sterol esters being present. 1, 2-Diacylglycerol acetates, 1, 3-diacylglycerol acetates and cholesterol acetates were also tentatively identified. Esterified steryl glycosides and steryl glycosides were major sugar-containing lipids, but monogalactosyl diglycerides, cerebrosides, digalactosyl diglycerides and sulfolipids were relatively minor components. Of the phospholipids, phosphatidyl cholines and serines, and phosphatidyl ethanolamines were the major components, comprising over 85% of this class. Phosphatidic acid and phosphatidyl inositols were also present. The major fatty acids in the total and three lipid classes were linoleic, palmitic, oleic and linolenic acid.

Sterol Composition and Phytoestrogen Activity of Safflower(Carthamus tinctorius L.) Seed (홍화(Carthamus tinctorius L.)씨의 sterol 및 Phytoestrogen 분석)

  • This study was done to investigated the phytosterol compositions of safflower (Carthamus tinctorius L.) seed. The phytoestrogen activity was also determined using CAT-ELISA Kit in ethanol extract of safflower seed. The phytosterol of safflower seeds was identified using gas chromatography-mass spectrometry after saponification of the oils. The phytosterol content and composition of safflower seed oils were 4% and identified stigmast-5-en-3-ol (3$\beta$, 24S)-form, ${\gamma}$-sitosterol (clionasterol) with Wiley MS spectrum library. The synergistic effect of human estrogen receptor (hER) has been investigated using a minimal chimeric promoters composed of the TATA region of the adenovirus-2 major late promoter (A22MLP) and two consensus perfectly polindromic Xenopus vitellogenin A2 gene estrogen responsive elements (XVEREl19). Transient transfection experiments in tile human breast adenocarcinoma cell line MCF-7, which is known to express the estrogen receptor endogenously, revealed that phytoestrogen from Carthamus tinctorius L. acts as estrogen. We have observed the transcriptional activities stimulated methanol and ethanol extract of safflower seed in MCF-7, were 0.43 and 0.37 respectively, compared to that by $\beta$-estradiol as 1.0. Our data showed that safflower seeds have estrogenic activity methanol and ethanol extracts and ethanol lower than that of $\beta$-estradiol. This result provides the first evidence that the beneficial effect of safflower seeds may be mediated, at least in part, by the stimulating effect of phytoestrogen ell bone-protecting.
