Effect of Stem Number and Propagation Method on Yield Components of Boxthorn(Lycium chines Mill.) (주당 적정줄기수와 번식방법이 구기자의 수량형질에 미치는 영향)

  • Experiments were conducted to investigate the proper stem number and the propagation method of Lycium Chinense Mill. from 1993 to 1994.The results obtained were summerized as follows. The most effective propagation method was stem cutting 20cm in which flowering time was 15days earlier and dry fruit weight was 30 percent heavier(91kg/10a) than in seed propagation. The optimal diameter of cutting was 0.9cm of which dry fruit weight was 227kg/10a and 28 percent heavier compared with that of 0.5cm. stem number 5 had the best top growth and the heaviest 100 fruit weight as 13.4g. Fruit number per plant was increased as 864 in stem number 5. Dry fruit yield of stem number 5 was 104kg/10a and 16 percent increased compared with that of control.

  • Since 1972 a series of experiments were conducted to reveal physiological function and biological activities involved in rooting of mulberry cuttings, and the behaviour of ether extractable growth control substances in leaves and stens. Measurements were made on various mulberry varieties for respiration of cuttings, suitable size for the production of cuttings, change of rootability of cuttings with growth of cutting shoots after cutting date, rooting tests on the green beans with ether leaf extract and ether stem extract, and rooting effect of cutting by NAA treatment.   • Ligusticum chuanxiong has been an important oriental crude drug and cultivated for a long time in Korea. Ligusticum chuanxiong has been vegetatively propagating crop without seed due to its sterility and would be an allodiploid derived from the hybridization of two allied species. Generally, vegetative propagation is more costly per unit propagule than sexual(seedling) propagation. This study was conducted to find out the techniques of mass propagation by stem cutting for reduced seedling cost. When cutted on August 30 with 1st to 2nd internode, its highest rooting percentage was 81 percent. Rhizome yield of plants cutted in 50 percent on September 1 decreased to 9 percent less than control (rhizome propagation). Rooting percentage of rooted cutting plants after field planting on October 25 was the highest being 87 percent. Rhizome yield with planting appeared 127 and 120kg/10a on September 25 and October 25, respectively.

Clonal Propagation in Commiphora Wightii (Arnott.) Bhandari

  • Studies were carried out to standardize and develop a suitable macro-propagation technology for large scale production of superior clonal stock through stem cuttings in Commiphora wightii Arnott (Bhandari), a data deficient medicinal plant of arid region. For the purpose, three experiments were conducted. The first experiment was tried to elucidate the impact of various cutting diameters (0.50-0.75 cm, 0.75-1.00 cm, 1.00-1.50 cm, and >1.50 cm) in combination with varying growing conditions (sunlight, shade house and mist chamber) on shoot sprouting and rooting without using exogenous plant growth regulators. Cutting diameter (size 0.75-1.00 cm) in mist chamber has shown maximum sprouting (90.00%) and rooting (73.33%), primary root (6.67) and secondary root (16.67) followed by 1.00-1.51 cm in mist chamber. Minimum sprouting (40.00%), rooting (33.33%), number of shoot (1.33), primary root (1.00) and number of secondary root (1.00) was recorded in cutting diameter (size >1.50 cm) in sunlight. Second experiment was performed to find out optimum growth regulator concentration of rooting hormone (100, 200, 500 and 1000 ppm) of Indole-3-acetic acid (IAA) and Indole-3-butyric Acid (IBA) on adventitious root formation on cuttings diameter (size 0.25-0.50 cm) in comparison to control. Maximum rooting percentage (93.33%) was recorded in 200 ppm followed by 500 ppm (86.66%) of IBA as compared to control, which showed only 60 per cent sprouting. Third experiment was performed with newly formed juvenile micro-cuttings treated with varying concentrations of IAA and IBA. The juvenile cuttings (size 6-10 cm, basal dia <0.25 cm) were selected as micro-cuttings. The cuttings treated with IBA (500 ppm) showed 64.30% rooting as compared to other treatments. Results of above experiments indicate that cuttings (size 0.75-1.00 cm dia) may be developed in mist chamber for better performance. While using heavier cuttings, no growth promoting hormones is required however; growth regulator 200 ppm concentration of IBA rooting hormone was observed optimum for promoting macro-propagation in stem cuttings of lower diameter class (0.25-0.50 cm).

  • To produce and supply the healthy potato, basic seed potatoes were produced by stem cutting, microtuberization and hydroponic culture. The total number of tubers and the total tuber weight per $\textrm{m}^2$ of potato were more in hydroponic culture as each products were 1, 152 and 4, 492g than in the stem cutting (75 and 4, 136g) or microtuberization (1, 080 and 1, 080g) using petridishes. The total yield per 10a in the field was propagated highly stem cutting > propagated microtubers > hydroponics > microtubers. The number of tubers per 10a produced by hydroponics (33, 064) was higher than any other methods. This indicated the hydroponic culture can be used in the multiplication of basic seed potatoes.

Quality Change of Garlic during Storage by Stem and Root Cutting Treatments (마늘의 줄기 밑 뿌리절단에 따른 저장 중 품질변화)

  • Physicochemical properties of garlic bulb during storage with different cutting treatments were estimated. Garlic harvested in early June was used in this experiments. Various physicochemical factors of garlic bulb such as weight loss rate, rotten rate, sprouting rate, moisture content, reducing sugar content and total pyruvate content were investigated. Bulbs with roots, bulb without roots and bulb with stem length of l cm, 3 cm, 5 cm were stored at 2$\^{C}$ for eight months. The weight loss increased sharply after 8 months for all treatments. Weight loss occurred most severely in treated garlics with bulbs with 5 cm stem length and progressed steadily at low rates in bulbs with 1 cm stem length. Also, less decay and internal sprouting were observed in bulbs with 1 cm stem length. Cutting treatments of roots were not significant for internal sprouting. Incidence of other chemical properties of bulbs, contents of total sugar, reducing sugar and pyruvic acid were not significant statistically. Moisture contents of treated garlics was remarkably reduced in longer leaving stems. As a result, top-clipped leaving stems fur long, seemed to have a beneficial effect on physicochemical properties of garlic stored at 2$\^{C}$ for 8 months possibly due to reduced weight loss, rotten decaying and sprouting rates. Utilization of such results in the processing industry can be effective.

Effect of Subirrigation of Dolrido on the Growth of Rose (토양 미생물제제인 돌리도의 관주처리가 장미의 생육에 미치는 영향)

  • This experiment was carried out to investigate the effect of subirrigation of Dolrido on the growth and development of stem cutting and young roes plant. Plant growth was measured at 20, 40, and 60 days after subirrigation of Dolrido. Plant height, stem diameter, number of leaves, leaf area and root length of cutting rose subirrigation were more affected than those of control. The growth of young seedling was significantly affected by subirrigation of Dolrido. Plant height, number of leaves, stem diameter, leaf area, fresh and dry weight were increased with subirrigation of Dolrido. However, T/R ratio was not affected by subirrigation of Dolrido. Growth of seedlings was promoted after 60 days treatment of Dolrido.

Effect of Nutrient Concentration and Plant Growth Regulators on Rooting in Coleus Stem Cuttings (코레우스의 삽목시 배양액과 생장조절제처리가 발근에 미치는 영향)

  • The effect of nutrient concentration and plant growth regulators on rooting of Coleus stem cuttings were investigated. In contrast to sand cutting, the nutrient solution or plant growth regulator treatment increased the root numbers and root length. The concentration of nutrient solution were 1, l/2 and 1/4 strength of the balanced nutrient solution developed by Japanese Horticultural Experimental Station and the highest root growth was obtained from 1/4 strength nutrient concentration. NAA or IBA treatment in culture solution stimulated the rooting and 0.01mg/l of NAA or IBA was effective in root development. Furthermore, days for rooting were shortened and root growth were promoted by mixing nutrient solutions with plant growth regulators and by using cutting with leaves. The rooting of Coleus was observed under microscope and the adventitious root formation initiated at 3 days after cutting with nutrient and auxin mixture.

Effect of BA and Physical Treatment on Axillary Bud Emergence of Hosta spp. (옥잠화(Hosta)속 식물의 액아 발생에 미치는 BA 및 물리적 처리의 효과)

  • This study was conducted to investigate the effects of BA, a growth regulator on enhancing propagation efficiency, and physical treatment on promotion of axillary bud generation in three hosta cultivars, 'Ellerbroek', 'Francee', and 'Halcyon'. These are cultivars suitable for cut foliage production among Hosta spp. because of their diverse leaf patterns and long vase life of cut foliage. The degree of axillary bud generation by treatment time and concentration of BA has not been proven to be extraordinarily effective as compared to not treated control. As a physical treatment, base wounding was more effective than stem cutting. In 'Ellerbroek', increase of axillary bud generation among treatment methods was not great. However, in 'Halcyon', base wounding had four more axillary buds per plant than not treated control. In addition, although number of axillary buds increased per plant by BA treatment and stem cutting was 2 and 1.8, respectively, it was 6.1 by base wounding, indicating the greatest effect on promotion of axillary bud generation among all treatments.

Studies on the Conditions of Softwood Cutting for Production of Mulgerry Sapling (뽕나무 묘목생산을 위한 신소삽목 조건 구명)

  • Most of the mulberry saplings in Korea have been produced by grafting. But the production of mulberry sapling by this method is very complex process and requires high level of technique and two years to produce saplings. Therefore, to develop the mulberry sapling production system by softwood cutting method which is suitable for mass production, several conditions on rooting and growth of cuttings were investigated. The rooting ability of cuttings varied according to mulberry varieties, showing the highest rooting ratio of 93.3% in cultivar of Shingwangppong. Of different soil texture as for rooting media, the clay was found to be the best; other media decreased in order as follows; sand, sand loam and vermiculite. The shading ratio by 70% with polyethylene film showed the highest rooting ratio of the cuttings. Optimum day of cutting was around 50th day after sprouting, whereas the ratio of stem thicker than 7mm at the base of saplings in diameter was highest at 44th day.
