합성얼음과 냉동얼음을 이용한 선형을 변화시킨 쇄빙상선의 저항특성 연구 (Comparative Study on Resistance Performance of Icebreaking Cargo Vessel according to Hull Form Variation by using Synthetic Ice and Refrigerated Ice)

  • The present paper deals with the comparative study of resistance performance with refrigerated ice and synthetic ice according to the variation of hull form characteristics. The resistance test has been conducted in pack ice condition in each concentration condition. Stem angle has been chosen as main parameters for the variation of hull form characteristics. The correlation of performance between with the refrigerated ice and with the synthetic ice has been shown according to the variation for stem angles. The present study show the possibility of ice test in general towing tank with synthetic ice for the time-consuming research such as hull form optimization although that is confined in pack ice condition. The more parametric study for the properties of synthetic ice is expected to be conducted to have more close correspondence for the test results of refrigerated ice in near future.

The Cancer Stem Cell Theory: Is It Correct?

  • The cancer stem cell hypothesis posits that tumor growth is driven by a rare subpopulation of cells, designated cancer stem cells (CSC). Studies supporting this theory are based in large part on xenotransplantation experiments wherein human cancer cells are grown in immunocompromised mice and only CSC, often constituting less than 1% of the malignancy, generate tumors. Herein, we show that all colonies derived from randomly chosen single cells in mouse lung and breast cancer cell lines form tumors following allografting histocompatible mice. Our study suggests that the majority of malignant cells rather than CSC can sustain tumors and that the cancer stem cell theory must be reevaluated.

Embryonic Stem Cells Lacking DNA Methyltransferases Differentiate into Neural Stem Cells that Are Defective in Self-Renewal

  • Background and Objectives: DNA methyltransferases (Dnmts) play an important role in regulating DNA methylation during early developmental processes and cellular differentiation. In this study, we aimed to investigate the role of Dnmts in neural differentiation of embryonic stem cells (ESCs) and in maintenance of the resulting neural stem cells (NSCs). Methods and Results: We used three types of Dnmt knockout (KO) ESCs, including Dnmt1 KO, Dnmt3a/3b double KO (Dnmt3 DKO), and Dnmt1/3a/3b triple KO (Dnmt TKO), to investigate the role of Dnmts in neural differentiation of ESCs. All three types of Dnmt KO ESCs could form neural rosette and differentiate into NSCs in vitro. Interestingly, however, after passage three, Dnmt KO ESC-derived NSCs could not maintain their self-renewal and differentiated into neurons and glial cells. Conclusions: Taken together, the data suggested that, although deficiency of Dnmts had no effect on the differentiation of ESCs into NSCs, the latter had defective maintenance, thereby indicating that Dnmts are crucial for self-renewal of NSCs.

NACA 처리에 따른 치주줄기세포 사멸 억제 효과 (Protective Effect of NACA on Periodontal Stem Cell)

  • Purpose :Periodontal ligament stem cells maintain tissue homeostasis in periodontal ligament. The purpose of this study was to determine the characteristics of periodontal ligament stem cells isolated from premolar teeth and observe protective effects against oxidative damage caused by Triethylene glycol dimethacrylate (TEGDMA) following treatment with N-acetylsysteine amide (NACA) drug known as enzymatic antioxidants. Methods : Primary periodontal ligament stem cell (PDSC) culture was performed from simply extracted human premolar of orthodontic patients. The characteristics of the primary cultured PDSCs was analyzed using the FACS system. PDSCs was incubated with TEGDMA and NACA. The cell proliferation and survival was determined using WST-1 assay. Collected data were analyzed using SPSS Window 20. Results : Primary cultured PDSCs grow on the floor and develop rapidly in a cluster form from up to 14 days. The morphology of PDSCs showed the spindle-shaped cells and grew directionally. FACS analysis, In addition, positive expression of visible cells were observed in mesenchymal stem cell biomarkers. PDLSCs cell viability was significantly decreased at high concentration in both 3 and 6 hours after TEGDMA treatment. We observed a decrease in the number of cells as well as a morphological change of PDLSCs. Antioxidative effect was notable since the death of PDLSC death was significantly inhibited compared to the control group at 24 and 48 hours after NACA treatment. Conclusion : Therefore, based on the results of this study, further research should be encouraged considering the development of clinical treatment methods using various antioxidants as well as regenerative engineering techniques utilizing periodontal ligament stem cells.

  • The present paper deals with characteristics of resistance performance according to the variation of synthetic ice thickness and hull form. The resistance test has been conducted with pack ice condition in Pusan National University towing tank. Stem angle has been chosen as main parameters for the variation of hull form characteristics, which is the most important factor especially in icebreaking cargo vessel. The serial comparisons of resistance test have been done with the variation of hull form parameter as well as with the different thickness of synthetic ice. The different trend of resistance performances with increasing of stem angle has been shown at each synthetic ice thickness. The present test results is expected to be confirmed by comparing the test results in ice tank in the near future.

제주지역에서 질소시비량 차이에 따른 양마의 생육특성, 수량 및 조성분 변화 (Effect of Nitrogen Rate on Agronomic characteristics, Forage Yield and Chemical Composition of Kenaf on Jeju Island)

  • In order to determine the influence of nitrogen on agronomic characters, forage yield and quality, a Kenaf was cultured on the volcanic ash soil at the Experimental Farm of Cheju national University under the seven levels of nitrogen rates(0, 50, 100, 150, 200, 250kg/ha) from April 25 to Dec. 25, 1999. The plant height increased by increase of nitrogen rate, showing longest 250kg/ha with 286.6cm and shortest at no application plot with 255.7cm. The difference on leaf number, leaf withering number, stem diameter and branches number by nitrogen rate showed a similar tendency to the plant height. Increasing N rate from 0 to 250kg/ha fresh forage yield form 55.8 to 99.8MT/ha, dry matter (DM) yield from 8.8 to 15.8MT/ha, crude protein(CP) yield form 1.2 to 3.1MT/ha, total digestible nutrients (TDN) yield from 3.8 to 8.6MT/ha. However, no significant differences in these yields were found between 200 and 250kg N/ha. Nitrogen uptake increased form 192.9 to 496.2 kg/ha but N use efficiency decreased form 95.0 to 66.6 kg DM$^{a}$ /kg N with increasing from 0 to 250 kg/ha. As N rate increased from 0 to 250kg/ha, leaf and stem out of CP, crude fat (CF), nitrogen free extract (NFE), TDN contents increased from 20.1% to 25.8% and from 9.7% to 12.4%, from 5.6% to 8.1% and from 3.3% to 4.4%, from 36.1% to 40.2% and from 21.9% to 32.4%, from 59.3% to 75.0% and from 32.2% to 38.2%, respectively, while leaf and stem out of crude fiber decreased from 18.5% to 16.7% and from 51.5% to 39.3%. Based on the these findings, the optimum N rate for forage production of kenaf seems to be about 200 kg/ha in atmospheric phenomena and volcanic ash soils of jeju island.

육상식물의 생산력추정을 위한 상대상장법의 이용에 대하여 (On the Utilization of the Allometric Method for an Estimation of the Productivity of the Terrestrial Plants)

  • In order to approach a reasonable method for the indirect estimation of the productivity in terrestrial plants, allometric method was studied with herbs, such as Boenmeria frutescens and Hibiscus mutabilis, and scrubs, such as Robinia pseud oacacia and Morus alba f. Hchiroe. Allometric relations between D230H, square of the stem(or shoot) diameter(D30) on 30cm height above ground multiplied by stem length(H), and the amount of the stem(Ws), orof the leaves per stem, were fitted to each material plant with linear regression. The allometric coefficient in relation of D230H to Ws was approximated between scrubs having different life form, and the allometric function in relation of DF2 to WL was common between herbs in spite of having different growth habitat. As the allometries between D230H and Ws or WL were changeable with the seasonal change it funciton should be calculated repeatedly whenever investigating the standing crop of such community. It is able to utilize an indirect value of D30 obtained from the allometry between D30 and H because direct measurement of D30 in the field is accompanied by troublesome and waste of much time.

  • Vertebrates regenerate tissues in three ways: proliferation of cells that maintain some or all of their differentiated structure and function, redifferentiation of mature cells followed by proliferation and redifferentiation into the same cell type or transdetermination to another cell type, and activation of restricted lineage stem cells, which have the ability to transdetermine to different lineages under the appropriate conditions. The behavior of the cells during regeneration is regulated by growth factors and extracellular matrix molecules. Some non-regenerating tissues are now known to harbor stem cells which, though they form scar tissue in vivo, are capable of producing new tissue-specific cells in vitro, suggesting that the injury environment inhibits latent regenerative capacity. Regenerative medicine seeks to restore tissues via transplantation of stem cell derivatives, implantation of bioartificial tissues, or stimulation of regeneration in vivo. These approaches have been partly successful, but several research issues must be addressed before regenerative medicine becomes a clinical reality.

Neural Transcription Factors: from Embryos to Neural Stem Cells

  • The early steps of neural development in the vertebrate embryo are regulated by sets of transcription factors that control the induction of proliferative, pluripotent neural precursors, the expansion of neural plate stem cells, and their transition to differentiating neural progenitors. These early events are critical for producing a pool of multipotent cells capable of giving rise to the multitude of neurons and glia that form the central nervous system. In this review we summarize findings from gain- and loss-of-function studies in embryos that detail the gene regulatory network responsible for these early events. We discuss whether this information is likely to be similar in mammalian embryonic and induced pluripotent stem cells that are cultured according to protocols designed to produce neurons. The similarities and differences between the embryo and stem cells may provide important guidance to stem cell protocols designed to create immature neural cells for therapeutic uses.

Expression of CD133, CD44, CK7, and OCT4 in Animal Cancers

  • Cancers are mainly sustained by a small pool of neoplastic cells, known as cancer stem cells or tumorinitiating cells. These cells possess the ability to self-renew and proliferate, and are thus able to form the tumor. In the present study cells that correspond to cancer stem cells in mammary and liver cancers in animals were identified by the expression of CD133, CD44, CK7, and OCT4 using immunochemistry. As a result, we found with CD133+ and CD44+ cancer stem cell-like phenotypes in mouse and canine hepatocellular carcinoma and canine mammary gland tumors. However, CK7+ and OCT4+ cells were not identified in animal mammary and liver cancer. CD133+ and CD44+ cells are wellknown stem cell lines and play key roles in development and metastasis in human cancer. These findings suggest that cancer stem cells are involved in animal tumorigenesis and may provide insight into mechanisms in cancer development as well as cancer diagnostics.
