MicroRNAs (miRNAs) are endogenous, non-coding, small RNA molecules consisting of 21-24 nucleotides (nts) that regulate target genes at the posttranscriptional level in plants and animals. In plants, miRNAs negatively regulate target mRNAs containing a highly complementary sequence by either mRNA cleavage or translational repression. MiRNAs are processed from single-stranded precursors containing stem-loop structures by a Dicer-like enzyme and are loaded into silencing complexes, where they act on target mRNAs. Although plant miRNAs were first reported in Arabidopsis 10 years later than animal miRNAs, numerous miRNAs have since been identified from various land plants ranging from mosses to flowering plants, and their roles in diverse aspects of plant developmental processes have been characterized. Furthermore, most of the annotated plant miRNAs are evolutionarily conserved in various plants. In particular, recent functional studies using Arabidopsis mutants have contributed a great deal of information towards establishing a framework for understanding miRNA biogenesis and functional roles. Extensive appraisal of miRNA-directed regulation during a wide array of plant development and plant responses to environmental conditions has confirmed the versatile roles of miRNAs as a key component of plant molecular biology.

E.coli methionyl tRNA synthetase consist of 676 amino acids and plays a key role in initiation of protein synthesis. The native form of this enzyme is a homodimer, but the monomeric enzyme truncated approximately C-terminal 120 amino acids retains the full enzymatic activities. X-ray crystal structure of the active monomeric enzyme shows that it has two domains. The N-terminal domain is thought to be a binding site for acceptor stem of tRNA, ATP, and methionine. The C-terminal domain is mainly a-helical and makes an interaction with the anticodon of $tRNA^{Met}$. Especially it is suggested that the region of helix-loop-helix including the tryptophan residue at the position 461 may be the essential for the interaction with anticodon of $tRNA^{Met}$. In this work the structure and function of E. coli methionyl-tRNA synthetase was studied by spectroscopic method (NMR, CD, Fluorescence). The importance of tryptophan residue at the position 461 was investigated by fluorescence spectroscopy. Tryptophan 461 is expected to be an essential site for the interaction between E. coli methionyl-tRNA synthetase and E. coli $tRNA^{Met}$. Proton and heteonuclear 2-dimensional NMR spectroscopy were also used to elucidate the protein-tRNA interaction.
The age-46.5 gene of Escherichia coli is induced by nitrate ion and regulated by Fnr, NarL, and NarP during anaerobic growth. aeg-46.5::lacZ fusion gene shows its maximum expression in narL host after two hours of aerobic to anaerobic switch in M9-Glc-nitrate medium. Fnr and NarP act as positive regulators, and NarL acts as a negative regulator. The control region of the aeg-46.5 was identified and the binding sites of regulator proteins have been predicted (Reznikoff and Choe (1993)). It has two symmetry regions. One is located at -52~-37 bp from the anaerobic mRNA 5'-end, which is the binding site of NarL and NarP. The other is located at +37~+56 bp from the 5'-end of mRNA. In this study, the downstream symmetry region from the mRNA 5'-end was investigated by site-directed mutagenesis. The destruction of the symmetry region increases the expression level of aeg-46.5. We propose that the symmetry region interferes with the expression of aeg-46.5 possibly by forming a stem-and-loop structure.
$Ni(II){\cdot}Gly$-Gly-His(Arg)COOH와 $Cu(II){\cdot}Gly$-Gly-His(Arg)COOH 형태의 금속펩타이드를 이용하여 P. alcaligenes에서 얻은 5S rRNA의 구조를 조사하였다. 그 결과 금속 펩타이드들은 5S rRNA의 줄기-고리 구조에서 염기쌍을 이루지 않거나 불안정하게 이루는 부분을 선택적으로 변형시켰다. 금속펩타이드의 선택성은 중심 금속이 Ni(II)인 경우와 Cu(II)인 경우에 차이가 거의 없었다. 금속펩타이드를 이용한 절단 결과를 금속 착물 M(II)CR을 이용한 결과와 비교하면 금속펩타이드에 의한 선택성이 더 크게 나타났다. 금속펩타이드와 금속착물을 이용한 절단 결과로부터 P. alcaligenes에서 얻은 5S rRNA의 이차구조를 살펴보았다.
Since molecular structure of hnRNP is not available in foreseeable future, it is best to construct a working model for hnRNP structure. A geometric problem, assembly of $700{\pm}20$ nucleotides with 48 proteins, is visualized by a frame work in which all the proteins participate in primary binding, followed by secondary, tertiary and quaternary binding with neighboring proteins without additional import. Thus, 40S hnRNP contains crown-like secondary structure (48 stemloops) and appearance of 6 petal (octamers) rose-like architectures. The proteins are wrapped by RNA. Co-transcriptional folding for RNP fibril of FMR1 gene can produce 2,571 stem-loops with frequency of 1 stem-loop/15.3 nucleotides and 53 40S hnRNP beaded structure. By spliceosome driven reactions, there occurs removal of 16 separate lariated RNPs, joining 17 separate beaded exonic structures and anchoring EJC on each exon junction. Skipping exon 12 has 5'GU, 3'AG and very compact folding pattern with frequency of 1 stem-loop per 12 nucleotides in short exon length (63 nucleotides). 5' end of exon 12 contains SS (Splicing Silencer) element of UAGGU. In exons 10, 15 and 17 where both regular and alternative splice sites exist, SS (hnRNP A1 binding site) is observed at the regular splicing site. End products are mature FMR-1 mRNP, 4 species of Pri-microRNAs derived from introns 7,9,15 and 3'UTR of exon17, respectively. There may also be some other regulatory RNAs containing ALU/Line elements as well.
Xanthomonas palargonii 5S rRNA의 일차구조 및 이차구조를 결정하고 에틸니트로소 우레아, Pb2+, 황산 이메틸 , 피로탄산 이에틸 들의 화학 탐침과 몇 가지 효소 탐침을 사용하여 고차원 구조를 분석하였다. 에틸니트로소 우레아는 삼차 상호작용에 관련되는 포스포디에스테르 결합을 조사하는데 사용되었다. Mg2+이 있을 때 에틸니트로소 우레아에 의한 변형에 대해서 저항성이 있는 자리는 불안정한 d 나선의 Nucleotides G72, A73, G75, A78, G98, G100, A101 들, c고리의 C36, C37, C39, C41들, 그리고 C 줄기의 A29, G33 들이다.이러한 결과와 Pb2+에 의한 가수분해 반응과 화학 탐침과 효소 탐침을 사용하여 얻은 결과들을 종합해 봄으로써 5S 의 b-C 구역과 d 나선 구역은 5S rRNA의 삼차 상호작용에서 돌쩌귀의 구실을 할 것으로 추정할 수 있었다.
구제역바이러스(FMDV)는 피코나바이러스 과에 속하는 바이러스로서 야생과 가축화된 소와 돼지에 감염된다. FMDV는 제어되기 어려워서 가축의 생산과 국제통상에 큰 장애가 되고 있다. FMDV RNA 게놈의 복제 과정에서 3D 중합효소가 특이적인 복제 기능을 담당하는데 게놈에 결합하는 부위가 매우 중요하다. 이 사실은 FMDV 게놈의 비코딩영역 내에서 3D 중합효소에 의해 인지되는 특이 RNA 구조가 관여함을 제시한다. 이 과정에서 cis-acting replication element (CRE)는 RNA 복제를 위한 개시에 필요하다. FMDV CRE는 15-17 뉴클레오티드의 고리와 이를 지지하는 이중가닥으로 형성된 줄기-고리 모양을 가지며 이들을 구성하는 RNA 뉴클레오티드 서열의 차이가 다른 RNA 이차구조를 생성한다. CRE 이외에 FMDV 복제를 위해서 많은 바이러스와 세포 인자들이 단백질-단백질 결합과 단백질-RNA 결합을 통해 협조적인 네트워크를 만들어낸다. 이 연구에서 국내에서 2010년부터 2017년 까지 구제역이 발생한 소와 돼지에서 FMDV를 분리하여 CRE 서열을 분석하였으며 이들 FMDV들은 A형과 O형의 유전자형을 가졌다. 흥미롭게 국내 FMDV들의 CRE RNA 이차구조의 변이들은 바이러스 간의 계통유전학적 상관관련성과 일치하며 특정 숙주 동물종의 FMDV 감염의 특이성을 밝혀주었다. 이를 토대로 국내 FMDV의 각 유전군의 분류는 독특한 기능적 CRE에 의해 특징지을 수 있으며 새로운 유전적 계통의 진화학적 연속성은 특징있는 CRE 모티프의 구분과 연관지을 수 있다. 그러므로 CRE의 특이적 RNA 구조는 숙주 동물종 의존적인 FMDV 부류의 부가적인 단서로 활용할 수 있음을 제안한다. 이들 연구 결과들은 향후 FMDV 대량감염이 발생할 때 숙주동물종의 특이성을 CRE 서열로 조기에 정확히 분석하는데 도움이 될 것이다.
Three isolates of Cucumber mosaic virus (CMV) from lily plants showing mosaic and distortion symptoms were detected by reverse-transcriptase polymerase chain reaction (RT-PCR) using primers specific to Cucumovirus genus namely, LK-CMV, LK4-CMV, and LKS-CMV. Restriction enzymes patterns of the RT-PCR products revealed that the lily isolates belonged to subgroup IA of CMV. In terms of biological properties, the lily isolates have highly similar but distinct pathogenicity as reported in other lily strains and ordinary strains of CMV. To characterize the molecular properties, cDNAs containing coat protein (CP) gene and 3' non-coding region (NCR) of RNA3 for the isolates were cloned and their nucleotide sequences were determined. The CP similarity (218 amino acids) was highly homologous (>97%) with that of subgroup I CMV strains. However, an additional 20-nulcleotide long segment was only present in 3' NCR of lily isolates, which form an additional stem-loop RNA structure. By using chimeric construct exchange cDNA containing 3'NCR of LK-CMV into the full-length cDNA clone of RNA3 of Fny-CMV, this additional segment may prove to be significant in the identification and fitness of the virus in lily plants. The pathology of zucchini squash infected by F1F2L3-CMV, a pseudorecombinant virus was showed to change drastically the severe mosaic and stunting symptom into a mild chlorotic spot on systemic leave, compared with Fny-CMV. To delimit the sequence of RNA3 affected the pathology, various RNA3 chimeras were constructed between two strains of CMV. The symptom determinants of F1F2L3-CMV were mapped to the positions amino acid 234, 239, and 250 in 3a movement protein (MP). RNA3 chimeras changed the sequences encoding three amino acids were resulted in alteration of systemic symptom.

Most follicles in the mammalian ovary undergo atresia. Granulosa cell apoptosis is a hallmark of follicle atresia. Our previous study using a microRNA (miRNA) microarray showed that the let-7 microRNA family was differentially expressed during follicular atresia. However, whether the let-7 miRNA family members are related to porcine (Sus scrofa) ovary follicular apoptosis is unclear. In the current study, real-time quantitative polymerase chain reaction showed that the expression levels of let-7 family members in follicles and granulosa cells were similar to our microarray data, in which miRNAs let-7a, let-7b, let-7c, and let-7i were significantly decreased in early atretic and progressively atretic porcine ovary follicles compared with healthy follicles, while let-7g was highly expressed during follicle atresia. Furthermore, flow cytometric analysis and Hoechst33342 staining demonstrated that let-7g increased the apoptotic rate of cultured granulosa cells. In addition, let-7 target genes were predicted and annotated by TargetScan, PicTar, gene ontology and Kyoto encyclopedia of genes and genomes pathways. Our data provide new insight into the association between the let-7 miRNA family in granulosa cell programmed death.
Cdc25B is a mitotic regulator that might act as a starter phosphatase to initiate the positive feedback loop at the entry into mitotic (M) phase. In the present study, distribution of Cdc25B mRNA in duodenal mucosa of the chicken was demonstrated by means of in situ hybridization histochemistry (ISHH) using sense and antisense digoxigenin (DIG)-labeled RNA probes. The results showed that there were many labeled cells distributing in the duodenal mucosa of the adult chicken. Of these labeled cells, 81.60${\pm}$9.63% of Cdc25B mRNA positive cells was distributed in the basilar part and mid-portion of the intestinal gland and 36.21${\pm}$8.81% in the middle and basilar portion of villi of the small intestine of the chicken, respectively. Most of these labeled cells were positive in the regions of the stem cell and proliferation. The signals of ISHH decreased from basilar to upper part in the crypt of Lieberkuhn and weakened in the inferior villi of the duodenum. Moreover, the positive signals were both in the cytoplasm and cell nucleus. However, the labeled cells were negative in both the lamina muscularis mucosae and muscular layer. The results of ISHH suggested the existence of Cdc25B mRNA and vigorous proliferation activities in the duodenal mucosa of adult chicken, replenishing the cells which had sloughed off from the superior part of the villus. Our results provide some molecular evidence for a regular pattern of avian intestinal epitheliosis and functional partition and provide an approach to further study of the locations of Cdc25B in the chicken.
