Stephania subpeltata H. S. Lo: 베트남 미기록종 (Stephania subpeltata H. S. Lo (Menispermaceae): A new record for the Flora of Vietnam)

  • Stephania subpeltata (방기과)의 베트남 분포를 처음으로 보고한다. 본 종은 형태적으로 S. japonica와 유사하지만 엽신이 방패모양에 가까운 난상삼각상 또는 넓은 삼각상이며, 엽병이 조금 더 짧고, 잎맥의 수가 적으며, 작은 화경에 산형모양의 취산화서가 달리지만 드물게는 복취산화서를 가지며, 꽃이 자색이고 내과피에 돌출된 열의 수가 적어 뚜렷하게 구별된다. 본 종의 분류학적 기재, 상세 분포 및 생태학적 정보와 함께 베트남에 분포하는 함박이속(Stephania) 식물의 종검색표를 제시하였다.

방기가 백서 중대뇌동맥 폐쇄에 의한 국소뇌허혈손상에 미치는 보호효과 (Protective Effects of Stephania tetrandra against Focal Cerebral Ischemic Damage by Middle Cerebral Artery Occlusion in Rats)

  • Objective : This study was performed to investigate the protective effect of Stephania tetrandra(ST) against ischemic brain damage after a middle cerebral artery(MCA) occlusion. The effect was evaluated using histological tests, neurobehavioral tests, and biochemical tests. Methods : Rats(Sprague-Dawley) were divided into four groups : sham operated group, MCA occluded group, post MCA occlusion Stephania tetrandra administrated (7.6mg/l00g) group, and normal group. The MCA was occluded by intraluminal method. Stephania tetrandra was administrated orally twice at 1 and 4 hours after MCA occlusion. The neurobehavioral test was performed at 3, 6, 9 and 24 hours after MCA occlusion by posture reflex test and swimming behavioral test. All groups were sacrificed then. The brain tissues were stained with 2% triphenyl tetrazolium chloride(TTC) or 1 % cresyl violet solution, to examine infarct size, volume and cell number. Tumor necrosis $factor-{\alpha}$ level was measured from sera using Enzyme-Linked Immunoabsorbent Assay(ELISA). The mRNA expression level of inflammatory cytokines and related receptor type I and II, $IL-1{\beta}$, IL-6, and IL-10 6hours after MCA occlusion were also studied by reverse transcriptase polymerase chain reaction(RTPCR). Results : The results showed that : Stephania tetrandra (1) reduced infarct size and total infarct volume by 52.2% compared to the control group; (2) attenuated significantly in neuronal death, which was shown by a decrease in cell number(P<0.01) and size(P<0.01) in the boundary area of the infarction; (3) significantly reduced serum $TNF-{\alpha}$ level, and increased the mRNA level of IL-10 in the cortex region(P<0.01). However, there was no significant effect on motor deficit in swimming behavioral test. Conclusions : In conclusion, Stephania tetrandra has protective effects against ischemic brain damage at the early stage of ischemia.

  • Stephania hernandifolia belonging to the family Menispermaceae is the biggest storehouse of diphenylbisbenzylisoquinoline (DBBI) alkaloids. Exhaustive chemical processing of the bulb of S. hernandifolia by the application of modern separation techniques yielded a DBBI alkaloid which was identified as cycleanine using spectroscopic methods (UV, IR, $^1HNMR$. $^{13}CNMR$, Mass). Cycleanine showed significant anti-inflammatory activity against carrageenin induced paw oedema, comparable to that produced by diclofenac sodium, used as standard drug. It exhibited potent analgesic effects against chemical and thermal noxious stimuli. It was also found to possess anticonvulsive activity in the strychnine induced convulsion model.

  • Stephania delavayi Diels. has been used as an immune activator or an anti-inflammatory drug in China. We examined the immune modulation effect and 7-days repeated-dose toxicity to validate its biological safety and efficiency. Mice were repeatedly administrated with 50 mg/kg S. delavayi Diels. daily by I.P for 7 days. S. delavayi Diels. induced B cell activation but had no effect on other immune cells such as T cell, natural killer (NK) cell, and macrophage ($M{\varphi}$). S. delavayi Diels.-treated group exhibited no statistical significance from the control group in physical conditions; body weight, complete blood count (CBC), serum biochemical indexes etc. There was no difference between the control group and S. delavayi Diels.-treated group in gross findings such as histopathological alteration. In conclusion, S. delavayi Diels. is safe above the dose of immune modulation.

목방기약침(木防己藥鍼)이 관절염(關節炎)에 미치는 영향(影響) (A Study on the Effect of Aqua-acupuncture with Stephania Tetrandra solution on Arthritis)

  • Objective : The purpose of this study is to observe the effect of Aqua-acupuncture with Stephania Tetrandra Solution (ST-AS) on arthritis. Methods : The author performde several experimental items : that isgene expression and secretion of IL-$1{\beta}$, IL-6, TNF-${\alpha}$, MMP-2, production of ROS, paw thickness, DTH, weight of spleen, expression of CD4+, CD8+, CD19+ in the spleen, production of IL-6, TNF-${\alpha}$, examination of histology. Results : The obstain results are summarized as follows. 1. IL-$1{\beta}$, IL-6, TNF-${\alpha}$ gene expression of hFLS were significantly inhibited in treatmentgroup, and gene expression of MMP-2 was not inhibited in treatmentgroup. 2. The secretion amount of IL-$1{\beta}$, IL-6, TNF-${\alpha}$ were significantly inhibited in treatmentgroup. 3. Expression of P-38 MAP kinase and production of ROS were inhibited in treatmentgroup. 4. Treatmentgroup were significantly inhibited the incidence of arthritis, hind paw edema, the index of arthritis and DTH of CIA (collagen II-induced arthritis) mice. 5. Treatmentgroup were significantly decreased splenetic weight and the number of CD4+, CD8+, CD19+ activated cells and secretion aroout of IL-6, TNF-${\alpha}$ of CIA (collagen II-induced arthritis) mice.. 6. Treatmentgroup were expressed form of new bone, synoviumin, new margine in histology imperison to controlgroup. Conclusions : Taking all these observations into account, ST-AS injection is considered to be effective in treating arthritis and put to practical use in future arthritis clinic.

Antifibrotic Effect of Stephania tetrandra on Experimental Liver Fibrosis Induced by Bile Duct Ligation and Scission in Rats

  • We examined the antifibrotic effect of a methanol extract from Stephania tetrandra (ST) on experimental liver fibrosis. liver fibrosis was induced by bile duct ligation and scission (BDL/S) in rats. In BDL/S rats, activity levels of aspartate transaminase (AST), alanine transaminse (ALT), alkaline phosphatase (ALP), concentration of total bilirubin in serum, and hydroxyproline content of the liver were significantly increased. The ST treatment (either 100 ${m}g/kg/day$ or 200 ${m}g/kg/day$, p.o. for 4 weeks) in BDL/S rats reduced the serum AST, ALT and ALP activity levels significantly (p<0.01). Similarly, when compared to the control group, the concentration of hydroxyproline in the livers of the BDL/S rats treated with 100${m}g$ or 200${m}g$ ST treated rats decreased by 40% and 33% respectively, when compared to the BDL/S control group (p<0.01). The morphological characteristics of fibrotic liver that were observed in the BDL/S control group, improved in the ST treated BDL/S group. In the fibrotic liver of BDL/S rats treated with ST, a marked reduction in the numbers of alpha smooth muscle cell actin positive stellate cells was observed. These results indicate that doses of either 100 or 200 ${m}g/kg/day$ of methanol extract from S. tetrandra, had an antifibrotic effect in rats with liver fibrosis induced by bile duct ligation and scission.

Genetic identification of Sinomenium acutum based on chloroplast gene ndhF sequences

  • Objectives : This study was conducted to identify the original Sinomini Caulis et Rhizoma plant among Stephania tetrandra, Cocculus trilobus, and Aristolochiae fangchi to develop the genetic marker for Sinomini Caulis et Rhizoma. Methods : Sinomenium acutum was identified by the classification and identification committee of the National Center for Standardization of Herbal Medicines. The chloroplast ndhF gene was amplified. We performed sequences alignment analysis of Sinomenium acutum, Stephania tetrandra, C. trilobus, and A. fangchi using BioEdit program. The SFR markers designed were consisted of SF01, SR04, and SR05 primers. Results : Many variations of Sinomeni Caulis et Rhizoma are currently commercialized as herbal medicine. We compared the base sequences of the ndhF intergenic space of chloroplast DNA with Sinomenium acutum, Stephania tetrandra, C. trilobus, and A. fangchi. According to the results, it showed that the nucleotide variations were seen in 30 genes of four species. Phylogenetic analysis revealed that 4 species were classified into five groups based on an inter-group divergence in nucleotide sequence of 9%. We developed SFR marker nucleotides enough to authenticate respective species and confirmed its application on the band size at 419 base pair. These sequence differences at corresponding positions were available genetic markers to identity the Sinomeni Caulis et Rhizoma. Conclusions : Base on these results, the ndhF region was effective in distinguishing Sinomini Caulis et Rhizoma The SFR genetic marker was useful for identifying Sinomini Caulis et Rhizoma with other species.

일문전(Stephania delavayi Diels.) 메탄올, 부탄올, 에틸아세테이트 분획물의 항산화 및 세포증식억제 효과 (Antioxidative Effects and Anti-proliferative Effects of MeOH, BuOH and Ethyl Acetate Fractionated from Stephania delavayi Diels)

  • 본 연구에서는 중국에서 급성 위장염에 대한 약제로 사용되어 오던 일문전의 메탄올 추출물을 이용하여 물, 에틸 아세테이트, 부탄올 층으로 분획한 뒤 항산화 활성과 항암활성을 측정하였다. DPPH 라디칼 소거능은 $1,000{\mu}g/mL$ 농도에서 부탄올(75.23%)> 메탄올(68.11%)> 에틸아세테이트 (63.58%)> 물(50.13%) 순으로 나타났으며 환원력의 경우 용매 분획물의 농도가 증가함에 따라 환원력이 증가하는 경향을 나타내었다. DPPH 라디칼 소거능 및 환원력 모두 부탄올 분획층에서 좋은 활성을 나타내었다. 항암활성 측정결과 메탄올 추출물과 부탄올 분획물, 에틸 아세테이트 분획물에서 MDA-MB-231 cell과 MCF-7 cell에 대한 세포증식억제효과를 나타내었다. 이상의 결과에서 일문전의 용매 분획물은 항암 후보물질로의 가능성이 있을 것으로 사료된다.

  • Tetrandrine is a bis-benzyl isoquinoline alkaloid derived from the root of Stephania tetrandra S. Moore. which was reported to elicit in vitro cytotoxic effect on HeLa cells and in vivo supprresive effects on mouse ascite tumor. Tetrandrine also induced apoptosis in a various cell lines. Recent studies have revealed that mitochondria has been shown to play an important role in the regulation of apoptotic processes. (omitted)

N-acetyl-${\beta}$-Dglucosaminidase(NAG). increase of urinary low molecular protein, ${\alpha}$1-microglobulin, ${\beta}$2-microglobulin. clara cell protein(CC16), retinol-binding protein(RBP) in clinical findings, and the proximal tubular atrophy, interstitial fibrosis. urothelial atrophy. glomerular sclerosis in histology, Because CHN was caused by Chinese herb contained in slimming preparation, western medical doctors have thought that all Herb medicine might have caused renal disease and prohibit the taking of any Herb medicine, However. CHN was actually caused by the aristolochic acid contained in some Herb medicines. Aristolochia manshuriensis, Aristolochia fang chi, which is the substitutions of Akebia quinata, Stephania tetrandra has being used in clinical. Aristolochia manshuriensis. Aristolochia fang chi were different with Akebia quinata. Stephania tetrandra in botany, and it have not been classified with medicines in Oriental medicine, That is, the aristolochic acid, not Herb medicines. causes CHN, So, Chinese Herb Nephropathy should be changed to Aristolochic acid Nephropathy.
