First Report on Pink Rot of Sunflower (Helianthus annuus) Caused by Trichothecium roseum in Korea (Trichothecium roseum에 의한 해바라기 분홍빛썩음병 발생 보고)

  • Pink rot appeared in greenhouse-grown sunflower (Helianthus annuus L.) in Jeonju, Korea. The symptoms appeared as brown discoloration of sunflower head and progressed into stem in advanced stage. In order to investigate the causal organism of this disease, we isolated a fungus from the infected seeds and maintained the isolated fungal culture on potato dextrose agar medium. Conidiophores were simple or branched, 62.5 to $123.1{\mu}m$ long. Conidia were produced in basipetal chains, ellipsoidal to pyriform with oblique and prominent truncate basal scars, two-celled, hyaline and measured $10.2-21.4{\times}7.5-12.6{\mu}m$. The fungus was inoculated to a new sunflower plant and showed the typical blight on the leaves. Study of morphological characters, pathogenicity tests and sequence analysis revealed that the isolated fungus is confirmed to be Trichothecium roseum. To our knowledge, this is the first report of pink rot on sunflower caused by T. roseum in Korea.

Scytalidium parasiticum sp. nov., a New Species Parasitizing on Ganoderma boninense Isolated from Oil Palm in Peninsular Malaysia

  • A mycoparasite, Scytalidium parasiticum sp. nov., isolated from the basidiomata of Ganoderma boninense causing basal stem rot of oil palm in Johor, Malaysia, is described and illustrated. It is distinct from other Scytalidium species in having smaller asci and ascospores (teleomorphic stage), longer arthroconidia (anamorphic stage), hyaline to yellowish chlamydospores, and producing a fluorescent pigment. The phylogenetic position of S. parasiticum was determined by sequence analyses of the internal transcribed spacers and the small-subunit ribosomal RNA gene regions. A key to identify Scytalidium species with teleomorphic stage is provided.

Detecting Seed-borne Fungi of Sesame and Virulance of Corynespora cassicola (참깨 종자전염진균(種子傳染眞菌)의 검정(檢定) 및 Corynespora cassicola의 병원성(病原性))

  • Seed samples of sesame (Sesamum indicum L.) from Chung-nam Provincial Office of Rural Development were infected by 12 fungi. Alternaria sesamicola, A. tenuis and Corynespora cassicola were the predominant fungi. Except C. cassicola, all fungi were almost completly reduced and wipid out the infections by pretreatment with chlorine. Plating of seed components also indicate that C. cassicola was well-established infections. In infection experiment C. cassicola produced severe leaf and stem spots and blights on sesame plants and showed severe seed and seedling rot.

Survey on Potato Parasitic Nematodes (감자 증산을 위한 기생선충 조사)

  • Survey on potato parasitic nematodes has been undertaken in order to find distribution of the nemic fauna. 41 soil samples were taken from potato fields in Gyeongbug, Gyeongnam and Gangweon provinces. Twenty four species belonging to 16 different genera were identified. Potato cyst nematode, Globodera rostochiensis was not found in the areas. Potato-rot nematode, Ditylenchus destructor and stem nematode, Ditylenchus dipsaci were found from several potato fields and population density high and showed damage to the crops. Root-lesion nematode, Pratylenchus minyus, Pratylenchus penetrans, Pratytenchus thornei and Pratylenchus vulnus were found and their population of these four species were high depending on the fields. Root-knot nematode, Meloidogyne hapla and Meloidogyne incognita were found. M. hapla was found only in Gangweon province and the population density was high. M. incognita was found at Milyang in Gyeongnam province. Spiral nematode, Heticotylenchus digonichus, Helicotylenchus dihystera, Helicotylenchus pseudorobustus, Rotylenchus orientalis and Rotylenchus pini were found. Aphelenchoides saprophilus, Criconemoides informis, Ditylenchus destructor, D. dipsaci, Helicotylenchus digonichus, H. dihystera, Hemicriconemoides intermedius, Meloidogyne hapla, Psilenchus hilarulus, Pratylenchus minyus, and Xiphinema americanum were first found from potato fields in Korea.

Studies on Simple Hydroponic Culture in Cultivation of Grafted Cactus for Export (수출용 접목선인장 재배시 간이양액재배 연구)

  • This study was carried out to investigate the effect of simple hydroponic culture in cultivation of grafted cactus. The results obtained are as follows: Growth indexes of grafted cactus cultivars, such as scion diameter, fresh weight, and scion hardness were improved in simple hydroponic culture compared with soil culture regardless of cultivars. Stem rot and rate of non-graft take were lower in simple hydroponic culture than in soil culture, but no significant difference was monitored between cultivars. Time for harvesting was fast in simple hydroponic culture compared with soil culture. Marketable yield increased in simple hydroponic culture.

A New Soybean Cultivar "Gaechuck#1" : Black Soybean Cultivar with Lipoxygenase2,3-free, Kunitz Trypsin Inhibitor-free and Green Cotyledon

  • Lipoxygenase and Kunitz trypsin inhibitor protein of mature soybean [Glycine max (L.) Merr.] seed are main anti-nutritional factors in soybean seed. A new soybean cultivar, "Gaechuck#1" with the traits of black seed coat, green cotyledon, lipoxygenase2,3 and Kunitz trypsin inhibitor protein free was developed. It was selected from the population derived the cross of "Gyeongsang#1" and C242. Plants of "Gaechuck#1" have a determinate growth habit with purple flowers, brown pubescence, black seed coat, black hilum, oval leaflet shape and brown pods at maturity. Seed protein and oil content on dry weight basis have averaged 39.1% and 16.2%, respectively. It has shown resistant reaction to soybean necrosis, soybean mosaic virus, Cercospora leaf spot and blight, black root rot, pod and stem blight, and soybean pod borer. "Gaechuck#1" matured on 5-10 October with a plant height of 50 cm. The 100-seed weight of "Gaechuck#1" was 23.2g. Yield of "Gaechuck#1" was averaged 2.2 ton/ha from 2005 to 2007.

A New Soybean Cultivar "Gaechuck#2": Yellow Soybean Cultivar with Lipoxygenase2,3-free and Kunitz Trypsin Inhibitor-free

  • Lipoxygenase and Kunitz trypsin inhibitor protein are the main antinutritional factor in mature soybean seed. A new soybean cultivar, "Gaechuck#2" with yellow seed coat, lipoxygenase2,3-free and Kunitz trypsin inhibitor protein-free was developed. It was selected from the population derived from the cross between "Jinpumkong2ho" and C242. Plants of "Gaechuck#2" have determinate growth habit with purple flowers, tawny pubescence, yellow seed coat, yellow hilum, oval leaflet shape and brown pods at maturity. Seed protein and oil content on a dry weight basis were 40.7% and 18.7%, respectively. It has shown a resistant reaction to soybean necrosis, soybean mosaic virus, Cercospora leaf spot and blight, black root rot, pod and stem blight, and soybean pod borer. Gaechuck#2 matured in 4 October with plant height of 54cm and a 100-seed weight of 24.4g. Average Yield of Gaechuck#2 was 230 - 250 kg/10a in 2005 - 2007.

Multiplex TaqMan qPCR Assay for Detection, Identification, and Quantification of Three Sclerotinia Species

  • White mold (or Sclerotinia stem rot), caused by Sclerotinia species, is a major air, soil, or seed-transmitted disease affecting numerous crops and wild plants. Microscopic or culture-based methods currently available for their detection and identification are time-consuming, laborious, and often erroneous. Therefore, we developed a multiplex quantitative PCR (qPCR) assay for the discrimination, detection, and quantification of DNA collected from each of the three economically relevant Sclerotinia species, namely, S. sclerotiorum, S. minor, and S. nivalis. TaqMan primer/probe combinations specific for each Sclerotinia species were designed based on the gene sequences encoding aspartyl protease. High specificity and sensitivity of each probe were confirmed for sclerotium and soil samples, as well as pure cultures, using simplex and multiplex qPCRs. This multiplex assay could be helpful in detecting and quantifying specific species of Sclerotinia, and therefore, may be valuable for disease diagnosis, forecasting, and management.
