Anti-hyperlipidemic Effect of Vitex doniana Ethanol Extract in Poloxamer Induced Hyperlipidemia

  • To determine the effect of Vitex doniana (leaves stem and root bark) ethanolic extracts on lipid profiles of Poloxamer 407 (P407) induced hyperlipidemic and normal rats. Fifty four mixed sex rats weighing 100-200g were divided into nine groups comprising six animals per group. At the end of the 21 day, the animals were sacrificed and blood sample were collected for determination of serum levels of: Total cholesterol (TC), Triacylglycerides (TAG), High-density lipoprotein cholesterol (HDL-c) and Low-density lipoprotein cholesterol (LDL-c). The studies showed that all induced treated groups significantly (P<0.05) lower serum levels of TC, TAG, LDL-c and significantly (P<0.05) increased HDL-c when compared to the P407 induced hyperlipidemic control. The normal treated groups showed no significant (P>0.05) difference in the serum levels of TC, TAG, LDL and HDL when compared to the normal control group. Calculation of atherogenic risk predictor indices of the induced treated groups showed that all the extracts significantly (P<0.05) lowered the LDL-c/HDL-c, log (TAG/HDL-c) and significantly (P<0.05) increased HDL-c/TC ratio when compared to the P407 induced hyperlipidemic control group. The atherogenic risk predictor indices of normal treated groups showed no significant difference (P>0.05) in LDL-c/HDL-c, Log (TAG/HDL-c) and HDL-c/TC ratio when compared to the normal control group. The study demonstrates the phytotherapeutic effect of Vitex doniana (leaves, stem and root bark) ethanolic extract in poloxamer 407 induced hyperlipidemia.

Alterations of Antioxidant Status and Mitochondrial Succinate Dehydrogenase Activity in the Liver of Wistar Strain Albino Rats Treated with by Ethanol Extracts of Annona senegalensis Pers (Annonaceae) Stem Bark

  • Numerous ethnomedicinal uses have been attributed to different parts of Annona senegalensis (ASE), including its uses as food and food additives. The present study investigated toxicological and antioxidant effects of 28 days administration of ethanol extracts of ASE stem bark to Wistar strain albino rats. Acute toxicity test was done to determine lethal dose in Wistar rats while sub-acute toxicity test was conducted on rats divided into four groups (A - control, B - 50 mg/kg, C - 100 mg/kg, D - 150 mg/kg, respectively and treated for 28 days. Oxidative stress markers in liver and kidney as well as hepatic succinate dehydrogenase activity in the mitochondrial and post mitochondrial fractions (PMF) were evaluated. The $LD_{50}$ value of ASE was > 2,000 mg/kg. White blood cell counts gradually increased, but red blood cell counts and haematocrits level decreased significantly (p < 0.05) by about 50%. Liver enzymes in the serum and mitochondrial succinate dehydrogenase activity increased significantly (p < 0.05). Superoxide dismutase and catalase activities also increased in liver mitochondria and PMF while malondialdehyde (MDA) and reduced glutathione levels increased only in the PMF. Furthermore, only MDA levels increased significantly in the kidney after 28 days extract administration. Histopathological examination showed hepatic necrosis and no obvious signs of nephrotoxicity. Anona senegalensis is relatively safe, but prolonged ingestion could induce oxidative stress and impair ATP synthesis through the modulation of the activity of mitochondrial succinate dehydrogenase.

소나무속(屬) 수목의 부위별 추출물의 항균활성 (Antifungal Activities of Extracts from the Various Parts of the Genus Pinus Trees)

  • 소나무속의 3수종, 소나무, 리기다소나무 및 잣나무의 부위별 추출물을 병원진균인 Colletotrichum gloeosporioides를 대상으로 항균활성을 조사하였다. 위 균주에 대하여 가장 우수한 항균활성은 잣나무 수피 및 근피 EtOAc 분획분에서 나타났는데 5.0 mg/l 농도에서 각각 98.8%와 100%로 조사되었다. 한편 두 분획분의 위 균주에 대한 $ED_{50}$(median effective dose)는 각각 469 ${\mu}g/ml$및 588 ${\mu}g/ml$로 조사되어 수피 추출물이 보다 낮은 값을 나타내었다. 잣나무 수피 EtOAc 분획분의 다른 병원진균 Alternaria brassicicola 및 Fusarium oxysporum에 대한 $ED_{50}$는 각각 533 ${\mu}g/ml$ 및 2,277 ${\mu}g/ml$로 나타나 이 분획분의 항균활성은 F. oxysporum보다 C. gloeosporiodes나 A. brassicicola이 더 우수한 것으로 조사되었다. 한편 잎 추출물의 경우를 보면, 소나무 잎 EtOAc 분획분은 C. gloeosporiodes에 대하여 5.0 mg/l 농도에서 39.6%의 활성을 보였으나 다른 두 수종의 잎 추출물에서는 추출 용액에 상관없이 거의 활성이 나타나지 않았다. 현재 주요 항균활성 화합물들은 분리, 동정 중에 있다.

Hepatoprotective activity of methanol extracts of Berberis tinctoria

  • The methanol extracts of the roots, root bark and stem of Berberis tinctoria, were investigated for their hepatoprotective activity against carbon tetrachloride $(CCl_4)$ induced toxicity in freshly isolated rat hepatocytes, HEp-G2 cells and animal models. The methanol extracts were able to significantly normalise the levels of aspartate amino transferase, alanine aminotransferase, alkaline phosphatase, triglycerides, total proteins, albumin, total bilirubin and direct bilirubin, which were altered due to $CCl_4$ intoxication in freshly isolated rat hepatocytes and also in animal models. The anti-hepatotoxic effect of the methanol extracts in vitro were observed at $600\;-\;1,000\;{\mu}g/ml$ concentrations. A dose dependent increase in the percentage viability was observed when $CCl_4$ exposed HEp-G2 cells were treated with different concentrations of the methanol extracts. The highest percentage viability of HEp-G2 was observed at a concentration of $1,000\;{\mu}g/ml$. The results from the present investigations also indicate good correlation between the in vivo and in vitro studies.

Preliminary antimicrobial and cytotoxic activities of Amoora cucullata extractives

  • Amoora cucullata (Meliaceae), a mangrove plant, has folkloric reputation as a medicinal agent in Bangladesh. In this study, the n-hexane, ethyl acetate and methanolic extracts of the stem bark of this plant were subjected to microbiological investigation and brine shrimp lethality bioassay. In case of antimicrobial screening, the ethyl acetate and methanolic extracts appeared to be potent in terms of both zone of inhibition and spectrum of activity showing the average zones of inhibition 8 - 14 mm and 9 - 16 mm, respectively. In the brine shrimp lethality bioassay, the methanolic extract demonstrated highest cytotoxicity having $LC_{50}$ of $0.549{\mu}g/ml$, whereas the ethyl acetate and n-hexane extract showed $LC_{50}$ of 7.943 and $17.180{\mu}g/ml$, respectively.

두충차 추출물의 Angiotensin Converting Enzyme 저해효과 (Inhibitory Effects of Eucommia ulmoides Extract on Angiotensin Converting Enzyme)

  • 두충의 기능성 식품으로서 이용 가능성을 평가하기 위하여, 두충의 항고혈압 활성 물질을 분리 및 확인하였고, 각 부위별(잎, 껍질, 줄기) 추출물의 관련물질의 함량 및 ACE 저해활성을 조사하였다. 두충에서 분리한 항고혈압성 물질로 추정되는 8A는 순도 95.64%의 pinoresinol-4,4'-di-O-${\beta}$-D-glucoside(PDG)로서 자연건조 및 볶은 껍질에서 가장 높았으며, 각각 135.13 mg%와 163.67 mg%였다. 또한 두충 추출물의 ACE 저해활성은 10 mg/mL 농도로 처리하였을 때 볶은 잎, 자연건조 껍질 및 볶은 껍질에서 각각 77.56%, 75.73%, 75.73%로 높은 활성을 나타내었으며, 줄기는 상당히 낮게 나타났다. 두충에서 분리한 PDG를 1 mg/mL 처리할 때, ACE 저해활성은 91.87%로서 $Enalapril^{(R)}$의 97.06%보다는 조금 낮았지만, $Captopril^{(R)}$의 90.32% 보다는 약간 높은 항고혈압 활성을 나타내었다.

국내 유통 중인 식용식물 추출물의 항산화효과 (Anti-oxidative Activities of Commercial Edible Plant Extracts Distributed in Korea)

  • 많은 식물 추출물들은 항산화 효능을 지니고 있으나 대량 생산 공정시 그들의 활성을 잃어버리는 경향이 있다. 따라서 본 본문에서는 대량 생산 공정을 거쳐 국내 유통 중인 43종의 식용식물 추출물을 대상으로 총 페놀 함량, DPPH 라디칼 포착 효능, 그리고 superoxide anion 라디칼 포착 효능을 측정하였다. 그 결과 개똥쑥(Artemisia annua, whole plant), 마태(Ilex paraguariensis leaf), 엉겅퀴(Silybum marianum, fruit 그리고 leaf), 유근피(Ulmus pumila, bark), 운지버섯(Coriolus versicolor, fruit) 그리고 울금(Curcuma longa, root 그리고 stem) 추출물은 70 mg/g 이상의 높은 페놀화합물을 함유하는 것으로 나타났다. DPPH 라디칼의 포착 효능에서 개똥쑥, 마테, 솔잎(Pinus densiflora, leaf), 엉겅퀴, 유근피, 그리고 울금 추출물의 $SC_{50}$ 값 측정 결과 각각 $53.96{\pm}0.81ppm,\;24.61{\pm}2.12ppm,\;35.96{\pm}1.11ppm,\;57.45{\pm}2.13ppm,\;55.25{\pm}1.65ppm$ 그리고 $12.99{\pm}1.67ppm$으로 나타났다(비타민 C의 $SC_{50}$값: $3.86{\pm}0.81ppm$). 또한 superoxide anion 라디칼 포착 효능에서는 개똥쑥, 계피(Cinnamomum zeylunicum, bark), 마테, 복분자(Rubus coreanus, fruit 그리고 leaf), 상엽(Morus alba, leaf), 솔잎, 엉겅퀴, 유근피, 운지버섯, 울금, 자소엽(Perilla frutescens var. acuta, leaf) 그리고 히비스커스(Hibiscus sabdariffa, leaf) 그리고 flower) 추출물의 $SC_{50}$ 값 측정 결과 각각 $53.21{\pm}1.83ppm,\;50.12{\pm}2.12ppm,\;5.59{\pm}0.84ppm,\;41.60{\pm}8.93ppm,\;20.19{\pm}0.97ppm,\;15.19{\pm}1.66ppm,\;21.20{\pm}1.88ppm,\;15.71{\pm}0.91ppm,\;55.48{\pm}2.42ppm,\;52.12{\pm}2.44ppm,\;23.80{\pm}1.98ppm$ 그리고 $11.14{\pm}0.51ppm$인 것으로 나타났다(비타민 C의 $SC_{50}$ 값:$9.61{\pm}0.93ppm$). 특히 마테 추출물과 솔잎 추출물은 총 페놀 함량이 높으면서 DPPH 라디칼과 superoxide anion 라디칼을 동시에 효율적으로 포착하는 효능을 지니고 있는 것으로 나타났다. 결론적으로 마테와 솔잎의 상업적인 추출물은 기능성 항산화제로서 유용한 소재로 사용 가능 할 것으로 사료된다.

분비나무 줄기 수피 추출물에 의한 HeLa Cell Line의 증식억제 효과와 RAW264.7 세포에서 Lipopolysaccharide에 의해 유도된 Nitric Oxide 생성 저해효과 (Inhibitory Effect on the Lipopolysaccharide-Induced Nitric Oxide Formation in RAW264.7 and on the Proliferation of HeLa Cell Lines by the Stem Bark Extracts of Abies nephrolepis)

  • Abies nephrolepis(Pinaceae) extracts were tested for determined immune system regulating activity based on antiinflammatory activity, antioxidant activity and anti-proliferative effect on HeLa cell line. The A. nephrolepis extracts increased dose-dependently anti-proliferation of HeLa cell line. The DM fraction of the extracts having anti-proliferatative effects of HeLa cell line was fractionalized four subfractions($D1{\sim}D4$). Inflammation-induced NO production was inhibited by D2 and D4 in LPS-activated RAW264.7 macrophages. And also, this fractions showed antioxidant activity examined by DPPH radical scavenging effects. These results suggest that the potential use of DM fraction of A. nephrolepis in chemoprevention and regulation overproduction of NO on pathogenic conditions. The mechanism of the inflammatory effects, however, must be evaluated through various parameters in the induction cascade of NO production.

Growth-inhibiting Effects of Brazilian and Oriental Medicinal Plants on Human Intestinal Bacteria

  • Methanol extracts of 27 Brazilian plant samples and 10 oriental medicinal plant samples (27 families), using spectrophotometric and paper disc agar diffusion methods under anaerobic conditions, were tested in vitro for their growth-inhibiting activities against Bifidobacterium longum, Bifidobacterium bifidum, Bifidobacterium adolescentis, Clostridium perfringens, and Bacteroides fragilis. The responses varied with bacterial strains, plant species, and tissues sampled. In a test with B. longum and B. bifidum(20 mg/disc), extracts of Acanthopanax sessilifolinus stem bark and Ampelozizyphus amazonicus leaves strongly inhibited the growth of B. longum, whereas other plant samples did not inhibit any intestinal bacteria tested. At 5 mg/disc, adding extracts of Aralia eleta, Euterpe oleracea, and Syzygium guineense to the media strongly inhibited the growth of C. perfringens and B. fragilis without growth inhibition of B. adolescentis, B. longum, and B. bifidum. Extracts of Jacaranda mimosifolia and Ulmus paraifolia significantly inhibited the growth of C. perfringens and B. fragilis as well as B. adolescentis. These results may be indications of at least one of the pharmacological actions of the five Brazilian plants but not oriental medicinal plants tested.

Inhibitory Effect of Rosmarinic acid Extrcted from Euonymus Alatus on Cyclooxygenase-2

  • Objectives and methods : Previous mechanistic studies suggest the cyclooxygenase-2 (COX-2) inhibitors represent the good candidates against tumor progression. MeOH extract of the stem barks of Euonymus alatus induced the strong inhibition of COX-2. A phenolic compound responsible for the anti- COX-2 known to involve in tumor adhesion and invasion has been studied through the methanol extracts. The compound, rosmarinic acid (ROS-A) was an ester of caffeic acid and 3,4-dihydroxyphenyllactic acid. ROS-A showed a strong inhibitory effect of COX-2 activity in a concentration-dependent manner. Then we have measured the IL-1${\beta}$, IL-6 and TNF-${\alpha}$ production related the immune regulation, induction of inflammatory related genes. Results and Conclusions :Hep3B cells produce proinflammatory cytokines of IL-1${\beta}$, IL-6 and TNF-${\alpha}$ while ROS A inhibited the cytokines production. Since IL-1${\beta}$, IL-6 and TNF-${\alpha}$ need the transcription factors such as nuclear factor- ${\kappa}$B (NF-${\kappa}$B) and activated protein-1 (AP-1), we measured the transcription factors. ROS-A inhibited the activation of p65, p50, c-Rel subunits of NF-${\kappa}$B and AP-1 transcription factors. These findings indicate that ROS A from the stem bark of E. alatus inhibits proliferation in metastatic cancer cells. It was suggested that stem barks of E. alatus could be suitable for anti-cancer drugs.
